Stimulated emission depletion (STED) microscopy enables ultrastructural imaging of biological samples with high spatiotemporal resolution. STED nanoprobes based on fluorescent organosilica nanohybrids featuring sub-2 nm size and near-unity quantum yield are presented. The spin–orbit coupling (SOC) of heavy-atom-rich organic fluorophores is mitigated through a silane-molecule-mediated condensation/dehalogenation process, resulting in bright fluorescent organosilica nanohybrids with multiple emitters in one hybrid nanodot. When harnessed as STED nanoprobes, these fluorescent nanohybrids show intense photoluminescence, high biocompatibility, and long-term photostability. Taking advantage of the low-power excitation (0.5 μW), prolonged singlet-state lifetime, and negligible depletion-induced re-excitation, these STED nanohybrids present high depletion efficiency (>96 %), extremely low saturation intensity (0.54 mW, ca. 0.188 MW cm−2), and ultra-high lateral resolution (ca. λem/28).  相似文献

Far‐red emitting fluorescent dyes for optical microscopy, stimulated emission depletion (STED), and ground‐state depletion (GSDIM) super‐resolution microscopy are presented. Fluorinated silicon–rhodamines (SiRF dyes) and phosphorylated oxazines have absorption and emission maxima at about λ≈660 and 680 nm, respectively, possess high photostability, and large fluorescence quantum yields in water. A high‐yielding synthetic path to introduce three aromatic fluorine atoms and unconventional conjugation/solubilization spacers into the scaffold of a silicon–rhodamine is described. The bathochromic shift in SiRF dyes is achieved without additional fused rings or double bonds. As a result, the molecular size and molecular mass stay quite small (<600 Da). The use of the λ=800 nm STED beam instead of the commonly used one at λ=750–775 nm provides excellent imaging performance and suppresses re‐excitation of SiRF and the oxazine dyes. The photophysical properties and immunofluorescence imaging performance of these new far‐red emitting dyes (photobleaching, optical resolution, and switch‐off behavior) are discussed in detail and compared with those of some well‐established fluorophores with similar spectral properties.  相似文献

The development of stimulated emission depletion (STED) microscopy represented a major breakthrough in cellular and molecular biology. However, the intense laser beams required for both excitation and STED usually provoke rapid photobleaching of fluorescent molecular probes, which significantly limits the performance and practical utility of STED microscopy. We herein developed a photoresistant fluorescent dye C‐Naphox as a practical tool for STED imaging. With excitation using either a λ=405 or 488 nm laser in protic solvents, C‐Naphox exhibited an intense red/orange fluorescence (quantum yield ΦF>0.7) with a large Stokes shift (circa 5900 cm?1). Even after irradiation with a Xe lamp (300 W, λex=460 nm, full width at half maximum (FWHM)=11 nm) for 12 hours, 99.5 % of C‐Naphox remained intact. The high photoresistance of C‐Naphox allowed repeated STED imaging of HeLa cells. Even after recording 50 STED images, 83 % of the initial fluorescence intensity persisted.  相似文献

Photostable and bright fluorescent dyes with large Stokes shifts are widely used as markers in far‐field optical microscopy, but the variety of useful dyes is limited. The present study introduces new 3‐heteroaryl coumarins decorated with a primary phosphate group (OP(O)(OH)2) attached to C‐4 in 2,2,4‐trimethyl‐1,2‐dihydroquinoline fragment fused with the coumarin fluorophore. The general synthetic route is based on the Suzuki reaction of 3‐bromocoumarines with hetarylboronic acids followed by oxidation of the methyl group at the C?C bond with SeO2 (to an aldehyde), reduction with NaBH4 (to an alcohol), and conversion into a primary phosphate. The 4 position in the coumarin system may be unsubstituted or bear a methyl group. Phosphorylated coumarins were found to have high fluorescence quantum yields in the free state and after conjugation with proteins (in aqueous buffers). In super‐resolution light microscopy with stimulated emission depletion (STED), the new coumarin dyes provide an optical resolution of 40–60 nm with a low background signal. Due to their large Stokes shifts and high photostability, phosphorylated coumarins enable to combine multilabel imaging (using one detector and several excitation sources) with diffraction unlimited optical resolution.  相似文献

Morphology evolution of sulfonic acid functionalized organosilica nanohybrids (Si(Et)Si‐Pr/ArSO3H) with a 1D tubular structure (inner diameter of ca. 5 nm), a 2D hexagonal mesostructure (pore diameter of ca. 5 nm), and a 3D hollow spherical structure (shell thickness of 2–3 nm and inner diameter of ca. 15 nm) was successfully realized through P123‐templated sol–gel cocondensation strategies and fine‐tuning of the acidity followed by aging or a hydrothermal treatment. The Si(Et)Si‐Pr/ArSO3H nanohybrids were applied in synthesis of alkyl levulinates from the esterification of levulinic acid and ethanolysis of furfural alcohol. Hollow spherical Si(Et)Si‐Pr/ArSO3H and hexagonal mesoporous analogues exhibited the highest and lowest catalytic activity, respectively, among three types of nanohybrids; additionally, the activity was influenced by the ?SO3H loading. The activity differences are explained in terms of different Brønsted acid and textural properties, reactant/product diffusion, and mass transfer rate, as well as accessibility of ?SO3H sites to the reactant molecules. The reusability of the nanohybrids was also evaluated.  相似文献

Lanthanide‐doped upconversion nanoparticles (UCNPs) have shown great promise in versatile bioapplications. For the first time, organosilica‐shelled β‐NaLuF4:Gd/Yb/Er nanoprobes with a rattle structure have been designed for dual‐modal imaging and photodynamic therapy (PDT). Benefiting from the unique rattle structure and aromatic framework, these nanoprobes are endowed with a high loading capacity and the disaggregation effect of photosensitizers. After loading of β‐carboxyphthalocyanine zinc or rose Bengal into the nanoprobes, we achieved higher energy transfer efficiency from UCNPs to photosensitizers as compared to those with conventional core–shell structure or with pure‐silica shell, which facilitates a large production of singlet oxygen and thus an enhanced PDT efficacy. We demonstrated the use of these nanoprobes in proof‐of‐concept X‐ray computed tomography (CT) and UC imaging, thus revealing the great potential of this multifunctional material as an excellent nanoplatform for cancer theranostics.  相似文献

We recently reported that fluorescent dye PB430, which consisted of a 2‐phenyl‐substituted benzophosphole P‐oxide skeleton that was reinforced by a methylene bridge, showed pronounced photostability and, thus, high utility for applications in super‐resolution stimulated emission depletion (STED) microscopy. Herein, we replaced the methylene bridge with another P=O group to 1) investigate the role of the bridging moieties; and 2) further modulate the fluorescence properties of this skeleton. We synthesized a series of phospholo[3,2‐b]phosphole‐based dyes—trans‐PO‐PB430, cis‐PO‐PB430, and trans‐PO‐PB460—all of which showed sufficient water solubility. Moreover, trans‐PO‐PB430 and trans‐PO‐PB460 exhibited intense green and orange fluorescence, respectively, and a high photostability that was comparable to that of PB430. In contrast, cis‐PO‐PB430 underwent rapid photobleaching upon continuous photoirradiation, which demonstrated the importance of steric shielding of the polycyclic skeleton by the substituents on the bridging moieties. The fluorescence properties of these dyes were insensitive to concentration, pH value, and polarity changes of the environment in solution. In addition, even in the solid state, these dyes showed strong green to orange emissions. These results demonstrate the potential utility of trans‐PO‐PB430 and trans‐PO‐PB460 as highly photostable fluorescent dyes.  相似文献

Exploration of low‐cost and earth‐abundant photocatalysts for highly efficient solar photocatalytic water splitting is of great importance. Although transition‐metal dichalcogenides (TMDs) showed outstanding performance as co‐catalysts for the hydrogen evolution reaction (HER), designing TMD‐hybridized photocatalysts with abundant active sites for the HER still remains challenge. Here, a facile one‐pot wet‐chemical method is developed to prepare MS2–CdS (M=W or Mo) nanohybrids. Surprisedly, in the obtained nanohybrids, single‐layer MS2 nanosheets with lateral size of 4–10 nm selectively grow on the Cd‐rich (0001) surface of wurtzite CdS nanocrystals. These MS2–CdS nanohybrids possess a large number of edge sites in the MS2 layers, which are active sites for the HER. The photocatalytic performances of WS2–CdS and MoS2–CdS nanohybrids towards the HER under visible light irradiation (>420 nm) are about 16 and 12 times that of pure CdS, respectively. Importantly, the MS2–CdS nanohybrids showed enhanced stability after a long‐time test (16 h), and 70 % of catalytic activity still remained.  相似文献

Drug toxicity is a long‐standing concern of modern medicine. A typical anti‐pain/fever drug paracetamol often causes hepatotoxicity due to peroxynitrite ONOO. Conventional blood tests fail to offer real‐time unambiguous visualization of such hepatotoxicity in vivo. Here we report a luminescent approach to evaluate acute hepatotoxicity in vivo by chromophore‐conjugated upconversion nanoparticles. Upon injection, these nanoprobes mainly accumulate in the liver and the luminescence of nanoparticles remains suppressed owing to energy transfer to the chromophore. ONOO can readily bleach the chromophore and thus recover the luminescence, the presence of ONOO in the liver leads to fast restoring of the near‐infrared emission. Taking advantages of the high tissue‐penetration capability of near‐infrared excitation/emission, these nanoprobes achieve real‐time monitoring of hepatotoxicity in living animals, thereby providing a convenient screening strategy for assessing hepatotoxicity of synthetic drugs.  相似文献

Low‐cost transparent counter electrodes (CEs) for efficient dye‐sensitized solar cells (DSSCs) are prepared by using nanohybrids of carbon nanotube (CNT)‐supported platinum nanoparticles as highly active catalysts. The nanohybrids, synthesized by an ionic‐liquid‐assisted sonochemical method, are directly deposited on either rigid glass or flexible plastic substrates by a facile electrospray method for operation as CEs. Their electrochemical performances are examined by cyclic voltammetry, current density–voltage characteristics, and electrochemical impedance spectroscopy (EIS) measurements. The CNT/Pt hybrid films exhibit high electrocatalytic activity for I?/I3? with a weak dependence on film thickness. A transparent CNT/Pt hybrid CE film about 100 nm thick with a transparency of about 70 % (at 550 nm) can result in a high power conversion efficiency (η) of over 8.5 %, which is comparable to that of pyrolysis platinum‐based DSSCs, but lower cost. Furthermore, DSSC based on flexible CNT/Pt hybrid CE using indium‐doped tin oxide‐coated polyethylene terephthalate as the substrate also exhibits η=8.43 % with Jsc=16.85 mA cm?2, Voc=780 mV, and FF=0.64, and this shows great potential in developing highly efficient flexible DSSCs.  相似文献

By using pentyl‐linked bis(rhodamine)‐derived tetra‐siloxane (PRh‐Si4) as the organosilica precursor, highly ordered PRh‐bridged periodic mesoporous organosilicas (PRhPMOs) were prepared. When excited at λ=500 nm, the PRhPMO suspension that contained metal ions showed two separate emission peaks at λ=550 and 623 nm. The first peak, located at λ=550 nm, was due to ring‐opening of the spiro structure in the rhodamine moiety and the second, located at λ=623 nm, originated from fluorescent aggregates of the PRh units embedded in the silica framework of the PRhPMO. By using the different intensity ratios of the two fluorescence signals (FI550/623), PRhPMOs could be used as turn‐ON type fluorescent ratiometric chemosensors for Cu2+. Furthermore, based on the single‐exciton theory, it was deduced that the fluorescent aggregates formed were of the J‐type and had a coplanar configuration. Consequently, PRhPMOs display a longer fluorescence lifetime and greater fluorescent quantum yield than the respective monomers dissolved in solution, which is consistent with the experimental results.  相似文献

In this study, surface‐functionalized, branched polyethylenimine (BPEI)‐modified YVO4:Bi3+,Eu3+ nanocrystals (NCs) were successfully synthesized by a simple, rapid, solvent‐free hydrothermal method. The BPEI‐coated YVO4:Bi3+,Eu3+ NCs with high crystallinity show broad‐band excitation in the λ=250 to 400 nm near‐ultraviolet (NUV) region and exhibit a sharp‐line emission band centered at λ=619 nm under excitation at λ=350 nm. The surface amino groups contributed by the capping agent, BPEI, not only improve the dispersibility and water/buffer stability of the BPEI‐coated YVO4:Bi3+,Eu3+ NCs, but also provide a capability for specifically targeted biomolecule conjugation. Folic acid (FA) and epidermal growth factor (EGF) were further attached to the BPEI‐coated YVO4:Bi3+,Eu3+ NCs and exhibited effective positioning of fluorescent NCs toward the targeted folate receptor overexpressed in HeLa cells or EGFR overexpressed in A431 cells with low cytotoxicity. These results demonstrate that the ligand‐functionalized, BPEI‐coated YVO4:Bi3+, Eu3+ NCs show great potential as a new‐generation biological luminescent bioprobe for bioimaging applications. Moreover, the unique luminescence properties of BPEI‐coated YVO4:Bi3+,Eu3+ NCs show potential to combine with a UVA photosensitizing drug to produce both detective and therapeutic effects for human skin cancer therapy.  相似文献

Well‐defined poly(vinyl acetate) (PVAc) and poly(N‐vinylpyrrolidone)‐co‐poly(vinyl acetate) (PNVP‐co‐PVAc) chains end‐capped by Co(acac)2 (acac=acetylacetonate) and prepared by cobalt‐mediated radical polymerization (CMRP) are grafted onto a fullerene. Homolytic Co? C bond cleavage of the polymer chain ends at 30 °C releases the polymeric radicals that add onto C60, thereby leading to the corresponding PVAc/C60 and PNVP‐co‐PVAc/C60 nanohybrids. The [polymer–Co(acac)2]/[C60] molar ratio was varied to adjust the structure of the nanohybrids, and more particularly the number of grafted arms. Finally, the potential of the hydrosoluble PVOH/C60 nanohybrids, which result from the methanolysis of the ester groups of PVAc/C60, and of the PNVP‐co‐PVAc/C60 nanohybrids as photosensitizers for photodynamic therapy (PDT), was approached. First, photobleaching tests demonstrated the ability of these nanohybrids to produce singlet oxygen upon irradiation, which can play a role in cell damage. Second, cell viability assays demonstrated that both types of nanohybrids are deprived of intrinsic cytotoxicity in the dark, whereas they promoted significant cell mortality when subjected to light treatment. The selective response of these materials to irradiation makes them promising compounds for PDT.  相似文献

Three new anthracene derivatives [2‐chloro‐9,10‐dip‐tolylanthracene (DTACl), 9,10‐dip‐tolylanthracene‐2‐carbonitrile (DTACN), and 9,10‐di(naphthalen‐1‐yl)anthracene‐2‐carbonitrile (DNACN)] were synthesized as triplet acceptors for low‐power upconversion. Their linear absorption, single‐photon‐excited fluorescence, and upconversion fluorescence properties were studied. The acceptors exhibit high fluorescence yields in DMF. Selective excitation of the sensitizer PdIIoctaethylporphyrin (PdOEP) in solution containing DTACl, DTACN, or DNA‐CN at 532 nm with an ultralow excitation power density of 0.5 W cm?2 results in anti‐Stokes blue emission. The maximum upconversion quantum yield (ΦUC=17.4 %) was obtained for the couple PdOEP/DTACl. In addition, the efficiency of the triplet–triplet energy transfer process was quantitatively studied by quenching experiments. Experimental results revealed that a highly effective acceptor for upconversion should combine high fluorescence quantum yields with efficient quenching of the sensitizer triplet.  相似文献

An organic‐based photocatalysis system for water oxidation, with visible‐light harvesting antennae, was constructed using periodic mesoporous organosilica (PMO). PMO containing acridone groups in the framework (Acd‐PMO), a visible‐light harvesting antenna, was supported with [RuII(bpy)32+] complex (bpy=2,2′‐bipyridyl) coupled with iridium oxide (IrOx) particles in the mesochannels as photosensitizer and catalyst, respectively. Acd‐PMO absorbed visible light and funneled the light energy into the Ru complex in the mesochannels through excitation energy transfer. The excited state of Ru complex is oxidatively quenched by a sacrificial oxidant (Na2S2O8) to form Ru3+ species. The Ru3+ species extracts an electron from IrOx to oxidize water for oxygen production. The reaction quantum yield was 0.34 %, which was improved to 0.68 or 1.2 % by the modifications of PMO. A unique sequence of reactions mimicking natural photosystem II, 1) light‐harvesting, 2) charge separation, and 3) oxygen generation, were realized for the first time by using the light‐harvesting PMO.  相似文献

A functionalized periodic mesoporous organosilica with incorporated chiral bis(cyclohexyldiamine)‐based NiII complexes within the silica framework was developed by the co‐condensation of (1R,2R)‐cyclohexyldiamine‐derived silane and ethylene‐bridge silane, followed by the complexation of NiBr2 in the presence of (1R,2R)‐N,N′‐dibenzylcyclohexyldiamine. Structural characterization by XRD, nitrogen sorption, and TEM disclosed its orderly mesostructure, and FTIR and solid‐state NMR spectroscopy demonstrated the incorporation of well‐defined single‐site bis(cyclohexyldiamine)‐based NiII active centers within periodic mesoporous organosilica. As a chiral heterogeneous catalyst, this functionalized periodic mesoporous organosilica showed high catalytic activity and excellent enantioselectivity in the asymmetric Michael addition of 1,3‐dicarbonyl compounds to nitroalkenes, comparable to those with homogeneous catalysts. In particular, this heterogeneous catalyst could be recovered easily and reused repeatedly up to nine times without obviously affecting its enantioselectivity, thus showing good potential for industrial applications.  相似文献

The synthesis of the C2‐symmetrical ligand 1 consisting of two naphthalene units connected to two pyridine‐2,6‐dicarboxamide moieties linked by a xylene spacer and the formation of LnIII‐based (Ln=Sm, Eu, Tb, and Lu) dimetallic helicates [Ln2? 1 3] in MeCN by means of a metal‐directed synthesis is described. By analyzing the metal‐induced changes in the absorption and the fluorescence of 1 , the formation of the helicates, and the presence of a second species [Ln2? 1 2] was confirmed by nonlinear‐regression analysis. While significant changes were observed in the photophysical properties of 1 , the most dramatic changes were observed in the metal‐centred lanthanide emissions, upon excitation of the naphthalene antennae. From the changes in the lanthanide emission, we were able to demonstrate that these helicates were formed in high yields (ca. 90% after the addition of 0.6 equiv. of LnIII), with high binding constants, which matched well with that determined from the changes in the absorption spectra. The formation of the LuIII helicate, [Lu2? 1 3], was also investigated for comparison purposes, as we were unable to obtain accurate binding constants from the changes in the fluorescence emission upon formation of [Sm2? 1 3], [Eu2? 1 3], and [Tb2? 1 3].  相似文献

Seven derivatives of 1,2‐dicarbadodecaborane (ortho‐carborane, 1,2‐C2B10H12) with a 1,3‐diethyl‐ or 1,3‐diphenyl‐1,3,2‐benzodiazaborolyl group on one cage carbon atom were synthesized and structurally characterized. Six of these compounds showed remarkable low‐energy fluorescence emissions with large Stokes shifts of 15100–20260 cm?1 and quantum yields (ΦF) of up to 65 % in the solid state. The low‐energy fluorescence emission, which was assigned to a charge‐transfer (CT) transition between the cage and the heterocyclic unit, depended on the orientation (torsion angle, ψ) of the diazaborolyl group with respect to the cage C? C bond. In cyclohexane, two compounds exhibited very weak dual fluorescence emissions with Stokes shifts of 15660–18090 cm?1 for the CT bands and 1960–5540 cm?1 for the high‐energy bands, which were assigned to local transitions within the benzodiazaborole units (local excitation, LE), whereas four compounds showed only CT bands with ΦF values between 8–32 %. Two distinct excited singlet‐state (S1) geometries, denoted S1(LE) and S1(CT), were observed computationally for the benzodiazaborolyl‐ortho‐carboranes, the population of which depended on their orientation (ψ). TD‐DFT calculations on these excited state geometries were in accord with their CT and LE emissions. These C‐diazaborolyl‐ortho‐carboranes were viewed as donor–acceptor systems with the diazaborolyl group as the donor and the ortho‐carboranyl group as the acceptor.  相似文献

By employing the subcomponent self‐assembly approach utilizing 5,10,15,20‐tetrakis(4‐aminophenyl)porphyrin or its zinc(II) complex, 1H ‐4‐imidazolecarbaldehyde, and either zinc(II) or iron(II) salts, we were able to prepare O‐symmetric cages having a confined volume of ca. 1300 Å3. The use of iron(II) salts yielded coordination cages in the high‐spin state at room temperature, manifesting spin‐crossover in solution at low temperatures, whereas corresponding zinc(II) salts led to the corresponding diamagnetic analogues. The new cages were characterized by synchrotron X‐ray crystallography, high‐resolution mass spectrometry, and NMR, Mössbauer, IR, and UV/Vis spectroscopy. The cage structures and UV/Vis spectra were independently confirmed by state‐of‐the‐art DFT calculations. A remarkably high‐spin‐stabilizing effect through encapsulation of C70 was observed. The spin‐transition temperature T 1/2 is lowered by 20 K in the host–guest complex.  相似文献

Significant charge recombination that is difficult to suppress limits the practical applications of hematite (α‐Fe2O3) for photoelectrochemical water splitting. In this study, Ti‐modified hematite mesocrystal superstructures assembled from highly oriented tiny nanoparticle (NP) subunits with sizes of ca. 5 nm were developed to achieve the highest photocurrent density (4.3 mA cm?2 at 1.23 V vs. RHE) ever reported for hematite‐based photoanodes under back illumination. Owing to rich interfacial oxygen vacancies yielding an exceedingly high carrier density of 4.1×1021 cm?3 for super bulk conductivity in the electrode and a large proportion of ultra‐narrow depletion layers (<1 nm) inside the mesoporous film for significantly improved hole collection efficiency, a boosting of multihole water oxidation with very low activation energy (Ea=44 meV) was realized.  相似文献
