Alpha-terthienyl is toxic toAedes aegypti larvae in the dark, but its activity is much enhanced in the presence of ultraviolet light. The development of first-instar larvae treated with alpha-terthienyl and ultraviolet light was followed until the emergence of adults. The LC50 value for first instars was about 0.002 ppm. Practically all the larvae which survived 24 hr reached adulthood. Fourth-instar larvae were also sensitive to photochemical treatment. When their development into adults was followed, the LC50 value was 0.45 ppm. Contrary to earlier reports, alpha-terthienyl was also phototoxic in pupae, but not when the adults were about to emerge. The LC50 value was ca. 0.06 ppm for pupae which were 1 or 2 days old. This is the first example where the activity of a photoinsecticide has been demonstrated in pupae. Alpha-terthienyl did not affect the hatching of eggs.  相似文献

Incorporation of certain phytoecdysones (ecdysterone, polypodine B, and ponasterone A) into a semisynthetic artificial diet induces pathophysiological effects in larvae of the leek-moth (Acrolepiopsis assectella Zell., Acrolepiidae). The effects include lethality of the newly hatched, first-instar larvae; special ecdysial failures associated with the appearance of larvae with two head capsules; and developmental anomalies during metamorphosis. The effective range of dietary ecdysteroid, as evaluated by larval mortality, varies from 25 to 250 ppm. The EC50 value is 100 ppm for polypodine B and 130 ppm for ecdysterone. The dietary effects of the phytoecdysones are similar to the previously observed effects caused by the dried flowers, but not leaves, of the leek plant. However, the active compound of the leek flowers is a saponin.  相似文献

Although metabolites of furanocoumarins have been characterized in a wide range of organisms, to date they have been identified in only a single insect species, Papilio polyxenes. Depressaria pastinacella, the parsnip webworm, like P. polyxenes a specialist on Apiaceae, routinely consumes plant tissues higher in furanocoumarin content than does P. polyxenes and is capable of faster cytochrome P-450-mediated detoxification of these compounds. In this study, we characterized metabolites of xanthotoxin, a linear furanocoumarin, and sphondin, an angular furanocoumarin, in midguts and frass of parsnip webworms. Two metabolites were isolated and identified from webworms fed artificial diet containing xanthotoxin. LC-ESI-MS analysis resulted in the determination of a MW of 266 for the compound in the frass and one of the compounds in the midgut; 1H NMR confirmed its structure as 6-(7-hydroxy-8-methoxycoumaryl)-hydroxyacetic acid (HCHA). The second compound from the midgut had a MW of 252 and was identified by 1H NMR and 13C NMR analysis as 6-(7-hydroxy-8-methoxycoumaryl)-hydroxyethanol) (HMCH). Whereas HCHA has been found in frass of Papilio polyxenes fed xanthotoxin, HMCH has not been reported previously in insects. Although the first step of metabolism of xanthotoxin in webworms as well as P. polyxenes is likely the formation of an epoxide on the furan ring, angular furanocoumarin metabolism in webworms appears to differ. The principal metabolite of sphondin was identified as demethylated sphondin (6-hydroxy-2H-furo[2,3-h]-1-benzopyran-2-one) by LC-ESI-MS and confirmed by 1H NMR and 13C NMR analyses. That webworms produce metabolites of xanthotoxin in common not only with other Lepidoptera (e.g., HCHA) but with other vertebrates (e.g., HMCH) suggests a remarkable conservatism in the metabolic capabilities of cytochrome P-450s and raises the possibility that insects may share other detoxification reactions with vertebrates with respect to toxins in foodplants.  相似文献

The free amino acids have been shown by isolational work and choice bioassays to be more important than all other factors evaluated in defining leaf-feeding resistance of corn (Zea mays L.) to fall armyworm (FAW) [(Spodoptera frugiperda J.E. Smith)] larvae. 6-MBOA (6-methoxybenzoxazolinone) and maysin, toxins present in corn, were shown not to be significant factors for leaf-feeding resistance to first-instar FAW larvae because of their low concentrations in the whorl. Amino acid analysis showed that while the ratios of the essential amino acids in susceptible (S) and resistant (R) lines were similar, there were differences in the nonessential amino acids, particularly aspartic acid, which was higher in R lines. Also, the ratio of essential amino acids to nonessential amino acids was important, being too low in expressed whorl leaf juice (obtained from V8–V10 growth stage plants) to support larval growth, although juice was stimulatory in choice tests. The total protein content of whorls in S lines was about 15% higher than in R lines, but the significance of this difference is uncertain, because nutritional tests showed that larval growth increased with total protein only up to 12% protein. Sugars were only slightly stimulatory. Thus, the amino acids along with higher hemicellulose content of R lines, established by us earlier, appear to explain much of the basis of resistance in corn to larval leaf-feeding of the FAW.  相似文献

Adult leaf beetles of the genusOreina are known to be defended either by autogenously produced cardenolides or by pyrrolizidine alkaloids (PAs) sequestered from the food plant, or both. In this paper we analyze larvae of differentOreina species and show that the larvae contain the same defensive toxins as the adults in quantities similar to those released in the adults' secretion. Both classes of toxins are found in the body and hemolymph of the larvae, despite their different origins and later distribution in the adults. Larvae of sequestering species differed in their PA patterns, even though they fed on the same food plants. The concentration in first-instar larvae of a PA-sequestering species was similar to that in fourth-instar larvae. In all stages examined, the amount of PAs per larva did not greatly exceed the estimated uptake of one day. Eggs of two oviparous species contained large concentrations of the adult's toxins, while neonates of a sequestering larviparous species had no PAs.  相似文献

Since several species of predatory ants show some kind of repulsion towards the first-instar larvae (FIL) ofDiaprepes abbreviatus L., the predatory behavior ofSolenopsis geminata (F.), a common ant in the citrus groves in Guadeloupe, was studied. Different extracts of larvae were disposed on egg masses ofD. abbreviatus and presented as prey to the ants, both in the field and in the laboratory. The ants are repelled by the FIL extracts. The allelochemicals involved are produced in large amounts, from 5 to 20 ng per larva. Physiochemical analyses have led to the identification of two sesquiterpenes of molecular weight 218 and 234, secreted in the respective proportions of 65 and 35%.  相似文献

Soybean plant volatiles, extracted as steam distillates, significantly affected the behavior and biology of the cabbage looper,Trichoplusia ni (Hübner) (Lepidoptera: Noctuidae). Distillates from the susceptible Davis variety attractedT. ni larvae and female adults, whereas those from resistant PI 227687 plants repelled them. When mixed in an artificial diet, steam distillates from PI 227687 plants caused mortality of first-instar larvae. Adults emerging from pupae topically treated with 5 g of such PI 227687 extractable showed developmental abnormalities. Larval feeding was significantly less on Davis leaves treated with PI 227687 volatiles as compared to solvent (acetone) or such Davis extractables. However, Davis volatiles on PI 227687 leaves did not increase larval feeding. HPLC analyses of steam distillates from susceptible Davis versus resistant PI 227687 indicated differences.  相似文献

Rice plant volatiles extracted as steam distillates significantly affected the behavior of the rice leaffolder,Cnaphalocrocis medinalis (Guenée). Leaffolder moths laid significantly fewer eggs on TN1 rice plants treated with extracts of resistant wild species of rice,Oryza officinalis andOryza punctata, than on TN1 plants treated with extracts of resistant cultivated rices. Extracts ofO. officinalis andO. punctata adversely affected egg hatchability.O. officinalis andO. punctata extracts were more toxic to first-instar larvae than extracts of other resistant varieties. Leaf area consumed by the larvae was reduced on TN1 plants treated withO. officinalis andO. punctata extracts compared with that on plants treated with extracts of resistant cultivated rice varieties.  相似文献

A phagostimulant, 6,10,14-trimethylpentadecan-1-one (phytone), was isolated and identified from Bermuda grass,Cynodon dactylon (L.). The phagostimulant activity of this isoprenoid ketone was established from bioassays of fall armyworm larvae,Spodoptera frugiperda (J.E. Smith). Larvae displayed increased body mass accumulation as well as preference to diet supplemented with this molecule. Neonate larvae fed diet supplemented with chromatographic isolates of phytone-containing fractions from six Bermuda grass cultivars showed a 10–40% increase in body mass accumulation as compared with controls. This variation in larval body mass accumulation seems attributable to a differential concentration of phytone in the cultivars, which ranged from 0.5 to 43 ppm. Additionally, first-instar larvae responded preferentially to diet pellets topically treated with phytone in concentrations as low as 0.1 ppm.  相似文献

First-instar larvae of the monarch butterfly, Danaus plexippus, a milkweed specialist, generally grew faster and survived better on leaves when latex flow was reduced by partial severance of the leaf petiole. The outcome depended on milkweed species and was related to the amount of latex produced. The outcome also may be related to the amount of cardenolide produced by the plants as a potential chemical defense against herbivory. Growth was more rapid, but survival was similar on partially severed compared with intact leaves of the high-latex/low-cardenolide milkweed, Asclepias syriaca, whereas both growth and survival were unaffected on the low-latex/low-cardenolide milkweed A. incarnata. On the low-latex/low-cardenolide milkweed A. tuberosa, both growth and survival of larvae were only marginally affected. These results contrast sharply to previous results with the milkweed, A. humistrata, in Florida, which has both high latex and high cardenolide. Larval growth and survival on A. humistrata were both increased by partially severing leaf petioles. Larval growth rates among all four milkweed species on leaves with partially severed petioles were identical, suggesting that latex and possibly the included cardenolides are important in first-instar monarch larval growth, development, and survivorship.  相似文献

Both biotic and abiotic selection pressures can contribute to geographic variation in allelochemical production in plants. We examined furanocoumarin production in western North American populations of Heracleum lanatum and Pastinaca sativa that, at different latitudes and altitudes, experience different ultraviolet (UV) light regimes. Total furanocoumarins and linear furanocoumarins of fruits were negatively correlated with UV irradiance, whereas amounts of angular furanocoumarins, which are generally less phototoxic, were not. Another factor potentially influencing furanocoumarin production is the presence of the parsnip webworm Depressaria pastinacella, (Lepidoptera: Oecophoridae), an herbivore that feeds on reproductive structures of both plant species. These insects sequester lutein from their host plants; this carotenoid acts to ameliorate furanocoumarin toxicity. Although the concentration of lutein in fruits did not vary with UV irradiance, lutein sequestration by sixth instars was positively correlated with UV irradiance. Webworm populations are variably infested with the polyembryonic webworm parasitoid Copidosoma sosares Walker (Hymenoptera: Encyrtidae). H. lanatum fruits from populations with webworms parasitized by C. sosares had lower concentrations of furanocoumarins, with the exception of sphondin, than fruits from plants infested with webworms free from parasitism. Lower levels of these furanocoumarins may reduce negative effects on the fitness of this parasitoid. In contrast with the variation in furanocoumarin content, the ability of webworms to metabolize furanocoumarins by cytochrome P450 did not differ significantly among populations from New Mexico to Alberta.  相似文献

The toxic effect of hypericin (HYP), a photodynamic quinone that occurs in certain species of the genusHypericum, towardsManduca sexta third-instar larvae was investigated. The LD50 of purified hypericin was 16 g/g larval initial fresh weight in constant light (22 W/m2). Reduced irradiance resulted in decreased mortality. Sublethal applications retarded larval growth (body fresh weight) in a dose-dependent manner. Toxicity had an absolute light dependence at the dose levels used, the active wavelengths being >500 nm. The phototoxic effect was rapidly lost when larvae were maintained in darkness (>8 hr) prior to irradiation. However, the potential for light-dependent mortality was retained if larvae were starved prior to irradiation.  相似文献

Jasmonic acid (JA) treatment of tomato plants induces several defense-related oxidative enzymes and increases pest resistance in a manner thought to simulate natural insect wounding. In a full-factorial greenhouse experiment, we examined the independent and interactive effects of plant age and exposure to wind-induced mechanical stress (MS), on the ability of JA to induce defense in tomato. In general, treatment of 4-, 6-, and 8-week-old tomato plants with 1 mM JA resulted in the induction of peroxidase and polyphenol oxidase activity and reduced the relative growth rate of first-instar Manduca sexta larvae fed treated leaves, in accordance with other studies. Peroxidase activity increased with plant age and was induced by JA most strongly in older plants. In contrast, polyphenol oxidase activity did not change with plant age and was induced by JA most strongly in young plants. While relative growth rates of M. sexta were lower on older plants overall, JA reduced growth rates most strongly in young plants, in which JA treatment enhanced polyphenol oxidase activity by more than 70%. MS enhanced the activity of peroxidase, but substantially reduced the activity of polyphenol oxidase; the latter most intensely on older plants. M. sexta tended to grow more slowly on MS-treated plants, although this effect was not significant. Thus, reduced polyphenol oxidase activity in MS-treated plants did not lead to an increase in growth rate of M. sexta, possibly because peroxidase activity was still elevated in MS-treated plants. Significant interactions between JA and MS and three-way interactions were not detected for any variable, although the inductive effects of both JA and MS interacted in complex ways with plant age. Our results indicate that resistance traits in tomato are differentially affected by JA and wind exposure and differ in their relative contribution to defense as plants age.  相似文献

Twenty esters, representing the biogenetically related tigliane, daphnane, ingenane, and lathyrane series of diterpenes, were screened for growth-inhibitory and insecticidal effects on newly hatched larvae of the North American cotton pest,Pectinophora gossypiella (pink bollworm). Among the tigliane derivatives tested, only 12-O-tetradecanoylphorbol-13-acetate and 12-O-(2-methyl)butyrylphorbol-13-decanoate, of seven phorbol diesters isolated from croton oil by a new procedure involving droplet countercurrent chromatography, were active againstP.gossypiella as both growth inhibitors and insecticides. The effects of the former compound were not significantly diminished by acetylation of its C-20 primary hydroxy group. Three other croton oil phorbol diester constituents, as well as daphnetoxin and daphnetoxin-5,20-diacetate, exhibited activity as growth inhibitors, but not as insecticidal agents, at the doses used. None of the ingenane or lathyrane derivatives investigated was active in either respect. 12-0-Tetradecanoylphorbol-13-acetate was found to cause 100% mortality on second-stadium larvae ofCulex pipiens at 0.6 ppm, but exhibited less significant effects onOncopeltus fasciatus (second-stadium nymphs) andTribolium confusion (adults) when applied at higher doses.  相似文献

The sesquiterpene lactone cnicin was extracted fromCentaurea maculosa andCentaurea vallesiaca. We examined its effects on the ovipositional response and larval development of generalist and specialist insect herbivores associated withC. maculosa. For the oviposition trials, three plant species (C. maculosa, Achillea millefolium, andCichorium intybus), half of which were sprayed with 3% of cnicin, were exposed to the specialist mothsStenodes straminea, Agapeta zoegana, andPterolonche inspersa in field cages. All three species significantly preferredC. maculosa to other plants andP. inspersa significantly preferred cnicin-sprayed plants to untreated plants for oviposition. Tested over all species, cnicin significantly increased the number of eggs laid on a given plant. A larval diet test examined the toxicity of cnicin for larvae of the generalist noctuid mothSpodoptera littoralis. Cnicin concentrations of 3% and 6% were lethal and 1% and 0.5% seriously inhibited growth and development. The larvae of theC. maculosa specialistStenodes straminea survived at 6% cnicin, but none of the pupae hatched.Agapeta zoegana was able to survive at 1% and 3% cnicin. Both specialists had difficulties with the artificial diet, but weight increase and survival was not further reduced when cnicin was present compared with on the control diet. In conclusion, cnicin influenced host recognition by the specialist species, and larvae of the generalist did not survive on natural levels of cnicin. Growth and survival of the specialist were not influenced by cnicin but were considerably hampered on artificial diet.  相似文献

The growth and survival of the Catalpa sphinx, Ceratomia catalpae (Sphingidae), were measured on five different species of Catalpa: C. bignonioides, C. bungeii, C. fargeseii, C. ovata, and C. speciosa. Larval growth varied significantly among these host plant species; however, survival did not differ. Quantification of the iridoid glycoside content of larvae, pupae, adults, larval frass, and leaves of the larval host plant, C. bignonioides, by gas chromatography showed that leaves contained both catalpol and catalposide; larvae, pupae, and frass contained only catalpol; and the adults contained no detectable iridoid glycosides. Amounts were highest in the larvae and declined in the pupal stage. Very small amounts of catalpol were detected in adults of the parasitoid, Cotesia congregata, and in the silken cocoons. The hemolymph in which the parasitoid larvae grew contained over 50% dry weight catalpol. Larvae of C. catalpae often regurgitate when disturbed. This may serve as a defense against predators. A comparison of the growth of larvae pinched with forceps to induce regurgitation with those that were not so treated showed that larvae that were pinched, and usually regurgitated, grew significantly more slowly than those that were not.  相似文献

Xanthotoxin is a phototoxic allomone found in many of the host plants of the black swallowtail,Papilio polyxenes (Lepidoptera: Papilionidae). When added to the diet of final instar larvae, xanthotoxin can induce the cytochrome P450 monooxygenase (P450) activity in midgut microsomes by which it is detoxified. Induction is dose-dependent, increasing sevenfold when larvae feed on parsley treated topically with xanthotoxin at 0.5 or 1.0% fresh weight. Although xanthotoxin exerts much of its toxic effects when photoactivated by ultraviolet light, induction of P450 activity did not differ in the presence or absence of ultraviolet light. Despite a 4.7-fold induction of xanthotoxin-metabolizing P450 activity, total P450 content measured in the same microsomal samples did not increase significantly. These data indicate that multiple forms of P450 exist in the black swallowtail midgut and that they are differentially induced by xanthotoxin.  相似文献

Larvae of the coccinellid beetle Cheilomenes sexmaculata (F.) produce an oviposition-deterring pheromone that inhibits egg laying of conspecific females on oviposition sites walked over by first-instar larvae. By use of bioassay-guided fractionation of larval extracts, (Z)-pentacos-12-ene was identified as an active component of the cuticular hydrocarbons of the larvae. Other compounds that occur in the active fractions, such as the alkaloid coccinelline and saturated hydrocarbons, were individually tested but proved to be inactive. Jojoba wax solutions were irradiated at wavelengths longer than 366 nm, in the presence of sensitizers, at room temperature.Cis-trans isomerization took place only with sensitizers with triplet energy greater than 68 kcal/mol. The role of the steroidal glycoalkaloid -tomatine in the hostplant resistance of tomato to the Colorado potato beetle,Leptinotarsa decemlineata (Say) was examined in short- (24 hr; using first- and fourth-instar larvae) and long-term (first-instar larvae reared through the prepupal stage) feeding experiments. Consumption rate, growth rate, efficiency of conversion of ingested food to body mass, and survival were compared forL. decemlineata provided foliage from susceptible (Lycopersicon esculetum Mill. cv. Walter), resistant (L. hirsutum f.Glabratum C.H. Mull accession PI 134417), and F1 hybrid plants. Values obtained for dietetic indices were regressed against corresponding values for -tomatine content of foliage provided to larvae. Differences in dietetic indices could not be attributed to variation in foliar -tomatine content despite a long-standing literature showing theex planta -tomatine inhibits feeding and growth byL. decemlineata.  相似文献
