It was hypothesized that human mesenchymal stromal cell (hMSC) can be guided by patterned and plain amorphous diamond (AD), titanium (Ti), tantalum (Ta) and chromium (Cr) coatings, produced on silicon wafer using physical vapour deposition and photolithography. At 7.5 h hMSCs density was 3.0–3.5× higher (P < 0.0003, except Ti) and cells were smaller (68 vs. 102 μm, P 0.000006–0.02) on patterns than on silicon background. HMSC-covered surface of the background silicon was lower on Ti than AD patterns (P = 0.015), but at 5 days this had reversed (P = 0.006). At 7.5 h focal vinculin adhesions and actin cytoskeleton were outgoing from pattern edges so cells assumed geometric square shapes. Patterns allowed induced osteogenesis, but less effectively than plain surfaces, except for AD, which could be used to avoid osseointegration. All these biomaterial patterns exert direct early, intermediate and late guidance on hMSCs and osteogenic differentiation, but indirect interactions exist with cells on silicon background.  相似文献

We have established the main post-translational modification of the major exoglucanase of Saccharomyces cerevisiae as the enzyme progresses through the secretory pathway. The protein portion of the enzyme accumulated by sec18 cells was about 2 kDa larger than that of the secreted enzyme. This precursor (form A) was stable when maintained in the endoplasmic reticulum but was processed to the mature form (form B) before the block imposed by the sec7 mutation. Sec7 cells, when incubated at 37 degrees C, accumulated form B first, but upon prolonged incubation, form A was preferentially accumulated. When the supply of newly synthesized exoglucanase was prevented by the addition of cycloheximide, the accumulated A was transformed into B in the presence of altered Sec7p that still prevented secretion. Conversion of A into B was prevented in the double mutant sec7 kex2-1, indicating that Kex2p is central to the in vivo processing. Consistent with this, a KEX2 deletion mutant secreted form A exclusively. Conversion of A into B was also prevented in sec7 cells by the presence of dinitrophenol, a poison that depletes ATP levels, indicating that processing is dependent upon intracellular transport which involves ER --> Golgi and/or, at least, one intra-Golgi step(s). It follows that this transport step(s) is independent of functional Sec7p.  相似文献

This paper presents an analysis and validation by advanced system simulation of compact and low-cost six-port transceivers for future wireless local area networks (WLANs) operating at millimeter-wave frequencies. To obtain realistic simulation results, a six-port model based on the measurement results of a fabricated V-band hybrid coupler, the core component, is used. A frequency-division multiplexing scheme is used by introducing four quadrature phase-shift keying (QPSK) channels in the wireless communication link. The data rate achieved is about 4 Gbit/s. The operating frequency is in the 60-64 GHz unlicensed band. Bit error rate (BER) results are presented, and a comparison is made between single-carrier and multicarrier architectures. The proposed wireless system can be considered an efficient candidate for millimeter-wave communication systems operating at quasi-optical data rates.  相似文献

Pooled versus separate measurements of tree-ring stable isotopes   总被引:1,自引:0,他引:1  
δ13C and δ18O of tree rings contain time integrated information about the environmental conditions weighted by seasonal growth dynamics and are well established as sources of palaeoclimatic and ecophysiological data. Annually resolved isotope chronologies are frequently produced by pooling dated growth rings from several trees prior to the isotopic analyses. This procedure has the advantage of saving time and resources, but precludes from defining the isotopic error or statistical uncertainty related to the inter-tree variability. Up to now only a few studies have compared isotope series from pooled tree rings with isotopic measurements from individual trees. We tested whether or not the δ13C and the δ18O chronologies derived from pooled and from individual tree rings display significant differences at two locations from the Iberian Peninsula to assess advantages and constraints of both methodologies. The comparisons along the period 1900-2003 reveal a good agreement between pooled chronologies and the two mean master series which were created by averaging raw individual values (Mean) or by generating a mass calibrated mean (MassC). In most of the cases, pooled chronologies show high synchronicity with averaged individual samples at interannual scale but some differences also show up especially when comparing δ18O decadal to multi-decadal variations. Moreover, differences in the first order autocorrelation among individuals may be obscured by pooling strategies. The lack of replication of pooled chronologies prevents detection of a bias due to a higher mass contribution of one sample but uncertainties associated with the analytical process itself, as sample inhomogeneity, seems to account for the observed differences.  相似文献

Small-scale gold mining in Portovelo-Zaruma, Southern Equador, performed by mercury amalgamation and cyanidation, yields 9-10 t of gold/annum, resulting in annual releases of around 0.65 t of inorganic mercury and 6000 t of sodium cyanide in the local river system. The release of sediments, cyanide, mercury, and other metals present in the ore such as lead, manganese and arsenic significantly reduces biodiversity downstream the processing plants and enriches metals in bottom sediments and biota. However, methylmercury concentrations in sediments downstream the mining area were recently found to be one order of magnitude lower than upstream or in small tributaries. In this study we investigated cyanide, bacterial activity in water and sediment and mercury methylation potentials in sediments along the Puyango river watershed, measured respectively by in-situ spectrophotometry and incubation with 3H-leucine and 203Hg2+.Free cyanide was undetectable (< 1 μg·L− 1) upstream mining activities, reached 280 μg·L− 1 a few km downstream the processing plants area and was still detectable about 100 km downstream. At stations with detectable free cyanide in unfiltered water, 50% of it was dissolved and 50% associated to suspended particles. Bacterial activity and mercury methylation in sediment showed a similar spatial pattern, inverse to the one found for free cyanide in water, i.e. with significant values in pristine upstream sampling points (respectively 6.4 to 22 μgC·mg wet weight− 1·h− 1 and 1.2 to 19% of total 203Hg·g dry weight− 1·day− 1) and undetectable downstream the processing plants, returning to upstream values only in the most distant downstream stations. The data suggest that free cyanide oxidation was slower than would be expected from the high water turbulence, resulting in a long-range inhibition of bacterial activity and hence mercury methylation. The important mercury fluxes resultant from mining activities raise concerns about its biomethylation in coastal areas where many mangrove areas have been converted to shrimp farming.  相似文献

The alarming raise of multi-drug resistance among human microbial pathogens makes the development of novel therapeutics a priority task. In contrast to conventional antibiotics, antimicrobial peptides (AMPs), besides evoking a broad spectrum of activity against microorganisms, could offer additional benefits, such as the ability to neutralize toxins, modulate inflammatory response, eradicate bacterial and fungal biofilms or prevent their development. The latter properties are of special interest, as most antibiotics available on the market have limited ability to diffuse through rigid structures of biofilms. Lipidation of AMPs is considered as an effective approach for enhancement of their antimicrobial potential and in vivo stability; however, it could also have undesired impact on selectivity, solubility or the aggregation state of the modified peptides. In the present work, we describe the results of structural modifications of compounds designed based on cationic antimicrobial peptides DK5 and CAR-PEG-DK5, derivatized at their N-terminal part with fatty acids with different lengths of carbon chain. The proposed modifications substantially improved antimicrobial properties of the final compounds and their effectiveness in inhibition of biofilm development as well as eradication of pre-formed 24 h old biofilms of Candida albicans and Staphylococcus aureus. The most active compounds (C5-DK5, C12-DK5 and C12-CAR-PEG-DK5) were also potent against multi-drug resistant Staphylococcus aureus USA300 strain and clinical isolates of Pseudomonas aeruginosa. Both experimental and in silico methods revealed strong correlation between the length of fatty acid attached to the peptides and their final membranolytic properties, tendency to self-assemble and cytotoxicity.  相似文献

Dioecious species differ in the pattern and intensity of male and female reproductive investments. We aimed to determine whether female shoots deprived of generative buds show biochemical features, indicating their less-pronounced reproductive effort. For this purpose, the same branches of mature Taxus baccata females were deprived of generative organs. In the second and third years of the experiment, measurements were made in every season from the control and bud-removed shoots of females and control males. Bud removal caused an increase in nitrogen concentration almost to the level detected in the needles of male specimens, but only in current-year needles. Moreover, differences between male and control female shoots were present in the C:N ratio and increment biomass, but they disappeared when bud removal was applied to females. Additionally, between-sex differences were observed for content of phenolic compounds, carbon and starch, and SLA, independent of the female shoot reproductive effort. The study revealed that nitrogen uptake in seeds and arils may explain the lower nitrogen level and consequently the lower growth rate of females compared to males. At the same time, reproduction did not disturb carbon level in adjacent tissues, and two hypotheses explaining this phenomenon have been put forward.  相似文献

The use of microwave synthesized sources improved significantly the performance of correlation reflectometry diagnostics for fusion plasmas providing broadband operation with high quality signals and fast frequency switching times. Recent new developments of those sources made at IPFN opened an all-new way to build correlation reflectometry diagnostics with improved performance and measuring capability. Some of the key features now attainable are independent probing and local oscillator signals generated by separated frequency synthesizers; very good frequency control and synchronization of both the local and the radio-frequency oscillators providing very high stability. The technological advances make it possible to further reduce the switching time between adjacent probing frequencies while keeping the ability to operate in a broad frequency range.Here we present a multichannel correlation system based on those novel technologies using several independent and self-contained fully synthesized broadband channels. With this type of system various type of measurements are possible: (i) fast stepwise radial scans to extract the radial profile of turbulence parameters (spectra, RMS, rotation from Doppler shift, etc.); (ii) radial scans of the plasma using two identical systems in parallel where one system operates in frequency steps and the other makes multiple small steps around each selected frequency of the first system. The proposed diagnostic is very powerful and versatile. With a system having n identical channels, all transmitting and receiving simultaneously, the number of simultaneous correlations measurements that can be performed is given by n × (n ? 1)/2. The minimal configuration uses two channels thus providing one (time domain) correlation signal channels can be easily added at any time increasing the measuring capability of the diagnostic. Several examples are given that illustrate the wide range of measurements that can be made using the novel approach for correlation reflectometry diagnostics.  相似文献

This study investigates the coupled effects of mild shear flow and temperature on the crystallization behavior of two thermoplastic polymers, namely, an isotactic polypropylene and an isotactic poly(1-butene). Rheological experiments are used to measure the crystallization induction time under isothermal, steady shear flow conditions. The experimental results clearly show the effects of the degree of undercooling on flow-induced crystallization (FIC). As temperature decreases, the corresponding increase in chain orientation at a given shear rate leads to an absolutely faster crystallization. At the same time, however, a temperature decrease makes the flow-induced driving force to crystallization relatively less influent with respect to the intrinsic kinetics. A FIC model based on the Doi-Edwards micro-rheological theory is shown to successfully describe the quantitative details of the observed experimental behavior.  相似文献
