The in vitro gastric digestion of several food allergens (beta-lactoglobulin (BLG), alpha-lactalbumin (LA) and peanut allergens (PE)) in the presence of a catechin-enriched polyphenol extract of green tea (GTC), oxidized polyphenols and phenol oxidase processed food allergens and GTC was investigated. Pepsin-resistant proteins, such as BLG, major peanut allergens, Ara h 1 and Ara h 2, degrade faster in the presence of catechin-enriched green tea polyphenols. Phenol oxidase polymerized GTC affected adversely protein digestion of BLG and LA, but not digestion of PE proteins. Protecting effect of polyphenols correlated well with the ability of proteins to form insoluble complexes with oxidized catechins. Cross-linking of proteins and polyphenols further extended the half-lives of BLG and LA in the in vitro digestion by pepsin. Catechin-enriched green tea polyphenols of food supplements facilitate pepsin digestion of major food allergens, but hamper their digestion if oxidized and polymerized by phenol oxidase.  相似文献

In this paper a fiber optic surface plasmon resonance (FO-SPR) sensor was used to analyze the melting process of DNA linked to silica nanoparticles. Real-time monitoring of a DNA melting process has rarely been studied using surface plasmon resonance (SPR), since most commercial SPR setups do not allow for dynamic and accurate temperature control above 50 °C. The FO-SPR sensor platform, with silica nanobead signal amplification, allows sensing inside a standard PCR thermocycler, which makes high resolution DNA melting curve analysis possible. This innovative combination was used to characterize the hybridization and melting events between DNA immobilized on the sensor surface and DNA probes on silica nanoparticles. At optimized hybridization conditions complementary DNA strands of different lengths could be distinguished. While the real-time FO-SPR analysis of DNA hybridization did not result in significant variances, the analysis of DNA melting determined the exact length of overlap and the matching Gibbs energy.  相似文献

Non-covalent interactions between β-lactoglobulin (BLG) and polyphenol extracts of teas, coffee and cocoa were studied by fluorescence and CD spectroscopy at pH values of the gastrointestinal tract (GIT). The biological implications of non-covalent binding of polyphenols to BLG were investigated by in vitro pepsin and pancreatin digestibility assay and ABTS radical scavenging activity of complexes formed. The polyphenol–BLG systems were stable at pH values of the GIT. The most profound effect of pH on binding affinity was observed for polyphenol extracts rich in phenolic acids. Stronger non-covalent interactions delayed pepsin and pancreatin digestion of BLG and induced β-sheet to α-helix transition at neutral pH. All polyphenols tested protected protein secondary structure at an extremely acidic pH of 1.2. A positive correlation was found between the strength of protein–polyphenol interactions and (a) half time of protein decay in gastric conditions (R2 = 0.85), (b) masking of total antioxidant capacity of protein–polyphenol complexes (R2 = 0.95).  相似文献

A series of aliphatic poly(ether–ester)s based on flexible poly(tetramethylene oxide) (PTMO) and hard poly (butylene succinate) (PBS) segments were synthesized by the catalyzed two‐step transesterification reaction of dimethyl succinate, 1,4‐butanediol, and α,ω‐hydroxy‐terminated PTMO (Mn = 1000 g/mol) in the bulk. The content of soft PTMO segments in the polymer chains was varied from 10 to 50 mass %. The effect of the introduction of the soft segments on the structure, thermal, and physical properties, as well as on the biodegradation properties was investigated. The composition and structure of the aliphatic segmented copolyesters were determined by 1H NMR spectroscopy. The molecular weights of the polyesters were verified by viscometry of dilute solutions and polymer melts. The thermal properties were investigated using DSC. The degree of crystallinity was determined by means of DSC and WAXS. Biodegradation of the synthesized copolyesters, estimated in enzymatic degradation tests on polymer films in phosphate buffer solution with Candida rugosa lipase at 37°C, was compared with hydrolytic degradation in the buffer solution. Viscosity measurements confirmed that there was no change in molecular weight of the copolyesters leading to the conclusion that the degradation mechanism of poly(ester–ether)s based on PTMO segments occurs through the surface erosion. © 2007 Wiley Periodicals, Inc. Olivine structure LiFePO4 attracted much attention as a promising cathode material for lithium-ion batteries. The overwhelming advantage of iron-based compounds is that, in addition to being inexpensive and naturally abundant, they are less toxic than Co, Ni, and Mn. Its commercial use has already started and there are several companies that base their business on lithium phosphate technology. Still, there is a need for a manufacturing process that produces electrochemically active LiFePO4 at a low cost. Therefore the interest in developing new approaches to the synthesis of LiFePO4 did not fade. Here is presented a review of the synthesis procedures used for the production of LiFePO4 powders along with the highlights of doped and coated derivatives. Apart from already established conventional routes of preparation, numerous alternative procedures are mentioned.  相似文献

The presence of a microgap along an implant–abutment connection (IAC) is considered the main disadvantage of two-piece implant systems. Its existence may lead to mechanical and biological complications. Different IAC designs have been developed to minimise microleakage through the microgap and to increase the stability of prosthodontic abutments. Furthermore, different sealing materials have appeared on the market to seal the gap at the IAC. The purpose of this study was to evaluate the antimicrobial efficacy and permeability of different materials designed to seal the microgap, and their behaviour in conical and straight types of internal IACs. One hundred dental implants with original prosthodontic abutments were divided into two groups of fifty implants according to the type of IAC. Three different sealing materials (GapSeal, Flow.sil, and Oxysafe gel) were applied in the test subgroups. The contamination of implant–abutment assemblies was performed by a joint suspension containing Candida albicans and Staphylococcus aureus. It was concluded that the IAC type had no significant influence on microleakage regarding microbial infection. No significant difference was found between the various sealing agents. Only one sealing agent (GapSeal) was found to significantly prevent microleakage. A complete hermetic seal was not achieved with any of the sealing agents tested in this study.  相似文献

Study of interaction of explosive reactive armor (ERA) with shaped charge jet is the basis for evaluation of the effectiveness of ERA. The physically based theoretical model of this interaction is given. It is incorporated in the NERA computer code. The influences of backward moving plate and forward moving plate thickness, explosive layer thickness, jet attack angle, and distance between ERA and main armor are investigated. The comprehensive analysis of a longitudinal point of impact effect on ERA efficiency, based on NERA code calculations, is presented. Computational results of NERA code are compared with experimental data. The computational and experimental results of penetration in the steel armour target are in good agreement. The developed code enables optimization of explosive reactive armor characteristics.  相似文献
