目的探讨表没食子儿茶素没食子酸酯(EGCG)对鼻咽癌细胞株CNE-1的生长抑制作用及其机制。方法不同浓度EGCG作用于鼻咽癌CNE-1细胞,用MTT法检测不同时间点的细胞生长抑制率,同时用RT-PCR法检测其survivin mRNA。结果EGCG作用后CNE-1细胞的生长抑制率升高,同时伴survivin mRNA表达下调,呈浓度和时间依赖性。结论EGCG可明显抑制鼻咽癌细胞株CNE-1的生长,其机制是下调该细胞survivin mRNA的表达。  相似文献

The aim of this work was to develop a novel targeted drug-loaded microbubble (MB) and to investigate its chemotherapy effect in vitro. Paclitaxel (PTX)-loaded lipid MBs were prepared by a mechanical vibration technique. The LyP-1, a breast tumor homing peptide, was coated onto the surface of PTX-loaded MBs through biotin-avidin linkage. The resulting targeted drug-loaded MBs were characterized and applied to ultrasound-assisted chemotherapy in breast cancer cells. Our results showed the ultrasonic MBs were able to achieve 43%-63% of drug encapsulation efficiency, depending on drug loading amount. The binding affinity assay indicated the attachment of targeted MBs to human MDA-MB-231 breast cancer cells was highly efficient and stable even with ultrasonic irradiation on. The cellular uptake efficiency of payload in targeted MBs was 3.71-, 4.95-, 7.43- and 7.66-fold higher than that of non-targeted MBs at the applied ultrasound time of 30, 60, 90 and 120 s, respectively. In addition, the cell proliferation inhibition assay showed the cell viability of targeted PTX-loaded MBs was significantly lower than that of non-targeted PTX-loaded MBs and non-targeted unloaded MBs when ultrasound was utilized. In conclusion, the study indicated the LyP-1-coated PTX-loaded MBs significantly increased the antitumor efficacy and can be used as a potential chemotherapy approach for ultrasound-assisted breast cancer treatment.(E-mail: hr.zheng@siat.ac.cn)  相似文献

Background

Biphenotypic acute leukemia is a rare disorder that is difficult to diagnose. It displays features of both myeloid and lymphoid lineage. There is still a lack of studies in biphenotypic acute leukemia in a Chinese population. We present here a comprehensive investigation of the clinical and biological characteristics, and outcome of biphenotypic acute leukemia in our hospital in over a seven year period.

Design and Methods

We retrospectively analyzed 452 adult acute leukemia patients diagnosed according to French-American-British (FAB) classification and biphenotypic acute leukemia diagnosed according to European Group for the Immunological Characterization of Leukemias (EGIL) classification, respectively. Biological characteristics, response to treatment, and outcome were examined in biphenotypic acute leukemia patients and compared with that in acute myeloid leukemia and acute lymphoblastic leukemia patients with complete follow-up profiles diagnosed in the same period.

Results

Of 452 acute leukemia patients, 21 cases (4.6%) were diagnosed as biphenotypic acute leukemia. Among them, 14 (66.7%) were B lymphoid and myeloid, 5 (23.8%) were T lymphoid and myeloid, one (4.8%) was T/B lymphoid and one (4.8%) was trilineage differentiation. When compared with acute myeloid leukemia and acute lymphoblastic leukemia, patients with biphenotypic acute leukemia showed significantly higher incidence of CD34 antigen expression, unfavorable karyotypes, and extramedullary infiltration (p<0.05). In this cohort of patients with biphenotypic acute leukemia, t(9;22) was the most common abnormality in chromosome structure. The median disease-free survival and overall survival in biphenotypic acute leukemia patients was five months and ten months, respectively, significantly shorter than those in acute myeloid leukemia and acute lymphoblastic leukemia patients (p<0.05).

Conclusions

The prognosis of biphenotypic acute leukemia patients is poor when compared with de novo acute myeloid leukemia or acute lymphoblastic leukemia. Biphenotypic acute leukemia patients showed a much higher incidence of CD34 antigen expression, complex abnormal karyotype, extramedullary infiltration, relapse, and resistance to therapy after relapse.  相似文献

目的 探讨美蓝还原酶法快速检测突发水污染微生物指标即菌落总数的可行性。 方法 通过对美蓝的褪色时间与GB/T5750.12-2006平板计数法的比较,对菌落总数进行检测。 结果 美蓝的还原褪色时间与菌量的多少呈反比,即菌量越大美蓝褪色时间越短。 结论 美蓝还原酶法操作简便,快速,结果准确可靠,可应用于突发水污染的菌落总数的快速检测。  相似文献

Aarskog-Scott综合征是一种罕见病,典型表现为特殊面容、矮小、性腺发育不良及骨骼畸形等。本文报道1例男性患儿,初次就诊年龄为3岁6个月,主因"发现身高增长缓慢3年"就诊。围生期正常。智力运动发育大致正常。身长90 cm(<第3百分位,-3 SD),特殊面容,关节过伸,双手短粗,脚趾球形。披肩样阴囊,左侧隐睾。FGD1基因第9~12外显子存在1.1 Mb缺失变异,来源于母亲,符合Aarskog-Scott综合征的诊断。患儿自5岁起开始重组人生长激素治疗,治疗18个月,身高增长17 cm,无明显不良反应。当矮小患者同时伴有特殊面容、骨骼畸型及性腺发育不良三联征时,应考虑Aarskog-Scott综合征的可能。  相似文献

背景:在体内条件下,细胞力学的功能研究因其所处生理环境的复杂性、实验条件的不易控制而很难得到满意结果。目的:在成功构建成肌细胞体外培养-力学刺激模型的基础上,研究p38MAPK信号通路在成肌细胞凋亡中的作用及其机制。方法:将体外培养的C2C12细胞分为对照组和SB203580组,SB203580组中加入20mmol/L的p38MAPK抑制剂SB203580。应用细胞应力加载装置FlecellStrainUnit-5000T给细胞提供15%的力值,分别施加0,6,12,24h的周期性张应力。每分钟10个循环,每循环包括3s牵张,3s松弛。Hoechst33258染色观察细胞的形态学变化;流式细胞仪检测细胞凋亡情况;RT-PCR法检测促凋亡基因baxmRNA的表达;Westernblot检测信号通路中p38MAPK和p-p38MAPK蛋白的表达。结果与结论:随着加力时间的延长,细胞逐渐出现核固缩及凋亡小体,凋亡率增加(P〈0.05),baxmRNA表达增多(P〈0.05);细胞p38MAPK和p-p38MAPK蛋白均在加力6h达到最低,此后逐渐升高。p38MAPK抑制剂SB203580可抑制加力引起的细胞凋亡,减少baxmRNA及p38MAPK和p-p38MAPK蛋白的表达(P〈0.05)。说明p38MAPK信号通路在应力介导的成肌细胞凋亡中起到重要的作用。  相似文献

AIM:To testify the immunogenicity of a conservative B-cell linear epitope of Helicobacter pylori (H pylori) flagellin A. METHODS: Different programs were used to analyze the secondary structure, molecular hydropathy, and surface accessibility of H pylori flagellin A. Linear B-cell epitopes were estimated based on the structural and physiochemical information. Analysis of residue divergence was proposed to screen a conservative linear epitope. The 29-peptide (Pep29mer) synthesized by chemical method, including the predicted conservative B-cell epitope and a known K2d compatible T-cell epitope, was used to immunize mice, and then H pylori-specific antibodies were detected by ELISA. RESULTS: Based on the analyses of divergent amino acid residues, structural and physiochemical characteristics, it was strongly suggested that the short fragment NDSDGR was the core of a conservative linear epitope in flagellin A. Animals immunized by Pep29mer acquired efficient immune response. In detail, serum H pylori-specific IgA and IgGl increased significantly in immunized group, while IgG2a only had an insignificant change. H pylori-specific IgA in gastrointestinal flushing fluid also increased significantly. CONCLUSION: The conservative short fragment NDSDGR is the core of a linear B-cell epitope of flagellin A.  相似文献
