Acacia angustissima has potential in agroforestry systems for forage production in tropical environments. However, feeding trials indicate that the leaves are potentially toxic. Signs of toxicity in sheep are similar to those presented by sheep fed flatpea (Lathyrus sylvestris) hay which contains a high concentration of the non‐protein amino acid 2,4‐diaminobutanoic acid (DABA). Cation exchange chromatography, high‐performance liquid chromatography (HPLC) and NMR were used to isolate and identify the non‐protein amino acid in A angustissima leaves. A quantitative HPLC method was used to determine the content of the non‐protein amino acid in leaves from 20 accessions. The non‐protein amino acid was identified as 4‐N‐acetyl‐2,4‐diaminobutanoic acid (ADAB), previously reported to occur in seeds. The average and range in ADAB content were 16.8 and 10.6–25.4 mg g?1 respectively. This content of ADAB is higher than the levels of DABA in flatpea hay that are toxic to sheep. © 2001 Society of Chemical Industry  相似文献

Destructive distillation of solar heat transfer fluids was conducted to determine the types of pyrolytic products which might be formed in solar collectors under conditions of stagnation or malfunction. The distillates were analyzed by gas chromatography/mass spectrometry (GC/MS) to determine the types of compounds which were present and the minor components which might be formed. Dehydration products were formed from ethylene and propylene glycols with ethylene oxide and propylene oxide being minor products. A high aromatic petroleum heat transfer fluid yielded distillation fractions enriched in quinolines or isoquinolines and methylated derivatives thereof. Fractionation of this aromatic heat transfer fluid showed that the basic fraction of unheated fluid also contained aza-arenes which exhibited mutagenic activity in the Ames' bioassay. GC/MS showed that the basic fraction from unheated heat transfer fluid also contained benzoquinolines which appeared to be lacking in heat degraded samples of the same fluid. Fractionation of heat transfer fluids, in particular petroleum-based fluids, may often be necessary in order to concentrate minor components so that they can be identified by GC/MS and in order to detect mutagenic activities without interference from cytotoxic components.  相似文献

Autoignition characteristics of n-heptane/air, gasoline/air, and ternary surrogate/air mixtures were studied behind reflected shock waves in a high-pressure, low-temperature regime similar to that found in homogeneous charge compression ignition (HCCI) engine cycles. The range of experiments covered combustion of fuel in air for lean, stoichiometric, and rich mixtures (Φ=0.5, 1.0, 2.0), two pressure ranges (15-25 and 45-60 atm), temperatures from 850 to 1280 K, and exhaust gas recirculation (EGR) loadings of (0, 20, and 30%). The ignition delay time measurements in n-heptane are in good agreement with the shock tube study of Fieweger et al. (Proc. Combust. Inst. 25 (1994) 1579-1585) and support the observation of a pronounced, low-temperature, NTC region. Strong agreement was seen between ignition delay time measurements for RD387 gasoline and surrogate (63% iso-octane/20% toluene/17% n-heptane by liquid volume) over the full range of experimental conditions studied. Ignition delay time measurements under fuel-lean (Φ=0.5) mixture conditions were longer than with Φ=1.0 mixtures at both the low- (15-25 atm) and high- (45-60 atm) pressure conditions. Ignition delay times in fuel-rich (Φ=2.0) mixtures for both gasoline and surrogate were indistinguishable in the low-pressure (15-25 atm) range, but were clearly shorter at high-pressures (45-60 atm). EGR loading affected the ignition delay times similarly for both gasoline and surrogate, with clear trends indicating an increase in ignition delay time with increased EGR loading. This data set should provide benchmark targets for detailed mechanism validation and refinement under HCCI conditions.  相似文献

Materials that respond to physiological stimuli are important in developing advanced biomaterials for modern therapies. The reversibility of covalent crosslinks formed by phenylboronate (PBA) and salicylhydroxamate (SHA) has been exploited to provide a pH‐responsive gel for application to the vaginal tract. Dynamic rheology reveals that the gel frequency‐dependent viscoelastic properties are modulated by pH. At pH 4.8 the viscous component dominates throughout most of the frequency range. As the pH increases, the characteristic relaxation time continues to increase while the GPlateau levels off above pH 6. At pH 7.5, the elastic component dominates throughout the frequency sweep and is predominately independent of frequency. Particle tracking assesses the transport of both fluorescently labeled HIV‐1 and 100‐nm latex particles in the PBA–SHA crosslinked gel as a function of pH. At pH 4.8 the ensemble‐averaged mean squared displacement at lag times greater than three seconds reveals that transport of the HIV‐1 and 100‐nm particles becomes significantly impeded by the matrix, exhibiting diffusion coefficients less than 0.0002 µm2 s?1. This pH‐responsive gel thus displays properties that have the potential to significantly reduce the transport of HIV‐1 to susceptible tissues and thus prevent the first stage of male‐to‐female transmission of HIV‐1.  相似文献

Is current research on computing by older adults simply looking at a short-term problem? Or will the technology problems that plague the current generation also be problematic for today’s tech-savvy younger generations when they become “old”? This paper considers age-related and experience-related issues that affect ability to use new technology. Without more consideration of the skills of older users, it is likely that applications and devices 20 years from now will have changed such that this “older” generation finds themselves confronting an array of technologies that they little understand and find generally inaccessible. Recent evidence suggests that older adults bring specific strengths to Web browsing. A fuller investigation of these strengths and how to design to optimize for strengths of older users has the potential to address the need for usable technology for this increasingly important demographic.  相似文献

It has long been recognized that a basic dimension to the lexical organization of the brain is semantic, and some brain mapping studies have indicated that the brain fields are distinctly different from some grammatical classes. Findings from the present investigation showed consistent relationships between 29 aphasic adults' performances on tasks involving graphic and gestural skills and those involving sequential recall of spoken words from different word categories. Each adult received the Porch Index of Communication Abilities which relies upon the physical manipulation of objects to assess verbal, gestural, and graphic abilities. Scores on a test requiring recall of word strings of nouns, verbs, adverbs, adjectives, or prepositions were used to predict the subscale scores from the Graphic and Gestural factors of the index. Recall scores for verb and preposition were predictive of the aphasic subjects' performances on the Graphic subscale, and noun and preposition scores were predictors of subjects' scores on the Gestural subscale. The results are related to other research showing that verb and preposition skills are predictive of fine motor abilities of children with communication disorders and brain-mapping studies. Some discussion centers on possible overlapping functions of brain activity involving word categories, language, and fine motor skills.  相似文献

BACKGROUND: Although inactivation of enveloped viruses transmitted by plasma derivatives has been successful, no methods for virus inactivation or removal have been established for platelet concentrates or red cell (RBC) components. Relatively little is known regarding the extent or significance of virus interactions with the cellular constituents in these components. STUDY DESIGN AND METHODS: Units of whole blood were collected from six HIV type 1 (HIV-1)-positive, asymptomatic individuals and separated into peripheral blood mononuclear cells (PBMNCs), cell-free plasma, white cell-reduced platelet concentrate, and white cell-reduced RBCs. DNA and RNA polymerase chain reaction and virus culture methods were used to study the compartmentalization of HIV-1 immediately after component preparation and after storage. RESULTS: As expected, HIV DNA and infectious virus were detected in fresh blood and in PBMNCs, and virion-associated RNA was detected in fresh plasma from all six donors. The levels of viral nucleic acids in these preparations remained relatively stable with 4 degrees C storage, whereas infectivity of PBMNCs was rapidly lost. Washed RBCs tested negative for HIV in all assays at all time points. Platelets retained high levels of HIV RNA (but not infectivity) after extensive washing, as well as after storage at 4 and 22 degrees C. High-level platelet-associated HIV-1 was also demonstrated in samples collected during early seroconversion. Periseroconversion and postseroconversion levels of platelet-associated HIV-1 correlated with the level of plasma viremia and with the rate of progression to AIDS. Cell-free virus from donor plasma and tissue culture fluid rapidly and firmly attached to platelets from noninfected donors. Infectivity of tissue culture virus bound to platelets was demonstrated in vitro. CONCLUSION: Significant levels of HIV-1 are associated with platelets during all stages of infection. Platelet-associated HIV could either mediate virus clearance or facilitate virus dissemination and expanded tropism. Finally, virus inactivation research must address virus associations with platelets.  相似文献
