DIMETHYL-1-CURINE DIMETHOCHLORIDE - AN ALTERNA- TIVE FOR d-TUBOCURARINE CHLORIDE

Dimethyll-curine dimethochloride was obtained by methylating the l-curine which was isolated from plants belonging to Genesia Cyclea widely distributed in Hainan Island, South China. It is a non-depolariz- ing post neuromuscular junctional blocker acting upon N2 receptor. Its chemical structural formula, physical properties, pharmacological actions and acute or chronic toxicities have been thoroughly studied. Its biological activities varied a great deal in different experimental animals but their clinical potency is uniform. Its activity is 51.8% as those of d-tubocura rine. Clinically, 1,449 cases, male and female, aged and young, were studied. Among them 422 were employed for endotracheal intubation, 468 for muscu Iar relaxation and 559 for both. Operation varied, including intracraneal, intra-abdominal and open heart surgery. Different kinds of anesthetics and adjuvants were used in association. I.V. dosage of 0.4-0.5 mg/kg or more was enough for intubation while 0.2-0.3 mg/kg for relaxation. Average duration of action was 30-40 minutes, but could be prolonged after heavy doses. Side effects were limited only to mild hypotension and temporal face freshness, while cardiovascular disturbances were very insig- nificant.

Developmental changes of glutamate acid decarboxylase 67 in mouse brain after hypoxia ischemia

Objective To study the developmental changes of glutamic acid decarboxylase-67 (GAD-67, a GABA synthetic enzyme) in normal and hypoxic ischemic (HI) brain. Methods C57/BL6 mice on postnatal day (P) 5, 9, 21 and 60, corresponding developmentally to premature, term, juvenile and adult human brain were investigated by using both Western blot and immunohistochemistry methods either in normal condition or after hypoxic ischemic insult. Results The immunoreactivity of GAD67 was up regulated with brain development and significant difference was seen between mature (P21, P60) and immature (P5, P9) brain. GAD67 immunoreactivity decreased in the ipsilateral hemisphere in all the ages after hypoxia ischemia (HI) insult, but, significant decrease was only seen in the immature brain. Double labeling of GAD67 and cell death marker, TUNEL, in the cortex at 8h post-HI in the P9 mice showed that (15.6±7.0)% TUNEL positive cells were GAD67 positive which was higher than that of P60 mice. Conclusion These data suggest that GABAergic neurons in immature brain were more vulnerable to HI insult than that of mature brain.     

THE INFLUENCE OF MULTIPLE FACTORS ON GALL BLADDER PRESSURE AFTER AURICULAR POINT STIMULATION

The study has explored 11 correlative variables which might affect the gallblad-der pressure GBP of patients with gall stone after auricular electrical stimulation by means of multiplestepwise regression. It was found that the size of gallbladder stone (X9), cholecystolithiasis (X4),sex (X1), and the baseline of GBP (X10) could affect the GBP change range (Y1) depressingly orreinforcingly. Also the baseline of GBP (Y1) could be affected by the variables X1 (sex), X2 (age),and X6 (long diameter of the gallbladder, LDGB), etc.. The result shows that the change in GBPafter auricular electrical stimulation is comprehensively influenced by multiple factors. And it providessome useful information for predicting the curative effect of auricular therapy on gallbladder stone inclinic.     

RETINOBLASTOMALACTIC DEHYDROGENASE OF AQUEOUS HUMOUR AND ITS ISOENZYMES

Lactic dehydrogenase (LDH) activity in aqueoushumour and serum was measured of 24 patients withretinoblastoma, senile cataract, glaucoma, and cho-roid melanoma in addition to LDH isoenzyme fractions. The results were compared with those of the20 normal children. The ratio of aqueous humourLDH to serum LDH was 4.32+2.80, and that ofisoenzymes LDH4 to LDHl was greater than l. Theresults of our study suggested that the total LDHactivity and isoenzyme fractions in the aqueoushumour is of value in the diagnosis of retinobla&toma.

Genetic polymorphism of UL144 open reading frame of human cytomegalovirus DNA detected in colon samples from infants with Hirschsprung''''s disease

AIM:To explore the genetic diversities of UL144 open reading frame (ORF) of cytomegalovirus DNA detected in colon tissue from infants with Hirschsprung's disease (HD) by sequencing UL144 DNA in 23 aganglionic colon tissue and 4 urine samples from 25 HD infants. METHODS:Nest PCR was performed for amplification of the UL144 gene. The UL144 gene was analyzed with softwares,such as DNAclub,BioEdit,PROSITE database,and DNAstar. RESULTS:The strains from HD patients were distributed among three genotypes of UL144:group 1A (64%),group 2 (24%),and group 3 (12%). The UL144 genotypes between strains from HD and control group were compared by chi square test (χ2 = 1.870,P = 0.393). Strains from the colon were sporadically distributed in UL144 genotypes. CONCLUSION:There are genetic diversities of UL144 ORF in colon tissue of infants with HD. However,cytomegalovirus UL144 genotypes are not associated with clinical manifestations of HD.
