3月龄成熟SD大鼠踮只,随机分卵巢切除组(oVX)及假手术组(ShamO),切除双侧卵巢或作假手术。术后1,3,5,7月时各分别处死,刮取出第4、5腰椎体,进行光镜、透射电镜、扫描电镜观察。发现OVX后椎体皮质骨及终板变薄,巾央区小梁稀少、变细、吸收、穿孔、中断,水平连结小梁丧失,骨小梁胶原排列明显不规则甚至断裂,并可见较多骨小梁显微骨折及细微骨痂结构。大量功能活跃的破骨细胞出现,成骨细胞功能相对减退,向骨细胞转化。提示OVX后骨小梁的结构强度明显下降,椎体的力学强度降低。  相似文献

In order to study the action of tiludronate on the changes in intraosseous vascularization induced by ovariectomy, and to link these effects to those observed in bone remodelling, 30 female Sprague-Dawley rats (age 40 weeks) were studied. Ten rats were sham-operated and treated by vehicle, 10 rats were ovariec-tomized and treated by vehicle, and 10 rats were ovariectomized and treated orally with tiludronate, 0.16 mmol/kg/per day, 3 days a week for 16 weeks, from the day following ovariectomy. The rats were killed after 4 months, and a histomorphometric study and quantification of intraosseous vessels carried out on the sixth lumbar vertebra. The area of the intraosseous sinusoidal capillaries increased after ovariectomy, which also induced a moderate increase in resorption surfaces and osteoid surfaces leading to a decrease of 40% in the trabecular bone volume at the lumbar spine level. This bone mineral loss was completely prevented by tiludronate, which normalized the bone turnover. However, tiludronate was without any effect on intraosseous vascularization. These results indicate that the surface area of the intraosseous sinusoidal capillaries was correlated positively with resorption surfaces and negatively with trabecular bone volume and the number of bone trabeculae. In these experimental conditions, an inhibitor of bone resorption can exert its positive effect on bone mass without normalization of vascularization.  相似文献

Gonadal hormone deficiency following ovariectomy and skeletal unloading by limb immobilization are useful models of osteopenia. The purpose of this study was to compare changes in cortical bone after ovariectomy (OVX) or immobilization (IMM) for 6 and 12 weeks. Comparisons were also made when rats were ovariectomized or immobilized for 6 weeks and then immobilized (OVX/IMM) and ovariectomized (IMM/OVX), respectively, for 6 more weeks. Tibias and femurs were collected and static and dynamic cortical bone indices were determined by morphometric methods. Femurs from animals OVX or IMM for 12 weeks were tested for bone stiffness by torsional testing. Six and 12 weeks after OVX, there were increases in the periosteal perimeter, cortical area, and periosteal bone formation indices, indicating that ovariectomy increased modeling-dependent bone gain on the periosteal envelope, relative to controls. Contrarily, 6 and 12 weeks after IMM, there were decreases, compared with controls, in periosteal perimeter, cortical bone area, and periosteal bone formation indices. This indicates that immobilization decreased modeling-dependent bone gain on the periosteal envelope. These differences in modeling between the animals that were OVX and IMM resulted in a smaller cortical width and minimum cortical width in the IMM compared with the OVX animals. There were significant decreases in cortical bone stiffness and minimum cortical width at the fracture site following mechanical testing in the animals IMM for 12 weeks. Both ovariectomy and immobilization increased endocortical resorption surface, endocortical perimeter and expansion of the marrow cavity. Because of suppressed periosteal bone formation with increased endocortical resorption, immobilization had a greater effect on bone loss and decreased bone stiffness than did ovariectomy. In the OVX/IMM or IMM/OVX groups, there were changes that reflected both conditions. Immobilization mitigated the increase in periosteal bone formation but tended to augment endocortical resorption following ovariectomy. These results show that ovariectomy and immobilization have envelope-specific effects on rat cortical bone.  相似文献

Influence of ovariectomy on bone metabolism in very old rats   总被引:2,自引:0,他引:2  
Twenty-five 30-month-old Lou rats fed a diet (6 g/100 g BW/day) containing 0.9% Ca and 0.8% Pi were divided into five groups. Four groups were surgically ovariectomized. From day 2 until day 29 after ovariectomy, they were S.C. injected either with 17 β estradiol (E2; 10 μg/kg BW/48 hours) or progesterone (P; 140 μg/kg BW/48 hours), or 17 β estradiol+progesterone (E2P) at the same doses, or solvent alone (OVX). The fifth group was sham operated (SH) and injected with solvent. Urine was collected in metabolic cages from day 24 to 29 after OVX, and urinary pyridinoline (PYD) and deoxypyridinoline (DPD) excretion (markers of bone resorption) was measured by HPLC. All animals were killed 30 days after ovariectomy. Serum was then collected for measurement of osteocalcin (OC), alka-line phosphatase (ALP), parathyroid hormone (PTH), and calcitonin (CT). At necropsy, the success of ovariectomy was checked by marked atrophy of the uterine horns. Left and right femur were harvested for densitometric and mineral analysis, respectively. Ovariectomy had no significant effect upon plasma calcium and PTH concentrations. E2 or E2P treatment significantly increased plasma PTH and calcitonin concentrations. Plasma OC concentration and ALP were not different in any of the groups. In contrast, urinary excretion of PYD and DPD was higher in OVX than in SH rats. Bone mineral density (BMD) of the distal femur was decreased by OVX, but was not different in the E2P and SH groups. A similar pattern was observed for the mineral or Ca content of whole femur. Thus, OVX decreased BMD and bone mineral content (BMC) in very old female rats. Plasma OC concentration and ALP activity failed to demonstrate any significant effect of OVX, whereas PYD and DPD were elevated. These results suggest that bone resorption is increased in OVX rats, even when supplemented with E2 or P alone. However, no significant difference was observed between SH and OVX rats treated with supplementation of both E2 and P. Thus, in very old rats, a combination of E2 and P is much more effective than E2 or P alone to prevent bone loss following ovariectomy.  相似文献

The effect of silicon (Si) supplement on preventing bone mass loss induced by ovariectomy (OVX) in rats was investigated. Three groups of 15, 100-day-old female Wistar rats each, with a mean initial weight of ∼260 g per animal, were selected for the present study. One of the experimental group consisting of 15 OVX rats was fed a diet supplemented with 500 mg of Si per kg of feed (Si + OVX). The other two groups consisting of 15 OVX and 15 sham-OVX rats did not receive these supplements. Morphometric (weight and length) and densitometric studies with dual-energy X-ray absorptiometry were performed on the whole femur and 5th lumbar vertebra of each animal 30 days after the experiment. The Si + OVX rats did not show a loss of bone mass induced by OVX at axial level (5th lumbar vertebra) or periphery (femur). Nonetheless, a significant increase (ANOVA with Bonferroni/Dunn post hocs test) of longitudinal development of the femur (P < 0.0001) was patent. These results, obtained through the measurements of axial and peripheral bones, warrant closer scrutiny in connection with the Si inhibitory effect on bone mass loss as well as the stimulatory effect on bone formation. Both actions, namely, inhibition of resorption and stimulation of formation, infer that Si may have a potential therapeutic application in the treatment of involutive osteoporosis. 目的研究大豆异黄酮对雌激素缺失状态下大鼠骨量丢失及骨转换的影响。方法 12月龄雌性SD大鼠40只随机分为4组,除假手术组大鼠行卵巢切除假手术外,其余均切除卵巢建立雌激素缺失模型。分组设计为:实验组[大豆异黄酮类40 mg/(kg.d)]及元素钙250 mg/(kg·d), 假手术组[蒸馏水)、实验对照组[元素钙250 mg/(kg·d)]及雌激素组[雌二醇30μg/100(g· 3d)及元素钙250mg/(kg·d)],除雌二醇皮下注射给予外,其余灌胃给予。28 d实验期满后处死大鼠,取股骨测定骨密度、股骨钙含量、股骨形态计量学相关指标,并测定血清碱性磷酸酶 (AKP)活性及血清骨钙素(OC)水平。结果实验组股骨骨密度、股骨钙含量稍高于假手术组和实验对照组,但未呈显著性差异(P>0.05);实验组骨小梁面积与假手术组无显著差异(P> 0.05);各组动物AKP活性无显著差异(P>0.05):实验组和雌激素组OC水平显著高于假手术组 (P<0.01)。结论大豆异黄酮具有弱雌激素样作用,可能通过促进成骨形成而减少雌激素缺失状态下的骨丢失,维持骨健康,预防绝经后骨质疏松。  相似文献

目的:观察补肾益骨方对去势大鼠血生化、骨量及其力学性能的影响;探讨补肾益气方能提高去势大鼠骨密度改善骨生物力学性能的机制。方法:取12周龄Wistar雌性大鼠40只,随机分为模型组(A)、治疗组(B)、阳性对照组(C)、正常对照组(D)4组,每组10只。D组行假手术(单纯剖腹后关闭),其余3组行完整双侧卵巢摘除术,术后常规饲养12周。A、D组以蒸馏水10ml/kg灌胃,B组以10ml/kg补肾益骨方提取液灌胃,C组以10ml/kgα-D3胶丸水溶液灌胃。用药12周后,取血清用电极法测定血清中Ca、P含量;用放免法测定血清中骨钙素(BGP)、降钙素(CT)水平;以双能X线密度测定仪测定骨密度;通过三点弯曲试验,记录载荷-变形曲线,测定骨生物力学指标。结果:治疗组血清中BGP、CT水平及骨矿含量、骨密度明显高于模型组,并改善了去势大鼠的骨生物力学性能。结论:补肾益骨方能提高去势大鼠骨密度、改善骨生物力学性能,推测激素BGP、CT水平变化是其重要的治疗机制之一。  相似文献

目的观察去卵巢大鼠骨质疏松模型复制情况并动态观察去卵巢后不同时间段骨丢失情况,为抗骨质疏松药物研究提供对照依据。方法4.5月龄SD雌性大鼠,按体重随机分组,大鼠摘除双侧卵巢;在实验的第0d(4.5月龄)、4w(5.5月龄)、12w(7、5月龄)、18w(9.5月龄)杀死大鼠取材;采用体内双荧光标记法,胫骨上段硬组织包埋切片及松质骨形态计量学分析处理,观察去卵巢后不同时间段骨丢失情况。结果大鼠去卵巢4w后骨量显著降低,骨结构变差,骨形成有增加,骨吸收增加,骨吸收大于骨形成,骨转换增加。去卵巢12w后,大鼠骨量进一步降低,骨小梁宽度先变窄(4w)后变宽(8、12、18w),骨形成和骨吸收均增加,去卵巢后大鼠在前一个月内(4w)骨量(Tb.Ar%)降低64.18%,差异有显著性(P〈0.05)。结论大鼠去卵巢4w后骨量降低,骨结构变差,骨形成和骨吸收增加,骨转换增加,去卵巢12w到18w骨量持续丢失,但较缓和,提示去卵巢后骨丢失在前4。最快更明显。因此,用于药物研究可选择去卵巢后4w时间进行。  相似文献

目的:探讨卵巢切除雌激素缺乏时鼠及异常应力鼠不同部位骨密度的变化规律。材料与方法:3个月龄SD雌性大鼠64只,双侧卵巢切除(OVX)及行假手术(Sham-O);于术后第1、3、5、7个月各处死8只动物,切取L1~L2椎体节段、股骨及胫骨,进行BMD(骨矿密度)检测;新生24~72小时雌性SD鼠48只,建立双后肢大鼠模型,椎体、股骨及胫骨均承受较高的应力。3月龄时再同法分组,术后第2、5、7个月时分别处死动物取上述部位进行BMD检测。结果:OVX后第1个月椎体BMD明显骨丢失(P<0.01),股骨丢失较为缓慢,7个月时方呈现差异(P<0.05)。松质骨区丢失时间提前(P<0.05)。胫骨1个月BMD即有差异性(P<0.05),松质骨区则呈持续丢失。结论:鼠OVX后BMD的丢失速度为腰椎>胫骨>股骨;腰椎体>胫骨上段>股骨上段。双后肢大鼠组亦呈现同样变化,但腰椎、股骨及胫骨上的应力增加,BMD的丢失较缓慢。  相似文献

目的 观察大鼠去除卵巢后不同部位骨的骨组织计量学参数与骨密度以及两之间的相关性。方法 分别进行卵巢去除和伪手术,56d时处死大鼠,取离体胫骨,股骨测量骨密度与组织学参数%Tb.Ar,Tb.Th,Tb.N。结果 OVX后5d时,股骨远端和胫胫骨近端大部分骨组织计量学参数下降;全股骨和股骨远端1/3处骨密度降低;股骨部分骨组织计量学参数与骨密度测量值之间存在正相关性且差异有显性。结论 切除大鼠卵巢56d时,大鼠骨组织计量学参数下降显,骨密度测量以股骨较为敏感,骨组织计量学与骨密度测量值之间的相关性以股骨明显。  相似文献

Summary Calcium homeostasis and bone pathology were studied in weanling rats fed a low (70 ppm) magnesium diet for 2–21 days. The rats developed significant, progressive hypercalcemia after 6 days on the diet. The increase in blood calcium was accompanied by progressive hypoactivity of the parathyroid gland (PTG), as determined by histologic and morphometric analyses. Thus hyperactivity of the PTG could not have been responsible for the hypercalcemia observed. Histologic examination of femora and humeri from magnesium-deficient rats showed progressive subperiosteal hyperplasia, consisting of undifferentiated osteoprogenitor cells and fibrous tissue, after 7 days of deficiency. The presence of unmineralized osteoid tissue in the metaphyses indicated that mineralization was not proceeding normally. The alterations in differentiation of osteoprogenitor cells, together with the failure of mineralization, resulted in significantly lower rates of bone formation (as measured by fluorochrome labeling) in the magnesium-deficient rats. Basophilic cementing lines and inactive osteocytes in the cortices of bones from magnesium-deficient rats indicated that bone resorption was also severely reduced in magnesium deficiency. We postulate that bone magnesium depletion (66% by day 21) has a direct negative effect on osteoblastic and osteocytic activity, and may explain, in part, the decreased responsiveness of bone to parathyroid hormone (PTH) that has been observed in magnesium-deficient animals.  相似文献

目的探讨使用microPET/CT扫描仪对骨质疏松非创伤性探查的可行性,分析去卵巢大鼠雌激素缺乏引起骨质疏松的骨代谢变化。方法 12只6月龄雌性未孕Sprague-Dawley大鼠,体重290 g~310 g,随机分为两组(n=6):去卵巢组(OVX)和假手术组(SHAM)。用microPET/CT扫描仪进行骨显像,检测大鼠第4腰椎椎体、左股骨近端,左股骨干以及左胫骨中段对核素显像剂18F-F-的摄取。骨显像检测后腹主动脉放血法处死大鼠,取子宫称重,收集第4腰椎、左侧股骨和胫骨,用双能X线骨密度检测仪(DXA)测定第4腰椎椎体、左股骨近端,左股骨干以及左胫骨干的骨密度,取大鼠右侧胫骨制备成不脱钙硬组织切片进行形态学观察。结果 OVX组第4腰椎椎体、左股骨近端,左股骨干以及左胫骨干的骨密度比SHAM组有显著性下降。与SHAM组比较,microPET/CT骨显像可见OVX组第4腰椎椎体、左股骨近端,左股骨干以及左胫骨干核素浓聚,去卵巢大鼠骨骼兴趣区PET影像强度增加。两组大鼠胫骨干的骨几何结构参数比较无显著性差异,OVX组大鼠胫骨近端松质骨的结构参数与SHAM组比较有明显变化。OVX组大鼠胫骨近端的骨形成参数矿化表面、矿物质沉积率和骨形成率比SHAM组有显著性升高。两组大鼠胫骨第4腰椎椎体、左股骨近端,左股骨干以及左胫骨干PET影像强度与其骨密度之间呈显著性负相关。结论 MicroPET/CT可用于骨质疏松大鼠的骨代谢研究,为临床诊断骨质疏松新方法研究提供了新思路。  相似文献

The following studies were undertaken to examine whether estrogen deficiency impairs calcium absorption in aged rats, and to determine whether impaired calcium absorption and the level of dietary calcium are related to the degree of bone loss due to estrogen deficiency. Sixty rats were sham operated (Sham) or ovariectomized (Ovx) to make them estrogen deficient and divided into three dietary groups of 10 rats per group: Group 1 (Sham) and Group 2 (Ovx) were maintained on a diet containing 0.5% calcium; Group 3 (Sham) and Group 4 (Ovx) were maintained on a diet containing 0.1% calcium; Group 5 (Sham) and Group 6 (Ovx) were maintained on a diet containing 0.02% calcium. Calcium absorption was measured in all animals at the beginning of the study and 2 weeks, 1 month, 2 months, and 3 months following surgery, then the animals were sacrificed. In Ovx rats fed 0.5% Ca diet, calcium absorption decreased progressively and the decrease became statistically significant 8 and 12 weeks following ovariectomy (P < 0.05). A similar ovariectomy-related impairment of calcium absorption was not observed in animals fed diets with lower calcium content, making the Ovx rat a tenuous model of intestinal calcium malabsorption. Low dietary calcium decreased cancellous bone mineral content and density at the proximal tibial metaphysis and the decrease was augmented by ovariectomy. 本实验目的是观察卵巢切除(OVX)大鼠皮质骨矿盐变化特点,研究其变化机理。选用SD.两月龄大鼠,摘除双侧卵巢,实验期60天。观察了OVX大鼠股骨干近、中和远1/3段矿盐含量的变化及股骨干近1/3段骨质中氨基多糖(GAG)、白蛋白和骨钙素(BGP)含量的变化。研究结果显示:OVX60天大鼠股骨干仅近1/3段矿盐含量显著降低,中和远1/3段矿盐含量无明显改变。股骨干近1/3段骨质中GAG和BGP含量显著增加,白蛋白含显显著降低。结果表明OVX60天大鼠股骨干近1/3段矿盐丢失与局部骨质内GAG含量增加,白蛋白含量和BGP质量降低有关。  相似文献

使用补肾健骨中药龟地散对切除卵巢的雌性大鼠进行不同剂量的药物治疗,并与模型对照组和钙剂组进行比较。实验结果表明:龟地散显提高模型对照组大鼬有密度值(P〈0.05)和骨矿含量(P〈0.05),从而抑制骨矿溶解和丢失。与模型对照组相比,龟地散明显增加骨质疏松大鼠的骨重(P〈0.01),并且对大鼠的骨长、骨直径、骨面积和体重也有一定的影响。龟地散还具有显提高大鼠骨强度(P〈0.01)等方面的作用。单  相似文献

The purpose of this study was to quantify in vivo the impact of different dietary Ca contents on the maternal total skeleton and skeletal sub-areas in adult rats during pregnancy and lactation, using DXA. Twenty-four female Wistar rats (approximately 5 months old) were mated and divided into three groups (n = 8) and fed one of the following diets, varying only in Ca content (LCD: 0.14%, NCD: 0.6% or HCD: 1.2%). Pups were adjusted to 8–9 per dam. Maternal ionic calcium and in vivo bone mineral density (BMD) were measured at the beginning, after delivery and after weaning. Regardless of the diet, ionized calcium decreased from onset to weaning (P < 0.05). At weaning, bone mass decreased 7.3% in NCD, 15% in LCD and 10.5% in HCD from initial values. Total skeleton, whole and proximal tibia and spine BMDs only decreased at delivery in the LCD group (P < 0.05) but, irrespective of the diet, at weaning, they were lower compared to delivery and initial values (P < 0.05). LCD group presented the lowest BMD in the proximal tibia and spine regions (P < 0.05). At birth, pups did not present differences, however, at weaning, LCD pups reached the lowest body weight (P < 0.05), NCD presented the highest body Ca content (P < 0.05) and there were no differences between LCD and HCD. This in vivo study showed that regardless of the dietary calcium content, the maternal skeleton is slightly affected by pregnancy but severely affected by lactation. However, the degree of such response appears to depend not only on dietary Ca content but also on dietary Ca/P molar ratio.  相似文献

The ovariectomized rat has proved to be a most useful model for preclinical testing of potential therapies for osteoporosis. We describe the immediate effects of a single treatment with salmon calcitonin (sCT) on calcium homeostasis and bone turnover markers in 6-month-old sham and ovariectomized (ovx) rats at 15 days postovariectomy. Rats were fasted for 24 h prior to and following administration of 0.3 µg/kg body weight sCT. Blood specimens were collected at 0 (pretreatment), 2, 4, and 8 h. Urine samples were collected during the intervening periods. sCT treatment produced a decrease in blood ionized calcium at 2 h posttreatment in sham and ovx rats (P < 0.001), which was exaggerated in the ovx rats (P < 0.001). Increased parathyroid hormone (PTH) levels (P < 0.001) accompanied the hypocalcemia in ovx rats. Furthermore, PTH levels were significantly higher in ovx rats compared with sham rats for the same ionized calcium range of 1.275–1.300 mmol/l (P < 0.05). sCT treatment in sham rats increased urine hydroxyproline (UHyp) at 6 h posttreatment (P < 0.01). In conclusion, the calcitonin-induced hypocalcemia and secondary hyperparathyroidism was more pronounced in the ovariectomized rats, consistent with the actions of calcitonin in states of increased bone turnover induced by estrogen deficiency. This study highlights the importance of considering the actions of PTH and estrogen status when interpreting changes in calcium homeostasis and bone turnover following treatment with calcitonin in rodent models and provides further evidence for a potential role of estrogen in parathyroid function.  相似文献

目的观察前交叉韧带切断(OA动物模型)对双侧卵巢切除大鼠(OVX绝经后骨质疏松模型)股骨骨折愈合的影响。方法12周龄SD大鼠共70只,分成基础对照组(Basal)、假手术组(Sham)、双侧卵巢切除术组(OVX)、卵巢切除 前交叉韧带切断术组(OVX OA)、假手术 骨折组(Sham F)、卵巢切除术 骨折组(OVX F)、卵巢切除 前交叉韧带切断 骨折组(OA OVX F),每组10只大鼠。所有受试大鼠在处死前第10d天和第4d天分别皮下注射盐酸四环素和钙黄绿素行双荧光标记。基础对照组在实验开始时杀死,其余6组在术后6W杀死,取大鼠右侧股骨标本。然后,分别进行CR摄片、组织形态学染色,以及应用Norland-XR36双能X线骨密度仪(DXA)测量右股骨远段骨密度和中段骨密度,并将股骨远段及骨折段骨痂进行硬组织包埋、切片,作骨组织形态计量学测量。结果①OVX OA组与OVX组比较,股骨远段。BMD和BWTV显著增加;②OVX F组与Sham F组比较,骨痂(股骨中段)BMD和BV/TV显著降低:③OVX OA F组与OVX F组比较,骨痂(股骨中段)BMD利BV/TV无统计学差异。结论①骨质疏松不仅延缓骨折愈合过程,而且降低骨折愈合质量;②在此动物模型中,骨性关节炎延缓股骨骨质疏松的发生,但是,对骨质疏松性骨折的愈合没有确切的促进作用。  相似文献
