人源Dickkopf1原核表达质粒的构建及表达产物初步鉴定

目的构建人源dickkopf1(dkk1)基因的原核表达质粒,表达、纯化、鉴定其表达产物。方法自足底皮肤组织提取成纤维细胞,抽提mRNA,RT-PCR获得人源dkk1基因片断,构建重组质粒PQE30-dkk1;转化至大肠杆菌内,诱导表达,超声裂菌、镍琼脂糖凝胶层析纯化表达产物,SDS-PAGE电泳、WesternBlot鉴定表达产物。结果RT-PCR获得片断与预期结果相符,双酶切鉴定、测序鉴定证实重组质粒PQE30-dkk1构建成功;SDS-PAGE电泳、抗Dkk1抗体WesternBlot鉴定表达产物为Dkk1蛋白。结论成功构建了人源dkk1的原核表达质粒,并表达、纯化、鉴定了该基因的原核表达产物,为下一步的Dkk1蛋白对皮肤黑素细胞的生物学活性研究奠定基础。     

30例血液学异常的2型糖尿病患者骨髓象分析

目的观察血液学异常的糖尿病患者的骨髓象特征,探讨其临床价值和意义。方法选取血液学异常的30例2型糖尿病患者进行骨髓穿刺,并对骨髓象进行分析。结果 30例糖尿病患者中骨髓诊断为粒系核左移伴继发性贫血17例(56.7%),缺铁性贫血3例(10.0%),铁粒幼红细胞性贫血1例(3.3%),溶血性贫血1例(3.3%),血小板减少性紫癜2例(6.6%),白血病4例(3.3%),多发性骨髓瘤1例(3.3%),干抽1例(3.3%)。结论通过骨髓检查有助于了解2型糖尿病合并血液学异常的病因和发病机制,帮助临床医生及时准确地对症治疗。

1000例老年人心律失常病因的临床研究

本文对1000例老年人心律失常的频率及病因进行了分析,并就其中低氧血症、钙化性主动脉瓣狭窄与老年人心律失常的关系进行了探讨,现报告如下。     

颅脑外伤后致鼻腔反复大出血经中西医结合保守治疗成活一例

患者男，35岁，市郊农民，于1985年1月10日因反复鼻衄严重贫血首次住本院。患者在84年9月3日建筑施工时，不慎从四楼跌下，掉至锅炉上后再落地，右侧头部先着地．当时昏迷半小时，送某医院就医．X片报告有颅底骨拆，治疗25天出院．出院后一周突发右鼻大出血约800毫升．其后3月内又先后4次出血，每次失血量约800—1200毫升。患者在我院就诊时，除严重失血性贫血的症状、体征外，全身系统检查无异常．双眼视力5．1，无搏动性突眼，右眼眶及前额部无血管杂音，双眼底正常。鼻中隔高度右偏。双耳听力正常。入院后一周突发右鼻出血如泉涌，失血1…     

神经干细胞移植延缓失神经肌萎的作用机制

Objective To study the effect and mechanism of transplanting spinal fetal neural stem cells (NSCs) into the peripheral nerve for delaying muscle denervation atrophy. Methods Spinal. fetal NSCs were separated from spinal cord of enceinte 10 to 12 days SD rats, cultured and purified. After three passages, the formed NSC spheres were blew into single cell suspension ( 106/μl×5 μl) and transplanted into the distal part of the transected tibial nerve. 5 μl of cell culture medium was injected into the distal tibial nerve in the control group. Three and 5 months after the transplantation, the distal part of the tibial nerve and the triceps suraes were harvested and identified with specific markers, by means of indirect immunofluorescent staining to evaluate survival and differentiation of transplanted NSCs in the nerve, and to observe the neuromuscular junctions.Results Compare to the control group, atrophy of the triceps suraes muscle was less severe 3 and 5 months after NSCs transplantation. Postsynaptic membrane was also better preserved in NSCs transplanted group. Five months after NSCs transplantation, new synapses (neuromuscular junction) formed in the denervated muscle.Conclusion NSCs transplantation can delay atrophy denervated muscles. NSCs transplantation can not only maintain the structure of postsynaptic membrane, but also form new synapse with the denervated muscle.     

CHARACTERIZATION OF VPI AS IMMUNODOMINANTANTIGEN OF ENTEROVIRUS TYPE 70 ABID ANTIGENICANALYSIS OF VIRUS STRAINS BY MONOCLONAL ANTIBODJES

The neutralizing antigens of eliterovirus type70(E70) are characterized immunochemically. It wasfound that all of the 7 neutralizing McAbs testedcould cambine with both naturally-occurring topcomponents (NTC) and virions in immunodot test,five of them could combine with VPl in the irmnuno-hlot system, and patient sera and antibodies fromimmune and hyperimmune ammals couId only cam-bine with VPl. Furthermore. 10 McAbs (includingthe mentioned 7) were used to analyse the antigenic differences of 8 E70 strains isolated in China in 1985 and the prototype virus J670/71. The results indicate that the 10 McAbs can neutralize all 9 viruses almostequally. The data indicate that VPl is immuno-dominant ancl stable enough to keep ycutralizingtem.     

STUDIES ON PERIPHERAL T LYMPHOCYTE SUBSETS IN PATIENTS WITH AUTOIMMUNE THYROID DISEASES

Peripheral T lymphocyte subsets in 60 cases of Graves disease (GD) and 16 cases of Hashimoto’s thyroiditis (HT) were determined by the use of OKT monoclonal antibodies (McAb). The results showed that in both diseases OKT3 cells (total T cells ) and OKT8 cells（suppressor T cells, TS cells ） were lower and the ratio of LKT4 cells (helper T cells, TH cells )/ OKT8 cells was much higher than those in normal controls. GD patients who were treated with antithyroid drugs (ATD) and whose serum T3, T4 recovered to normal, could be further divided into subgroups of high OKT4 / OKT8 cell ratio and of normal ratio. The pathogenesis, treatment and relapse of autoimmune thyroid diseases (AITD) were discussed in this paper on the basis of the results.
