The present study investigates the antioxidant mechanism of grape procyanidins and, in particular, their aptitude to establish redox interactions with two important components of the endogenous antioxidant system of muscle tissues, α-tocopherol (α-TOH) and ascorbic acid (AA). To this end, the progress of lipid oxidation was monitored in fish muscle supplemented with grape procyanidins at the concentrations usually employed in antioxidant food applications, and then related to the redox stability of the endogenous α-TOH and AA. In addition to the lipid oxidation protective effect, the incorporation of procyanidins also provided an improvement of the redox stability of the endogenous components in a straight procyanidinic concentration-dependent manner. Results showed the capacity of procyanidins to repair oxidised α-TOH at medium-long term, and to delay the AA depletion. Therefore, such cooperative redox interaction of exogenous procyanidins adequately complements the natural α-TOH regenerative system supplied by AA that is efficient at the early post mortem stages.  相似文献

Historically, cell-signaling pathways have been studied as the compilation of isolated elements into a unique cascade that transmits extracellular stimuli to the tumor cell nucleus. Today, growing evidence supports the fact that intracellular drivers of tumor progression do not flow in a single linear pathway, but disseminate into multiple intracellular pathways. An improved understanding of the complexity of cancer depends on the elucidation of the underlying regulatory networks at the cellular and intercellular levels and in their temporal dimension. The high complexity of the intracellular cascades causes the complete inhibition of the growth of one tumor cell to be very unlikely, except in cases in which the so-called “oncogene addiction” is known to be a clear trigger for tumor catastrophe, such as in the case of gastrointestinal stromal tumors or chronic myeloid leukemia. In other words, the separation and isolation of the driver from the passengers is required to improve accuracy in cancer treatment. This review will summarize the signaling pathway crossroads that govern renal cell carcinoma proliferation and the emerging understanding of how these pathways facilitate tumor escape. We outline the available evidence supporting the putative links between different signaling pathways and how they may influence tumor proliferation, differentiation, apoptosis, angiogenesis, metabolism and invasiveness. The conclusion is that tumor cells may generate their own crossroads/crosstalk among signaling pathways, thereby reducing their dependence on stimulation of their physiologic pathways.  相似文献

This study reports on the intestinal permeability of salt‐containing mixtures of amino acids extracted from codfish salting wastewater. Permeability was evaluated in vitro using the Caco‐2 cell line model; cell integrity during exposure to mixtures of amino acids was estimated by measuring the transepithelial electrical resistance (TEER). The effect of salt (NaCl) on the permeability and on the intestinal cell's integrity was also examined. Permeation rate (i.e. transport) was ≥95% for all amino acids except for creatine, for which it was 6%. Values for apparent permeability coefficients, Papp > 10?5 cm s?1, for mixture with isotonic concentration of NaCl suggest that amino acids are very likely to be absorbed in humans. Mixture with a hypertonic level of NaCl exerts a cytotoxic effect in intestinal cells resulting in a loss of epithelium integrity. Results show that isotonic mixture of amino acids extracted from codfish salting wastewater could be used in food, feed, cosmetic and pharmaceutical formulations. These applications could contribute to the fish industry sustainability.  相似文献

One of the most common effects among aphasia patients is the difficulty to recall names or words. Typically, word retrieval problems can be treated through word naming therapeutic exercises. In fact, the frequency and the intensity of speech therapy are key factors in the recovery of lost communication functionalities. In this sense, speech and language technology can have a relevant contribution in the development of automatic therapy methods. In this work, we present an on-line system designed to behave as a virtual therapist incorporating automatic speech recognition technology that permits aphasia patients to perform word naming training exercises. We focus on the study of the automatic word naming detector module and on its utility for both global evaluation and treatment. For that purpose, a database consisting of word naming therapy sessions of aphasic Portuguese native speakers has been collected. In spite of the different patient characteristics and speech quality conditions of the collected data, encouraging results have been obtained thanks to a calibration method that makes use of the patients’ word naming ability to automatically adapt to the patients’ speech particularities.  相似文献

In this research, several plant compounds were in vitro evaluated against Salmonella enterica ser. Saintpaul and Escherichia coli O157:H7 through disc’s method. Among them Panax ginseng (PG) showed the highest antimicrobial activity in comparison to other natural antimicrobials. Combinations of PG (2?% v/v) with malic acid (MA) (0.5?% v/v) and/or potassium sorbate (PS) (0.05?% v/v) were made for evaluating their effects on S. Saintpaul and E. coli O157:H7 populations in sterile and fresh mango and orange juices stored at 5?°C. The best combination of antimicrobial compounds on native flora (during 21?days) as well as sensory attributes (0?day) of fresh juices were evaluated. The antimicrobial compounds added into mango and orange juices were more effective against S. Saintpaul than against E. coli O157:H7. The combination of PG (2?% v/v), MA (0.5?% v/v) and PS (0.05?% v/v) showed the highest antimicrobial effectiveness against both pathogenic microorganisms in both juices, in addition to a higher microbiological inhibition during storage (21?days). Sensory attributes such as aroma, color and taste were enhanced, but acidity was notably affected in both juices. In conclusion, the combination of PG, MA and PS could be an effective method in the food industry for ensuring the microbial safety and quality in mango and orange juices.  相似文献

Chestnuts are widely consumed around the world, especially in China, which is the major producer. Portugal is the fifth biggest producer, reaching an income of 17 M €, with particular relevance for Trás-os-Montes region, which is responsible for 81 % of Portuguese production. During postharvest storage, a number of pests tend to attack chestnuts, contributing to high economic losses. Since 2010, the most effective postharvest treatment, i.e., fumigation with methyl bromide, was banned in the European Union, urging producers to seek effective and reasonable alternatives. One alternative could be irradiation with gamma rays or electron beam, which is used in food commodities, legally regulated and allows outstanding results. Our research group has tested both irradiation types in chestnuts and studied the nutritional, antioxidant, and other chemical parameters, obtaining promising results. Herein, we extended these studies to selected cultivars from Portugal and Italy in order to validate this technique as a viable alternative to fumigation. The selected irradiation dose (1 kGy) was chosen following previous results where it proved to be effective without causing remarkable changes in chemical or antioxidant profiles. To obtain a global knowledge about how each cultivar reacts to irradiation, principal component analysis was performed using all the measured parameters. Despite the detected differences among cultivars, which differentiated particularly Palummina and Cota, it was verified that irradiation did not cause changes in chemical and antioxidant parameters that could enable defining distinctive features among irradiated and non-irradiated chestnuts. Hence, the results herein reported might be seen as a new step toward the completion of irradiation as feasible conservation technology, independently of chestnuts origin.  相似文献

The oxidation kinetics, under different pressures, of soot samples obtained from different liquid fuels and two standards (a commercial black carbon sample and a reference diesel soot) was studied. Soot samples were generated in a flat-flame, premixed burner under heavily-sooting conditions and captured on a water-cooled stabilization plate located above the burner surface. The collected soot was oxidized using a high-pressure thermogravimetric analyzer (HTGA). TGA operation was optimized to reduce mass transfer effects by adjusting the oxidizer flow rate and initial sample mass. Further corrections for mass transfer were accomplished by computing the effectiveness factors for intraparticle, interparticle, and external mass transfer. Two pressures were evaluated (1 and 10 atm) and O2 concentration was varied between 10 and 21%.  相似文献

The association between in vitro antioxidant capacity of dark chocolates with different cocoa percentage and the in vivo response on antioxidant status was investigated. In a randomized crossover design, 15 healthy volunteer consumed 100 g of high antioxidants dark chocolate (HADC) or dark chocolate (DC). In vitro, HADC displayed a higher Total Antioxidant Capacity (TAC) than DC. In vivo, plasma TAC significantly peaked 2 h after ingestion of both chocolates. TAC levels went back to zero 5 h after DC ingestion whilst levels remained significantly higher for HADC. HADC induced a significantly higher urinary TAC in the 5-12 h interval time than DC. No change was detected in urinary excretion of F2-isoprostanes. Plasma thiols and triacylglycerol (TG) levels significantly increased for both chocolate with a peak at 2 h remaining significantly higher for DC after 5 h respect to HADC. Results provide evidence of a direct association between antioxidant content of chocolate and the extent of in vivo response on plasma antioxidant capacity.  相似文献

Total Diet Studies to estimate dietary exposure to food contaminants need to evaluate laboratory measurements data variance. In this process it is critical that data from analytical methods are reliable to correctly scrutinize and compare values over time and between countries. In Europe it is widely recognized that the evaluation of measurement uncertainty is an important parameter when assessing the sources of analytical data variability. Two approaches are considered to estimate uncertainty in analytical measurement. Arsenic, Lead, Chromium and Cadmium, content in several food matrix determined by Inductively Coupled Mass Spectrometry (ICP-MS) microwave digestion assisted, are used as examples. The aim of the present research work is to compare both approaches accepted by Eurolab and GUM: Mathematical modeling to assess uncertainty components based on a classical model (bottom up) and an empirical method (top down), based on either experimental data obtained from a single laboratory validation data or inter-laboratory data from Proficiency Testing schemes. Relative expanded uncertainty calculated by both approaches agree when U (%) <20%. These values are concordant with RSDR reported in collaborative studies of EN 15763 (2009), which were assumed as target uncertainty. The top down approach described is simple and easy to use when compared with the mathematical modeling approach providing considerable benefits to those who assess data produced by several laboratories.  相似文献
