为了给运动心脏研究提供更新的理论依据,在不同强度运动训练动物模型的基础上,用免疫印迹检测心肌组织中AMPK的表达,探讨不同运动训练强度对大鼠心肌AMPK分泌功能的影响.实验结果显示:中等强度运动组(ME组)和大强度运动组(HE组)AMPK含量表达升高.大强度运动组(HE组)升高较明显;中等强度运动组单磷酸腺苷激酶(ME组)呈升高趋势,但没有统计学意义,(p〉0.05)而大强度运动组(HE组)与中等强度运动组(ME组)相比,有统计学意义,(p〈0.05).经过分析认为:大鼠经不同强度运动训练后,心肌组织中AMPK表达随运动训练强度的增加,呈现逐渐递增趋势,相对于低强度运动训练,高强度训练对于大鼠心肌AMPK的影响更大.提示心肌AMPK表达对不同强度运动产生适应性改变,心肌通过增加AMPK表达适应强度运动.  相似文献

To research juglone's immunology regulation, several perspectives including immunology regulation, lymphocyte proliferation, and cytokines were presented.The index of thymus and spleen, total supperoxide dismutase activity (T-SOD), serum malondialdehyde (MDA), content of nitrogen oxide (NO), the anti-superoxide anion ability, the suppressing hydroxy radical ability, contents of reduced glutathione (GSH) in thymus, total antioxidationability (T-AOC) and the level of mouse lysozyme (LZM) in plasma were measured. LPS-induced B thymocytes proliferation was measured by MTT assay.In the low-immunity model group, MDA and NO levels were decreased (p<0.001). Juglone improved lysozyme (LZM), GSH contents in thymus, T-AOC, T-SOD activity, the anti-superoxide anion ability (anti- O2-) (p<0.001).In the stimulation immunity model group, MDA and NO levels (p<0.05), anti- O2- and T-SOD activity (p<0.05) were up-regulated, whereas LZM, T-AOC contents were down-regulated.juglone has a significant effect, which promotes cell regeneration and function recovery, also may alleviate oxidative damage; juglone possesses a dual regulating effect on humoral immunity in mice.  相似文献

Constipation is a global epidemic.To evaluate the ameliorating effect of camellia oil on constipation,two slow transit costive mice models were established by water deprivation or sucralfate gavage.Administration of camellia oil (4.0 and 8.0 mL/kg/day) significantly shortened the defecation time,increased defecation mass and fecal water content,with efficacies comparable to that of hemp seed oil.Intestinal propulsion test showed that camellia oil significantly enhanced the propelling rates of the charcoal powder in the intestine.Meanwhile,camellia oil also significantly decreased the serum levels of nitric oxide (NO),nitric oxide synthase (NOS) and vasoactive intestinal peptide (VIP) and increased serum substance P in costive animals.These results suggested that camellia oil can largely relieve slow transit constipation and enhance the gastrointestinal motility through modulation of serum gastrointestinal motility key factors such as NO,NOS and VIP and substance P.  相似文献

为研究一氧化氮(NO)与金属铜对海洋微藻生长的影响,选用中肋骨条藻(Skeletonema costatum)和裸甲藻(Gymnodinium sp.)进行了不同NO和Cu浓度、不同添加方式的室内培养实验,初步探讨了它们的作用机制.结果表明:一次性或每天两次加入不同浓度NO(10-5~10-10mol/L),对微藻的生长表现出不同程度的促进或抑制作用,呈"两面性".一次性加入1.4、0.001 4μmol/L NO或每天两次加入0.001 4μmol/L NO,对处于正常生长或铜胁迫下微藻的生长都有促进作用,表现出NO能"抵御"铜对微藻生长的抑制作用.NO对微藻的生长具有重要的调控作用,这可能与NO所具有的抗氧化作用有关.  相似文献

研究当归对高脂喂养家兔内皮素和一氧化氮水平的影响。取健康家兔随机分为正常对照组(普通饲料喂养);高脂组(高脂饲料喂养)和当归组(高脂饲料 当归注射液)。喂养10周末处死动物取血和主动脉,检测血浆内皮素浓度,并用Griess反应法检测主动脉条一氧化氮(NO)的含量。当归组主动脉NO含量高于高脂组而接近于正常组;当归组血浆内皮素(ET)含量明显低于高脂组而接近正常组。当归能够拮抗高脂饮食所致家兔主动脉NO水平的降低以及血浆ET水平的升高,从而推测该药有可能具有抗动脉粥样硬化作用。  相似文献

为探究复方黄芪口服液对皮肤衰老的延缓功效,将SD大鼠随机分为含药血清组与不含药血清组,分别采用10 mL/kg复方黄芪口服液和生理盐水灌胃SD大鼠7 d,每天1次,获取血清后将10%血清分别作用于200 nmol丝裂霉素(MMC)干预的人成纤维细胞衰老模型,验证了含药血清可有效促进细胞增殖. 再利用组织工程皮肤Episkin表皮模型和三维真皮衰老模型,分别通过免疫荧光染色与Elisa试剂盒评价血清干预后表皮模型丝聚蛋白与三维真皮中人胶原蛋白I、Ⅲ型的含量. 实验结果表明,服用复方黄芪口服液有助于表皮层丝聚蛋白与真皮层人胶原蛋白I、Ⅲ型蛋白的表达,从而提高皮肤屏障与保水能力,提升皮肤支撑力,减少皱纹生成,达到延缓皮肤衰老的功效.  相似文献

针对轴承加速寿命试验中多应力加速模型欠缺、失效规律认识不到位等科学问题,首先,深入分析真空、温度、预紧力、转速、微重力环境下,液体润滑轴承的界面微观失效模式及机理;然后,推导和建立考虑上述多应力综合作用机理的微观接触统一润滑模型,研究给出基于改进牛顿-拉夫逊法的数值解算方法;其次,设计5因素5水平的正交仿真方案,开展真空、温度、预紧力、转速、微重力的失效规律仿真与分析,获得微观接触界面膜厚、压力峰值、摩擦系数、最大下表面应力随应力水平及接触区尺寸变化的失效响应值;最后,建立了基于响应面方法辨识真空、温度、预紧力、转速、微重力水平与失效响应值之间的多应力加速模型,并对其进行试验验证及工程应用,奠定了空间轴承加速寿命试验的模型基础.  相似文献

本试验选用8头处于泌乳早期的高产奶牛,分对照组和试验组进行能量应激和酮病预防试验。试验组奶牛日粮中添加6克烟酸/头·日,对照组则不加烟酸,所用日粮根据《中国奶牛饲养标准》配制。 结果表明,泌乳早期奶牛日粮中添加烟酸可显著降低血清游离脂肪酸和丙酮量(P<0.05)。对血糖、胰岛素、乙酰乙酸、β—羟丁酸及酮体总量无显著性影响(P>0.05)。但对分娩后1~4周奶牛血糖有升高趋势( 10.92%),面对分娩后3~5周β—羟丁酸及酮体总量有降低趋势。  相似文献

为了探讨氯化镧(LaCl3)染毒对子代大鼠回避学习记忆能力的影响。采用健康成年雌性Wistar大鼠24只,随机分为对照组(饮用蒸馏水);低剂量染镧组(饮用含0.25%LaCl3的蒸馏水溶液)、中剂量染镧组(饮用含0.5%LaCl3的蒸馏水溶液)和高剂量染镧组(饮用含1.0%LaCl3的蒸馏水溶液),每组6只动物。自雌鼠受孕日至仔鼠断乳后1个月以自由饮水的方式摄入镧。通过穿梭箱实验判断仔鼠的主被动逃避学习能力。结果显示,中剂量组仔鼠主动逃避潜伏期和电击次数显著高于低剂量组和对照组(P0.05),电击时间显著高于对照组(P0.05);高剂量组仔鼠主动逃避潜伏时间、电击次数和电击时间均显著高于中剂量组、低剂量组和对照组(P0.05)。LaCl3染毒造成大鼠主动逃避能力下降,LaCl3染毒组仔鼠表现出剂量依赖的回避学习能力损伤(P0.05)。  相似文献

利用旋转细胞培养系统模拟微重力场,考察模拟微重力场及添加不同质量密度葡萄糖对小球藻(Chlorella sp. )生长情况的影响. 研究结果表明,以BG-11为基础培养基,模拟微重力环境对小球藻生长有明显促进作用. 当小球藻细胞接种密度为1.42×107 个/毫升,培养时间为12 d时,普通环境下培养的小球藻最高细胞密度仅为5.80×107 个/毫升,而在相同培养条件下模拟微重力环境下生长的小球藻细胞密度达到1.58×108 个/毫升. 在培养基中添加葡萄糖作为额外碳源后,微藻生长周期显著缩短,适宜小球藻生长的葡萄糖质量密度为10 g/L. 当添加10 g/L葡萄糖,小球藻接种密度为5.76×107 个/毫升,培养时间为5 d时,普通环境和模拟微重力环境下培养的小球藻的最高细胞密度分别可达3.85×108 个/毫升和5.42×108 个/毫升.  相似文献

用干烧黄芪鱼掺和到饲料中饲养实验性衰老小鼠40天,测定小鼠脑、肾超氧化物歧化酶(SOD)活力、小鼠肝谷胱甘肽过氧化物酶(GSH-Px)活力,小鼠睾丸、脑中一氧化氮(NO)含量和睾丸、血清中一氧化氮合成酶(NOS)活力.结果表明:添加干烧黄芪鱼的实验组相比与空白对照组,在以上各项指标上均存有显著性或十分显著性差异.即药膳干烧黄芪鱼具有一定的抗自由基作用,并对雄性衰老小鼠生殖系统NO含量及NOS活力有一定影响.  相似文献

In order to accelerate the chronic wounds healing, we investigated the healing effects of bioactive glass and Yuunan baiyao ointments in streptozotocin-induced diabetic rats. The ointments were prepared by mixing 45S5 bioactive glass powder (16% weight) with Vaseline and different weight percentages of Yurman baiyao. Full-thickness defect wounds were created on the back of 130 SD rats and were randomly divided into 8 groups. The wound healing rates were calculated at 4, 7, 10, 14 and 21 days after surgery. The samples were harvested for further observations. Considering the wound closure rate, group 6 (with 5% Yuunan baiyao) has better wound healing performance than other diabetic groups. The lower inflammatory response was observed by gross observation and confirmed by the results of H&E staining and TEM observation. Besides, the proliferation of fibroblasts, the formation of granulation tissue, as well as the vascularization, were improved in group 6 compared to other diabetic groups. All results suggest that bioactive glass and Yunnan baiyao ointments can accelerate the recovery of diabetes-impaired skin wounds, and comparing to other diabetic groups, group 6 (with 5% Yunnan baiyao) has better healing effect.  相似文献

通过角叉菜胶致小鼠足肿胀法检测炎症渗出液中前列腺素E2(PGE2)、丙二醛(MDA)、组胺(Histamine)和5-羟色胺(5-HT)的含量变化,以及血清中过氧化氢酶(CAT)、超氧化物歧化酶(SOD)活性和一氧化氮(NO)含量的变化,研究迷迭香酸抗炎的作用机制.结果表明,连续给小鼠灌胃7d,迷迭香酸能明显抑制角叉菜胶致小鼠后足肿胀,并且显著降低炎症渗出液中前列腺素E2、丙二醛、组胺和5-羟色胺的含量,显著提高血清中过氧化氢酶、超氧化物歧化酶活性,显著降低血清中一氧化氮含量.说明迷迭香酸能够通过抑制炎症部位炎症因子的合成或释放而起到抗炎作用,为迷迭香酸更好地应用于营养食品、保健药品中提供理论依据.  相似文献

The anti-inflammatory effect and mechanism of Usnic acid (UA) were explored on lipopoly-saccharide (LPS)-stimulated RAW264.7 cell line.The effects of UA on pro-inflammatory cytokines including tumor necrosis factor-alfa (TNF-α),interleukin-6 (IL-6) and interleukin-1 beta (IL-1β),pro-inflammatory mediators such as nitric oxide (NO),inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were studied by sandwich ELISA,real-time PCR and western blot analyses.Similarly,the effect of UA on anti-inflammatory cytokine interleukin-10 (IL-10) and anti-inflammatory mediator heme oxygenase-1 (HO-1) were also studied following the same methods.Furthermore,nuclear factor-κB (NF-κB) was assayed by immunocytochemistry.The results showed that UA has anti-inflammatory effect by down-regulatinng iNOS,COX-2,IL-1β,IL-6 and TNF-α,COX-2 gene expression through the suppression of NF-κB activation and increasing anti-inflammatory cytokine IL-10 and anti-inflammatory mediator HO-1 production.  相似文献
