Graves’眼病患者血清IL-2、sIL-2R和TGF-β测定的临床意义   总被引:1,自引:1,他引:0  
目的:探讨Graves’眼病患者血清IL-2、sIL-2R和TGF-β水平的变化、病情进展和病因学的关系。方法:对照组30名和Graves眼病30例患者的血清TGF-β含量均采用放射免疫分析;血清IL-2、sIL-2R均采用酶联免疫吸附试验。结果:30例Graves’眼病患者血清IL-2治疗前水平非常显著低于对照组(P〈0.01);经免疫抑制剂治疗6个月后水平较治疗前升高非常显著,与对照组比较已无显著性差异(P〉0.05);sIL-2R含量治疗前非常显著的高于治疗后组和对照组(P〈0.01);经用上述方案治疗后与对照组比较下降非常显著,但仍存在显著性差异(P〈0.05);血清TGF-β浓度在治疗前非常显著地高于对照组(P〈0.01);经治疗后显著下降,但与对照组比较差异仍非常显著(P〈0.01)。结论:Graves’眼病患者血清IL-2、sIL-2R和TGF-β三项血清指标的测定对于了解和认识其发病机理及预估病情有帮助。  相似文献

探讨格雷夫斯病(GD)患者131I治疗前后免疫功能的变化及其与甲状腺功能的关系.用化学发光法对27例GD患者在131I治疗前后分别测定FT3、FT4、TSH和sIL-2R;同时测定37名对照组的sIL-2R.GD患者131I治疗前组血清sIL-2R水平为810±248 kU/L,高于治疗后组(453±302 kU/L)和对照组(410±130kU/L);治疗前组FT3、FT4与sIL-2R的相关系数分别为0.75,0.57,治疗后组FT3、FT4与sIL-2R的相关系数分别为0.71,0.80.GD患者血清sIL-2R水平明显升高,与FTs、FT4有良好的相关性,经131I治疗后,患者血清sIL-2R水平随着甲状腺激素水平的下降而降低.  相似文献

本研究应用酶联免疫吸附试验(ELISA)检测了103例乙型肝炎各型患者及26倒乙肝病毒(HBV)携带者血清中可溶性白细胞介素2受体(sIL-2R)水平,并结合临床资料进行分析。结果发现:各型患者及HBV 携带者sIL-2R 水平均明显高于正常对照(P<0.01~0.001),其升高程度与病情轻重呈正比,在去除危重患者酶—胆分离时谷丙转氨酶(ALT)过低的影响后,sIL-2R 水平与ALT 有良好的正相关关系(r=0.38.p<0.05);作者认为sIL-2R 能够较好地反映疾病的活动状况,动态观察血清sIL-2R 水平的变化,对判断和评价HBV 感染后的病情演变及免疫失衡状态具有重要意义。  相似文献

ELISA 法检测27例急性早幼粒细胞白血病患者(APL)血清白细胞介素2受体(sIL-2R)水平.结果表明APL 患者血清sIL-2R 高于正常对照(P<0.01),经维甲酸治疗后逐渐降低,但治疗中期患者血清sIL-2R 仍高于正常和治疗后(缓解期)患者血清sIL-2R(P<0.05)。治疗后血清sIL-2R 虽略高于正常对照。却已恢复正常范围(P<0.05)。同时发现治疗中WBC 却明显升高,治疗后又下降,L-CFU 数随治疗逐渐降低并与病情同步变化。经相关分析,L-CFU 与血清sIL-2R 成正向相关(r=0.737)(P<0.01),而与WBC总数无明显相关性(P>0.05)。  相似文献

目的探讨sIL-2R在溃疡性结肠炎(UC)中的表达水平。方法采用特异性ELISAsIL-2R检测试剂盒,对25例溃结患者PBMCs体外PHA诱导产生的sIL-2R水平进行测定。结果溃结患者PHA诱导48h的细胞培养上清液中sIL-2R的含量为319.63±164.70U/ml,明显低于正常对照组的451.15±246.93U/ml(P<0.05);而未经PHA诱导的sIL-2R含量,两组无明显差异(UC组263.78±115.05U/ml,对照组236.47±139.10U/ml,P>0.05)。结论溃结患者体外PHA诱导的sIL-2R水平下降,提示其淋巴细胞的免疫活化功能障碍。  相似文献

目的:研究CRF患者血清IL-18、IL-10、TNF-α、sIL-2R的水平变化及临床意义。方法:分别采用RIA和ELISA法检测CRF患者血清IL-18、IL-10、TNF-α、sIL-2R含量,并与正常对照组比较分析。结果:CRF患者血清IL-18、IL-10、TNF-α、sIL-2R明显高于正常对照组(P〈0.01)。结论:CRF患者多种细胞因子水平升高,血清IL-18和TNF-α水平升高提示了CRF的发生发展过程,而IL-10和sIL-2R升高则对其肾脏功能有一定的保护作用。  相似文献

应用ELISA 方法于术前、术后2天、10天、30天动态观察了16例肾移植患者血清中sIL-2R 水平,并与正常人进行了比较。结果显示,患者术前sIL-2R 水平明显高于正常人;术后发生排斥反应者sIL-2R 水平仍很高,未排斥者sIL-2R 下降明显,二者有显著性差异(P<0.01)。以术前水平为参照,动态检测sIL-2R 下降百分率,对早期发现肾移植急性排斥反应具有重要意义。  相似文献

目的:探讨系统性红斑狼疮( SLE)患者治疗前后肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和可溶性白细胞介素-2受体(sIL-2R)的水平变化及对免疫功能的影响。方法:采用固相酶标记化学发光免疫分析技术检测40例SLE患者激素治疗前后血清TNF-α、IL-6和sIL-2R水平,应用免疫速率散射比浊法测定免疫球蛋白G(IgG)和补体3(C3)水平。以30例体检正常者作对照。结果:40例SLE患者治疗前活动期血清IgG、TNF-α、IL-6和sIL-2R水平明显高于正常对照组,而C3水平明显低于正常对照组,均P0.01。经激素治疗后(稳定期),SLE患者血清IgG、TNF-α、IL-6和sIL-2R水平显著下降,C3显著升高,均P0.01,但IgG、C3、TNF-α和IL-6水平仍与正常对照组有显著统计学差异(P0.01)。结论:SLE患者TNF-α、IL-6和sIL-2R表达增加,提示SLE的发病过程涉及免疫紊乱和细胞因子失平衡。  相似文献

The purpose of this study is to demonstrate the feasibility of dynamic renal R2/R2′/R2* measurements based on a method, denoted psMASE‐ME, in which a periodic 180° pulse‐shifting multi‐echo asymmetric spin echo (psMASE) sequence, combined with a moving estimation (ME) strategy, is adopted. Following approval by the institutional animal care and use committee, a block design of respiratory challenge with interleaved air and carbogen (97% O2, 3% CO2) breathing was employed in nine rabbits. Parametrical R2/R2′/R2* maps were computed and average R2/R2′/R2* values were measured in regions of interest in the renal medulla and cortex. Bland–Altman plots showed good agreement between the proposed method and reference standards of multi‐echo spin echo and multi‐echo gradient echo sequences. Renal R2, R2′ and R2* decreased significantly from 16.2 ± 4.4 s?1, 9.8 ± 5.2 s?1 and 25.9 ± 5.0 s?1 to 14.9 ± 4.4 s?1 (p < 0.05), 8.5 ± 4.1 s?1 (p < 0.05) and 23.4 ± 4.8 s?1 (p < 0.05) in the cortex when switching the gas mixture from room air to carbogen. In the renal medulla, R2, R2′ and R2* also decreased significantly from 12.9 ± 4.7 s?1, 15.1 ± 5.8 s?1 and 27.9 ± 5.3 s?1 to 11.8 ± 4.5 s?1 (p < 0.05), 14.2 ± 4.2 s?1 (p < 0.05) and 25.8 ± 5.1 s?1 (p < 0.05). No statistically significant differences in relative R2, R2′ and R2* changes were observed between the cortex and medulla (p = 0.72 for R2, p = 0.39 for R2′ and p = 0.61 for R2*). The psMASE‐ME method for dynamic renal R2/R2′/R2* measurements, together with the respiratory challenge, has potential use in the evaluation of renal oxygenation in many renal diseases  相似文献

本文对30例鼻咽癌患者的四项免疫学指标进行了检测,发现mIL-2R表达减少,sIL-2R和TNF水平升高,NK活性与正常对照无显著差异,提示患者免疫功能抑制。上述指标与鼻咽癌病期的关系有待进一步研究。  相似文献

对新生儿脐血T细胞IL-2R表达率和血清sIL-2R水平进行检测,结果显示:①早产儿脐血T细胞IL-2R自然表达率明显低于足月新生儿和正常儿童组,而后两者间无差异;②经PHA活化后足月新生儿和早产儿T细胞IL-2R表达率皆显著低于正常儿童组,早产儿组也明显低于足月新生儿组;③足月新生儿和早产儿脐血血清sIL-2R水平均明显高于正常儿童组,但前两者间无明显差异;④新生儿组(包括足月和早产儿)脐血T细胞IL-2R自然表达率与血清sIL-2R水平呈直线负相关,而正常儿童组二者间呈直线正相关。  相似文献

Incomplete T cell antigen receptor-mediated signaling induces an unresponsive state known as anergy. Previously, we had shown that anergy can be induced in antigen-primed but not naive T cells. In this report, we found that in vitro primed T cells from IL-2R alpha-deficient mice were resistant to anergy induction in contrast to comparably treated wild-type T cells. This resistance persisted even after proliferation of IL-2R alpha chain-deficient CD4 T cells with high-dose IL-2-IL-2R beta gamma chains interaction. Thus, antigen activation, and/or progression through cell cycle are not sufficient to induce anergy susceptibility in T cells. The high-affinity IL-2-IL-2R interaction appears to play a critical role in this process.  相似文献

T_Ⅱ是中草药雷公藤的一类粗提物,临床治疗类风湿关节炎和系统性红斑狼疮等免疫性疾病效果显著。T_4是进一步从T_Ⅱ中提取出的单体成分。以正常人外周血单个核细胞(PBMC)为实验材料,测定了不同浓度的T_Ⅱ(0.098~3.13U/ml)和T_4(0.039~5ng/ml)对于T细胞功能的影响。结果表明,T_Ⅱ和T_4对T细胞表面IL-2R的表达剂量相关的抑制作用。结果还显示,在药物作用下,IL-2对IL-2R表达的调节作用是很有限的。  相似文献

High serum level of immunoreactive but not biologically active IL-2 was detected 1 day after surgery in patients undergoing major operation (abdominal, open-heart), in proportion to the tissue injury caused by surgical trauma. IL-2 values were highest in those patients who underwent open-heart surgery and received blood transfusions. In all patients they declined in the third and fourth post-operative days. Elevated serum levels of soluble IL-2 receptors (sIL-2R) were already present 1 day after operation, and peaked in the third and fifth post-operative days after mitogen triggering. Blood lymphocytes derived from operated patients secreted reduced amounts of both IL-2 and sIL-2R compared with control lymphocytes. The extent and duration of this reduction were also proportional to the tissue trauma and were affected by blood transfusions. Based on these data we suggest that early post-operative systemic immunological activation (appearance of IL-2 in the serum) is followed by elevation of sIL-2R, which then interferes with IL-2-dependent immunity. Blood lymphocytes are probably not involved in the post-operative immunological activation. The trigger for and the site of IL-2/sIL-2R synthesis are not yet clear.  相似文献

Immunological tolerance to self antigens is a tightly regulated process. Recent work has demonstrated that the forkhead family member Foxp3 is a critical element in the differentiation and function of mouse CD4(+)CD25(+) regulatory T cells (Treg). Recent work has suggested an important role for IL-2 in the development and maintenance of Treg. To directly assess the effect of IL-2 signaling on Treg development and function, we analyzed mice that were genetically deficient in components of the IL-2 receptor (IL-2R). Mice lacking CD25 (IL-2Ralpha) displayed a slight decrease in Treg within the thymus, while peripheral numbers are unchanged. In contrast, we found that mice deficient in CD122 (IL-2Rbeta) had a profound reduction in both thymic and peripheral Treg, coinciding with more rapid development of a fatal lymphoproliferative disease. Expression of a Foxp3 transgene restored Treg and protected against the onset of autoimmunity. Thus, a signal mediated by IL-2Rbeta is essential for the development and homeostasis of Foxp3(+) Treg in vivo.  相似文献
