Prostate apoptosis response-4 (Par-4), an anticancer protein that interacts with cell surface receptor GRP78, can selectively suppress proliferation and induce apoptosis of cancer cells. The core domain of Par-4 (aa 137–195), designated as SAC, is sufficient to inhibit tumor growth and metastasis without harming normal tissues and organs. Nevertheless, the anticancer effects of SAC have not been determined in ovarian cancer cells. Here, we developed a novel method for producing native SAC in Escherichia coli using a small ubiquitin-related modifier (SUMO) fusion system. This fusion system not only greatly improved the solubility of target protein but also enhanced the expression level of SUMO-SAC. After purified by Ni-NTA affinity chromatography, SUMO tag was cleaved from SUMO-SAC fusion protein using SUMO protease to obtain recombinant SAC. Furthermore, we simplified the purification process by combining the SUMO-SAC purification and SUMO tag cleavage into one step. Finally, the purity of recombinant SAC reached as high as 95% and the yield was 25 mg/L. Our results demonstrated that recombinant SAC strongly inhibited proliferation and induced apoptosis in ovarian cancer cells SKOV-3. Immunofluorescence analysis and competitive binding reaction showed that recombinant SAC could specifically induce apoptosis of SKOV-3 cells through combination with cell surface receptor, GRP78. Therefore, we have developed an effective strategy for expressing bioactive SAC in prokaryotic cells, which supports the application of SAC in ovarian cancer therapy. 相似文献

Bimolecular coupling/unimolecular cyclization strategies, including McMurry- and Glaser-type homocoupling reactions, were utilized to synthesize two shape-persistent elliptic macrocycles, which consist of polycyclic aromatic hydrocarbon units. A new class of pi-conjugated dendrimers G0, G1, and G2 was developed through a double-stage divergent/convergent growth approach, in which 5,5,10,10,15,15-hexahexyltruxene was employed as the node and oligo(thienylethynylene)s (OTEs) with different lengths as the branching moieties. The dendrimers were fully characterized by (1)H and (13)C NMR, elemental analysis, gel permeation chromatography, and MALDI-TOF MS. Also, by using atomic force microscopy, it was observed that dendrimer G2 laid nearly flat on the mica surface as a single molecule. Dynamic light scattering results showed that the molecule retained its relatively flat shape in solution. To our best knowledge, dendrimer G2, with a radius approaching 10 nm and a molecular weight of 27 072 Da, was the largest among reported second generation dendrimers. The energy gradient in G2 was constructed by linking OTEs of increasing effective conjugation lengths from the dendritic rim to the core. The intramolecular energy transfer process was studied using steady-state UV-vis absorption and photoluminescent spectroscopies, as well as time-resolved fluorescence spectroscopy. Our structurally extended dendrimers showed an excellent energy funneling ability (their energy transfer efficiencies were all over 95%). All results demonstrate that these dendrimers are promising candidates as light-harvesting materials for optoelectronic devices. 相似文献

The aim of this review article is to introduce recent studies on an emergent class of singlet oxygen photosensitizers of potential applications to the photodynamic therapy,with a primary focus on the cyclometalated transition-metal complexes.Singlet oxygen photosensitization performances of various cyclometalated Ir and Pt scaffolds are reviewed,and the general photophysical properties of relevant systems and the mechanisms of singlet oxygen production via photo-sensitization are also briefly discussed.Thus far,investigations of singlet oxygen sensitization by such Ir and Pt complexes are mainly carried out in organic solvents and under non-physiological conditions,while some research efforts have been made at examining the feasibility of applying pertinent cyclometalated complexes to photodynamic therapy. 相似文献

To analyse the ionization and acceleration properties of an inductive plasma excited by a pulsed current flowing through the planar coil, the extended GLM formulations of the MHD (EGLM‐MHD) model, combined with the high‐temperature thermodynamic and transport model, is employed to simulate the characteristics of the flow. The two‐dimensional axisymmetric calculation captures the generation, growth, and acceleration of the current sheet, and the process is completed in the first half period. The sheet is mainly comprised of lower ionization level ions in the front and higher level ions at the back, and the density is one order higher than that of the residual plasma on the coil surface. As the abscissa value of the sheet is larger than the decoupling distance, a reversed flow emerges, generating a backward impulse, and the negative velocity can be more than 15 km/s at peak intensity B0 = 0.5 T. In the second 1/4 period, the magnetic field and current density distribute non‐linearly on the surface and regularly in the sheet, caused by the reversing of the changing rate of the magnetic field and the particles' radial diffusion. The results at different intensities show that, for the same coil size, the time at which the maximum velocity Vmax appears is advanced as the intensity increases, and Vmax can be greater than 20 km/s above 0.5 T. 相似文献
