We previously demonstrated increased apolipoprotein B (apoB) mRNA editing, elevated levels of mRNA for the catalytic component of the apoB mRNA editing complex, apobec-1, and increased secretion of the product of the edited mRNA, apoB48, in very low density lipoproteins (VLDL) in primary cultures of Sprague-Dawley rat hepatocytes following insulin treatment. In order to determine the effect of in vivo hyperinsulinemia on these processes, we determined apoB mRNA editing, apobec-1 expression, hepatic expression of mRNA for apoB and other VLDL apoproteins, and the quantity and composition of plasma VLDL in the hyperinsulinemic fatty Zucker rat. Total apoB mRNA content of the livers of the fatty rats and lean littermates did not differ, however, edited apoB message coding for hepatic apo B48, and abundance of mRNA for the catalytic subunit of the apoB mRNA editing complex, apobec-1, was increased by 1.7-and 3.3-fold, respectively, in fatty rats. ApoCIII mRNA abundance was increased in livers of fatty rats as well, but the abundance of hepatic apoE mRNA in the fatty animal was not different from that of the lean rat. Hepatic apoAI mRNA abundance was also increased in the fatty rats. Associated with increased apoB mRNA editing, was the 1.7-fold increase in the fraction of apoB in plasma as apoB48 in fatty rats. VLDL-triglyceride and-apoB in plasma were 15-and 3-fold higher, respectively, in fatty Zucker rats compared to lean littermates, indicating both enrichment of VLDL with triglycerides and increased accumulation of VLDL particles. Increased hepatic expression of mRNA for apoCIII and apoAI was associated with increased content of apoC (and relative depletion of apoE) in VLDL of fatty rats, and plasma apoAI was increased in fatty Zucker rats, primarily in the HDL fraction. The current study provides further evidence that chronic exposure to high levels of insulin influences both the quantity of and lipid/apoprotein composition of VLDL in plasma. The increased apoC and decreased apoE (as well as increased triglyceride) content of VLDL in the fatty Zucker rat observed in the current study may affect VLDL clearance and therefore may be a factor in the observed accumulation of VLDL in the plasma of the fatty hyperinsulinemic Zucker rats.  相似文献

The subcritical growth of fatigue cracks under (tension-tension) cyclic loading is demonstrated for ceramic materials, based on experiments using compact C(T) specimens of a MgO-partially-stabilized zirconia (PSZ), heat-treated to vary the fracture toughness K c from ∼3 to 16 MPa·m1/2 and tested in inert and moist environments. Analogous to behavior in metals, cyclic fatigue-crack rates (over the range 10−11 to 10−5 m/cycle) are found to be a function of the stress-intensity range, environment, fracture toughness, and load ratio, and to show evidence of fatigue crack closure. Unlike toughness behavior, growth rates are not dependent on through0-thickness constraint. Under variable-amplitude cyclic loading, crack-growth rates show transient accelerations following low-high block overloads and transient retardations following high-low block overloads or single tensile overloads, again analogous to behavior commonly observed in ductile metals. Cyclic crack-growth rates are observed at stress intensities as low as 50% of K c , and are typically some 7 orders of magnitude faster than corresponding stress-corrosion crack-growth rates under sustained-loading conditions. Possible mechanisms for cyclic crack advance in ceramic materials are examined, and the practical implications of such "ceramic fatigue" are briefly discussed.  相似文献

Hydrodynamic studies were conducted in gas-liquid-solid systems (0.1 m ID, 2 m high) of 3.0 mm glass beads and of 2.1 mm polypropylene low-density particles, with particles densities of 2471 and 1290 kg/m3, respectively. Simultaneous measurement of differential pressure and bubble conductivity probe signals sampled at 500 Hz for 60 s enabled the investigation of the change in flow structure in relation to the flow regime transitions. Superficial gas velocities ranged between 0.010 and 0.052 m/s for polypropylene particles, and extended to 0.12 m/s for glass beads, while the superficial liquid velocities covered the ranges of 0.0007-0.045 m/s for polypropylene particles, and ranged up to 0.056 m/s for glass beads.Spectral analysis of the pressure fluctuations revealed a transition from dispersed to coalesced bubbling flow with decreasing liquid velocity for a given superficial gas velocity. The use of a conductivity probe facilitated characterization of the local flow structure in terms of bubble movement. The measurements were extensively analyzed using fractals and chaos, power spectra frequency analysis and wavelet decomposition in addition to the standard statistical analyses. The coefficient of variation of the bubble probe signals was found to be the most effective in deducing the transition velocity between coalesced and dispersed bubbling flow regimes, while wavelet energy confirmed the similarity in the distribution between two axial positions once operated in the dispersed flow regime. Comparison of the flow structure between glass beads and polypropylene particles showed that both the minimum liquid fluidization velocity and the transition velocity between the bubble flow regimes were much higher for the glass beads than for the lighter polypropylene particles. Furthermore, the standard deviations of the decomposed bubble probe signals through wavelet transformation successfully highlighted the difference between the two systems of particles.  相似文献

The structure of a suite of high-volatile, bituminous Surat Basin, Queensland coals has been investigated by a combination of analytical probes. These included elemental analyses, pyrolysis-gas chromatography-mass spectroscopy and Fourier transform infrared spectroscopy, together with a study of their liquefaction products in both tetralin and solvent free-tin catalysed hydrogenations. Samples were gathered across a 300 m depth interval intersected by the sampling well. Most techniques revealed clear but subtle differences in structure as a function of depth. The oils produced by solvent free-tin catalysed hydrogenation, however, showed very distinct dependence with depth and the waxy content, as indicated by 1H-NMR, could be correlated with the coal seam gas content.  相似文献

The entrenched position now taken by participatory planning in urban planning practice has made the call for community participation in the planning process now higher than ever. Community participation has been well-acknowledged to give local people a voice in planning decisions. This paper assessed the level of participation by local people in the management of urban green spaces. Using a case study research approach, Kumasi city, once the garden city of Africa was selected as the study area. The Kumasi city authorities, and residents including opinion leaders, and officials of allied bodies on green spaces constituted the study’s target population. A blend of qualitative research techniques such as in-depth interviews, focus group discussion, and personal observation was employed whilst theoretical sampling technique was adopted. It was found out that the participation of the local people in the management of green space was low. The local people were neither consulted nor informed on green space projects (parks and gardens) by the city authorities. They were also not empowered to self-facilitate initiatives on green spaces and passively involved in final decisions on green spaces. To correct the situation, it is recommended that features of the communicative planning theory such as dialogue, consensus building, and facilitative leadership should be given attention in the urban planning system of Kumasi especially on green space projects.  相似文献

Whole-genome sequencing (WGS) data present a readily available resource for mitochondrial genome (mitogenome) haplotypes that can be utilized for genetics research including population studies. However, the reconstruction of the mitogenome is complicated by nuclear mitochondrial DNA (mtDNA) segments (NUMTs) that co-align with the mtDNA sequences and mimic authentic heteroplasmy. Two minimum variant detection thresholds, 5% and 10%, were assessed for the ability to produce authentic mitogenome haplotypes from a previously generated WGS dataset. Variants associated with NUMTs were detected in the mtDNA alignments for 91 of 917 (~8%) Swedish samples when the 5% frequency threshold was applied. The 413 observed NUMT variants were predominantly detected in two regions (nps 12,612–13,105 and 16,390–16,527), which were consistent with previously documented NUMTs. The number of NUMT variants was reduced by ~97% (400) using a 10% frequency threshold. Furthermore, the 5% frequency data were inconsistent with a platinum-quality mitogenome dataset with respect to observed heteroplasmy. These analyses illustrate that a 10% variant detection threshold may be necessary to ensure the generation of reliable mitogenome haplotypes from WGS data resources.  相似文献

The nitric oxide–guanylyl cyclase-1–cyclic guanylate monophosphate (NO–GC-1–cGMP) pathway is integral to the control of vascular tone and morphology. Mice lacking the alpha catalytic domain of guanylate cyclase (GC1−/−) develop retinal ganglion cell (RGC) degeneration with age, with only modest fluctuations in intraocular pressure (IOP). Increasing the bioavailability of cGMP in GC1−/− mice prevents neurodegeneration independently of IOP, suggesting alternative mechanisms of retinal neurodegeneration. In continuation to these studies, we explored the hypothesis that dysfunctional cGMP signaling leads to changes in the neurovascular unit that may contribute to RGC degeneration. We assessed retinal vasculature and astrocyte morphology in young and aged GC1−/− and wild type mice. GC1−/− mice exhibit increased peripheral retinal vessel dilation and shorter retinal vessel branching with increasing age compared to Wt mice. Astrocyte cell morphology is aberrant, and glial fibrillary acidic protein (GFAP) density is increased in young and aged GC1−/− mice, with areas of dense astrocyte matting around blood vessels. Our results suggest that proper cGMP signaling is essential to retinal vessel morphology with increasing age. Vascular changed are preceded by alterations in astrocyte morphology which may together contribute to retinal neurodegeneration and loss of visual acuity observed in GC1−/− mice.  相似文献

The laminar burning velocity is a fundamental property of a fuel that affects many aspects of its combustion behaviour. Experimental values are required to validate kinetic simulations, and also to provide input for models of flashback, minimum ignition energy and turbulent combustion.A constant volume vessel (rated at 3.4 MPa) in conjunction with a multi-zone model was used to calculate burning velocity from pressure and schlieren data, allowing the user to select data uncorrupted by heat transfer or cellularity. Using the pressure rise data allows measurements to be derived for much higher pressures and temperatures than when the constant pressure data are used. A 12 term correlation for burning velocity was fitted to the data.n-Heptane, iso-octane, toluene, ethylbenzene and ethanol were tested over a wide range of initial pressures (50, 100, 200 and 400 kPa), temperatures (310, 380 and 450 K) and equivalence ratios (0.7–1.4), along with tests using combustion residuals at mole fractions of up to 0.3. The results compared favourably with the limited data already published, especially at high pressures. Conditions at the onset of cellularity are given for iso-octane.  相似文献

Destructive distillation of solar heat transfer fluids was conducted to determine the types of pyrolytic products which might be formed in solar collectors under conditions of stagnation or malfunction. The distillates were analyzed by gas chromatography/mass spectrometry (GC/MS) to determine the types of compounds which were present and the minor components which might be formed. Dehydration products were formed from ethylene and propylene glycols with ethylene oxide and propylene oxide being minor products. A high aromatic petroleum heat transfer fluid yielded distillation fractions enriched in quinolines or isoquinolines and methylated derivatives thereof. Fractionation of this aromatic heat transfer fluid showed that the basic fraction of unheated fluid also contained aza-arenes which exhibited mutagenic activity in the Ames' bioassay. GC/MS showed that the basic fraction from unheated heat transfer fluid also contained benzoquinolines which appeared to be lacking in heat degraded samples of the same fluid. Fractionation of heat transfer fluids, in particular petroleum-based fluids, may often be necessary in order to concentrate minor components so that they can be identified by GC/MS and in order to detect mutagenic activities without interference from cytotoxic components.  相似文献
