Androgens repress expression of many genes, yet the mechanism of this activity has remained elusive. The cytokine, interleukin-6, is active in a variety of biological systems, and its expression is repressed by androgens. Accordingly we dissected the mechanism of androgen's ability to inhibit interleukin-6 expression at the molecular level. In a series of co-transfection assays, we found that 5alpha-dihydrotestosterone, through the androgen receptor, repressed activation of the interleukin-6 promoter, in part, by inhibiting NFkappaB activity. It did not appear that 5alpha-dihydrotestosterone inhibited NFkappaB by activating the androgen receptor to compete for the NFkappaB response element as we could not detect androgen receptor binding to the IL-6 promoter by DNase I footprinting assay. However, by electrophoretic mobility shift assay we found that 5alpha-dihydrotestosterone repressed formation of NFkappaB middle dotNFkappaB response element complex formation. In LNCaP prostate carcinoma cells, 5alpha-dihydrotestosterone achieved this effect through maintenance of IkappaBalpha protein levels in the face of phorbol ester, a stimulus that results in IkappaBalpha degradation. Finally, we confirmed that IkappaBalpha inhibits NFkappaB-mediated activation of the interleukin-6 promoter. These data suggest that maintenance of IkappaBalpha levels may represent the first identified mechanism for androgen-mediated repression of a natural androgen-regulated gene. 相似文献

Contrast effects occur when people judge the behavior and attitudes of others relative to their own. We tested a motivational account suggesting that these effects arise because people tailor their judgments of others to affirm their own self-worth. Consistent with that interpretation, participants displayed more egocentric contrast in their judgments of another person's intelligence (i.e., their evaluation of his score on the Scholastic Aptitude Test was more negatively related to their own score) after their self-esteem was threatened than after it was bolstered (Studies 1 and 2). High-self-esteem individuals displayed more judgmental contrast overall than did their low-esteem counterparts (Study 2). Strongly pro-choice participants whose esteem was threatened also displayed more contrast in their judgments of another person's attitude on abortion, relative to esteem-bolstered participants (Study 3). Elevated concentrations of intracellular calcium ([Ca]i) have been implicated as an important signalling event during attaching and effacing (A/E) lesion formation by enteropathogenic Escherichia coli (EPEC). The highly localized nature of the cytoskeletal and cell surface alterations occurring during A/E lesion formation suggests that there should be equally localized EPEC-induced signalling events. To analyze further the calcium responses to infection of HEp-2 cells by EPEC, we employed calcium-imaging fluorescence microscopy, which allows both temporal and spatial measurements of [Ca]i in live cells. Using this imaging technique, not only were we unable to detect any significant elevation in [Ca]i at sites of A/E EPEC adhesion, but, with several different classical EPEC and enterohemorrhagic E. coli (EHEC) strains and three different infection procedures, each of which resulted in extensive A/E bacterial adhesion, we were unable to detect any significant alterations in [Ca]i in infected cells compared to uninfected cells. In addition, chelation of intracellular free calcium with bis-(aminophenoxy)-ethane-N,N,N',N'-tetraacetic acid (BAPTA) did not, as previously reported, prevent A/E lesion formation. The relation of childhood personality to the development of friendship understanding and moral judgment in adolescence was considered in a longitudinal study. Personality at age 7, assessed with the California Child Q-Set, was characterized in terms of ego-resiliency and ego-control. IQ and social class were also measured. Friendship understanding was assessed when the participants were ages 7, 9, 12, 15, and 19, and moral judgment was elicited when the participants were 12, 15, and 19. Ego-resiliency was found to predict social-cognitive development in adolescence, even after the effects of IQ and childhood measures of social-cognitive development were controlled for. Analyses indicate that the effects of ego-resiliency on social-cognitive development are largely unmediated by the ability to focus attention or by social participation. 相似文献

Many objects in images of natural scenes are so complex and erratic, that describing them by the familiar models of classical geometry is inadequate. In this paper, we exploit the power of fractal geometry to generate global characteristics of natural scenes. In particular we are concerned with the following two questions: 1) Can we develop a measure which can distinguish between different global backgrounds (e.g., mountains and trees)? and 2) Can we develop a measure that is sensitive to change in distance (or scale)? We present a model based on fractional Brownian motion which will allow us to recover two characteristics related to the fractal dimension from silhouettes. The first characteristic is an estimate of the fractal dimension based on a least squares linear fit. We show that this feature is stable under a variety of real image conditions and use it to distinguish silhouettes of trees from silhouettes of mountains. Next we introduce a new theoretical concept called the average Holder constant and relate it mathematically to the fractal dimension. It is shown that this measurement is sensitive to scale in a predictable manner, and hence, provides the potential for use as a range indicator. Corroborating experimental results are presented. 相似文献

With more than 25 million people affected, heart failure (HF) is a global threat. As energy production pathways are known to play a pivotal role in HF, we sought here to identify key metabolic changes in ischemic- and non-ischemic HF by using a multi-OMICS approach. Serum metabolites and mRNAseq and epigenetic DNA methylation profiles were analyzed from blood and left ventricular heart biopsy specimens of the same individuals. In total we collected serum from n = 82 patients with Dilated Cardiomyopathy (DCM) and n = 51 controls in the screening stage. We identified several metabolites involved in glycolysis and citric acid cycle to be elevated up to 5.7-fold in DCM (p = 1.7 × 10−6). Interestingly, cardiac mRNA and epigenetic changes of genes encoding rate-limiting enzymes of these pathways could also be found and validated in our second stage of metabolite assessment in n = 52 DCM, n = 39 ischemic HF and n = 57 controls. In conclusion, we identified a new set of metabolomic biomarkers for HF. We were able to identify underlying biological cascades that potentially represent suitable intervention targets. 相似文献

Extraction of heavy metals by accumulating plants is a method which is currently in development for the "gentle" remediation of contaminated agricultural soils (phytoremediation, see [2]). Areal contaminant mass flux balances are basic criteria for the design of such remediations and their control. A framework for the integration of contaminant balances relating the field scale of remediation with the regional scale of soil monitoring networks is provided by the recently developed method PROTERRA [3]. The objective of the study presented in this paper was to test the suitability of PROTERRA for planning and monitoring gentle soil remediations. For this purpose we applied the PROTERRA method to the contaminated agricultural land in and around Dornach, Switzerland, to assess copper flux balances.The calculations showed that atmospheric deposition and the application of pesticides and manure are important pathways for the inputs of copper. The copper export with a special maize cultivar accumulating heavy metals would be about three times higher than the average metal export with crops. A moderate increase of sewage sludge application would lead to a substantial increase of the copper input. Decisions to remediate the soil should take the uncertainty of mass flux balances both on field scale as well as on regional scale into account. Therefore, an important need of further development of the PROTERRA method is the integration of uncertainty analysis on both scales. 相似文献
