The development of artificial vesicles into responsive architectures capable of sensing the biological environment and simultaneously signaling the presence of a specific target molecule is a key challenge in a range of biomedical applications from drug delivery to diagnostic tools. Herein, the rational design of biomimetic DNA-grafted quatsome (QS) nanovesicles capable of translating the binding of a target molecule to amphiphilic DNA probes into an optical output is presented. QSs are synthetic lipid-based nanovesicles able to confine multiple organic dyes at the nanoscale, resulting in ultra-bright soft materials with attractiveness for sensing applications. Dye-loaded QS nanovesicles of different composition and surface charge are grafted with fluorescent amphiphilic nucleic acid-based probes to produce programmable FRET-active nanovesicles that operate as highly sensitive signal transducers. The photophysical properties of the DNA-grafted nanovesicles are characterized and the highly selective, ratiometric detection of clinically relevant microRNAs with sensitivity in the low nanomolar range are demonstrated. The potential applications of responsive QS nanovesicles for biosensing applications but also as functional nanodevices for targeted biomedical applications is envisaged.  相似文献

This paper presents a full formalization of the semantics of definite programs, in the calculus of inductive constructions. First, we describe a formalization of the proof of first-order terms unification: this proof is obtained from a similar proof dealing with quasi-terms, thus showing how to relate an inductive set with a subset defined by a predicate. Then, SLD-resolution is explicitely defined: the renaming process used in SLD-derivations is made explicit, thus introducing complications, usually overlooked, during the proofs of classical results. Last, switching and lifting lemmas and soundness and completeness theorems are formalized. For this, we present two lemmas, usually omitted, which are needed. This development also contains a formalization of basic results on operators and their fixpoints in a general setting. All the proofs of the results, presented here, have been checked with the proof assistant Coq.  相似文献

Local electric stimulation of tissues and cells has gained importance as therapeutic alternative in the treatment of many diseases. These alternatives aim to deliver a less invasively stimuli in liquid media, making imperative the development of versatile micro‐ and nanoscale solutions for wireless actuation. Here, a simple microfabrication process to produce suspended silicon microphotodiodes that can be activated by visible light to generate local photocurrents in their surrounding medium is presented. Electrical characterization using electrical probes confirms their diode behavior. To demonstrate their electrochemical performance, an indirect test is implemented in solution through photoelectrochemical reactions controlled by a white‐LED lamp. Furthermore, their effects on biological systems are observed in vitro using mouse primary neurons in which the suspended microphotodiodes are activated periodically with white‐LED lamp, bringing out observable morphological changes in neuronal processes. The results demonstrate a simplified and cost‐effective wireless tool for photovoltaic current generation in liquid media at the microscale.  相似文献

Suspended planar‐array (SPA) chips embody millions of individual miniaturized arrays to work in extremely small volumes. Here, the basis of a robust methodology for the fabrication of SPA silicon chips with on‐demand physical and chemical anisotropies is demonstrated. Specifically, physical traits are defined during the fabrication process with special focus on the aspect ratio, branching, faceting, and size gradient of the final chips. Additionally, the chemical attributes augment the functionality of the chips with the inclusion of complete coverage or patterns of selected biomolecules on the surface of the chips with contact printing techniques, offering an extremely high versatility, not only with the choice of the pattern shape and distribution but also in the choice of biomolecular inks to pattern. This approach increases the miniaturization of printed arrays in 3D structures by two orders of magnitude compared to those previously demonstrated. Finally, functional micrometric and sub‐micrometric patterned features are demonstrated with an antibody binding assay with the recognition of the printed spots with labeled antibodies from solution. The selective addition of physical and chemical attributes on the suspended chips represents the basis for future biomedical assays performed within extremely small volumes.  相似文献

We propose a hybridizable discontinuous Galerkin (HDG) method to numerically solve the Oseen equations which can be seen as the linearized version of the incompressible Navier-Stokes equations. We use same polynomial degree to approximate the velocity, its gradient and the pressure. With a special projection and postprocessing, we obtain optimal convergence for the velocity gradient and pressure and superconvergence for the velocity. Numerical results supporting our theoretical results are provided.  相似文献

In this work heteroepitaxial stabilization with nanoscale control of the magnetic Co2FeO4 phase at 250 °C is reported. Ultrasmooth and pure Co2FeO4 thin films (5–25 nm) with no phase segregation are obtained on perovskite SrTiO3 single crystal (100) and (110) oriented substrates by atomic layer deposition (ALD). High resolution structural and chemical analyses confirm the formation of the Co‐rich spinel metastable phase. The magneto‐crystalline anisotropy of the Co2FeO4 phase is not modified by stress anisotropy because the films are fully relaxed. Additionally, high coervice fields, 15 kOe, and high saturation of magnetization, 3.3 μB per formula unit (at 10 K), are preserved down to 10 nm. Therefore, the properties of the ALD‐Co2FeO4 films offer many possibilities for future applications in sensors, actuators, microelectronics, and spintronics. In addition, these results are promising for the use of ALD compared to the existing thin‐film deposition techniques to stabilize epitaxial multicomponent materials with nanoscale control on a wide variety of substrates for which the processing temperature is a major drawback.  相似文献

Several methods to determine the color gamut of any digital camera are shown. Since an input device is additive, its color triangle was obtained from their spectral sensitivities, and it was compared with the theoretical sensors of Ives‐Abney‐Yule and MacAdam. On the other hand, the RGB digital data of the optimal or MacAdam colors were simulated to transform them into XYZ data according to the colorimetric profile of the digital camera. From this, the MacAdam limits associated to the digital camera are compared with the corresponding ones of the CIE‐1931 XYZ standard observer, resulting that our color device has much smaller MacAdam loci than those of the colorimetric standard observer. Taking this into account, we have estimated the reduction of discernible colors by the digital camera applying a chromatic discrimination model and a packing algorithm to obtain color discrimination ellipses. Calculating the relative decrement of distinguishable colors by the digital camera in comparison with the colorimetric standard observer at different luminance factors of the optimal colors, we have found that the camera distinguishes considerably fewer very dark than very light ones, but relatively much more colors with middle lightness (Y between 40 and 70, or L* between 69.5 and 87.0). This behavior is due to the short dynamic range of the digital camera response. © 2006 Wiley Periodicals, Inc.
