Inferring the breed of origin of dairy products can be achieved through molecular analysis of genetic markers with a population-specific pattern of segregation. The goal of the current work was to generate such markers in goats by resequencing several pigmentation genes [melanocortin 1 receptor (MC1R), v-kit Hardy-Zuckerman 4 feline sarcoma viral oncogene homolog (KIT), tyrosinase (TYR), and tyrosinase-related protein 2 (TYRP2)]. This experiment revealed 10 single nucleotide polymorphisms (SNP), including 5 missense mutations and 1 nonsense mutation. These markers were genotyped in 560 goats from 18 breeds originally from Italy, the Iberian Peninsula, the Canary Islands, and North Africa. Although the majority of SNP segregated at moderate frequencies in all populations (including 2 additional markers that were used as a source of information), we identified a c.764G>A SNP in MC1R that displayed highly divergent allelic frequencies in the Palmera breed compared with the Majorera and Tinerfeña breeds from the Canary Islands. Thus, we optimized a pyrosequencing-based technique that allowed us to estimate, very accurately, the allele frequencies of this marker in complex DNA mixtures from different individuals. Once validated, we applied this method to generating breed-specific DNA profiles that made it possible to detect fraudulent cheeses in which Palmero cheese was manufactured with milk from Majorera goats. One limitation of this approach, however, is that it cannot be used to detect illegal manufacturing where Palmero dairy products are produced by mixing milk from Palmera and Majorera goats, because the c.764G>A SNP segregates in both breeds. 相似文献

Raw materials with different technological properties found in the same geological region can constitute a ceramic formulation upon proper mixing. This study aims to demonstrate the best way to develop a mixture using a mixture design method, emphasizing the geology of three clays in the Rio Bonito Formation/Paraná Basin. The mineralogical and chemical characteristics of these clays were investigated by X-ray diffraction and X-ray fluorescence, respectively, along with elementary (CNH), differential thermal and thermogravimetric analysis, and dilatometry. Ten formulations and their technological properties, including apparent density, shrinkage, water absorption, and mechanical resistance, were investigated. The results showed the potential of the studied mineral deposit clays for application in the ceramic industry in different materials. Clay 1 showed refractory characteristics, and Clay 3 was more plastic. The mixtures with higher contents of Clay 1 showed ideal for monoporosa tile recipes. The mixtures with higher contents of Clay 3 were ideal for porcelain tile pastes. 相似文献

Recently, MV-algebras with product have been investigated from different points of view. In particular, in [EGM01], a variety resulting from the combination of MV-algebras and product algebras (see [H98]) has been introduced. The elements of this variety are called ŁΠ-algebras. Even though the language of ŁΠ-algebras is strong enough to describe the main properties of product and of ukasiewicz connectives on [0,1], the discontinuity of product implication introduces some problems in the applications, because a small error in the data may cause a relevant error in the output. In this paper we try to overcome this difficulty, substituting the product implication by a continuous approximation of it. We investigate the resulting algebras, called ŁΠq
-algebras, and we show that these algebras constitute a quasivariety which is generated by the class of all ŁΠ

Increasing experimental and clinical evidence points toward a very important role for the gut microbiome and its associated metabolism in human health and disease, including in cardiovascular disorders. Free fatty acids (FFAs) are metabolically produced and utilized as energy substrates during almost every biological process in the human body. Contrary to long- and medium-chain FFAs, which are mainly synthesized from dietary triglycerides, short-chain FFAs (SCFAs) derive from the gut microbiota-mediated fermentation of indigestible dietary fiber. Originally thought to serve only as energy sources, FFAs are now known to act as ligands for a specific group of cell surface receptors called FFA receptors (FFARs), thereby inducing intracellular signaling to exert a variety of cellular and tissue effects. All FFARs are G protein-coupled receptors (GPCRs) that play integral roles in the regulation of metabolism, immunity, inflammation, hormone/neurotransmitter secretion, etc. Four different FFAR types are known to date, with FFAR1 (formerly known as GPR40) and FFAR4 (formerly known as GPR120) mediating long- and medium-chain FFA actions, while FFAR3 (formerly GPR41) and FFAR2 (formerly GPR43) are essentially the SCFA receptors (SCFARs), responding to all SCFAs, including acetic acid, propionic acid, and butyric acid. As with various other organ systems/tissues, the important roles the SCFARs (FFAR2 and FFAR3) play in physiology and in various disorders of the cardiovascular system have been revealed over the last fifteen years. In this review, we discuss the cardiovascular implications of some key (patho)physiological functions of SCFAR signaling pathways, particularly those regulating the neurohormonal control of circulation and adipose tissue homeostasis. Wherever appropriate, we also highlight the potential of these receptors as therapeutic targets for cardiovascular disorders. 相似文献

Protein expression from the berry skin of four red grape biotypes with varying hybrid character was compared at a proteome-wide level to identify the metabolic pathways underlying divergent patterns of secondary metabolites. A bottom-up shotgun proteomics approach with label-free quantification and MaxQuant-assisted computational analysis was applied. Red grapes were from (i) purebred Vitis vinifera (Aglianico cv.); (ii) V. vinifera (local Sciascinoso cv.) grafted onto an American rootstock; (iii) interspecific hybrid (V. vinifera × V. labrusca, Isabel), and (iv) uncharacterized grape genotype with hybrid lineage, producing relatively abundant anthocyanidin 3,5-O-diglucosides. Proteomics supported the differences between hybrids and purebred V. vinifera grapes, consistently with distinct phenotypic metabolite assets. Methanol O-anthraniloyltransferase, which catalyses the synthesis of methyl anthranilate, primarily responsible for the “foxy” odour, was exclusive of the Isabel hybrid grape. Most of the proteins with different expression profiles converged into coordinated biosynthetic networks of primary metabolism, while many possible enzymes of secondary metabolism pathways, including 5-glucosyltransferases expected for hybrid grapes, remained unassigned due to incomplete protein annotation for the Vitis genus. Minor differences of protein expression distinguished V. vinifera scion grafted onto American rootstocks from purebred V. vinifera skin grapes, supporting a slight influence of the rootstock on the grape metabolism. 相似文献

Research has suggested that negative emotions are associated with problematic Internet use. This study investigated the role of metacognitions as a mediator of the relationship between negative emotions and problematic Internet use. A sample of 97 university students completed the following questionnaires: Hospital Anxiety and Depression Scale, Boredom Proneness Scale, Metacognitions Questionnaire 30, and Internet Addiction Test. All dimensions of metacognition were found to be positively and significantly correlated with problematic Internet use. Positive and significant correlations were also observed between problematic Internet use and negative emotions (anxiety, depression and boredom). Structural equation modeling was used to test a mediation model in which negative emotions predicted metacognitions which in turn predicted problematic Internet use. The results supported the hypothesis that the relationship between negative emotions and problematic Internet use is fully mediated by metacognitions, suggesting that metacognitive theory may be relevant to understanding problematic Internet use. The implications of these findings are discussed. 相似文献

BACKGROUND: The functional significance of the anterograde and retrograde filling of coronaries on angiography is controversial. METHODS AND RESULTS: Eighteen patients with 27 severe lesions (> 85% diameter stenosis) after previous extensive myocardial infarction were selected. The left ventricle was divided into 33 segments for regional comparison of epicardial flow (as assessed by angiography) and tissue perfusion as well as metabolism (as measured by 13NH3- and 18FDG-PET). Viability was defined as normal perfusion (> 80% relative of maximum 13NH3 activity) or mismatch defect (> 1.2 metabolism/flow ratio). A method has been developed to register the 'lesion predicted region', determined on the basis of angiography, in the same polar map as derived from the positron emission tomography data. Distal to the lesion, the anterograde epicardial flow was evaluated by Thrombolysis in Myocardial Infarction (TIMI) criteria (TIMI flow 0-3), and retrograde filling was graded on a 0-3 scale (collateral grade 0-3). TIMI flow grade and retrograde collateral grade in every lesion predicted region segment were summed to indicate the total segmental epicardial flow. Out of the 594 segments, 369 were associated with a severe lesion. Among them, significantly higher average perfusion and metabolic activities were found in segments of good epicardial filling (summed epicardial flow > or = 3) than in the territories of limited epicardial flow (summed score < 3): 65.4 +/- 17% vs 45.6 +/- 10 (P = 0.001%) and 68.6 +/- 16% vs 47.4 +/- 11% (P = 0.0004), respectively. However, when we analysed the predictive value of angiographically detectable good epicardial flow for positron emission tomography viability criteria then the positive predictive value was found to be as low as 0.5, while the negative predictive value was considerably higher (0.82). CONCLUSION: After myocardial infarction, angiographically detectable limited epicardial flow reveals scarred segments while good epicardial contrast filling does not necessarily indicate maintenance of nutritive function. 相似文献
