Erwinia amylovora was shown to secrete DspE, a pathogenicity factor of 198 kDa and a functional homolog of AvrE of Pseudomonas syringae pv. tomato. DspE was identified among the supernatant proteins isolated from cultures grown in an hrp gene-inducing minimal medium by immunodetection with a DspE-specific antiserum. This paper summarises the levels and composition of chlorobenzenes, chlorinated pesticides, coplanar polychlorinated biphenyls (PCBs) and the chlorinated compounds octachlorostyrene (OCS), hexachlorocyclobutadiene (HCBD) and pentachloro-anisole (PCA) in biota from the terrestrial, freshwater and marine environment of Greenland. The data were obtained during the second phase of the Arctic Monitoring and Assessment Programme (AMAP). Of the chlorobenzenes, hexachlorobenzene was the main constituent detected in almost all samples. The chlorobenzenes accumulate in the marine food web in a similar manner to the better-studied persistent organic pollutants, with maximum concentrations in beluga, minke whale and narwhal. However, concentrations in ringed seals and kittiwakes were lower than in marine fish, contradicting biomagnification. Of the organochlorine pesticides, the drin pesticides (aldrin, endrin, dieldrin) and heptachlor had increasing concentrations along the food chain, whilst biomagnification was less pronounced for endosulfan, methoxychlor and mirex. Endosulfan and methoxychlor are pesticides still in use and considered less persistent than other organochlorine pesticides. Their occurrence in Arctic biota is of particular concern, also given the high acute toxicity of endosulfan to fish. Chlorobenzene and pesticide concentrations tended to be lower in the Greenland samples than in the same animals from the Canadian Arctic, whilst their concentrations were similar to samples from Svalbard and Iceland. However, temporal trends might overlap the geographical differences. Coplanar chlorobiphenyls (CBs) were found in all samples analysed, with the maximum concentrations found in marine mammals such as beluga and narwhal. Biota from the terrestrial environment appeared to be less contaminated. The main contributor on a TEQ basis was CB126. OCS, HCBD and PCA were detected in biota from Greenland, although at very low concentrations. OCS seems to have the widest occurrence and the highest potential for biomagnification of the three compounds analysed.  相似文献

Biogenic amines (histamine, tyramine, putrescine, cadaverine, agmatine, spermidine and spermine) are nitrogenous compounds. They occur naturally in living organisms and are involved in many biological processes. Nonetheless, high amounts in food may be hazardous to human health. The diamines putrescine and cadaverine in food can potentiate the effects of simultaneously ingested histamine. In protein-rich foods, high concentrations of these diamines are indicative for hygienic deficiencies in the food chain. Even though being formed endogenously and being essential for some physiological metabolic pathways, both diamines are known as precursors for carcinogenic nitrosamines. Putrescine also plays a certain role in tumour growth. Nevertheless, no tolerable levels in foods have been established so far. The present study suggests tolerable levels in cheese, fermented sausages, fish, sauerkraut and seasonings that are based on toxicological threshold levels, occurrence of diamines in foods and food consumption in Austria. Average daily intake of putrescine via fermented food was calculated to be 6.8 (female adults) and 8.8 (male adults) mg per person. Respective numbers for cadaverine were 9.8 and 11.6 mg per person and day. For putrescine, proposed maximum tolerable levels for sauerkraut, fish, cheese, fermented sausages and seasonings are 140, 170, 180, 360 and 510 mg/kg, respectively. Likewise, for cadaverine, in sauerkraut, fish, cheese, fermented sausages and seasonings, maximum tolerable levels are 430, 510, 540, 1,080 and 1,540 mg/kg, respectively. These limits can be met by current manufacturing practices, as ascertained from the results of our own studies and from literature. Admittedly, only few data are published on toxicological threshold levels of these diamines, which mean that these tolerable levels are associated with some uncertainty.  相似文献

Air was sampled for one year in the central valley of Costa Rica using an active high-volume sampler as well as passive samplers (PAS) based on polyurethane foam (PUF) disks and XAD-resin filled mesh cylinders. Extracts were analyzed for pesticides that are either banned or currently used in Costa Rican agriculture. Sampling rates for PUF-based passive air samplers, determined from the loss of depuration compounds spiked on the disks prior to deployment averaged 5.9 +/- 0.9 m3 x d(-1) and were higher during the windier dry season than during the rainy season. Sampling rates for the XAD-based passive sampler were determined from the slopes of linear relationships that were observed between the amount of pesticide sequestered in the resin and the length of deployment, which varied from 4 months to 1 year. Those sampling rates increased with decreasing molecular size of a pesticide, and their average of 2.1 +/- 1.5 m3 x d(-1) is higher than rates previously reported for temperate and polar sampling sites. Even though the trends of the sampling rate with molecular size and temperature are consistent with the hypothesis that molecular diffusion controls uptake in passive samplers, the trends are much more pronounced than a direct proportionality between sampling rate and molecular diffusivity would suggest. Air concentrations derived by the three sampling methods are within a factor of 2 of each other, suggesting that properly calibrated PAS can be effective tools for monitoring levels of pesticides in the tropical atmosphere. In particular, HiVol samplers, PUF-disk samplers, and XAD-based passive samplers are suitable for obtaining information on air concentration variability on the time scale of days, seasons and years, respectively. This study represents the first calibration study for the uptake of current use pesticides by passive air samplers.  相似文献

Between 1976 and 1978, 248 harp seals were sampled from 5 locations in the Northwest Atlantic and Arctic for organochlorine (OC) residue analysis in tissue. Blood, kidney, brain, muscle and blubber samples were analysed for PCB, DDT, dieldrin, hexachlorobenzene (HCB), and heptachlor epoxide. Levels were generally greatest in blubber tissue. Overall, mean levels of DDT and PCB were greater than those of other OC. A barrier prevented OC compounds from accumulating in the brain to the level one might expect in lipid tissues. Positive correlations were found between 1: DDT and PCB, 2: DDT and dieldrin, and 3: PCB and deildrin.Male harp seals bioaccumulated DDT, PCB, heptachlor epoxide and dieldrin with age. As a group, adult males sampled in the Gulf of St. Lawrence carried the highest concentrations of all OC, except that chlordane did not appear to bioaccumulate and levels of hexachlorobenzene were minimal. Females generally ceased exhibiting significant bioaccumulation once breeding age was reached, due to transplacental and transmammary residue transfer of OC from mother to pup. There were some significant differences in OC levels between locations with DDT: Gulf > Front; PCB: Gulf > Front; dieldrin: Gulf > Front; heptachlor epoxide: Front > Gulf. No significant differences were found, however, when northern locations (Grise Fiord, Northwest Greenland, Pangnirtung) were compared to southern (Gulf, Front) on a group basis. The latter is not surprising in view of the harp seals extensive cyclical annual migration between the southern breeding grounds and the High Arctic.  相似文献

Food- and waterborne viruses, such as human norovirus, hepatitis A virus, hepatitis E virus, rotaviruses, astroviruses, adenoviruses, and enteroviruses, are major contributors to all foodborne illnesses. Their small size, structure, and ability to clump and attach to inanimate surfaces make viruses challenging to reduce or eliminate, especially in the presence of inorganic or organic soils. Besides traditional wet and dry methods of disinfection using chemicals and heat, emerging physical nonthermal decontamination techniques (irradiation, ultraviolet, pulsed light, high hydrostatic pressure, cold atmospheric plasma, and pulsed electric field), novel virucidal surfaces, and bioactive compounds are examined for their potential to inactivate viruses on the surfaces of foods or food contact surfaces (tools, equipment, hands, etc.). Every disinfection technique is discussed based on its efficiency against viruses, specific advantages and disadvantages, and limitations. Structure, genomic organization, and molecular biology of different virus strains are reviewed, as they are key in determining these techniques effectiveness in controlling all or specific foodborne viruses. Selecting suitable viral decontamination techniques requires that their antiviral mechanism of action and ability to reduce virus infectivity must be taken into consideration. Furthermore, details about critical treatments parameters essential to control foodborne viruses in a food production environment are discussed, as they are also determinative in defining best disinfection and hygiene practices preventing viral infection after consuming a food product.  相似文献

Self-adaptive service-oriented Applications (Self-Apps) must be able to understand themselves or the environment in which they are executed, and propose solutions to meet changing conditions. The development of these applications is not a trivial task, since it encompasses issues from different research areas. Despite the importance of frameworks for Self-Apps, there is a lack of comprehensive analysis of how the design of such applications is performed, and regarding the standardization of concepts and coverage of minimum requirements for Self-Apps. The main contribution of this article is to present this comprehensive analysis, providing the state of the art for this subject. This analysis was built through a Systematic Mapping Study, based on a total of 65 studies, from which we identify the main attributes for Quality of Service (QoS), search strategies, and service management strategies employed in the design of frameworks for Self-Apps. The main aspects of requirements involved in the design of Self-Apps were pointed out to stakeholders. For example, these applications must implement a method for evaluation of QoS based on metrics. We also put forward the S-Frame, a modular solution that brings together the main features for the design of Self-Apps, and describe the main challenges concerning these applications.  相似文献

The pentafluorosulfanyl (-SF5) functional group is of increasing interest as a bioisostere in medicinal chemistry. A library of SF5-containing compounds, including amide, isoxazole, and oxindole derivatives, was synthesised using a range of solution-based and solventless methods, including microwave and ball-mill techniques. The library was tested against targets including human dihydroorotate dehydrogenase (HDHODH). A subsequent focused approach led to synthesis of analogues of the clinically used disease modifying anti-rheumatic drugs (DMARDs), Teriflunomide and Leflunomide, considered for potential COVID-19 use, where SF5 bioisostere deployment led to improved inhibition of HDHODH compared with the parent drugs. The results demonstrate the utility of the SF5 group in medicinal chemistry.  相似文献

Tuberculosis represents one of the ten most common courses of death worldwide and the emergence of multidrug-resistant M. tuberculosis makes the discovery of novel anti-tuberculosis active structures an urgent priority. Here, we show that (+)-floyocidin B representing the first example of a novel dihydroisoquinoline class of fungus-derived natural products, displays promising antitubercular hit properties. (+)-Floyocidin B was identified by activity-guided extract screening and its structure was unambiguously determined by total synthesis. The absolute configuration was deduced from a key synthesis intermediate by single crystal X-ray diffraction analysis. A hit series was generated by the isolation of further natural congeners and the synthesis of analogs of (+)-floyocidin B. Extensive biological and physicochemical profiling of this series revealed first structure-activity relationships and set the basis for further optimization and development of this novel antitubercular scaffold.  相似文献
