The release of metoclopramide hydrochloride (very water soluble cationic drug) and diclofenac sodium (sparingly soluble anionic drug) from pellets coated with hydroxypropylmethylcellulose (HPMC; water-soluble polymer) or ethylcellulose aqueous dispersion (Surelease; water-insoluble polymer) at different coating loads was investigated. The release rates of either drug decreased as the coating load of HPMC increased, but overall, the release was fast, and the majority of both drugs released in about 1 hr, even at the highest coating load. The drug release mechanism for either drug was not affected by the coating load of HPMC or by the type of drug used, and it was found to be mainly diffusion controlled. Diclofenac sodium released slightly more slowly than metoclopramide hydrochloride from HPMC-coated pellets. This was attributed to the lower water solubility of the former drug. The release rate of either drug decreased greatly as the coating load of Surelease increased. The release of both drugs was sustained over 12 hr as the coating load of Surelease increased, and only about 70% of either drug was released after this period at the highest coating load (20%). The mechanism of release of metoclopramide hydrochloride was independent of coating load, and it was predominantly diffusion controlled. However, the mechanism of diclofenac sodium release was dependent on the coating load of Surelease. At low coating loads, diffusion of drug was facilitated due to the presence of more pores at the surface of the coated pellets; therefore, the rate of dissolution of the drug particles was the rate-limiting step. However, at high coating loads, drug release was mainly diffusion controlled. Despite its lower water solubility, diclofenac sodium released slightly faster than metoclopramide hydrochloride from Surelease-coated pellets at equivalent coating loads.  相似文献

Although the microcomputer and the Internet continue to advance rapidly in Western cultures, developing nations, especially those in Africa, are lagging behind—a situation that continues to widen the gap between the haves and the have-nots of the world. This article offers the rare opportunity to get a glimpse of Africa on the information highway and documents the use of computers in the educational systems of three African nations: Ghana, Kenya, and Egypt. It documents the progress these countries have made in exposing their citizens to information technology and reveals the challenges they face in closing the digital divide that Africans continue to experience. Trans fatty acid consumption is associated with an increased risk of coronary heart disease. This increased risk has been attributed to decreased levels of HDL cholesterol and increased levels of LDL cholesterol. However, the mechanism by which trans fatty acid modulates cholesterol transit remains poorly defined. ATP-binding cassette transporter A1 (ABCA1)-mediated macrophage cholesterol efflux is the rate-limiting step initiating apolipoprotein A-I lipidation. In this study, elaidic acid, the most abundant trans fatty acid in partially hydrogenated vegetable oil, was shown to stabilize macrophage ABCA1 protein levels in comparison to that of its cis fatty acid isomer, oleic acid. The mechanism responsible for the disparate effects of oleic and elaidic acid on ABCA1 levels was through accelerated ABCA1 protein degradation in cells treated with oleic acid. In contrast, no apparent differences were observed in ABCA1 mRNA levels, and only minor changes were observed in Liver X receptor/Retinoic X receptor promoter activity in cells treated with elaidic and oleic acid. Efflux of both tracers and cholesterol mass revealed that elaidic acid slightly increased ABCA1-mediated cholesterol efflux, while oleic acid led to decreased ABCA1-mediated efflux. In conclusion, these studies show that cis and trans structural differences in 18 carbon n-9 monoenoic fatty acids variably impact cholesterol efflux through disparate effects on ABCA1 protein degradation.  相似文献

Following an infection, Kaposi's sarcoma-associated herpes virus (KSHV) exists predominantly in its latent state, with only 1-2% of infected cells undergoing lytic reactivation. We have previously demonstrated along with others a relationship between lytic reactivation and cell cycle progression (Bryan et al., 2006. J. Gen. Virol. 87: 519; McAllister et al., 2005. J. Virol. 79: 2626). Infected cells in the S phase are much more likely to undergo lytic reactivation when compared to those in G(0)/G(1) phase. Through the use of scanning electron microscopy (SEM), we analyzed changes occurring on the surface of cells undergoing KSHV reactivation. KSHV reactivation was observed predominantly in cells with smoother surface topology; a hallmark of cells derived from S phase. Interestingly, during the late stages of the reactivation process, we observed KSHV particles to egress cells through budding. Taken together, based on scanning electron microscopy and transmission electron microscopy evidences, we demonstrate for the first time the existence of a direct link between cell surface topology, cell cycle progression and KSHV reactivation.  相似文献

Cholesteryl esters (CE) are important lipid storage molecules. The present study demonstrates that sodiated adducts of CE molecular species form positive ions that can be detected in both survey scan mode as well as by exploiting class-specific fragmentation in MS/MS scan modes. A common neutral loss for CE is the loss of cholestane (NL 368.5), which can be used to specifically quantify tissue CE molecular species. Using this MS/MS technique, CE molecular species were quantified in mouse monocyte-derived macrophages (J774 cells) incubated with either linoleic (18:2) or arachidonic acid (20:4). These studies revealed that arachidonic acid was not only incorporated into the CE pool, but also was elongated resulting in the accumulation of 22:4 and 24:4 CE molecular species in macrophages. Additionally, this technique was used to quantify CE molecular species present in crude lipid extracts from plasma of female mice fed a Western diet, which led to an enrichment in CE molecular species containing monounsaturated fatty acids compared to female mice fed a normal chow diet. Last, NL 368.5 spectra revealed the oxidation of the aliphatic fatty acid residues of CE molecular species containing polyunsaturated fatty acids. Taken together, these studies demonstrate the utility of using sodiated adducts of CE in conjunction with direct infusion electrospray ionization tandem mass spectrometry to rapidly quantify CE molecular species in biological samples.  相似文献

Determining the size of nanoparticles accurately, quickly and easily is becoming more and more important as the use of such particles increases. One of the common techniques for measuring the size of particles in suspension is dynamic light scattering (DLS). In principle, DLS is able to estimate the hydrodynamic particle diameter and its intensity-weighted distribution. However, the measured correlation function or power spectrum must be inverted to obtain this size distribution. The inversion is an ill-posed mathematical problem, and only under certain assumptions can the distribution be determined reliably. Suspensions containing bimodal (or multi-modal) particle size distributions are particularly challenging. This study reports on DLS measurements on a range of bimodal distributions of latex spheres with varying ratios of particle sizes. To determine the efficacy of different inversion techniques, the data has been analyzed both with the algorithms implemented in the DLS instrument’s proprietary analysis software and with other inversion routines based on simple analytical models of the particle size distribution. In addition, the results of the DLS analysis have been compared to scanning and transmission electron microscopy (SEM and TEM) measurements.  相似文献

Fiber imaging bundles have been investigated for use in endoscopic optical coherence tomography (OCT) systems, to obviate the requirement for scanning components within the endoscope probe section. Images have been acquired using several optical configurations, two of which are common path in design. Configurations have been selected as having potential for miniaturization and inclusion in endoscopic-type systems, since the advantages of employing imaging bundles are most clearly seen in this type of system. The various types of bundle available are described, and the properties of the leached bundles used here are discussed in detail, with reference to their effect upon the performance of OCT systems. Due to changes in their lending pattern, along with wider social and economic change, building societies face increasing risk, signified by growing default. All creditors have ultimate legal sanctions against debtors but typically first initiate a non‐judicial debt collection process. This paper outlines policy and practice with regard to the pre‐judicial management for the recovery of mortgage arrears, and indicates a substantial gulf between the two. In particular the content of the ‘recovery’ strategies and relationships is examined, using qualitative data from a study of 40 defaulters followed over a nine‐month period. Procedures designed to keep borrowers out of court and within owner occupation had the opposite effect in practice in almost half the cases examined. Emphasis is placed upon late intervention, poor communication and the formulation of a recovery strategy that treats the mortgage debt in isolation from current income and expenditure. The potential contribution of managers’ underlying attitudes to debtors is raised. In the face of rising default, a number of possible explanations for an ‘underdeveloped’ arrears management process are examined.  相似文献
