Direct dissimilar joining of various Al alloy (A1050, A3004, A5052 and A5083) plates and a polyamide 6 plate was performed using friction lap joining (FLJ) with the Al alloy plate as a top and the polyamide 6 plate as a bottom. The effect of Mg content in Al alloys on the joining strength was investigated. TEM analysis made clear that the polyamide 6 and Al alloys were joined via oxide layer consisting of Al2O3 and MgO. The results of XPS analysis indicated that MgO was formed by the heating during FLJ, and the quantity of MgO was increased with increasing Mg content in Al alloys. The tensile shear strengths of A3004, A5052 and A5083 joints were saturated to about 2 kN because of the tensile fracture at the polyamide 6 plate outside the tool-passed zone, which were higher than that of the A1050 joint fractured at the joint interface. The peel strength of the joint was increased with increasing Mg content in Al alloys, and the fracture occurred both at the joint interface and at the polyamide 6 plate on the joint area. The substrate specificity of rat brain neurolysin was rapidly modified by semirational mutagenesis coupled with a yeast molecular display system. Neurolysin mainly recognizes substrates with sequential six residues close to the scissile bond in polypeptides, cleaving a peptide bond in the center position of the six residues. To alter the recognition of the P2' amino acid of substrates by neurolysin, six residues of neurolysin, Asp467, Arg470, Glu510, Tyr606, Tyr610 and Tyr611, which might be involved in the formation of the neurolysin S2' subsite, were individually and comprehensively substituted. The protein libraries of mutant neurolysins comprising 120 species were displayed on the yeast cell surface and screening was carried out using two fluorescence-quenching peptides, the matrix metalloproteinase-2/9- (MMPs-2/9-) and MMP-3-specific substrates, which consisted of similar amino acids, except for alanine (for MMPs-2/9) or glutamic acid (for MMP-3) at the P2' amino acid position. Among mutant neurolysins, the Y610L mutant neurolysin exhibited a marked change in substrate specificity. Steady-state kinetic analysis of the purified Y610L mutant neurolysin revealed that the binding efficiency toward the MMP-3-specific substrate was about 3-fold higher than that toward the MMP-2/9-specific substrate. These results indicate that Tyr610 of neurolysin is the important residue to recognize the P2' amino acid of substrates. 相似文献

An experimental and analytical study was conducted to investigate the ductility of concrete-encased steel piers, referred to as “steel-reinforced concrete (SRC) construction.” Based on the cyclic lateral loading tests of SRC column specimens, the restorable and ultimate limit states are defined as the point when concrete cover spalling occurs (equivalent to longitudinal bar buckling) and the point when flange buckling of the H-shaped steel occurs, respectively. To estimate the lateral displacement capacity at both the restorable and ultimate limit states, the curvature distribution of the column was calculated based on the buckling analysis of the longitudinal bar, which was restrained by a concrete cover and transverse reinforcement, and of the steel flange encased in concrete. The lateral displacement was obtained by integrating the curvature distribution. Comparison of the computed results with experimental results, including other writers’ reports, confirmed that the proposed method can appropriately estimate the lateral displacement at the restorable and ultimate limit states, and it can accurately evaluate the buckling characteristics of the longitudinal bar and steel flange components of SRC column specimens. 相似文献

The purpose of this study is to develop electrodes for electrical stimulation of the nervous system using conductive polymers, polypyrrole (PPy) and/or poly(3,4-ethylenedioxythiophene), PEDOT. We evaluated biocompatibility in fibroblast and/or myoblast of mouse. Cultured cells on PPy and/or PEDOT extended their neuritis and survived over a week. These experiments have demonstrated that conductive polymers such as PPy, PEDOT, etc. have high biocompatibility, and PPy and/or PEDOT are applicable to nerve stimulation electrodes. 相似文献

Single Event Effects (SEEs) triggered by energetic heavy ions traversing a sensitive parts of electric devices have been studied using high-energy heavy ion microbeams connected with Transient Ion Beam Induced Current (TIBIC) measuring system at the Japan Atomic Energy Agency (JAEA) Takasaki Ion Accelerators for Advanced Radiation Applications (TIARA) facility. In the TIBIC system, SEE for semiconductor device, that is fast charge collection, has been observed in timescales of the order of picoseconds. In this paper, we show successful demonstration of the performance of the system, in which clear images of TIBIC map have been observed for Si pin photodiodes irradiated by 260 MeV 20Ne7+ and also by 520 MeV 40Ar14+ microbeams. 相似文献

Recently, multifilamentary superconducting wires with very low ac losses have been produced and practical applications will now be considered. To realize actualsize power machines and apparatuses, it is necessary to develop 1 - 10 kA ac conductors. However, the critical currents of multifilamentary wires at 1 T are several tens of A, and therefore it is necessary to use multistrand conductors consisting of several tens or several hundreds of strands. Such conductors sometimes show ac current degradation because of such factors as (1) nonuniform current distribution, (2) wire motion, (3) temperature increase, (4) longitudinal magnetic field effect, etc. Formerly, a coreless transformer was considered unpractical because of its large exciting current. However, Yamamoto and others proposed that a coreless superconducting transformer could be used as a stepdown autotransformer at the receiving side, utilizing its large exciting current as the reactive power source to cancel the charging current of an underground transmission line or UHV line, and therefore the shunt reactors could be eliminated. In this paper, development of ac-superconducting conductors aimed at prevention of current degradation are discussed, as well as quench test results of two small coils made with these conductors. In these conductors, low ac low strands with ultrafine NbTi filaments are twisted around a central bundle of stainless steel wires. One of the coils has been designed as a model coreless autotransformer, and its test result is also described. 相似文献

This study was performed to evaluate p53 overexpression as a predictor of the response to chemotherapy of patients with gastric cancer. The subjects comprised 20 patients with Stage IV gastric cancer and three with locally recurrent lesions, all of whom were treated with 5-fluorouracil (5-FU) plus cisplatin (CDDP) for 4 weeks. Of the total 23 patients there were 10 responders; 2 showing complete response (CR) and 8, partial response (PR). Specimens obtained by endoscopic biopsy were immunohistochemically stained using anti-p53 protein and bcl-2 protein antibody. Of the 10 responders, 7 demonstrated negative p53 staining, and of the 13 nonresponders, 11 demonstrated positive p53 staining (P = 0.013). Tissue from 3 of the responders and 7 of the nonresponders that stained for bcl-2 were positive prior to chemotherapy; however, there was no association between bcl-2 staining and chemotherapeutic effect. In conclusion, immunohistochemical identification of p53 in pretreatment tissue may represent a useful predictor for chemotherapeutic outcome in patients with gastric cancer. 相似文献

At least four genes involved in DNA mismatch repair (MMR), hMSH2, hMLH1, hPMS1 and hPMS2, have been cloned and characterized. These genes have been demonstrated to be altered in the germline of patients with hereditary non-polyposis colorectal cancer (HNPCC). HNPCC is an autosomal dominant disease characterized by a preponderance of proximal colon, young age of onset, increased multiplicity, and improved stage-specific survival. In this study, we examined the expression of hMSH2 protein in sporadic colorectal cancer (CRC). As a result, the frequency of right-sided CRC and multiple CRCs were significantly higher in the patients with hMSH2-negative CRC than in those with hMSH2-positive CRC. The rate of p53 positivity was significantly lower in the hMSH2-negative tumours than that in the hMSH2-positive tumours. The disease-free survival rate tended to be higher in the patients with hMSH2-negative CRC than in the patients with hMSH2-positive CRC. Our findings suggest that both the clinicopathological and biological features of hMSH2-negative sporadic CRC seemed to be similar to those of HNPCC. To clarify the mechanism of carcinogenesis in HNPCC and sporadic CRC, further investigations of genetic alterations caused by MMR genes will be needed. 相似文献
