An enzyme-linked immunometric assay for cortisol based on idiotype-anti-idiotype reactions

Cortisol levels in body fluids are useful for monitoring the function of the pituitary-adrenal axis. Here, we established an “enzyme-linked immunometric assay” (a noncompetitive-type ELISA) for cortisol based on idiotype-anti-idiotype reactions. Six different anti-idiotype monoclonal antibodies that recognized the variable regions of a newly established anti-cortisol antibody were generated using hybridoma technology; these were two β-type and four α-type anti-idiotype antibodies, recognizing the paratope and framework regions, respectively. An immunometric assay was established using a combination of a selected α-type and a selected β-type antibody. The analyte (cortisol) was captured by an excess amount of anti-cortisol antibody immobilized on microplates, and the unoccupied paratope was saturated with the β-type antibody. Hapten-occupied anti-cortisol antibody, with less steric hindrance, was then selectively bound by the α-type antibody, labeled with biotin. The amount of biotin residue on the microplates was colorimetrically monitored using a peroxidase-labeled streptavidin. This assay had an approximately threefold higher sensitivity (detection limit: 90 pg = 248 fmol cortisol) than a competitive ELISA using the same anti-cortisol antibody, as well as a practical specificity for providing reasonable determination of normal urinary cortisol levels.     

Organic high-spin systems: Synthesis,electrochemical and spectroscopic studies of π-extended tetraaryl m-phenylenediamines

Three highly π-extended tetraaryl-1,3-phenylenediamines for charged high-spin molecular systems were successfully synthesized using Suzuki coupling. Their electrochemical and spectroscopic properties were also examined. They showed blue fluorescence. The dications derived from highly π-extended tetraaryl-1,3-phenylenediamines were generated and characterized by electron transfer stopped-flow method.     

The anomalous behaviour of Gd atoms (Fe1−xMnx)2 compounds

The educed Gd atoms in the X-structure (Th₆Mn₂₃-type) of Gd(Fe_1−xMn_x)₂ were magnetically investigated by comparing with Gd₆(Fe_1−yMn_y)₂₃ whose structure is Th₆Mn₂₃-type. The magnetic properties of Gd(Fe_1−xMn_x)₂ (0.4≤x≤0.7) were observed to be quite similar to those of Gd₆(Fe_1−yMn_y)₂₃ (0.4≤y≤0.7).     

Simulation of flux dynamics in a superconducting bulk magnetized by multi-pulse technique

We have simulated the time and spatial dependence of local field B_z(t, r) and temperature T(t, r) on the superconducting bulk during pulsed field magnetization (PFM) using the finite element method (FEM). A modified multi-pulse technique with step-wise cooling (MMPSC) was performed to the cryo-cooled bulk, which was experimentally confirmed to the effective PFM technique to enhance the trapped field B_z higher than 5 T. In the simulation, the B_z value at the center of the bulk surface was enhanced at the 2nd stage of the MMPSC method, in which the results of the simulation reproduced the experimental ones. The enhancement of B_z results from the reduction in the temperature because of the already trapped flux in the bulk at the 1st stage of the MMPSC method.     

Magnetic behavior of Fe0.2Mg0.8TiO3 and Fe0.3Mg0.7TiO3 probed by Mössbauer timescale

Magnetization and Mössbauer measurements have been made on a diluted antiferromagnets Fe_xMg_1?xTiO₃ with x=0.2, 0.3 and 0.7. It has been demonstrated that this system exhibits a spin glass or a reentrant spin glass like behavior in the samples with x around the percolation concenrration x_c≈0.25. It has been shown that various sizes of magnetic clusters are formed well above the transition temperatures determined by the magnetization measurements in the samples with x near x_c. The behavior of Fe_0.2Mg_0.8TiO₃ is discussed as a typical cluster glass.     

Stereospecific synthesis and redox properties of ruthenium(II) carbonyl complexes bearing a redox-active 1,8-naphthyridine unit

Two stereoisomers of cis-[Ru(bpy)(pynp)(CO)Cl]PF₆ (bpy = 2,2′-bipyridine, pynp = 2-(2-pyridyl)-1,8-naphthyridine) were selectively prepared. The pyridyl rings of the pynp ligand in [Ru(bpy)(pynp)(CO)Cl]⁺ are situated trans and cis, respectively, to the CO ligand. The corresponding CH₃CN complex ([Ru(bpy)(pynp)(CO)(CH₃CN)]²⁺) was also prepared by replacement reactions of the chlorido ligand in CH₃CN. Using these complexes, ligand-centered redox behavior was studied by electrochemical and spectroelectrochemical techniques. The molecular structures of pynp-containing complexes (two stereoisomers of [Ru(bpy)(pynp)(CO)Cl]PF₆ and [Ru(pynp)₂(CO)Cl]PF₆) were determined by X-ray structure analyses.     

Magnetic circular dichroism in the A,B and C absorption bands of KBr: In+

The magnetic circular dichroism in the A, B and C bands of KBr:In⁺ has been measured as a function of temperature between 4 K and 260 K. Results for the A band agree with those reported previously at one temperature only. Our experimental data on the B and C bands are compared with recent theoretical calculations.     

Characterization of polymer-coated silica particles by microelectrophoresis

Electrophoretic mobility measurements have been used to characterize monodispersed colloidal particles of silica, silica coated with alumina (cores), of these cores incorporating a dye (pigments), and finally of pigments coated with polymers. The latter consisted of poly(divinylbenzene), of poly(vinylbenzyl chloride), and of their copolymers, synthesized directly on the core or pigment particles, with and without subsequent sulfonation.     

Thermal and photochemical reactions of a stable thiobenzaldehyde, 2,4,6-tri-t-butylthiobenzaldehyde

Thermal and photochemical reactions of the title compound gave benzothiolane $\text{2}$ in high yield; the reaction with free radicals also afforded $\text{2}$ along with some other products.
