With more than 25 million people affected, heart failure (HF) is a global threat. As energy production pathways are known to play a pivotal role in HF, we sought here to identify key metabolic changes in ischemic- and non-ischemic HF by using a multi-OMICS approach. Serum metabolites and mRNAseq and epigenetic DNA methylation profiles were analyzed from blood and left ventricular heart biopsy specimens of the same individuals. In total we collected serum from n = 82 patients with Dilated Cardiomyopathy (DCM) and n = 51 controls in the screening stage. We identified several metabolites involved in glycolysis and citric acid cycle to be elevated up to 5.7-fold in DCM (p = 1.7 × 10−6). Interestingly, cardiac mRNA and epigenetic changes of genes encoding rate-limiting enzymes of these pathways could also be found and validated in our second stage of metabolite assessment in n = 52 DCM, n = 39 ischemic HF and n = 57 controls. In conclusion, we identified a new set of metabolomic biomarkers for HF. We were able to identify underlying biological cascades that potentially represent suitable intervention targets.  相似文献

Deformation due to two different surface-machining conditions—grinding (126 μm diamond) and polishing (3 μm diamond)—in an uniaxial hot-pressed Al2O3–30%-SiC-whisker composite has been investigated. A Warren–Averbach analysis of grazing incidence X-ray diffractometry data shows that the deformation is localized to the very top surface zone. The cell size and the root mean square of the strain show a gradient in the deformed layer. Transmission electron microscopy studies, in cross-sectional view, also show a near-surface deformation zone containing dislocations, twins, and cracks. This is seen for both machining procedures, but the depth of the zone and the degree of deformation, in terms of dislocation density and number of cracks, is much higher in the roughly ground specimen than in the polished one. For comparison, a monolithic Al2O3 sample also has been studied after grinding. The deformation zone is very similar to the Al2O3–SiC sample with the same grinding condition, but cracks and dislocations are present at a slightly larger depth. The deformation depth for the polished Al2O3–SiC sample is ∼50 nm. In the ground Al2O3–SiC sample, the deformation depth is 1–1.5 μm and corresponds to the grain size. The deformation zone in the ground monolithic Al2O3 sample is 1.5–2 μm deep. The observed grain-boundary cracks are almost parallel to the surface and may originate from nonaccommodated plastic flow between grains.  相似文献

This paper presents a system enabling robotic helicopters to fly safely without user interaction at low altitude over unknown terrain with static obstacles. The system includes a novel reactive behavior‐based method that guides rotorcraft reliably to specified locations in sparsely occupied environments. System dependability is, among other things, achieved by utilizing proven system components in a component‐based design and incorporating safety margins and safety modes. Obstacle and terrain detection is based on a vertically mounted off‐the‐shelf two‐dimensional LIDAR system. We introduce two flight modes, pirouette descent and waggle cruise, which extend the field of view of the sensor by yawing the aircraft. The two flight modes ensure that all obstacles above a minimum size are detected in the direction of travel. The proposed system is designed for robotic helicopters with velocity and yaw control inputs and a navigation system that provides position, velocity, and attitude information. It is cost effective and can be easily implemented on a variety of helicopters of different sizes. We provide sufficient detail to facilitate the implementation on single‐rotor helicopters with a rotor diameter of approximately 1.8 m. The system was extensively flight‐tested in different real‐world scenarios in Queensland, Australia. The tests included flights beyond visual range without a backup pilot. Experimental results show that it is feasible to perform dependable autonomous flight using simple but effective methods. © 2013 Wiley Periodicals, Inc.  相似文献

In this paper, we extend the single relaxation time Lattice-Boltzmann Method (LBM) to the 3D body-centered cubic (BCC) lattice. We show that the D3bQ15 lattice defined by a 15 neighborhood connectivity of the BCC lattice is not only capable of more accurately discretizing the velocity space of the continuous Boltzmann equation as compared to the D3Q15 Cartesian lattice, it also achieves a comparable spatial discretization with 30 percent less samples. We validate the accuracy of our proposed lattice by investigating its performance on the 3D lid-driven cavity flow problem and show that the D3bQ15 lattice offers significant cost savings while maintaining a comparable accuracy. We demonstrate the efficiency of our method and the impact on graphics and visualization techniques via the application of line-integral convolution on 2D slices as well as the extraction of streamlines of the 3D flow. We further study the benefits of our proposed lattice by applying it to the problem of simulating smoke and show that the D3bQ15 lattice yields more detail and turbulence at a reduced computational cost.  相似文献

The Body-Centered Cubic (BCC) and Face-Centered Cubic (FCC) lattices have been analytically shown to be more efficient sampling lattices than the traditional Cartesian Cubic (CC) lattice, but there has been no estimate of their visual comparability. Two perceptual studies (each with N = 12 participants) compared the visual quality of images rendered from BCC and FCC lattices to images rendered from the CC lattice. Images were generated from two signals: the commonly used Marschner-Lobb synthetic function and a computed tomography scan of a fish tail. Observers found that BCC and FCC could produce images of comparable visual quality to CC, using 30-35 percent fewer samples. For the images used in our studies, the L(2) error metric shows high correlation with the judgement of human observers. Using the L(2) metric as a proxy, the results of the experiments appear to extend across a wide range of images and parameter choices.  相似文献

In this paper, we develop an interactive analysis and visualization tool for probabilistic segmentation results in medical imaging. We provide a systematic approach to analyze, interact and highlight regions of segmentation uncertainty. We introduce a set of visual analysis widgets integrating different approaches to analyze multivariate probabilistic field data with direct volume rendering. We demonstrate the user's ability to identify suspicious regions (e.g. tumors) and correct the misclassification results using a novel uncertainty‐based segmentation editing technique. We evaluate our system and demonstrate its usefulness in the context of static and time‐varying medical imaging datasets.  相似文献

In this paper we present a novel anisotropic diffusion model targeted for 3D scalar field data. Our model preserves material boundaries as well as fine tubular structures while noise is smoothed out. One of the major novelties is the use of the directional second derivative to define material boundaries instead of the gradient magnitude for thresholding. This results in a diffusion model that has much lower sensitivity to the diffusion parameter and smoothes material boundaries consistently compared to gradient magnitude based techniques. We empirically analyze the stability and convergence of the proposed diffusion and demonstrate its de-noising capabilities for both analytic and real data. We also discuss applications in the context of volume rendering.  相似文献
