Enzootic bovine granulomatosis (EBG) is a disease that affects beef calves in the flooded Savanna regions of Venezuela. Although Pythium insidiosum was originally the suspected etiologic agent, nothing was done to demonstrate its presence in the infected animals. The objective of this study was to investigate the etiologic agent causing cutaneous lesions in a group of 63 calves diagnosed with EBG. The collected samples were analyzed by histopathology, immunohistochemistry, serology, and mycological techniques. The clinical manifestations of BEG included ulcerated and swollen granulomatous lesions on the limbs of young calves. Histopathological studies revealed an eosinophilic inflammatory reaction with neutrophils, giant cells and branching hyphae with the Splendore-Hoeppli phenomenon. Immunofluorescence analysis identified the branching hyphae as elements of the fungal-like organism P. insidiosum. Wet mounts in 10% KOH revealed hyaline slender branching hyaline coenocytic hyphae, but cultures from those samples were negative. Anti-P. insidiosum immunoglobulins were also demonstrated in the collected sera of the affected bovines using latex agglutination, immunodiffusion, ELISA, and Western Blot. The histopathological, immunohistochemical, and serological tests showed for the first time that P. insidiosum could be involved in epizootic bovine pythiosis. 相似文献

The aims were (a) to quantify the number of Duddingtonia flagrans chlamydospores per gram of faeces (CPG) recovered from sheep administered with different oral doses and, (b) to describe the relationship between CPG and eggs per gram of faeces (EPG) on the efficacy to reduce Haemonchus contortus infective larvae. Three doses of chlamydospores per kg BW were orally administered during seven days: (T1) non treated control group, (T2) 1 × 106, (T3) 2.5 × 106 and (T4) 5 × 106. Three lambs, infected with H. contortus, were used per group. Faeces were obtained from the rectum of each lamb during the fungal administration period (days 0–6) and for six days after that period. Four coproculture replicates were made from each animal in days 2, 4, 6, 8 and 10. A higher chlamydospore dose produced higher CPG in faeces (p < 0.05), but a clear dose dependent effect was not found either in the larvae reduction or in the CPG:EPG ratio. When ratios were re-analyzed, independently of the treatment groups of origin, a better efficacy was obtained with a ratio from 5 to 10 CPG:EPG and a higher ratio (>10 per egg) showed a lower reduction efficacy (p < 0.05). The binomial analysis showed that for each unit of increment in CPG:EPG ratio there was a reduction of larvae number until a point (between 5 and 10 CPG:EPG) where no further reduction was detected. The surface response test indicated that the number of larvae was reduced by CPG until possible saturation. The highest CPG:EPG ratios did not necessarily improve efficacy of D. flagrans. 相似文献

Biofilm formation where bacterial cells adhere to a surface and surround themselves in a polysaccharide matrix is thought to be an important factor in disease initiation and persistence for many bacterial species. We have examined biofilm formation by Mycoplasma mycoides subsp. mycoides small colony using a simple model without an air/liquid interface and have found that adherent Mmm SC was more resistant to many stresses, including heat, osmotic shock and oxidative stress. Biofilms of Mmm SC also exhibited remarkable persistence and were able to survive for up to 20 weeks in stationary phase. Significant variation was seen between Mmm SC strains in their ability to form a biofilm and the morphology of the biofilm produced with some strains unable to produce microcolonies. Proteomic analysis found that a number of proteins linked to adherence were over-expressed in biofilms compared with planktonic cells. 相似文献

Crimean–Congo haemorrhagic fever (CCHF) is a widespread tick‐borne viral zoonosis with a case‐fatality rate ranging from 9% to 50% in humans. Although a licensed vaccine to prevent infection by the CCHF virus (CCHFV) exists, its ability to induce neutralizing antibodies is limited and its efficacy against CCHFV remains undetermined. In addition, controlling CCHF infections by eradication of the tick reservoir has been ineffective, both economically and logistically, and the treatment options for CCHF remain limited. In this review, we first critically discuss the existing animal models to evaluate therapeutics for CCHF. We then review the therapeutic options for CCHF that have been investigated in human cases, followed by investigational drugs that have been evaluated in pre‐clinical studies. We highlight the importance of understanding human prognostic factors in developing an animal model for CCHF that recapitulates hallmarks of human disease and its implication for selecting therapeutic candidates. 相似文献

The effects of intensive banana production with high mineral‐fertilizer application and of extensive pastures were compared regarding water quality in a lowland region of SE Mexico. We monitored NO$ _3^- $ , NO$ _2^- $ , and PO43– concentrations in groundwater (80 m depth), subsurface water (5 m depth), and surface water (open‐ditch drainage) at monthly intervals for a one‐year period. Irrespective of the land use, the NO$ _3^- $ concentrations in all water bodies were lower than the threshold value for drinking water and aquatic life. Particularly in areas with intense banana production, the NO$ _2^- $ contents in water exceeded the safety thresholds for drinking water of 1.0 mg L–1 (WHO, 2006) and aquatic ecosystems of 0.2 mg L–1 (OATA, 2008). Water from pastureland showed significantly higher PO43– concentration than that from the banana plantation, indicating a high risk of eutrophication. There is a need to provide recommendations for optimal time and amount of N application in commercial banana production and for limitation of P inputs in pasturelands to avoid further contamination of water bodies. 相似文献

In the present work, land cover and land use changes between 2003 and 2008 were assessed in the Cupatitzio River sub‐basin located in the Trans‐Mexican Volcanic System and Balsas Depression in Mexico. The effects of land cover conversion from temperate forest to avocado orchards on soil properties were determined. Land cover and land use databases were built for the years 1975, 2003, and 2008 using available cartographic information and interpretation of panchromatic SPOT images for the year 2008. Additionally, soil sampling was carried out in 16 representative sites in the higher parts of the sub‐basin, where avocado plantation dynamics have caused important changes in areas covered by temperate forests. Results show that (i) temperate coniferous and tropical low deciduous forests were reduced at an annual rate of 0·9 per cent (1, 001 ha y−1) between 1975 and 2008; (ii) cropland areas expanded at an annual rate of 0·7 per cent between 1975 and 2008, representing a growth of 553 ha y−1; (iii) wet soil aggregate stability, mechanical resistance to penetration and nitrate (N‐NO3) concentration in the topsoil layer of Andosols were significantly affected by land use conversion to avocado orchards. Other soil properties such as organic matter, interchangeable K, and available P showed degradation tendencies. These results suggest the importance of implementing strategies for mitigating the accelerated process of transformation and degradation of natural resources in the Cupatitzio River sub‐basin. Turkeys are extremely sensitive to aflatoxin B1 (AFB1) which causes decreased growth, immunosuppression and liver necrosis. The purpose of this study was to determine whether probiotic Lactobacillus, shown to be protective in animal and clinical studies, would likewise confer protection in turkeys, which were treated for 11 days with either AFB1 (AFB; 1 ppm in diet), probiotic (PB; 1 × 1011 CFU/ml; oral, daily), probiotic + AFB1 (PBAFB), or PBS control (CNTL). The AFB1 induced drop in body and liver weights were restored to normal in CNTL and PBAFB groups. Hepatotoxicity markers were not significantly reduced by probiotic treatment. Major histocompatibility complex (MHC) genes BG1 and BG4, which are differentially expressed in liver and spleens, were not significantly affected by treatments. These data indicate modest protection, but the relatively high dietary AFB1 treatment, and the extreme sensitivity of this species may reveal limits of probiotic-based protection strategies. 相似文献

Ensiling fresh dairy cattle manure (CM), corn stover, molasses (MO) or bakery by-products (BBP), as a viable method to produce good quality silage was evaluated. Silages composition was as follows: 1) molasses silage: CM, corn stover and molasses; and 2) BBP silages: CM, corn stover, and BBP. A low (250 g/kg dry matter) and a high (500 g/kg dry matter) level of CM with MO or BBP silages were incorporated in diets, and compared with a control diet (without CM silage) in lambs. There was a linear increase (P < 0.01) of dry matter (DM) intake and a quadratic increase of fiber digestion. Linear decrease (P < 0.05) in acetate to propionate ratio was observed with increased concentrations of MO or BBP in silages. Increasing MO silage caused a linear reduction of N intake and retained N, as well as a linear increase of fecal N. There were quadratic effects of BBP silage level on N intake, urine and retained, being the lowest values observed with the low level of BBP silage. Addition of BBP silages (250 or 500 g/kg DM) bring higher DM intake and total tract digestion of fiber, as compared to those diets without CM silage. 相似文献
