Apical membrane antigen 1 is a microneme protein which plays an indispensable role during Apicomplexa parasite invasion. The detailed mechanism of AMA-1 molecular interaction with its receptor on bovine erythrocytes has not been completely defined in Babesia bovis. This study was focused on identifying the minimum B. bovis AMA-1-derived regions governing specific and high-affinity binding to its target cells. Different approaches were used for detecting ama-1 locus genetic variability and natural selection signatures. The binding properties of twelve highly conserved 20-residue-long peptides were evaluated using a sensitive and specific binding assay based on radio-iodination. B. bovis AMA-1 ectodomain structure was modelled and refined using molecular modelling software. NetMHCIIpan software was used for calculating B- and T-cell epitopes. The B. bovis ama-1 gene had regions under functional constraint, having the highest negative selective pressure intensity in the Domain I encoding region. Interestingly, B. bovis AMA-1-DI (100YMQKFDIPRNHGSGIYVDLG119 and 120GYESVGSKSYRMPVGKCPVV139) and DII (302CPMHPVRDAIFGKWSGGSCV321)-derived peptides had high specificity interaction with erythrocytes and bound to a chymotrypsin and neuraminidase-treatment sensitive receptor. DI-derived peptides appear to be exposed on the protein’s surface and contain predicted B- and T-cell epitopes. These findings provide data (for the first-time) concerning B. bovis AMA-1 functional subunits which are important for establishing receptor-ligand interactions which could be used in synthetic vaccine development. 相似文献

The preferences adopted by individuals are constantly modified as these are driven by new experiences, natural life evolution and, mainly, influence from friends. Studying these temporal dynamics of user preferences has become increasingly important for personalization tasks in information retrieval and recommendation systems domains. However, existing models are too constrained for capturing the complexity of the underlying phenomenon. Online social networks contain rich information about social interactions and relations. Thus, these become an essential source of knowledge for the understanding of user preferences evolution. In this work, we investigate the interplay between user preferences and social networks over time. First, we propose a temporal preference model able to detect preference change events of a given user. Following this, we use temporal networks concepts to analyze the evolution of social relationships and propose strategies to detect changes in the network structure based on node centrality. Finally, we look for a correlation between preference change events and node centrality change events over Twitter and Jam social music datasets. Our findings show that there is a strong correlation between both change events, specially when modeling social interactions by means of a temporal network. 相似文献

A coal-water slurry is an econonical substitute for premium liquid fuels, but the amount of sulfur in coal is usually higher than in petroleum liquids. Lime has been used to capture sulfur in fluidized coal combustion as CaSO4, but the latter decomposes above 1273 K. We report here on the possible capture of sulfur in the ash as CaS, which has higher thermal stability than CaSO4. This can be accomplished using a fuel-rich first stage in a coal-water slurry combustor, or under the conditions normally encountered in some coal gasifiers. Thermodynamic calculations and some preliminary experiments suggest that the concept is promising for effective sulfur removal and, by extension, for NOx, control. 相似文献

Epichloë fungi (Ascomycota) live within aboveground tissues of grasses and can have important implications for natural and managed ecosystems through production of alkaloids. Nonetheless, vertebrate herbivores may possess traits, like oral secretions, that mitigate effects of alkaloids. We tested if sheep saliva mitigates effects of Epichloë alkaloids on a beetle pest of perennial ryegrass (Lolium perenne L.) in a New Zealand pasture setting. Plants with one of several fungal isolates were clipped with scissors, grazed by sheep, or clipped with sheep saliva applied to cut ends of stems. We then assessed feeding damage by Argentine stem weevils on blade segments collected from experimental plants. We found that clipping plants induced synthesis of an alkaloid that reduces feeding by beetles and that sheep saliva mitigates this effect. Unexpectedly, the alkaloid (perloline) that explains variation in beetle feeding is one produced not by the endophyte, but rather by the plant. Yet, these effects depended upon fungal isolate. Such indirect, complex interactions may be much more common in both managed and natural grassland systems than typically thought and could have implications for managing grazing systems. 相似文献

The fate of anthocyanins and ellagitannins in rats was monitored following ingestion of raspberry juice. After 1 h low nM concentrations of unmetabolised anthocyanins were present in plasma but these declined by 2 h and after 4 h they were no longer detectable. For the first 2 h there was an almost full recovery of anthocyanins as they passed from the stomach through the duodenum/jejunum and into the ileum. After 3 h less than 50% were recovered, and the levels declined rapidly thereafter. Excretion of raspberry anthocyanins in urine over a 24 h period was equivalent to 1.2% of the amount ingested. Trace quantities of anthocyanins were detected in the caecum, colon and faeces and they were absent in extracts of liver, kidneys and brain. Urine also contained a number of phenolic acids but most were present in quantities well in excess of the 918 nmol of anthocyanins present in the ingested juice. These findings indicate that raspberry anthocyanins per se are poorly absorbed, probably prior to reaching the ileum, and that substantial amounts pass from the small to the large intestine where they are degraded by colonic bacteria. Low-grade chronic inflammation plays a pivotal role in the pathogenesis of insulin resistance (IR), and skeletal muscle has a central role in this condition. NLRP3 inflammasome activation pathways promote low-grade chronic inflammation in several tissues. However, a direct link between IR and NLRP3 inflammasome activation in skeletal muscle has not been reported. Here, we evaluated the NLRP3 inflammasome components and their role in GLUT4 translocation impairment in skeletal muscle during IR. Male C57BL/6J mice were fed with a normal control diet (NCD) or high-fat diet (HFD) for 8 weeks. The protein levels of NLRP3, ASC, caspase-1, gasdermin-D (GSDMD), and interleukin (IL)-1β were measured in both homogenized and isolated fibers from the flexor digitorum brevis (FDB) or soleus muscle. GLUT4 translocation was determined through GLUT4myc-eGFP electroporation of the FBD muscle. Our results, obtained using immunofluorescence, showed that adult skeletal muscle expresses the inflammasome components. In the FDB and soleus muscles, homogenates from HFD-fed mice, we found increased protein levels of NLRP3 and ASC, higher activation of caspase-1, and elevated IL-1β in its mature form, compared to NCD-fed mice. Moreover, GSDMD, a protein that mediates IL-1β secretion, was found to be increased in HFD-fed-mice muscles. Interestingly, MCC950, a specific pharmacological NLRP3 inflammasome inhibitor, promoted GLUT4 translocation in fibers isolated from the FDB muscle of NCD- and HFD-fed mice. In conclusion, we found increased NLRP3 inflammasome components in adult skeletal muscle of obese insulin-resistant animals, which might contribute to the low-grade chronic metabolic inflammation of skeletal muscle and IR development. 相似文献

Main modelling approaches used for investigating the Photonic bandgap (PBG) devices are reviewed. In particular, the model based on Leaky Mode Propagation (LMP) method is described. A complete analysis of the propagation characteristics, including the determination of modal propagation constants, electromagnetic field harmonics and total field distribution, transmission and reflection coefficients, total forward and backward power flow in the structure, guided and radiated power, and total losses, can be carried out by a computer program based on the LMP approach. The numerical results have been validated by comparisons with those obtained by using other more complex and expensive models. The new model shows some significant advantages in terms of very low computational time, absence of any a priori theoretical assumptions and approximations, capability of simulating the actual physical behaviour of the device and fast determination of the bandgap position.Copyright © 2003 John Wiley & Sons, Ltd. 相似文献

In this paper, an analytical model for the electrothermal solution to the non‐linear 3‐D heat flow equation for multilayer structure electronic devices is proposed. Compared with previous models presented in literature, it is general and can be easily applied to a large variety of integrated devices, provided that their structure can be represented as an arbitrary number of superimposed layers with a 2‐D embedded thermal source, so as to include the effect of the package. The proposed method is independent of the specific physical properties of the layers, hence GaAs MESFETs and HEMTs as well as silicon and silicon‐on‐insulator MOSFETs and heterostructure LASERs can be analysed. Moreover, it takes into account the dependence of the thermal conductivity of all the layers on the temperature; the heat equation is solved coupled with the device current–voltage relation in order to give physical consistence to the experimental evidence that a temperature increase causes a degradation of the electrical performances and that the electrical power is not uniformly distributed.
