Coagulation involves a complex set of events that are important in maintaining hemostasis. Biochemical interactions are classically known to regulate the hemostatic process, but recent evidence has revealed that mechanical interactions between platelets and their surroundings can also play a substantial role. Investigations into platelet mechanobiology have been challenging however, due to the small dimensions of platelets and their glycoprotein receptors. Platelet researchers have recently turned to microfabricated devices to control these physical, nanometer-scale interactions with a higher degree of precision. These approaches have enabled exciting, new insights into the molecular and biomechanical factors that affect platelets in clot formation. In this review, we highlight the new tools used to understand platelet mechanobiology and the roles of adhesion, shear flow, and retraction forces in clot formation.  相似文献

A combination of bioceramics and polymeric nanofibers holds promising potential for bone tissue engineering applications. In the present study, hydroxyapatite (HA), bioactive glass (BG), and tricalcium phosphate (TCP) particles were coated on the surface of electrospun poly(L-lactic acid) (PLLA) nanofibers, and the capacity of the PLLA, BG-PLLA, HA-PLLA, HA-BG-PLLA, and TCP-PLLA scaffolds for bone regeneration was investigated in rat critical-size defects using digital mammography, multislice spiral-computed tomography (MSCT) imaging, and histological analysis. Electrospun scaffolds exhibited a nanofibrous structure with a homogeneous distribution of bioceramics along the surface of PLLA nanofibers. A total of 8 weeks after implantation, no sign of complication or inflammation was observed at the site of the calvarial bone defect. On the basis of imaging analysis, a higher level of bone reconstruction was observed in the animals receiving HA-, BG-, and TCP-coated scaffolds compared to an untreated control group. In addition, simultaneous coating of HA and BG induced the highest regeneration among all groups. Histological staining confirmed these findings and also showed an efficient osseointegration in HA-BG-coated nanofibers. On the whole, it was demonstrated that nanofibrous structures could serve as an appropriate support to guide the healing process, and coating their surface with bioceramics enhanced bone reconstruction. These bioceramic-coated scaffolds can be used as new bone-graft substitutes capable of efficiently inducing osteoconduction and osseointegration in orthopedic fractures and defects.  相似文献

In order to achieve early detection of diabetic retinopathy (DR) for the sake of preventing from blindness, regular screening using retinal photography is necessary. Abnormalities of DR do not have uniform distribution over the retina. Certain types of abnormalities usually occur in specific areas on the retina. The distance between lesions, such as micro-aneurysms, and the foveal avascular zone (FAZ) is a useful feature for later analysis and grading of DR. In this paper, a new fully automatic system is presented to find the location of FAZ in fundus fluorescein angiogram photographs. The method is based on two procedures: digital curvelet transform (DCUT) and morphological operations. Firstly, end points of vessels are detected based on vessel segmentation using DCUT. By connecting these points in the selected region of interest, FAZ region is extracted. Secondly, vessels are subtracted from the retinal image, and morphological dilatation and erosion are applied on the resulted image. By choosing an appropriate threshold, FAZ region is detected. The final FAZ region is extracted by performing logical AND between two segmented FAZ. Our experiments show that the system achieves, respectively, the specificity and sensitivity of (>98 and >96 %) for normal stage, for mild/moderate non-proliferative DR (NPDR) (>98, and >95 %) and for Sever NPDR + PDR (>97 and >93 %).  相似文献

The study investigates the relative degree and timing of cortical activation in parietal, temporal, and frontal regions during simple arithmetic tasks in children who experience math difficulties. Real-time brain activity was measured with magnetoencephalography during simple addition and numerosity judgments in students with math difficulties and average or above average reading skills (MD group, N = 14), students with below average scores on both math and basic reading tests (MD/RD group, N = 16) and students with above average scores on standardized math tests (control group, N = 25). Children with MD showed increased degree of neurophysiological activity in inferior and superior parietal regions in the right hemisphere compared to both controls and MD/RD students. Left hemisphere inferior parietal regions did not show the expected task-related changes and showed activity at a significant temporal delay. MD students also showed increased early engagement of prefrontal cortices. Taken together, these findings may indicate increased reliance on a network of right hemisphere parietal (and possibly frontal areas as well) for simple math calculations in students who experience math difficulties but perform within normal range in reading. Whole Nonpareil variety almonds were inoculated with Salmonella Enteritidis PT 30 and stored at 4 or 23 °C for up to 48 wk. At 1, 12, 24, 37, and 48 wk of storage, almonds were heated by immersion in 121 °C oil. After heating for 0.5 to 2.5 min, almonds were drained, transferred to tryptic soy broth, and mixed with a stomacher prior to plating onto tryptic soy and bismuth sulfite agars. Over the 48 wk of storage, Salmonella declined by 0.5 and 2.1 log CFU/g at 4 and 23 °C, respectively. The survivor inactivation curves were upwardly concave with rapid initial reductions in the levels of Salmonella. For up to 24 wk of storage, the mean counts of the survivors after treatment were not significantly different. The Weibull model predicted 4- and 5-log reductions of Salmonella in 0.85 ± 0.16 and 1.8 ± 0.43 min, respectively, for almonds stored at 4 °C, and in 1.6 ± 0.53 and 3.2 ± 1.0 min, respectively, for almonds stored at 23 °C. Refrigerated storage had little impact on heat resistance of Salmonella that were inoculated on almonds. PRACTICAL APPLICATION: This research provides information of value in performing or evaluating validation studies for thermally processed almonds. The sensitivity of Salmonella to oil roasting is demonstrated during typical commercial almond storage times and temperatures.  相似文献

Preterm labour (PTL) is the most important cause of neonatal morbidity and mortality. While some causes have been identified, the mechanisms involved remain elusive. This study investigates whether term labour (TL) is an appropriate model for PTL by examining pro-labour gene expression, using quantitative rtPCR, and protein synthesis, using Western analysis, in preterm and term myometrial samples obtained from the upper and lower uterine segments before and after the onset of labour. In the lower segment, the levels of prostaglandin H synthase type-2 (PGHS-2), interleukin-1beta (IL-1beta), IL-6 and IL-8 mRNA expression were significantly higher in TL compared with PTL samples. Compared with non-labour controls, the expression of IL-1beta and IL-8 mRNA was increased in both PTL and TL samples and the expression of PGHS-2 and IL-6 mRNA was increased in TL samples only. In the upper segment, there were no differences between PTL and TL samples and the mRNA expression of PGHS-2 and IL-1beta was increased in TL compared with term no labour samples. No effect of PTL or TL was seen on either oxytocin receptor or connexin-43 mRNA expression or protein levels. The multiple regression analysis and studies in primary cultures of uterine myocytes suggest that the inflammatory cytokines, IL-1beta and tumour necrosis factor-alpha, are the most important regulators of PGHS-2 and IL-8. Our data show that preterm and term labouring myometrium are significantly different and that the most marked labour-induced changes in gene expression are in the lower segment. These changes may occur in response to the release of inflammatory cytokines by the labour-associated inflammatory infiltration.  相似文献

Intervention-related changes in spatiotemporal profiles of regional brain activation were examined by whole-head magnetoencephalography in 15 children with severe reading difficulties who had failed to show adequate progress to quality reading instruction during Grade 1. Intensive intervention initially focused on phonological decoding skills (for 8 weeks) and, during the subsequent 8 weeks, on rapid word recognition ability. Clinically significant improvement in reading skills was noted in 8 children who showed "normalizing" changes in their spatiotemporal profiles of regional brain activity (increased duration of activity in the left temporoparietal region and a shift in the relative timing of activity in temporoparietal and inferior frontal regions). Seven children who demonstrated "compensatory" changes in brain activity (increased duration of activity in the right temporoparietal region and frontal areas, bilaterally) did not show adequate response to intervention. Administered the Children's Report of Parent Behavior Inventory (CRPBI) and an SES scale to 1,949 13–18 yr olds from 2 cities in India and to 364 age-matched Ss from 1 city in Canada. Indian Ss were designated as traditional or transitional according to the values they were taught (traditional Indian vs modern Western); Canadian Ss were designated as modern. Factor analyses of CRPBI scales revealed 3 replicable factors: Acceptance, Firm Control (FC), and Psychological Control (PC). ANOVAs were used to test the effects of culture, sex, age, and SES on the 3 CRPBI factors. Younger Ss consistently reported more PC and FC than did older ones. Modern Ss reported the most FC, and traditional Ss reported the most PC. Even though higher SES correlated with lower PC and FC, statistical SES control did not attenuate the strong cultural differences for PC. Routine antenatal hepatitis B surface antigen (HBsAg) screening and immunization of risk babies is very effective in preventing perinatal transmission of hepatitis B virus (HBV). We studied 1,800 parturients attending a public hospital to assess the rationale for such vaccination in Bangladesh. In one in every 29 deliveries (63 of 1,800 or 3.5%), the mother was found to be HBsAg positive. All were asymptomatic and many (41 of 63 or 65%) without risk factors would remain undetected if HBsAg screening were performed on selected groups. Most of the HBsAg-positive mothers (54 of 63 or 85.7%) were found to be chronic carriers and 30.2% (19 of 63) were also hepatitis B e antigen (HBeAg) positive, indicating high infectivity. Although 23 cord blood were positive for HBsAg or HBeAg, none were positive for IgM antibody to hepatitis B core antigen (IgM anti-HBc), suggesting transplacental transmission of the antigens rather than intrauterine infection. These findings are discussed in relation to the cost-effectiveness of routine prenatal screening and immunization of risk babies compared with universal infant immunization.  相似文献

Reviews the book, Research with the locus of control construct, vol. 3: Extensions and limitations edited by Herbert M. Lefcourt (1984). Herbert Lefcourt's third edited volume on the topic of research with the locus of control construct is directed to discussions of extensions and limitations of the construct. It is a very timely book, and therefore of interest to researchers on the locus of control (LOC). In Part I, comprehensive reviews are presented in three areas: work and retirement, psychophysiology, and cross-cultural researches. In Part II, two chapters describe the limitations and suggest alternatives. The five chapters are extensive and critical, as well as constructive in presenting ideas and models for future research. The editor and the authors have done an excellent job. As a reference source the book eminently meets the needs of researchers in the area. The editor has provided a succinct introduction and epilogue, which are very helpful in orienting the reader and maintaining a balanced view of the appropriate uses of the concept. The editor and authors of the book are to be congratulated for a thorough, provocative, and balanced outlook presented in this volume. I recommend the book highly.
