The authors examined associations among parental and child adjustment, child syncope, somatic, and school problems. Participants were children (N = 56) ages 7-18 years with syncope. Measures included syncope severity, parental distress, and children's internalizing symptoms. For children diagnosed negative for neurocardiogenic syncope (NCS), their fathers' and their own psychological symptoms were positively associated with the severity of syncope, whereas their mothers' functioning was negatively associated with the severity of syncope. Also, for the negative NCS group, fathers' psychological functioning was associated with children's nonsyncope somatic complaints but not with their school problems. For the positive NCS group, few significant father-child associations were found, but several significant positive associations were revealed between mothers' psychological symptoms and their children's syncope as well as somatic and school problems. Gut microbiota alterations are intimately linked to chronic constipation upon aging. We investigated the role of targeted changes in the gut microbiota composition in the relief of constipation symptoms after rhubarb extract (RE) supplementation in middle-aged volunteers. Subjects (95% women, average 58 years old) were randomized to three groups treated with RE at two different doses determined by its content of rhein (supplementation of 12.5 mg and 25 mg per day) vs. placebo (maltodextrin) for 30 days. We demonstrated that daily oral supplementation of RE for 30 days was safe even at the higher dose. Stool frequency and consistency, and perceived change in transit problem, transit speed and difficulty in evacuating, investigated by validated questionnaires, were improved in both groups of RE-treated volunteers compared to placebo. Higher abundance of Lachnospiraceae (mainly Roseburia and Agathobacter) only occurred after RE treatment when present at low levels at baseline, whereas an opposite shift in short-chain fatty acid (SCFA) levels was observed in both RE-treated groups (increase) and placebo (decrease). Fecal Lachnospiraceae and SCFA were positively correlated with stool consistency. This study demonstrates that RE supplementation promotes butyrate-producing bacteria and SCFA, an effect that could contribute to relieving chronic constipation in middle-aged persons.  相似文献

The authors modeled depressive and anxiety symptom data from 1,391 participants in a longitudinal study of middle-aged and older Swedish twins (M age?=?60.9 years, SD?=?13.3). Although anxiety and depression were highly correlated, a model with distinct Anxiety and Depression factors fit the data better than models with Positive and Negative Affect factors or a single Mental Health factor. Lack of well-being was associated with anxiety rather than depression. Over two 3-year intervals, anxiety symptoms led to depressive symptoms, but the relationship was not reciprocal. Anxiety symptoms were more stable than depression. These findings provide additional support for the idea that anxiety symptoms may reflect a personality trait such as neuroticism more than do depressive symptoms and suggest that low positive affect may not be as specific to depression among older adults as in younger people. In 3 studies, the authors tested the hypothesis that Chinese participants would view social groups as more entitative than would Americans and, as a result, would be more likely to infer personality traits on the basis of group membership--that is, to stereotype. In Study 1, Chinese participants made stronger stereotypic trait inferences than Americans did on the basis of a target's membership in a fictitious group. Studies 2 and 3 showed that Chinese participants perceived diverse groups as more entitative and attributed more internally consistent dispositions to groups and their members. Guided by culturally based lay theories about the entitative nature of groups, Chinese participants may stereotype more readily than do Americans when group membership is available as a source of dispositional inference. Seed size is often considered to be an important trait for seed quality, i.e., vigour and germination performance. It is believed that seed size reflects the quantity of reserve material and thus the C and N sources available for post-germinative processes. However, mechanisms linking seed size and quality are poorly documented. In particular, specific metabolic changes when seed size varies are not well-known. To gain insight into this aspect, we examined seed size and composition across different accessions of barrel medic (Medicago truncatula Gaertn.) from the genetic core collection. We conducted multi-elemental analyses and isotope measurements, as well as exact mass GC–MS metabolomics. There was a systematic increase in N content (+0.17% N mg−1) and a decrease in H content (–0.14% H mg−1) with seed size, reflecting lower lipid and higher S-poor protein quantity. There was also a decrease in 2H natural abundance (δ2H), due to the lower prevalence of 2H-enriched lipid hydrogen atoms that underwent isotopic exchange with water during seed development. Metabolomics showed that seed size correlates with free amino acid and hexoses content, and anticorrelates with amino acid degradation products, disaccharides, malic acid and free fatty acids. All accessions followed the same trend, with insignificant differences in metabolic properties between them. Our results show that there is no general, proportional increase in metabolite pools with seed size. Seed size appears to be determined by metabolic balance (between sugar and amino acid degradation vs. utilisation for storage), which is in turn likely determined by phloem source metabolite delivery during seed development.  相似文献

Prostate specific membrane antigen targeted radionuclide therapy (PSMA-TRT) is a promising novel treatment for prostate cancer (PCa) patients. However, PSMA-TRT cannot be used for curative intent yet, thus additional research on how to improve the therapeutic efficacy is warranted. A potential way of achieving this, is combining TRT with poly ADP-ribosylation inhibitors (PARPi), which has shown promising results for TRT of neuroendocrine tumor cells. Currently, several clinical trials have been initiated for this combination for PCa, however so far, no evidence of synergism is available for PCa. Therefore, we evaluated the combination of PSMA-TRT with three classes of PARPi in preclinical PCa models. In vitro viability and survival assays were performed using PSMA-expressing PCa cell lines PC3-PIP and LNCaP to assess the effect of increasing concentrations of PARPi veliparib, olaparib or talazoparib in combination with PSMA-TRT compared to single PARPi treatment. Next, DNA damage analyses were performed by quantifying the number of DNA breaks by immunofluorescent stainings. Lastly, the potential of the combination treatments was studied in vivo in mice bearing PC3-PIP xenografts. Our results show that combining PSMA-TRT with PARPi did not synergistically affect the in vitro clonogenic survival or cell viability. DNA-damage analysis revealed only a significant increase in DNA breaks when combining PSMA-TRT with veliparib and not in the other combination treatments. Moreover, PSMA-TRT with PARPi treatment did not improve tumor control compared to PSMA-TRT monotherapy. Overall, the data presented do not support the assumption that combining PSMA-TRT with PARPi leads to a synergistic antitumor effect in PCa. These results underline that extensive preclinical research using various PCa models is imperative to validate the applicability of the combination strategy for PCa, as it is for other cancer types.  相似文献

Objective: To quantify and compare the nutrient‐density of commonly consumed snacks using two nutrient‐density measures, Nutrient Rich Foods Indices 9.3 (NRF 9.3) and 15.3 (NRF 15.3). Design: Identify commonly consumed categories of snacks and individual snack foods, calculate NRF 9.3 and 15.3 scores, rank snacks by category and by individual food based on nutrient density, compare and contrast scores generated by the two NRF Indices. Main Outcome Measures: NRF 9.3 and 15.3 scores. Analysis: Averages and standard deviations of nutrient‐density scores for each snack category. Results: Vegetables and coffee/tea received the highest category scores on both indices. Cakes/cookies/pastries and sweets had the lowest category scores. NRF 9.3 scores for individual snacks ranged from –46 (soda) to 524 (coffee). NRF 15.3 scores ranged from –45 (soda) to 736 (coffee). Conclusions and Implications: If added to food labels, NRF scores could help consumers identify more nutritious choices. The differences between NRF 9.3 and 15.3 scores generated for the same foods and the limitations of these indices highlight the need for careful consideration of which nutrient‐density measure to include on food labels as well as consumer education.  相似文献

With the objective of evaluating the potential effects of sodium bicarbonate or a magnesium-based product on rumen pH and milk performance of dairy cattle exposed to a dietary challenge, 30 lactating Holstein cows (648 ± 67 kg of body weight; 44.4 ± 9.9 kg/d of milk yield; 155 ± 75 d in milk) were blocked by parity (9 primiparous and 21 multiparous) and randomly distributed to 3 treatment groups. One group received a total mixed ration (TMR) that acted as a control (CTR), a second group (SB) received the same TMR but with an additional supplementation of 0.8% of sodium bicarbonate, and a third group (MG) received the same TMR as CTR but an additional supplementation of 0.4% of a magnesium-based product (pHix-Up, Timab, Dinard, France). After 1 wk of exposure to this TMR, all 3 rations were supplemented with 1 kg/d of barley, which was then increased 1 kg/wk until reaching 3 kg/d of barley during wk 4 of the study. Every kilogram of barley replaced 1 kg of forage in the diet. Individual feed intake and behavior were monitored using electronic feed bins. Seven cows per treatment were equipped with an intraruminal bolus that recorded pH every 15 min. As the severity of the barley challenge increased, dry matter intake decreased, but this decrease was more pronounced in SB cows than in MG cows, with an intermediate response for CTR cows. The MG cows produced more milk when challenged with 2 or 3 kg/d of additional barley than when challenged with 1 kg/d, whereas CTR cows produced less milk with the 3 kg/d challenge compared with 1 or 2 kg/d, and the SB cows maintained milk production. Milk fat content decreased with barley challenges, with CTR cows experiencing a more severe decrease than SB cows, which maintained stable butterfat values throughout the study, and MG cows showed a decline in milk fat content only with the 3 kg/d of additional barley. Meal size was also reduced as the severity of barley challenge increased, and this reduction was more modest in MG cows than in SB cows. The number of daily meals consumed by SB and MG cows was more constant than that recorded in CTR cows. Cows on the CTR and SB treatments showed a marked decrease in rumen pH with the 3 kg/d of additional barley, whereas MG cows maintained stable rumen pH during the barley challenges and had greater average rumen pH (5.93 ± 0.04) than CTR cows (5.83 ± 0.04) with the 3 kg/d of additional barley; SB cows showed intermediate values (5.85 ± 0.04). Last, MG cows spent less time (32.3 ± 6.1%) with rumen pH ≤5.8 when exposed to the 3 kg/d of barley challenge than CTR and SB cows (50.7 ± 5.02%). In conclusion, supplementation with MG prevents the decline in dry matter intake and milk production induced by a rumen challenge, whereas supplementation with SB prevents the decay in milk production but does not prevent the decrease in feed intake. These changes were probably due to the ability of the MG treatment to prevent a reduction in rumen pH when challenging cows with 3 kg/d of additional barley in the ration.  相似文献

An observational study was conducted to estimate prevalence and risk factors for Salmonella and Campylobacter spp. carcass contamination in broiler chickens. Eighty-two lots were sampled in four slaughterhouses located in the province of Québec, Canada, over a 10-month period. Carcass contamination was evaluated by the carcass rinse technique for about 30 birds per lot. Exposure to potential risk factors was evaluated based on data from questionnaires, meteorology, and cecal cultures. Multivariable binomial negative regression models were used for risk factor analysis at the lot level. The prevalence of Salmonella-positive carcasses was 21.2% (95% confidence interval: 15.7 to 26.7%). Significant risk factors (P < 0.05) associated with a higher proportion of positive carcasses within lots were Salmonella-positive cecal culture, low rainfall during transportation to the slaughterhouse, temperature of > or = 0 degree C during transportation to the slaughterhouse, and a > or = 4-h waiting period in shipping crates before slaughtering. The prevalence of Campylobacter-positive carcasses was 35.8% (95% confidence interval: 27.1 to 44.5%). Lots containing birds with Campylobacter-positive cecal culture results, lots of birds that were slaughtered at the end of the week, and lots with at least 20% of birds with digestive contents detected in the jejunum at time of slaughtering had a significantly higher proportion (P < 0.05) of contaminated carcasses. These results support the importance of preharvest control measures implemented during rearing to reduce contamination of the final product. Weather during transportation to slaughter and the day of the week that birds were slaughtered also were associated with carcass contamination; further studies are needed to determine the underlying mechanisms by which these factors influence carcass contamination.  相似文献

Vitamin A deficiency is a public health issue in developing countries and promoting dietary carotenoids as precursors is a promising strategy. However, carotenoids present in numerous fruits and vegetables are unstable and poorly bioaccessible. This study evaluated these two parameters during in vitro digestion of carotenoids and retinoids from carrot juice, raw and cooked spinach, micronutrient-fortified flour and standards without food matrix. Standards were unstable whereas vitamin A from fortified flour and native food carotenoids were generally better protected by the food matrix (30–100% remaining versus 7–30% for standards). Hydrothermal cooking did not influence spinach carotenoid digestive stability but decreased their contents, phenomenon compensated by a significantly better micellarisation from 15-fold for β-carotene to 72-fold for lutein. Finally, carrot juice provided the greatest amount of bioaccessible provitamin A with 1850 μg/100 g dry matter (DM) versus 790 and 80 μg/100 g DM in cooked and raw spinach, respectively.  相似文献
