Current methods for the detection of Mycobacterium tuberculosis (Mtb) are either time consuming or require expensive instruments and are thus are not suitable for point‐of‐care diagnosis. The design, synthesis, and evaluation of fluorogenic probes with high specificity for BlaC, a biomarker expressed by Mtb, are described. The fluorogenic probe CDG‐3 is based on cephalosporin with substitutions at the 2 and 7 positions and it demonstrates over 120 000‐fold selectivity for BlaC over TEM‐1 Bla, the most common β‐lactamase. CDG‐3 can detect 10 colony‐forming units of the attenuated Mycobacterium bovis strain BCG in human sputum in the presence of high levels of contaminating β‐lactamases expressed by other clinically prevalent bacterial strains. In a trial with 50 clinical samples, CDG‐3 detected tuberculosis with 90 % sensitivity and 73 % specificity relative to Mtb culture within one hour, thus demonstrating its potential as a low‐cost point‐of‐care test for use in resource‐limited areas.  相似文献

In this paper, we show that the discrete GI/G/1 system with Bernoulli retrials can be analyzed as a level-dependent QBD process with infinite blocks; these blocks are finite when both the inter-arrival and service times have finite supports. The resulting QBD has a special structure which makes it convenient to analyze by the Matrix-analytic method (MAM). By representing both the inter-arrival and service times using a Markov chain based approach we are able to use the tools for phase type distributions in our model. Secondly, the resulting phase type distributions have additional structures which we exploit in the development of the algorithmic approach. The final working model approximates the level-dependent Markov chain with a level independent Markov chain that has a large set of boundaries. This allows us to use the modified matrix-geometric method to analyze the problem. A key task is selecting the level at which this level independence should begin. A procedure for this selection process is presented and then the distribution of the number of jobs in the orbit is obtained. Numerical examples are presented to demonstrate how this method works.  相似文献

We show that the discrete time MAP/D/k presented by Chaudhry et al. (Oper. Res. Lett. 30(3) (2002) 174) has a special structure which results in a simple and more efficient computational scheme than they have presented. Specifically, we show that the computational efforts for the matrix G at each iteration can be reduced from O(d3k3m3) to O(dk3m3) by rearranging the state space and then capitalizing on the resulting structure. Two electron rich Bodipy dyes with strong absorptivities in the visible region were designed and synthesized as potential electron donors in bulk heterojunction photovoltaic constructs. Overall efficiency is above 1%, with impressive responsiveness at both UV and near-IR ends of the visible spectrum. We consider the M/G/1 and GI/M/1 types of Markov chains for which their one step transitions depend on the times of the transitions. These types of Markov chains are encountered in several stochastic models, including queueing systems, dams, inventory systems, insurance risk models, etc. We show that for the cases when the time parameters are periodic the systems can be analyzed using some extensions of known results in the matrix-analytic methods literature. We have limited our examples to those relating to queueing systems to allow us a focus. An example application of the model to a real life problem is presented.  相似文献

Hybrid solar cells have been fabricated using perylene monoimide–monoanhydride dyes with nanorod zinc oxide electrodes as electron transporting layers. We have investigated the influence of the spacer alkyl chain length of perylene monoimide–monoanhydride (PMIMA) dyes on the device performance in hybrid solar cells using nanorod zinc oxide electrodes. Nanorod zinc oxide electrodes with 50–150 nm of diameter were synthesized in the presence of PEG400 by using microwave heating method. We observed that the dyes with longer and brunched alkyl chains exhibit higher efficiencies in hybrid solar cells. We report the highest efficiency obtained with zinc oxide nanorods under standard conditions for perylene monoimide–monoanhydride derivative with PMIMA_1 that performs 400 mV open circuit voltage, 2.81 mA/cm2 short-circuit current and 0.59% overall conversion efficiency.  相似文献
