Supercritical carbon dioxide, with and without a methanol modifier, was used to extract sinomenine from Sinomenium acutum (Thumb) Rehd et Wils. Sinomenine determinations were carried out using high-performance liquid chromatography (HPLC). The results show that the yield obtained after 2.5 h extraction with methanol-modified supercritical carbon dioxide was the highest (7.47 mg/g), while that obtained with only supercritical carbon dioxide was the lowest (0.17 mg/g). The recovery obtained with supercritical carbon dioxide, with and without a methanol modifier, could not be increased greatly by the method of the alkalinization of sample. Higher recoveries were obtained than extraction using methanol in Soxhlet extractor.  相似文献

Hydrocarbons, dialkyl alkylphosphonates and alkyl alkylphosphonic acids are selectively extracted from spiked soils by successive implementation of supercritical carbon dioxide, supercritical methanol-modified carbon dioxide and pressurized water. More than 95% of hydrocarbons are extracted during the first step (pure supercritical carbon dioxide extraction) whereas no organophosphorus compound is evidenced in this first extract. A quantitative extraction of phosphonates is achieved during the second step (methanol-modified supercritical carbon dioxide extraction). Polar phosphonic acids are extracted during a third step (pressurized water extraction) and analyzed by gas chromatography under methylated derivatives (diazomethane derivatization). Global recoveries for these compounds are close to 80%, a loss of about 20% occurring during the derivatization process (co-evaporation with solvent). The developed selective extraction method was successfully applied to a soil sample during an international collaborative exercise.  相似文献

A preparative supercritical fluid extraction system is described and was used with supercritical carbon dioxide to extract the active insecticide components pyrethrin I (PI) and pyrethrin II (PII) successfully from pyrethrum flower. A high-performance liquid chromatography method was developed and was used to separate and analyze the supercritical carbon dioxide extracts. Extraction efficiencies under several different extraction conditions were examined. Under the conditions examined, the most effective extractions of PI and PII (140 +/- 18 mg and 55 +/- 9 mg per 100 g of dry pyrethrum flower powder) were performed at 40 degrees C and 1200 psi. The results showed that extraction efficiencies of supercritical carbon dioxide are much better than those of n-hexane for pyrethrins I and II. During the extraction process, the most efficient extraction period was the first 3 h of the experiment.  相似文献

Summary Chromatographic methods for the qualitative and quantitative analysis of thyme (Thymus vulgaris L.) extracts (essential oil obtained by steam distillation and extracts obtained by carbon dioxide supercritical fluid extraction and methylene chloride) are described. The composition of extracts obtained at different pressures (from 80 bar to 400 bar) and constant temperature (40°C) is discussed. The extraction system thyme— supercritical carbon dioxide was modelled by empirical equations defining the dependence of the total extract (TE) solubility and thymol solubility in CO2 on the density of carbon dioxide.  相似文献

In the extraction of spiked PCB from soil, three extracting fluids were investigated: supercritical carbon dioxide (CO2), supercritical sulfur hexafluoride (SF6) and subcritical water. Among the tested fluids SF6 appeared to be appropriate especially for the extraction of low polar PCB. CO2 and water were found to be suitable for the quantitative extraction of all PCB. Water was judged as the best because of its low price, good availability and environmental safety.  相似文献

Polycyclic aromatic hydrocarbons (PAH) and alkyl-PAH in the lower mg/kg range were extracted from soil and fly ash. Extraction yields were measured for toluene Soxhlet, supercritical carbon dioxide, toluene modified carbon dioxide, and toluene modified dimethylether (DME) extractions. Pure DME similarly to toluene Soxhlet extraction enhances extraction yields (32 mg/kg) as compared to pure carbon dioxide (21 mg/kg). In particular, higher molecular weight PAH are extracted with pure DME. 5% Toluene modified carbon dioxide (43 mg/kg) and toluene modified DME (50 mg/kg) yield much better extraction yields than Soxhlet (32 mg/kg) or pure supercritical fluid extractions. The objective of this work is to track the amount of metal complexes distributed in the extraction cell, collection vial, and tubing used in supercritical fluid extraction (SFE) systems after progressive removal of metal ions in supercritical carbon dioxide (SC-CO2). Sodium diethyldithiocarbamate (NaDDC) and dibutylammonium dibutyldithiocarbamate (DBDC) ligands were used to form complexes with Cd, Cu, Pb, and Zn and CO(2)/5% methanol as a supercritical fluid. The mass balance of metal complexes were obtained before and after extraction, and metals in different locations in the system were flushed out using an organic solvent and nitric acid (HNO3). These results infer that the stability constant (beta) of the metal-ligand complex has a strong correlation with SFE. Because of the composition of the stainless-steel cell, Fe, Cr, and Ni or other trace elements in the cell might interfere with the mass balance of metal complexes in SFE due to an exchange mechanism taking place between the cell and the sample.  相似文献

Supercritical carbon dioxide has been considered an appropriate alternative for extraction and purification process of cosmetics, pharmaceuticals, food supplements and natural products. Solubility information of biological compounds is essential for choosing supercritical fluid (SCF) processes. The solubility of iodopropynyl butylcarbamate (IPBC), a fungicide and anti-dandruff agent, was measured in supercritical carbon dioxide with a high pressure apparatus equipped with a variable volume view cell at 313.15, 323.15, and 333.15 K and at pressure between 80 and 35 MPa. The experimental data were correlated well with Peng–Robinson equation of state (PR EOS) and quasi-chemical nonrandom lattice fluid model.  相似文献

Supercritical fluid extraction (SFE) is a promising technique for the extraction of 2,3,7,8-tetrachlorodibenzo-p-dioxin (2,3,7,8-TCDD) from environmental matrices such as contaminated sediments. The ability of SFE to solubilize many organic contaminants is well documented in industrial processes but its analytical applications were exploited just recently. In this study supercritical carbon dioxide and nitrous oxide and their mixtures with 2% methanol were used to extract 2,3,7,8-TCDD from aquatic sediments. An attractive feature of this process is that the carbon dioxide, being a virtually inert fluid, leaves no solvent residue on the processed sediment. Almost 100% of the 2,3,7,8-TCDD can be extracted from a sediment spiked with 200 μg/kg 2,3,7,8-TCDD in 30 minutes by using supercritical carbon dioxide + 2% methanol. Cleanup procedure is compared with the Soxhlet extraction procedure currently used as a standard method for extracting dioxins from sediment samples.  相似文献

Fungicides are used to suppress the growth of fungi for crop protection. The most widely used fungicides are succinate dehydrogenase inhibitors (SDHIs) that act by blocking succinate dehydrogenase, the complex II of the mitochondrial electron transport chain. As recent reports suggested that SDHI-fungicides could not be selective for their fungi targets, we tested the mitochondrial function of human cells (Peripheral Blood Mononuclear Cells or PBMCs, HepG2 liver cells, and BJ-fibroblasts) after exposure for a short time to Boscalid and Bixafen, the two most used SDHIs. Electron Paramagnetic Resonance (EPR) spectroscopy was used to assess the oxygen consumption rate (OCR) and the level of mitochondrial superoxide radical. The OCR was significantly decreased in the three cell lines after exposure to both SDHIs. The level of mitochondrial superoxide increased in HepG2 after Boscalid and Bixafen exposure. In BJ-fibroblasts, mitochondrial superoxide was increased after Bixafen exposure, but not after Boscalid. No significant increase in mitochondrial superoxide was observed in PBMCs. Flow cytometry revealed an increase in the number of early apoptotic cells in HepG2 exposed to both SDHIs, but not in PBMCs and BJ-fibroblasts, results consistent with the high level of mitochondrial superoxide found in HepG2 cells after exposure. In conclusion, short-term exposure to Boscalid and Bixafen induces a mitochondrial dysfunction in human cells.  相似文献

The solubilities of phenylalanine and tyrosine in supercritical carbon dioxide (SCCO2) were measured after derivatization as the N-acetyl amino acid ethyl ester, N-carbobenzoxy amino acid and N-acetyl amino acid. Using an SCCO2 flow system, a measuring method of the saturated solubilities of the derivatized amino acids was established in which the contact height of the extraction cell, i.e. a packed column, is increased till the concentration of a derivatized amino acid at the exit of the cell reaches a plateau. The solubilities of N-acetyl phenylalanine ethyl ester (APEE) exceeded 0.001 mole fraction, which is higher than those of caffeine produced in industrial SCCO2 processes. A possible way of separating the amino acid mixtures using polarity differences in different amino acid side chains was demonstrated using the solubility data of the N-acetyl-amino acid ethyl esters in SCCO2, as the solubilities of APEE are higher than those of N-acetyl tyrosine ethyl ester by two orders of magnitude.  相似文献

Davidone C is a newly discovered flavonoid compound purified from the ethyl acetate-soluble fraction of Sophora davidii (Franch.) Skeels. This study explored the anti-tumor activity of davidone C on hepatocellular carcinoma HepG2 and Bel-7402 cells and its mechanism through MTT method, morphological observation, flow cytometry and Western blotting. The results showed that davidone C significantly inhibited the proliferation of HepG2 and Bel-7402 cells in a time- and dose-dependent manner. The morphological changes of apoptotic cells can be observed under an inverted microscope, such as cell floating, chromosome condensation, apoptotic bodies, and other phenomena. The expressions of Bax, cleaved caspase-9, cleaved caspase-3 and cleaved PARP increased with the increase of dosage while Bcl-2 decreased, suggesting that the apoptotic mechanism might be related to the mitochondrial apoptotic pathway. Moreover, davidone C administration can down-regulate the expression of Grp78, and simultaneously up-regulate the expression of caspase-7 and caspase-12, indicating that the apoptotic mechanism might be related to the ERS pathway. In addition, davidone C can down-regulate the expression of p62, and simultaneously up-regulate the expression of LC3-I and LC3-II with a quantitative dependence, suggesting that the mechanism of apoptosis may be related to the autophagy signal pathway. All these results showed davidone C has potential effects on hepatocellular carcinoma.  相似文献

In this work, a simulation procedure of a supercritical extraction process was developed through the use of the commercial simulator HYSYSTM (Hyprotech Ltd.), adapting the existing units to the operating conditions typical of the supercritical extraction process. The objective is to recover provitamin A (β-carotene) from palm oil (esterified) using carbon dioxide/ethanol as the supercritical mixed solvent. This example characterizes the problem for recovering high added value product from natural sources, as the palm oil, which is desired by the market. Owing to the fact that esterified palm oil is a complex mixture, made by several components, in order to characterize this system in the simulator, it was necessary to create hypothetical components using the UNIFAC (universal function-group activity coefficients model) group contribution, because they are not present in a conventional database and, then, their physical properties must be estimated and/or predicted before the simulation. The optimization was carried out in each simulation for each equipment, in terms of operating conditions (temperature and pressure), in order to obtain the maximum recovery of carotenes. According to the results, it was possible to concentrate carotenes through two cycles of supercritical extraction with high yield. Furthermore, ethyl esters (biodiesel) were also obtained, as a byproduct of the proposed process, which can also be used as an alternative fuel, with the important characteristic that it is renewable.  相似文献

The use of four dithiocarbamates and three fluorinated β-diketones as potential chelating agents for three transition metal ions (Cd2+, Pb2+, and Hg2+) extracted from spiked sand and filter paper samples by supercritical fluid extraction (SFE) was investigated. The extractions were performed at 45°C and 250 atm for spiked sand samples and at 60°C and 200 atm for filter paper samples using supercritical carbon dioxide modified with 5% methanol. At 250 atm and using carbon dioxide modified with 5% methanol, the recoveries of Cd2+, Pb2+, and Hg2+ ions from spiked sand samples were 95% with lithium bis(trifluoroethyl)dithiocarbamate (LiFDDC) as the chelating agent; they ranged from 83–97% with diethylammonium diethyldithiocarbamate and from 87–97% with sodium di-ethyldithiocarbamate as chelating agents, and from 68–96% with trifluoracetylacetone, hexafluoroacetylacetone, and thenoylfluoroacetone as chelating agents. Ammonium pyrrolidinedithiocarbamate was not effective in the chelation SFE of Cd2+, Pb2+, and Hg2+ ions from either spiked sand or spiked filter paper samples under the extraction conditions used. Supercritical carbon dioxide alone gave consistently lower analyte recoveries than supercritical carbon dioxide modified with 5% methanol. The results suggest that the solubility of the metal chelate in the supercritical fluid plays a more important role than the solubility of the chelating agent in the supercritical fluid, as long as sufficient chelating agent is present in the fluid phase. Fluorination of the chelating agent, as in the case of LiFDDC, increases the solubility of the metal chelate, and subsequently enhances the extraction efficiency for the metal ions.  相似文献

A dual-beam thermal lens spectrometer (TLS) with a pulse pump–probe coaxial configuration was coupled to commercially available supercritical fluid extraction equipment using a high-pressure flow cell interface. Because of its feasible critical parameters, moderate cost and good thermooptical properties, carbon dioxide was used as supercritical solvent. Using trans-β-carotene as analyte, several factors related with the extraction process under continuos flow conditions were studied in order to establish their influence in the thermal lens signal magnitude. Under the optimum experimental conditions, the relative TLS signal area showed a linear relationship with the concentration of trans-β-carotene from 1.5×10−6 to 8×10−8 M (n=5, r=0.998) in the supercritical phase. Thus, the viability of on-line detection for supercritical fluid extraction with a pulse thermal lens spectrometer was demonstrated.  相似文献

Supercritical fluid extraction using carbon dioxide modified with methanol, methanol-diethylamine, or methanol-triethylamine was used to extract vinblastine from the aerial portions of Catharanthus roseus. An HPLC-electrospray ionization (ESI)/MS analysis method was also developed to quantify the alkaloids in these extracts. Of the supercritical solvents evaluated, carbon dioxide-methanol-triethylamine (80 : 18 : 2) at 80 degrees C and 34.0 MPa greatly improved the supercritical fluid extraction (SFE) yield of vinblastine by as much as 76.4% over methanol extraction, while the other solvent conditions extracted the compound at yields less than 25% that of a methanol extraction. These results were confirmed by the robust HPLC-ESI/MS analytical method developed in this study.  相似文献
