Cdc7 kinase is essential for initiation of DNA replication. Cdc7(-/-) mouse embryonic stem (ES) cells are non-viable but their growth can be rescued by an ectopically expressed transgene (Cdc7(-/-)tg). Here we report that, despite the normal growth capability of Cdc7(-/-)tg ES cells, the mice with the identical genetic background exhibit growth retardation. Concomi tantly, Cdc7(-/-)tg embryonic fibroblasts (MEFs) display delayed S phase entry and slow S phase progression. Furthermore, spermatogenesis of Cdc7(-/-)tg mice is disrupted prior to pachytene stage of meiotic prophase I. The impairment in spermatogenesis correlates with the extremely low level of Cdc7 protein in testes, and is rescued by introducing an additional allele of transgene, which results in increase of Cdc7 expression. The increased level of Cdc7 also recovers the growth of Cdc7(-/-)tg MEFs and mice, indicating that the developmental abnormalities of Cdc7(-/-)tg mice are due to insufficiency of Cdc7 protein. Our results indicate the requirement of a critical level of a cell-cycle regulator for mouse development and provide genetic evidence that Cdc7 plays essential roles in meiotic processes in mammals.  相似文献

Populations of CuI ground state, resonance, and metastable levels have been measured in CuBr laser with and without hydrogen additives. Temporal evolution and spatial distribution populations have been studied during the excitation pulse and also during the interpulse period of time. Addition of hydrogen results in a smaller rate of resonance and metastable levels excitation and smaller depopulation of the ground state during the excitation pulse. Repopulation of the ground state after the excitation pulse with hydrogen is much faster. CuBr molecules have been detected only in the periphery of the active zone. The density of the CuBr molecules had no noticeable change either during the excitation pulse or in the interpulse period  相似文献

A series of experiments examining the effects of welded boundaries on the localised blast load response of mild steel plates is presented in this short communication. Two types of welding are examined, TIG and MIG welding. The welded boundary response is compared with clamped boundary and straight and chamfer machined boundary responses. The overall trend of midpoint deflection–thickness ratio versus dimensionless damage number for all the plates follows previously reported empirical trends. The tearing threshold (Mode II*) in terms of the damage number is lowest for the machined plates followed by the welded plates with the clamped plates having the highest resistance to tearing.  相似文献

We tested the effect of caffeine, on hyperoxia-induced seizures. Thirty-seven rats with chronic cortical electrodes were injected i.p. with caffeine (1.25, 2.5, and 10 mg/kg) or vehicle before exposure to 0.5 MPa oxygen and 17 rats to oxygen with 5% CO2 at 0.5 MPa. EEG monitoring and spectral analysis of EEG activity were carried out. Caffeine significantly prolonged the latent period to the onset of seizures (P < 0.05 in ANOVA), in a dose-related manner. Our results suggest that caffeine may be used in low doses for protection against hyperoxia-induced seizures.  相似文献

The process of mixing and solid-liquid mass transfer in a one-fifth scale water model of a 100-ton Creusot-Loire Uddeholm (CLU) converter was investigated. The modified Froude number was used to relate gas flow rates between the model and its protoype. The influences of gas flow rate between 0.010 and 0.018 m3/s and bath height from 0.50 to 0.70 m on mixing time were examined. The results indicated that mixing time decreased with increasing gas flow rate and increased with increasing bath height. The mixing time results were evaluated in terms of specific energy input and the following correlation was proposed for estimating mixing times in the model CLU converter: T mix=1.08Q −1.05 W 0.35, where Q (m3/s) is the gas flow rate and W (tons) is the model bath weight. Solid-liquid mass-transfer rates from benzoic acid specimens immersed in the gas-agitated liquid phase were assessed by a weight loss measurement technique. The calculated mass-transfer coefficients were highest at the bath surface reaching a value of 6.40 × 10−5 m/s in the sprout region. Mass-transfer coefficients and turbulence parameters decreased with depth, reaching minimum values at the bottom of the vessel.  相似文献
