We analysed the antimicrobial susceptibility, biofilm formation and genotypic profiles of 27 isolates of Staphylococcus haemolyticus obtained from the blood of 19 patients admitted to a hospital in Rio de Janeiro, Brazil. Our analysis revealed a clinical significance of 36.8% and a multi-resistance rate of 92.6% among these isolates. All but one isolate carried the mecA gene. The staphylococcal cassette chromosome mec type I was the most prevalent mec element detected (67%). Nevertheless, the isolates showed clonal diversity based on pulsed-field gel electrophoresis analysis. The ability to form biofilms was detected in 66% of the isolates studied. Surprisingly, no icaAD genes were found among the biofilm-producing isolates.  相似文献

Experiments were carried out to analyze the biological characteristics of two sympatric isolates of Schistosoma mansoni derived from humans and murines in a low endemic transmission area (Sumidouro county, state of Rio de Janeiro, Brazil). Sympatric reared-laboratory Biomphalaria glabrata and C3H/He mice were used as experimental hosts. Parameters assessed comprised: precercarial period, infectivity and mortality (snails), prepatent period, infectivity (percentage of cercariae maturation into adult worm) and intestinal egg count (mice). The murine isolate showed a shorter precercarial period and higher infectivity than human isolate (p<0.05). This biological heterogenicity did not correspond to the vertebrate data because any biological parameter presented significant difference (p>0.05). These data suggest that both isolates are local sub-populations, providing support for the hypotheses that in a same biotope mixed populations or sub-populations circulate among their main host (human beings) and/or rodent as an anfixenous infection.  相似文献

The karyotypes of Biomphalaria tenagophila collected from Rio de Janeiro, Brazil were studied using the air-drying method. Somatic cells of this species had 2n=36. The 18 chromosome pairs were identified and classified into 3 groups. The diploid cell has 7 pairs of metacentric, 8 pairs of submetacentric, and 3 pairs of subtelocentric chromosomes. Observed chromosomes ranged from 2.4 to 6.4 µm, and the total length was 122.3 µm. This is the first report on the chromosome of B. tenagophila.  相似文献

The economic importance of Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) is mainly due to its wide host range, variable kinds of damage, and great intraspecific variation. The delineation of two African biotypes of this pest has been carried by molecular, ecological, and host‐affiliation approaches, with largely consistent results. However, an understanding of its intricate host–pest interaction is necessary as a basis of its sustainable integrated control. This study investigated the host preference and suitability of cassava and okra biotypes of B. tabaci, based on multiple‐choice landing and oviposition preference assays and stage‐specific survival on eight common whitefly hosts. The cassava biotype significantly preferred cassava, Manihot esculenta, for landing and oviposition, but did not oviposit on okra, Abelmoschus esculentus. The okra biotype preferred okra, oviposited on eggplant, Solanum melongena, tomato, Lycopersicon esculentum, garden egg, Solanum integrifolium, and cowpea, Vigna unguiculata, but did not oviposit on cassava. The okra biotype developed on all hosts except cassava, but only survived marginally on cabbage, Brassica oleracea, and pepper, Capsicum annum var. grosum, while the cassava biotype did not develop on okra, cabbage, or pepper. Thus the observed host acceptance of the two biotypes is wider than earlier reported by host transfer experiments and molecular genetic surveys. Mortality was highest in the first instar nymphal stage, during which total mortality occurred on non‐hosts. Development time was slightly longer on marginal hosts than on the preferred hosts. Cowpea, garden egg, and tomato are additional common hosts of the two biotypes, whose role as reservoir hosts and biotype interbreeding grounds should be investigated further.  相似文献

The influence of various cropping sequences on population densities of Meloidogyne hapla and carrot yield was studied in organic soil under microplot-and field conditions. Spinach, radish, barley, oat, and wheat were poor or nonhosts for M. hapla. Population densities of M. hapla were maintained or increased on cabbage, celery, lettuce, leek, marigold, and potato. Marketable percent-age and root weight of carrots were greater following spinach, oat, radish, and fallow-onion than those following two crops of onion or carrot in microplots. Under field conditions, the carrot-onion-oat-carrot cropping sequence decreased M. hapla population densities and provided a 282% increase in marketable yield of carrot compared to a carrot monoculture. Two consecutive years of onion increased M. hapla population densities causing severe root galling and a 50% yield loss in the following crop of carrot. Based on root-gall indices, carrots could be grown economically for 2 years following radish, spinach, and oat, but not following onion and carrot without the use of nematicides.  相似文献

The identification and characterisation of Cryptosporidiumgenotypes and subtypes are fundamental to the study of cryptosporidiosis epidemiology, aiding in prevention and control strategies. The objective was to determine the genetic diversity ofCryptosporidium in samples obtained from hospitals of Rio de Janeiro, Brazil, and Buenos Aires, Argentina. Samples were analysed by microscopy and TaqMan polymerase chain reaction (PCR) assays forCryptosporidium detection, genotyped by nested-PCR-restriction fragment length polymorphism (RFLP) analysis of the 18S rRNA gene and subtyped by DNA sequencing of the gp60 gene. Among the 89 samples from Rio de Janeiro, Cryptosporidium spp were detected in 26 by microscopy/TaqMan PCR. In samples from Buenos Aires,Cryptosporidium was diagnosed in 15 patients of the 132 studied. The TaqMan PCR and the nested-PCR-RFLP detected Cryptosporidium parvum, Cryptosporidium hominis, and co-infections of both species. In Brazilian samples, the subtypes IbA10G2 and IIcA5G3 were observed. The subtypes found in Argentinean samples were IbA10G2, IaA10G1R4, IaA11G1R4, and IeA11G3T3, and mixed subtypes of Ia and IIa families were detected in the co-infections. C. hominis was the species more frequently detected, and subtype family Ib was reported in both countries. Subtype diversity was higher in Buenos Aires than in Rio de Janeiro and two new subtypes were described for the first time.  相似文献

The effects of eight summer rotation crops on nematode densities and yields of subsequent spring vegetable crops were determined in field studies conducted in north Florida from 1991 to 1993. The crop sequence was as follows: (i) rotation crops during summer 1991; (ii) cover crop of rye (Secale cereale) during winter 1991-92; (iii) ''Lemondrop L'' squash (Cucurbita pepo) during spring 1992; (iv) rotation crops during summer 1992; (v) rye during winter 1992-93; (vi) ''Classic'' eggplant (Solanum melongena) during spring 1993. The eight summer crop rotation treatments were as follows: ''Hale'' castor (Ricinus communis), velvetbean (Mucuna deeringiana), sesame (Sesamum indicum), American jointvetch (Aeschynomene americana), weed fallow, ''SX- 17'' sorghum-sudangrass (Sorghum bicolor x S. sudanense), ''Kirby'' soybean (Glycine max), and ''Clemson Spineless'' okra (Hibiscus esculentus) as a control. Rotations with castor, velvetbean, American jointvetch, and sorghum-sudangrass were most effective in maintaining the lowest population densities of Meloidogyne spp. (a mixture of M. incognita race 1 and M. arenaria race 1), but Paratrichodorus minor built up in the sorghum-sudangrass rotation. Yield of squash was lower (P ≤ 0.05) following sorghum-sudangrass than after any of the other treatments except fallow. Yield of eggplant was greater (P ≤ 0.05) following castor, sesame, or American jointvetch than following okra or fallow. Several of the rotation crops evaluated here may be useful for managing nematodes in the field and for improving yields of subsequent vegetable crops.  相似文献

The genetic variability of 12 burrowing nematode (Radopholus sp.) isolates from Central America, the Caribbean, and Florida, and one isolate from Ivory Coast were compared with RAPD analysis. A high degree of genetic similarity (>0.82) was determined for isolates from the Western Hemisphere. Genome similarity was greatest among isolates collected within a country. Among isolates collected in Central America and the Caribbean, burrowing nematodes from Belize and Guatemala were genetically more distant. However, the genome of the isolate from Ivory Coast was most dissimilar (>0.30). These results suggest that African and American burrowing-nematode isolates may have had different origins or that they have been geographically isolated for a sufficient amount of time to have accumulated genetic changes detectable by RAPD analysis. No relationship was found between the genomic similarity and extent of reproduction or damage to banana or citrus roots. Morphometric analysis involving eight of the isolates indicated that they were morphologically identical and values for morphometric parameters were well within the range previously published for banana and citrusparasitic burrowing nematodes.  相似文献

Background and Aims

Cereals have two root systems. The primary system originates from the embryo when the seed germinates and can support the plant until it produces grain. The nodal system can emerge from stem nodes throughout the plant''s life; its value for yield is unclear and depends on the environment. The aim of this study was to test the role of nodal roots of sorghum and millet in plant growth in response to variation in soil moisture. Sorghum and millet were chosen as both are adapted to dry conditions.

Methods

Sorghum and millet were grown in a split-pot system that allowed the primary and nodal roots to be watered separately.

Key Results

When primary and nodal roots were watered (12 % soil water content; SWC), millet nodal roots were seven times longer than those of sorghum and six times longer than millet plants in dry treatments, mainly from an 8-fold increase in branch root length. When soil was allowed to dry in both compartments, millet nodal roots responded and grew 20 % longer branch roots than in the well-watered control. Sorghum nodal roots were unchanged. When only primary roots received water, nodal roots of both species emerged and elongated into extremely dry soil (0·6–1·5 % SWC), possibly with phloem-delivered water from the primary roots in the moist inner pot. Nodal roots were thick, short, branchless and vertical, indicating a tropism that was more pronounced in millet. Total nodal root length increased in both species when the dry soil was covered with plastic, suggesting that stubble retention or leaf mulching could facilitate nodal roots reaching deeper moist layers in dry climates. Greater nodal root length in millet than in sorghum was associated with increased shoot biomass, water uptake and water use efficiency (shoot mass per water). Millet had a more plastic response than sorghum to moisture around the nodal roots due to (1) faster growth and progression through ontogeny for earlier nodal root branch length and (2) partitioning to nodal root length from primary roots, independent of shoot size.

Conclusions

Nodal and primary roots have distinct responses to soil moisture that depend on species. They can be selected independently in a breeding programme to shape root architecture. A rapid rate of plant development and enhanced responsiveness to local moisture may be traits that favour nodal roots and water use efficiency at no cost to shoot growth.  相似文献

Greenhouse and growth room experiments were conducted to investigate the effect of host plant in relation to different nematode inoculum levels, and temperature fluctuations on the development of Pasteuria penetrans. Host plant affected the development of P. penetrans indirectly through its effect on nematode development. Endospores collected from Meloidogyne javanica females reared on different hosts did not show any differences in subsequent attachment and infectivity. The numbers of endospores produced per infected female were reduced with increasing numbers of females parasitizing okra and tomato roots. Fluctuating temperatures retarded the development of P. penetrans. The life cycle of the parasite was completed faster at approximately constant temperatures close to 30 °C than when the temperature fluctuated away from 30 °C. The temperature of irrigation water did not affect the duration of life cycle of P. penetrans.  相似文献

Analyses of MLEE, RAPD and LSSP-PCR were used to compare the panel of american tegumentary leishmaniasis (ATL) isolates obtained from lesions of patients with rare clinical manifestations of the disease and typical lesions. All of the 34 samples analyzed by MLEE demonstrated similar electromorphic profiles with Leishmania (Viannia) braziliensis reference strain. Through the RAPD analysis, nine genetic profiles (genotypes) were identified. LSSP-PCR corroborates the initial screening and phenetic analysis has grouped the isolates into two major clusters comprising the nine different genotypes. Prevalent genotype defined as LbmtDNAgen1 was detected in the largest number of isolates. There was no association between genotypes and clinical symptoms. However, two different genotypes could be identified in the initial (LbmtDNAGen9) and reactivated lesion (LbmtDNAGen3) of the same patient. Our results support the idea of a less pronounced genotypic diversity among L. (V.) braziliensis circulating in the State of Rio de Janeiro and demonstrate the useful application of these molecular markers in genetics variability studies.  相似文献

The genetic diversity and structure of the ant Atta robusta were assessed by ISSR (inter-simple sequence repeats) in 72 colonies collected from 10 localities in the Brazilian states of Espírito Santo (48 colonies) and Rio de Janeiro (24 colonies). The ISSR pattern included 67 bands, 51 of them (76.1%) polymorphic. Analysis of molecular variance (AMOVA) revealed a high level (57.4%) of inter-population variation, which suggested a high degree of genetic structure that was confirmed by UPGMA (unweighted pair-group method using an arithmetic average) cluster analysis. The significant correlation between genetic and geographic distances (r = 0.64, p < 0.05) indicated isolation that reflected the distance between locations. Overall, the populations were found to be genetically divergent. This finding indicates the need for management plans to preserve and reduce the risk of extinction of A. robusta.  相似文献

The continual detection of a slow (I1) esterase band in greenhouse cultures of Meloidogyne konaensis isolated from the field led to a hypothesis that the nematode may be polymorphic for esterase. A survey of coffee fields demonstrated at least four esterase phenotypes were present in Meloidogyne recovered. An F1 phenotype predominated (60% of the females), but an I1 phenotype was also common (30% of samples). A series of greenhouse and laboratory experiments were undertaken to understand this polymorphism. Esterase phenotype was not affected by development at 22º, 25º, or 33 ºC on tomato. Two different esterase phenotypes (I1 and F1-I1) were detected after M. konaensis was grown on tomato for several generations, even in single-egg-mass lines derived from an F1 female. Three isolates of M. konaensis differing in esterase phenotype (F1, I1, and F1-I1) did not differ morphologically but did differ in their parasitic ability. Only the F1 isolate parasitized Coffea arabica. The F1-I1 isolate had greater reproduction on Lycopersicon esculentum and Cucumis sativus than either the I1 or F1 isolate. The mechanism of the development of the polymorphism has yet to be determined. However, the F1 esterase may be useful as a marker for future research on parasitism of coffee by M. konaensis.  相似文献

Seven populations of Heterodera trifolii from Arkansas, Kentucky, Pennsylvania, and Australia plus 3 or 4 single-cyst isolates (SCI) from each population were tested for reproduction on seven species of plants to compare the host preferences among and within populations. Common lespedeza, Kummerowia striata cv. Kobe, was a good host for all populations and isolates. Therefore, a plant was considered to be a host if the number of females produced on it was 10% or more of the number on Kobe. All seven populations reproduced on Trifolium repens and T. pratense. None reproduced on Beta vulgaris or Glycine max. One single-cyst isolate from the Australian population produced a few females on T. pratense. The Australian population maintained on carnation, Dianthus caryophyllus, produced females on carnation but not on curly dock, Rumex crispus. However, its subpopulation maintained on T. repens produced females on R. crispus but not on carnation. Four of the other six populations produced females on R. crispus, and four produced females on carnation. Differences in host range were observed among seven of the mother populations and their SCI, and among isolates within each population. Five host range patterns were found in populations and SCI of H. trifolii. Significant quantitative differences occurred among populations in the numbers of females on most hosts, between isolates and their original populations, and among isolates from the same population. SCI selected from white clover produced fewer females on a series of test hosts and had host ranges the same as or narrower than those of the original populations. However, SCI selected from Kobe lespedeza had more females on some hosts and had host ranges the same as or wider than those of the original populations. The host ranges of all populations and SCI of H. trifolii were different from those of populations and SCI of race 3 of H. glycines and H. lespedezae.  相似文献

Interbreeding potential, chromosome number, and host range were compared among several isolates and species of Bursaphelenchus from diverse geographic areas. Some isolates from North America, Japan, and France had a wide-ranging interbreeding potential, whereas others were restricted in their potential to hybridize with other isolates. Although interbreeding occurred in the laboratory between some "M" and "R" forms of B. xylophilus, interbreeding of B. xylophilus and B. mucronatus was rare. The hybrids had the pathogenicity of the parent with the broader host range. This fact suggests that virulence may be inherited as a dominant character or that increased virulence may have resulted from differences in hybrid vigor. The haploid chromosome number of the different isolates separated the isolates into three groups and distinguished B. xylophilus from B. mucronatus. The findings suggest that the pinewood nematode species complex consists of sibling species that have evolved by reproductive isolation, that the French isolate is a new species, and that B. xylophilus and B. mucronatus have evolved from a common ancestor.  相似文献

Population development of isolates of Heterodera lespedezae from Illinois and North Carolina was compared on Korean, sericea, and striate lespedezas and red clover at soil temperatures of 14, 18, 22, 26, 30, 34 C (± 1 C) and in a greenhouse where temperatures ranged from 19 to 40 C (av. 25 C). Cyst production on all plants, but not at all temperatures, was significantly different between the two isolates. Males of the Illinois isolate were recovered from red clover and striate lespedeza at 22 and 26 C and at greenhouse temperatures. No males of the North Carolina isolate were found on any host. Both isolates retarded growth of striate lespedeza but had no effect on growth of the other species tested.  相似文献

Microorganisms produce volatile organic compounds (VOCs) which mediate interactions with other organisms and may be the basis for the development of new methods to control plant-parasitic nematodes that damage coffee plants. In the present work, 35 fungal isolates were isolated from coffee plant rhizosphere, Meloidogyne exigua eggs and egg masses. Most of the fungal isolates belonged to the genus Fusarium and presented in vitro antagonism classified as mutual exclusion and parasitism against the nematode-predator fungus Arthrobotrys conoides (isolated from coffee roots). These results and the stronger activity of VOCs against this fungus by 12 endophytic bacteria may account for the failure of A. conoides to reduce plant-parasitic nematodes in coffee fields. VOCs from 13 fungal isolates caused more than 40% immobility to Meloidogyne incognita second stage juveniles (J2), and those of three isolates (two Fusarium oxysporum isolates and an F. solani isolate) also led to 88-96% J2 mortality. M. incognita J2 infectivity decreased as a function of increased exposure time to F. oxysporum isolate 21 VOCs. Gas chromatography-mass spectrometry (GC-MS) analysis lead to the detection of 38 VOCs produced by F. oxysporum is. 21 culture. Only five were present in amounts above 1% of the total: dioctyl disulfide (it may also be 2-propyldecan-1-ol or 1-(2-hydroxyethoxy) tridecane); caryophyllene; 4-methyl-2,6-di-tert-butylphenol; and acoradiene. One of them was not identified. Volatiles toxic to nematodes make a difference among interacting microorganisms in coffee rhizosphere defining an additional attribute of a biocontrol agent against plant-parasitic nematodes.  相似文献

The burrowing nematode Radopholus similis is one of the major constraints to banana (Musa spp.) production worldwide. Resource-poor farmers can potentially manage R. similis by using naturally occurring banana endophytes, such as nonpathogenic Fusarium oxysporum, that are inoculated into tissue culture banana plantlets. At present, it is unclear at what stage in the R. similis infection process the endophytes are most effective. In this study, the effect of three endophytic F. oxysporum isolates (V5w2, Eny1.31i and Eny7.11o) on R. similis host preference of either endophyte-treated or untreated banana plants was investigated. No differences were observed between the proportion of nematodes attracted to either root segments excised from endophyte-treated or untreated plants, or in experiments using endophyte-treated and untreated tissue culture banana plantlets. These results imply that the early processes of banana plant host recognition by R. similis are not affected by endophyte infection.  相似文献

Trichomonad species inhabit a variety of vertebrate hosts; however, their potential zoonotic transmission has not been clearly addressed, especially with regard to human infection. Twenty-one strains of trichomonads isolated from humans (5 isolates), pigs (6 isolates), rodents (6 isolates), a water buffalo (1 isolate), a cow (1 isolate), a goat (1 isolate), and a dog (1 isolate) were collected in Indonesia and molecularly characterized. The DNA sequences of the partial 18S small subunit ribosomal RNA (rRNA) gene or 5.8S rRNA gene locus with its flanking regions (internal transcribed spacer region, ITS1 and ITS2) were identified in various trichomonads; Simplicimonas sp., Hexamastix mitis, and Hypotrichomonas sp. from rodents, and Tetratrichomonas sp. and Trichomonas sp. from pigs. All of these species were not detected in humans, whereas Pentatrichomonas hominis was identified in humans, pigs, the dog, the water buffalo, the cow, and the goat. Even when using the high-resolution gene locus of the ITS regions, all P. hominis strains were genetically identical; thus zoonotic transmission between humans and these closely related mammals may be occurring in the area investigated. The detection of Simplicimonas sp. in rodents (Rattus exulans) and P. hominis in water buffalo in this study revealed newly recognized host adaptations and suggested the existence of remaining unrevealed ranges of hosts in the trichomonad species.  相似文献
