Osteocytes—the central regulators of bone remodeling—are enclosed in a network of microcavities (lacunae) and nanocanals (canaliculi) pervading the mineralized bone. In a hitherto obscure process related to aging and disease, local plugs in the lacuno‐canalicular network disrupt cellular communication and impede bone homeostasis. By utilizing a suite of high‐resolution imaging and physics‐based techniques, it is shown here that the local plugs develop by accumulation and fusion of calcified nanospherites in lacunae and canaliculi (micropetrosis). Two distinctive nanospherites phenotypes are found to originate from different osteocytic elements. A substantial deviation in the spherites' composition in comparison to mineralized bone further suggests a mineralization process unlike regular bone mineralization. Clearly, mineralization of osteocyte lacunae qualifies as a strong marker for degrading bone material quality in skeletal aging. The understanding of micropetrosis may guide future therapeutics toward preserving osteocyte viability to maintain mechanical competence and fracture resistance of bone in elderly individuals.  相似文献

This paper investigates Swedish recycling centres from the users' perspective. The aim was to describe the characteristics and experiences of the users and their activities when sorting and disposing of waste, and to identify improvements for the users. The typical recycling centre user is a recently retired man, living in a house with a garden, having travelled 5 km alone in his own car. The users requested longer opening hours and better information available at home and at the recycling centre. The major difficulty for the users is to understand which fraction their waste belongs to, and consequently into which container they should throw it. The most important sources of sorting information, in addition to experience from earlier visits, are signs and asking the personnel. Although the service at recycling centres is perceived positively by a majority of users, substantial improvements can still be made, and a number of such suggestions are given.  相似文献

In this installment of Semantics and Services, we further develop the idea of spatial, temporal, and thematic (STT) processing of semantic Web data and describe the Web infrastructure needed to support it. 23 highly hypnotizable undergraduates (Harvard Group Scale of Hypnotic Susceptibility) underwent 2 specially constructed 7-item hypnotic inductions. Over the 14 items, the main finding was of a relationship between E. R. Hilgard's (1973, 1977, 1979) "hidden observer" effect and 2 aspects of hypnotic age regression. Ss reporting a hidden observer experienced duality during regression to age 5, in which they were aware of being both adult and child. When asked to write a complex sentence, most did so, usually without spelling errors. The Ss not reporting this effect experienced quasi-lateral age regression in which they had the exclusive feeling of being 5 yrs old, with no sense of an adult identity. Most of these Ss were unable to write the same complex sentence when requested to during age regression. The study replicated Hilgard's finding of the hidden observer phenomenon in terms of its incidence and obtained similar verbal reports from Ss experiencing it. At the same time, the results suggest that a neodissociation account of hypnosis may need some modifications to accommodate these additional findings. Upper-, middle-, and lower-class parents were asked to explain a proverb which had both "growth" and "stability" interpretations to their 11- and 16-yr-old sons. The growth interpretation was overwhelmingly endorsed by most parents. With 16-yr-olds, lower-middle-class parents continued to endorse the growth interpretation; most upper-class parents endorsed the stability interpretation. A selenium depletion diet (.025 ppm selenium) was fed to two male Holstein calves for its effect on concentrations of selenium in serum. By 90 days, selenium in serum was depressed from .022 ppm to .013 ppm, and one of two calves had died of "possible white muscle disease." Repletion was rapid in that a diet containing .200 ppm selenium increased selenium in serum to .028 ppm within 2 wk. Dietary .280 ppm selenium increased it to .051 ppm in 1 wk. Eleven cows and eleven calves showed averages of .030 ppm in serum (range of .013 to .051 ppm) fed a diet of corn silage (.039 ppm) and supplement (.090 ppm). A sampling of feedstuffs showed a wide range of selenium among feedstuffs (.023 for corn cobs to 2.663 ppm for dried egg albumin) and a wide range within one type feedstuff (shelled corn, .017 to .219 ppm).  相似文献

The fine structure and cellular associations of the large pigment cells (LPC's) of the compound eye of the house fly were studied with high voltage and conventional electron microscopy. Depending on the sector of the compound eye, the facets are either rectangular or hexagonal. The underside of each facet has indentations exactly aligned with those on top into which inserts an angulated sleeve of LPC's. Under the rectangular lens facet 6 or 8 small compact (in cross section) LPC's join four elongate LPC's. Clusters of compact cells alternate in this ring with elongate ones. Compact cells compress together and become quadrangular (in cross section) several microns below their insertion into the lens and form "building block" corners while elongate cells form "side rails" for the rectangular type of distal pseudocone enclosure. Beneath hexagonal facets all LPC's are rather elongate with out corner cells. In both facet types LPC's enclose the pseudocone for a longitudinal distance of 4 mum and then are displaced as bordering cells by a sleeve of two corneal pigment cells (CPC's), each of which encloses half of the proximal pseudocone. For the following 6 mum of longitudinal distance these concentric sleeves of CPC's and LPC's form a double layer around the pseudocone. At about 10 mum below lens base the two sleeves separate; LPC's become attenuated and extend cable-like to the basement membrane and CPC's enclose the proximal pseudocone, Semper cells and distal retinula. The junction between lens and LPC's has critical structural value in that (1) this is the sole anchorage to the lens by the lengthy remainder of the ommatidium, and (2) LPC's enclose the semiliquid pseudocone in the most distal portion of the pseudocone. In addition to vertical support, the LPC's send out numerous lateral processes that make structural contact among themselves, with the corneal pigment cells and the photoreceptor cells. The structural features of this array are discussed relative to possible physiological roles.  相似文献

We give a distributed approximation algorithm for job scheduling in a ring architecture. In contrast to many other parallel scheduling models, the model we consider captures the influence of the underlying communications network by specifying that task migration from one processor to another takes time proportional to the distance between those two processors in the network. As a result, our algorithm must balance computational load and communication time. The algorithm is simple, requires no global control, and yields schedules of length at most 4.22 times optimal. We also give a lower bound on the performance of any distributed algorithm and the results of simulation experiments which suggest better performance than does our worst-case analysis.  相似文献

A disintegrin and metalloproteinase (ADAM) family proteins, acting as sheddases, are important factors in a number of pathologies, including cancer, and have been suggested as promising therapeutic targets. The study presented focuses on the involvement of ADAM10 and ADAM17 in retinoblastoma (RB), the most common malignant intraocular childhood tumor. A significant correlation between ADAM17 expression levels and RB laterality and RB staging was observed. Levels of ADAM10 or ADAM17 regulating miRNAs miR-145, -152, and -365 were significantly downregulated in RB cell lines, and reduced miR levels with simultaneously upregulated ADAM10 and ADAM17 expression were found in RB patients. The involvement of both ADAMs analyzed in ectodomain shedding of the neuronal cell adhesion molecule L1 (L1CAM), shown to induce pro-tumorigenic effects in RB, was confirmed. Lentiviral ADAM10 and ADAM17 single or ADAM10/17 double knockdown (KD) induced caspase-dependent apoptosis and reduced cell viability, proliferation, growth, and colony formation capacity of RB cells. Moreover, differential phosphorylation of the serine/threonine kinase AKT was observed following ADAM17 KD in RB cells. Chicken chorioallantoic membrane (CAM) assays revealed that ADAM17 and ADAM10/17 depletion decreases the tumorigenic and migration potential of RB cells in vivo. Thus, ADAMs are potential novel targets for future therapeutic RB approaches.  相似文献
