For viruses made of nucleic acid and protein, the structure of the protein outer shell has, in the past, been found to be uniquely determined by the viral genome. However, here, non-denaturing agarose gel electrophoresis of bacteriophage T7 reveals two states of the mature T7 capsid; the conditions of growth are found to alter the population by T7 of these two electrophoretically defined states. Both states have been previously observed for a genetically altered T7 and they are observed here for wild-type T7. The average electrical surface charge density of a bacteriophage particle (delta) determines its state; the delta of particles in both states is negative. For a given condition of growth, the population of these two states is influenced by the extent to which the major T7 outer shell protein, p10A, is accompanied by its minor readthrough variant, p10B. Comparison of the two electrophoretic states reveals the following. (1) No difference in radius is present in the outer shell (+/-2%). (2) As the pH of electrophoresis is either increased or decreased from neutrality, the state becomes more highly populated for which delta is greater in magnitude (state 1). By changing the pH, some T7 particles are made to change state. (3) Particles in state 1 adsorb less quickly to host cells than do the particles in the alternative state (state 2). This latter observation suggests the hypothesis that state 1 evolved to reduce the probability of re-initiating an infection when conditions are not favorable for growth. This hypothesis is supported by the observation that, as conditions of growth become apparently more unfavorable, progeny increasingly populate state 1.  相似文献

Rate of passage from the rumen was estimated from samples from the rumen, duodenum, ileum, and rectum after four crossbred heifers were dosed with cobalt ethylenediaminetetraacetic acid and ytterbium-labeled alfalfa or corn grain as digesta markers. Diets consisted of chopped alfalfa hay or an 80% concentrate mixture fed twice daily at 2.0% of body weight (dry matter per day) in two trials. Rates of particulate passage were calculated by two mathematical models, and influence of site of sampling on rate of ruminal passage was appraised. Rate of fluid passage calculated from ruminal samples was more rapid than rates from samples at the duodenum, ileum, and rectum for both the alfalfa diet (10.6 versus 6.3, 5.6, and 5.6% X h-1) and the 80% concentrate diet (10.3 versus 6.8, 7.2, and 6.1% X h-1). With the alfalfa diet, rates of fluid passage estimated from ruminal and rectal samples were correlated .98 despite the marked disparity of means. Rates of passage of particulates estimated from samples from the duodenum, ileum, and rectum were not correlated. Estimates of rate of digesta passage from the rumen differed with postruminal site of sampling.  相似文献

We have extended the empirical work of Vano et al.[1] relating the slope of the detector efficiency curve to the active volume for Ge detectors. The analysis was carried out using Monte Carlo techniques and covered a wide range of incident energies (200 keV-20 MeV) and active volumes (19.6 cm3–396 cm3). It is shown that the expression of Vano et al.[1] is only valid over the energy range 200 keV-3 MeV for active volumes <50 cm3. The upper bound decreases to 2 MeV for volumes of a few hundred cm3. The usable energy range can, however, be extended to 6 MeV by introducing higher order terms into the polynomial. Above this energy, the shape of the efficiency curve is better described by a non-linear function since linear forms fail simultaneously to fit large active volumes and high energies. We therefore propose a composite function which reduces to the form given in Vano et al. in the low energy/active volume limit. By comparison with the Monte Carlo results, it is estimated that relative efficiencies can be calculated to within 6% over the energy range 200 keV-20 MeV and active volumes 20 cm3–400 cm3. Since the largest errors occur for the smallest volumes, we recommend that for energies <3 MeV a two-fold approach be followed, i.e. using the expression of Vano et al.[1] for active volumes less than 50 cm3 and the proposed non-linear form for larger volumes. For high energy work (E > 3 MeV), we advocate the non-linear form. In this way, average errors can be kept 3%. Finally, we point out that the real power of the expression of Vano et al. lies not in predicting efficiencies, but active volumes.  相似文献

Transgenic mice carrying a hybrid gene consisting of ovine beta-lactoglobulin gene sequences and human gamma-interferon (hIFN-g) cDNA were produced. hIFN-g expression in the mammary gland of two lactating transgenic founder females was found. The concentration of active hIFN-g in the milk was estimated as being ca. 1800 IU/ml. The hIFN-g ability to express in the mammary gland was found in the progeny of transgenic founder male.  相似文献

Molecular orbital calculations of the ionization potential of single wall carbon nanotubes having donor NH2 and acceptor NO2 groups bonded to the side walls and ends and boron and nitrogen substituted for carbon show substantial increases in ionization potential compared to carbon nanotubes with no functional groups and no carbon substitutions. The presence of a carbon vacancy on the side wall also causes a substantial increase in the ionization potential. The effect of tube length on the ionization energy is also calculated. The calculations also suggest that at appropriate levels of boron and nitrogen doping the armchair carbon nanotubes could be high temperature organic ferromagnetic materials.  相似文献

The Gram-positive bacterium Leuconostoc mesenteroides, ATCC 8293, is intrinsically resistant to the antibiotic vancomycin. This phenotype correlates with substitution of D-Ala-D-lactate (D-Ala-D-Lac) termini for D-Ala-D-Ala termini in peptidoglycan intermediates in which the depsipeptide has much lower affinity than the dipeptide for vancomycin binding. Overproduction of the L. mesenteroides D-Ala-D-Ala ligase (LmDdl) 2 in E. coli and its purification to approximately 90% homogeneity allow demonstration that the LmDdl2 does have both depsipeptide and dipeptide ligase activity. Recently, we reported that mutation of an active site tyrosine (Tyr), Tyr216, to phenylalanine (Phe) in the E. coli DdlB leads to gain of D-Ala-D-Lac depsipeptide ligase activity in that enzyme. The vancomycin-resistant LmDdl2 has a Phe at the equivalent site, Phe261. To test the prediction that a Tyr residue predicts dipeptide ligase while an Phe residue predicts both depsipeptide and dipeptide ligase activity, the F261Y mutant protein of LmDdl2 was constructed and purified to approximately 90% purity. F216Y LmDdl2 showed complete loss of the ability to couple D-Lac but retained D-Ala-D-Ala dipeptide ligase activity. The Tyr-->Phe substitution on the active site omega-loop in D-Ala-D-Ala ligases is thus a molecular indicator of both the ability to make D-Ala-D-Lac and intrinsic resistance to the vancomycin class of glycopeptide antibiotics.  相似文献
