Lipid peroxidation of the erythrocyte membrane caused by stimulated polymorphonuclear leukocytes in the presence of ferritin 总被引:1,自引:0,他引:1
Lipid peroxidation of erythrocyte membrane was caused by phorbol myristate acetate (PMA)-stimulated polymorphonuclear leukocytes (PMN) in the presence of ferritin. PMN themselves were not peroxidized. A lag period was observed before the start of the peroxidation reaction. In contrast, ferritin iron was continuously released by PMA-stimulated PMN, suggesting that accumulation of free iron in the reaction system was important for proceeding of the peroxidation reaction. Superoxide dismutase, catalase, hydroxyl radical scavengers and an iron chelator, diethylenetriaminepenta-acetic acid, inhibited the lipid peroxidation, indicating that the lipid peroxidation is initiated by a hydroxyl radical generated from the interaction of H2O2 with ferrous iron released from ferritin. 相似文献

Analysis of plasma catecholamines (norepinephrine, epinephrine and dopamine) by high-performance liquid chromatography using 1,2-diphenylethylenediamine as a fluorescent reagent is described. We have developed an automatic catecholamine analyser, based on pre-column fluorescence derivatization and column switching. The analysis time for one assay was 15 min. The correlation coefficients of the linear regression equations were greater than 0.9996 in the range 10-10,000 pg/ml. The detection limit, at a signal-to-noise ratio of 3, was 2 pg/ml for dopamine. A new method of sample preparation for the pre-column fluorescence derivatization of plasma catecholamines was used. In order to protect the catecholamines from decomposition, an ion-pair complex between boric acid and the diol group in the catecholamine was formed at a weakly alkaline pH. The stabilities of plasma catecholamines were evaluated at several temperatures. After complex formation, the catecholamines were very stable at 17 degrees C for 8 h, and the coefficients of variation for norepinephrine, epinephrine and dopamine were 1.2, 4.2 and 9.3%, respectively. 相似文献

The compound 5,12‐diacetyl‐5,12‐dihydroquinoxalino[2,3‐b]quinoxaline 1 a and its derivatives were prepared, and their solid‐ and solution‐state spectroscopic properties were studied; 1 a shows stronger fluorescence in solution than in the solid state due to aggregation caused by self‐quenching. Phenyl‐ or alkoxy‐substituted derivatives 1 b – d show solid‐state fluorescence with moderate quantum yields of about Φ=0.12–0.15, although the corresponding values are 0.01–0.07 in solution. The spectroscopic properties of alkoxy‐substituted derivatives were hardly changed compared to 1 a and 1 b , although 1 a and 1 b have similar absorption and fluorescence maxima in solution and in the solid state. DFT calculations indicate that orbital switching occurs between HOMO and HOMO‐1 and HOMO‐2 due to orbital interactions with introduced substituents. Crystal structure analysis revealed that the molecules have bent structures around tertiary nitrogen atoms and form a characteristic dimeric structure. 相似文献

Lanthanum(III) has been found to catalyse the reaction of thiosulphate with cyanide. The effects of pH, reaction time and temperature, amount of cyanide and lanthanum, and order of addition of reagents have been investigated. Temperature has little effect in the range 10–30°, but at both 35° and 40° the rate is noticeably faster. Reaction is complete at room temperature in 1.5 hr at pH 9.3–9.6, and in 3 hr at pH 9.1–9.6. The method can be applied to the determination of 1.0 × 10−5−6.0 × 10−4M thiosulphate, with a relative standard deviation of 0.3% for 4 × 10−4M thiosulphate. 相似文献

A surface plasmon resonance (SPR) biosensor that carries DNA-binding small ligands has been developed for the detection of single-nucleotide polymorphisms (SNPs). 3,5-Diaminopyrazine derivatives, with a hydrogen-bonding profile fully complementary to the thymine base, were utilized as recognition elements on the sensor surface, and a target single-stranded DNA sequence was hybridized with a DNA probe containing an abasic site to place this site opposite a nucleobase to be detected. In a continuous flow of sample solutions buffered to pH 6.4 (0.25 M NaCl), the 3,5-diaminopyrazine-based SPR sensor can detect an orphan nucleobase in the duplex with a clear selectivity for thymine over cytosine, guanine, and adenine (5'-GTT GGA GCT GXG GGC GTA GGC-3'/3'-CAA CCT CGA CNC CCG CAT CCG-5'; X=abasic site, N=target nucleobase G, C, A, or T). The SPR response was linear in the concentration range 10-100 nM. Allele discrimination is possible based on the combination of different binding surfaces in a flow cell of the SPR system, which is demonstrated for the analysis of the thymine/cytosine mutation present in 63-meric polymerase chain reaction (PCR) amplification products (Ha-ras gene, codon 12, antisense strand). Comparison with a bulk assay based on 3,5-diaminopyrazine/DNA binding shows that the immobilization of 3,5-diaminopyrazine derivatives on the SPR sensor allows more sensitive detection of the target DNA sequence, and binding selectivity can be tuned by controlling the salt concentration of sample solutions. These features of the DNA-binding small-molecule-immobilized SPR sensor are discussed as a basis for the design of SPR biosensors for SNP genotyping. 相似文献

We measured the affinity of more than 20 sugars with concanavalin A (ConA) by an optical biosensor (surface plasmon resonance sensor) using asialofetuin (ASF) as an immobilized binding partner of ConA. We determined kinetic parameters of the effects of sugars on the dissociation of ConA from ASF quantitatively, and the structural requirements of the functional groups of sugars for binding with ConA. We found that the affinity of ConA for sugars is dependent on its conformation induced by interaction with the binding partner. In addition, the results showed that optical biosensor system is well mimics the interaction of ConA with sugars in biomembrane. 相似文献

Novel vibrational circular dichroism (VCD) studies in the CH region of a series of methyl glycosidic carbohydrates were examined. The specific CH stretching VCD band predicts absolute stereochemistries of their anomeric positions. The C-1 chiral information was extracted to the methoxy substituent as a probe. The concept of the vibrational chirality probe from a single chiral center in the presence of numerous such centers might be useful in determining the absolute configuration, when a multiplicity of chiral centers is present in a molecule such as a carbohydrate. 相似文献

In present study the relevance of using the time-dependent density functional theory (DFT) within the adiabatic approximation for computing oscillator strengths (f) is assessed using different LDA, GGA, and hybrid exchange-correlation (XC) functionals. In particular, we focus on the lowest-energy valence excitations, dominating the UV/visible absorption spectra and originating from benzenelike HOMO(pi)-->LUMO(pi(*)) transitions, of several aromatic molecules: benzene, phenol, aniline, and fluorobenzene. The TDDFT values are compared to both experimental results obtained from gas phase measurements and to results determined using several ab initio schemes: random phase approximation (RPA), configuration interaction single (CIS), and a series of linear response coupled-cluster calculations, CCS, CC2, and CCSD. In particular, the effect of the amount of Hartree-Fock (HF) exchange in the functional is highlighted, whereas a basis set investigation demonstrates the need of including diffuse functions. So, the hybrid XC functionals--and particularly BHandHLYP--provide f values in good agreement with the highly correlated CCSD scheme while these can be strongly underestimated using pure DFT functionals. These results also display systematic behaviors: (i) larger f and squares of the transition dipole moments (mid R:mumid R:(2)) are associated with larger excitation energies (DeltaE); (ii) these relationships present generally a linear character with R>0.9 in least-squares fit procedures; (iii) larger amounts of HF exchange in the XC functional lead to larger f, R:mumid R:(2), as well as DeltaE values; (iv) these increases in f, mid R:mumid R:(2), and DeltaE are related to increased HOMO-LUMO character; and (v) these relationships are, however, not universal since the linear regression parameters (the slopes and the intercepts at the origin) depend on the system under investigation as well as on the nature of the excited state. 相似文献
