High pressure processing (HPP) reduces the glycolytic activity of lactic acid bacteria (LAB) and provides a means to control further production of acidic metabolites in fermented dairy products during storage. However, there is limited information on the effects of HPP on specific enzymes of dairy starter bacteria responsible for the metabolism of lactose. The aim of this study was to determine pressure-induced inactivation of glycolytic enzymes in Lactococcus lactis subsp. lactis C10, Streptococcus thermophilus TS1 and Lactobacillus acidophilus 2400. Cultures were grown for 16 h in M17 or MRS broth containing 5% (w/v) lactose at pH 6.5 (maintained by addition of 10 M NaOH). The cells were harvested by centrifugation, washed and resuspended in 100 mM phosphate buffer (pH 6.5) and pressure-treated at 300 and 600 MPa (≤ 22 °C, 5 min). The ability of pressure-treated resting cells of Lactococcus, incubated with 5% (w/v) lactose at 30 °C, to ferment lactose was evaluated by determining titratable acidity (TA) during incubation. The activities of phospho-β-galactosidase (P-β-gal), β-galactosidase (β-gal) and lactate dehydrogenase (LDH) were determined in cell-free extracts of untreated and pressure-treated cells. Resting cells of Lactococcus treated at 600 MPa had a substantially lower rate of acidification than the controls and those treated at 300 MPa. Both P-β-gal and β-gal were significantly inactivated (p < 0.01) in the starter cultures treated at 300 or 600 MPa. The LDH in Lactococcus and Lactobacillus was highly resistant to pressure treatment at 300 MPa. In contrast, the LDH in Streptococcus was almost completely inactivated at ≥ 300 MPa.Industrial relevanceContinuing production of acidic metabolites in fermented dairy products during storage can be a technological challenge that adversely affects product quality. The current study demonstrates that high pressure processing (HPP) offers the potential of controlling this problem by inactivation of glycolytic enzymes in various mesophilic and thermophilic starter cultures. The findings of this research will assist in establishing optimised operating parameters for HPP treatment of cultured products to extend shelf-life, by reducing acid production during storage.  相似文献

BACKGROUND: The effectiveness of chitosan films containing Zataria multiflora Boiss essential oil (ZEO) (5 and 10 g kg?1) and grape seed extract (GSE) (10 g kg?1) on lipid oxidation and microbial (lactic acid bacteria, aerobic mesophiles and inoculated Listeria monocytogenes) characteristics of mortadella sausage at 4 °C for 21 days was evaluated. The release of total phenolics (TPs) into sausage was also assessed. RESULTS: All films exhibited antibacterial activity against L. monocytogenes on agar culture media. Chitosan films containing ZEO were the most effective on the growth of bacteria. The growth of L. monocytogenes was significantly inhibited by ZEO‐GSE containing films especially during storage of the sausages for 6 days. Aerobic mesophiles and lactic acid bacteria were the most sensitive and resistant groups to films by 0.1–1.1 and 0.1–0.7 log cycles reduction, respectively. Sausages wrapped by 10 g kg?1 GSE + 10 g kg?1 ZEO films had the lowest degrees of lipid oxidation, which was 23% lower than the control. The TPs of ZEO films decreased to zero after 6 days, whereas TPs of GSE films followed a slight decrease during the storage. CONCLUSION: Antimicrobial/antioxidant chitosan film could be developed by incorporating GSE and ZEO for extending the shelf life of mortadella sausage. Extraction of ferulic acid from sugar beet pulp was carried out using three extraction solvents, sodium hydroxide (0.5, 1, 2 M), methanol and their mixture (alkaline methanolic solvent). The Ferulic acid extracted by each solvent was identified and quantified by HPLC method and the effects of solvent type, concentration and reaction time on ferulic acid solubilisation were assessed. There were differences in the contents of products extracted in the experiment conditions. The minimum amount of ferulic acid was obtained from methanolic extract while the highest concentrations (957.4 mg/L ferulic acid) were obtained employing the highest NaOH concentration (2 M), and reaction time (12 h), so phenolic compounds are better released with alkaline hydrolysis than in methanol conditions. Finally a simple procedure for the purification of ferulic acid from the alkaline extracts is presented and evaluated by FT-IR spectrum.  相似文献

In this paper, we propose a hybrid approach using genetic algorithm and neural networks to classify Peer-to-Peer (P2P) traffic in IP networks. We first compute the minimum classification error (MCE) matrix using genetic algorithm. The MCE matrix is then used during the pre-processing step to map the original dataset into a new space. The mapped data set is then fed to three different classifiers: distance-based, K-Nearest Neighbors, and neural networks classifiers. We measure three different indexes, namely mutual information, Dunn, and SD to evaluate the extent of separation of the data points before and after mapping is performed. The experimental results demonstrate that with the proposed mapping scheme we achieve, on average, 8% higher accuracy in classification of the P2P traffic compare to the previous solutions. Moreover, the genetic-based MCE matrix increases the classification accuracy more than what the basic MCE does.  相似文献

The present paper evaluates the use of software agents to identify relevance of information, called awareness. This evaluation is based on existing policies and scenarios in the context of wireless communication of a hospital in Norway. The study is to address the lack of literature for experimental studies on a method to employ software agents for awareness identification. Research in computer supported cooperative work indicates the significant contributions of software agents to assist individuals. There are bodies of work that show awareness provides the means for software agents in which effective cooperation can take place. In addition, the role of the methods to identify awareness is emphasized in the literature of both computer supported cooperative work and software agents. This paper explains a step-wise process, called Policy-based Awareness Management, which allows agents to use policies as a source to identify awareness and thus change their behaviors accordingly. The contribution of this method is based on the concepts proposed by the logic of general awareness. The present study applies Directory Enabled Networks-next generation as the policy structure for the method. The paper evaluates the process via its application to identify the relevance of information in wireless communication scenarios in a hospital. The present study conducts observations, interviews and discussions on the wireless communication system of the hospital to identify the different scenarios happening in the system. The paper presents a set of simulations on these scenarios and concludes that the method is effective and cost-efficient.  相似文献

Large‐scale aerial sensing missions can greatly benefit from the perpetual endurance capability provided by high‐performance low‐altitude solar‐powered unmanned aerial vehicles (UAVs). However, today these UAVs suffer from small payload capacity, low energetic margins, and high operational complexity. To tackle these problems, this paper presents four individual technical contributions and integrates them into an existing solar‐powered UAV system: First, a lightweight and power‐efficient day/night‐capable sensing system is discussed. Second, means to optimize the UAV platform to the specific payload and to thereby achieve sufficient energetic margins for day/night flight with payload are presented. Third, existing autonomous launch and landing functionality is extended for solar‐powered UAVs. Fourth, as a main contribution an extended Kalman filter (EKF)‐based autonomous thermal updraft tracking framework is developed. Its novelty is that it allows the end‐to‐end integration of the thermal‐induced roll moment into the estimation process. It is assessed against unscented Kalman filter and particle filter methods in simulation and implemented on the aircraft's low‐power autopilot. The complete system is verified during a 26 h search‐and‐rescue aerial sensing mock‐up mission that represents the first‐ever fully autonomous perpetual endurance flight of a small solar‐powered UAV with a day/night‐capable sensing payload. It also represents the first time that solar‐electric propulsion and autonomous thermal updraft tracking are combined in flight. In contrast to previous work that has focused on the energetic feasibility of perpetual flight, the individual technical contributions of this paper are considered core functionality to guarantee ease‐of‐use, effectivity, and reliability in future multiday aerial sensing operations with small solar‐powered UAVs.  相似文献

This paper presents an iterative mathematical decision model for organizations to evaluate whether to invest in establishing information technology (IT) infrastructure on‐premises or outsourcing IT services on a multicloud environment. This is because a single cloud cannot cover all types of users’ functional/nonfunctional requirements, in addition to several drawbacks such as resource limitation, vendor lock‐in, and prone to failure. On the other hand, multicloud brings several merits such as vendor lock‐in avoidance, system fault tolerance, cost reduction, and better quality of service. The biggest challenge is in selecting an optimal web service composition in the ever increasing multicloud market in which each provider has its own pricing schemes and delivers variation in the service security level. In this regard, we embed a module in the cloud broker to log service downtime and different attacks to measure the security risk. If security tenets, namely, security service level agreement, such as availability, integrity, and confidentiality for mission‐critical applications, are targeted by cybersecurity attacks, it causes disruption in business continuity, leading to financial losses or even business failure. To address this issue, our decision model extends the cost model by using the cost present value concept and the risk model by using the advanced mean failure cost concept, which are derived from the embedded module to quantify cloud competencies. Then, the cloud economic problem is transformed into a bioptimization problem, which minimizes cost and security risks simultaneously. To deal with the combinatorial problem, we extended a genetic algorithm to find a Pareto set of optimal solutions. To reach a concrete result and to illustrate the effectiveness of the decision model, we conducted different scenarios and a small‐to‐medium business IT development for a 5‐year investment as a case study. The result of different implementation shows that multicloud is a promising and reliable solution against IT on‐premises deployment.  相似文献
