Inactivation of phage T5 by lysed cells after phage multiplication is prevented by a phage-encoded lipoprotein (Llp) that inactivates the FhuA outer membrane receptor protein (K. Decker, V. Krauel, A. Meesmann, and K. Heller, Mol. Microbiol. 12:321-332, 1994). Using FhuA derivatives carrying insertions of 4 and 16 amino acid residues and point mutations, we determined whether FhuA inactivation is caused by binding of Llp to FhuA and which regions of FhuA are important for inactivation by Llp. Cells expressing Llp were resistant not only to phage T5 but to all FhuA ligands tested, such as phage phi 80, colicin M, and albomycin, and they were strongly reduced in the uptake of ferrichrome. Most of the FhuA derivatives which were not affected by Llp were, according to a previously published FhuA transmembrane topology model, located in periplasmic turns and in the TonB box close to the periplasm. Since the ligands bind to the cell surface, interaction of FhuA with Llp in the periplasm may induce a FhuA conformation which impairs binding of the ligands. This conclusion was supported by the increase rather than decrease of colicin M sensitivity of two mutants in the presence of Llp. The only Llp-resistant FhuA derivatives with mutations at the cell surface contained insertions of 16 residues in the loop that determines the permeability of the FhuA channel and serves as the principal binding site for all FhuA ligands. This region may be inactivated by steric hindrance in that a portion of Llp penetrates into the channel. Outer membranes prepared with 0.25% Triton X-100 from cells expressing Llp contained inactivated FhuA, suggesting Llp to be an outer membrane protein whose interaction with FhuA was not abolished by Triton X-100. Llp solubilized in 1.1% octylglucoside prevented T5 inactivation by FhuA dissolved in octylglucoside.  相似文献

Titania photocatalysts were synthesised by the molten salts method by reaction of a titanium precursor with three different alkali metal nitrates (LiNO3, NaNO3, KNO3). The titania powders obtained have been characterised using TEM, XRD, BET and UV-Vis absorption spectroscopy. Their photoactivities were evaluated by degrading a commercial sulfonylurea herbicide, Prosulfuron®. It has been found that the rate constants increased in the following order: Li2 prepared in KNO3, with a rate constant 1.4 times smaller than that of P25. The effect of calcination was also studied, especially for the sample prepared in NaNO3 (TiO2[Na]). An improvement in photocatalytic activity was observed when TiO2[Na] was calcined in the region of 700–800 °C. Even though the photocatalytic activities obtained did not exceed or even equal to that of Degussa P25, nevertheless, this study constitutes the first attempt to synthesise titania photocatalysts by the molten salts method.  相似文献

In the present work, multi-cation-doped (Sr2+–Mg2+) SiAlON ceramics were investigated. MgO and SrO were used in 100:0 and 50:50 molar ratios. The mixture was sintered at 1800° and 1830°C for 1 h in a gas pressure-sintering furnace. The results showed that sintered samples were composed of mainly α- and β-SiAlON phases and small amounts of some Sr-containing phases and SiAlON polytypes. According to Rietveld analysis of X-ray diffraction patterns, Mg is incorporated into the α-SiAlON structure. However, the incorporation of Sr is limited.  相似文献

Choosing an appropriate control scheme to alleviate nonlinearities and uncertainties is not a trivial task, especially when models are not easily available and practical evaluation provides the only means for actual performance assessment. Various factors can contribute to these nonlinearities and uncertainties, such as friction and stiction. Thus, this article investigates four different control schemes, namely PID, adaptive, conventional sliding mode control (SMC) and integral sliding mode control (ISMC) which are implemented in the Bristol Elumotion Robot Hand (BERUL) to analyse and overcome the aforementioned problems. The hand has five fingers with 16 joints and all fingers are underactuated. The implementation of the proposed control schemes are challenging since the BERUL fingers have significant friction, stiction and unknown parameters. The fingers are light in weight and fragile. Comparative performance characteristics have shown that the ISMC is the most suitable candidate to provide good experimental trajectory following and positioning control for underactuated BERUL fingers.  相似文献

Electrical conductivity measurements on EUROCAT V2O5–WO3/TiO2 catalyst and on its precursor without vanadia were performed at 300°C under pure oxygen to characterize the samples, under NO and under NH3 to determine the mode of reactivity of these reactants and under two reaction mixtures ((i) 2000 ppm NO + 2000 ppm NH3 without O2, and (ii) 2000 ppm NO + 2000 ppm NH3 + 500 ppm O2) to put in evidence redox processes in SCR deNOx reaction.It was first demonstrated that titania support contains certain amounts of dissolved W6+ and V5+ ions, whose dissolution in the lattice of titania creates an n-type doping effect. Electrical conductivity revealed that the so-called reference pure titania monolith was highly doped by heterovalent cations whose valency was higher than +4. Subsequent chemical analyses revealed that so-called pure titania reference catalyst was actually the WO3/TiO2 precursor of V2O5–WO3/TiO2 EUROCAT catalyst. It contained an average amount of 0.37 at.% W6+dissolved in titania, i.e. 1.07 × 1020 W6+ cations dissolved/cm3 of titania. For the fresh catalyst, the mean amounts of W6+ and V5+ ions dissolved in titania were found to be equal to 1.07 × 1020 and 4.47 × 1020 cm−3, respectively. For the used catalyst, the mean amounts of W6+ and V5+ ions dissolved were found to be equal to 1.07 × 1020 and 7.42 × 1020 cm−3, respectively. Since fresh and used catalysts have similar compositions and similar catalytic behaviours, the only manifestation of ageing was a supplementary progressive dissolution of 2.9 × 1020 additional V5+ cations in titania.After a prompt removal of oxygen, it appeared that NO alone has an electron acceptor character, linked to its possible ionosorption as NO and to the filling of anionic vacancies, mostly present on vanadia. Ammonia had a strong reducing behaviour with the formation of singly ionized vacancies. A subsequent introduction of NO indicated a donor character of this molecule, in opposition to its first adsorption. This was ascribed to its reaction with previously adsorbed ammonia strongly bound to acidic sites. Under NO + NH3 reaction mixture in the absence of oxygen, the increase of electrical conductivity was ascribed to the formation of anionic vacancies, mainly on vanadia, created by dehydroxylation and dehydration of the surface. These anionic vacancies were initially subsequently filled by the oxygen atom of NO. No atoms, resulting from the dissociation of NO and from ammonia dehydrogenation, recombined into dinitrogen molecules. The reaction corresponded to
. In the presence of oxygen, NO did not exhibit anymore its electron acceptor character, since the filling of anionic vacancies was performed by oxygen from the gas phase. The electrical resistivities of high purity lanthanum, cerium, praseodymium, and neodymium were investigated as a function of temperature over the range from room temperature to near the melting points of the metals.
