以铁皮石斛为原料,采用超声-微波协同法提取铁皮石斛多糖。选择液料比、微波时间、微波功率、超声时间为单因素,以铁皮石斛多糖提取率为评价指标,通过响应面试验对超声-微波协同法提取铁皮石斛多糖进行优化。确定最佳工艺参数为液料比50∶1(mL/g)、微波时间120 s、微波功率370 W、超声时间30 min,在此条件下铁皮石斛多糖提取率为30.56% 。根据体外抗氧化活性评价表明,铁皮石斛多糖对羟自由基的清除能力低于维生素C,对α-葡萄糖苷酶表现出明显的抑制能力,具有较好的体外降血糖作用;另外,铁皮石斛多糖对α-淀粉酶也有一定的抑制作用。  相似文献

优化铁皮石斛多糖的提取工艺。分别考察单因素料液比、微波功率、提取时间和提取温度的影响,并在单因素基础上进行了四因素三水平正交设计试验。结果表明,铁皮石斛多糖的最佳提取工艺为料液比为1∶45(g/mL),微波功率为900 W,提取时间30 min,提取温度95℃。在最佳条件下,铁皮石斛多糖的提取率达到9.77%。该提取工艺简单、快速、高效,可以应用于从铁皮石斛中提取多糖物质。  相似文献

应用微波-超声波辅助乙醇法和水提法分别提取枣渣总黄酮和多糖。实验结果表明:枣渣黄酮提取的较优条件为:提取温度65℃、超声提取时间30min、固液比1∶20、乙醇浓度70%,在此工艺条件下黄酮含量为1.50%。枣渣多糖最佳提取工艺为:浸提温度90℃,浸提时间3h,固液比1∶20,粗多糖提取率2.17%,多糖含量达63.64%。  相似文献

对豆渣中水溶性大豆多糖进行超声辅助萃取,以获得其超声辅助萃取大豆豆渣多糖的优化工艺。以豆渣为原料,在pH值为6.0的条件下,从提取时间、液固比、超声功率及提取温度等因素分析其对豆渣可溶性大豆多糖得率的影响,并通过正交试验优化超声波辅助萃取豆渣中可溶性大豆多糖的工艺。最佳萃取工艺条件为:液固比20∶1、温度60℃、时间30min及功率70W。此条件下豆渣中可溶性大豆多糖提取率可达到最大,为0.019%。在影响大豆豆渣多糖提取率的4个因素(提取温度、液固比和提取时间、超声功率)中,液固比、提取时间为主要因素,其次是温度和超声功率。超声波辅助萃取可有效提高豆渣中可溶性大豆多糖得率。  相似文献

研究了微波辅助法提取羽状蕨藻中蕨藻红素的工艺条件。通过单因素实验探究了微波温度、乙醇浓度、微波时间、微波功率、料液比五个因素对蕨藻红素提取率的影响,并经过正交实验对影响蕨藻红素提取率的参数条件进行优化。确定微波提取最佳工艺条件为:提取溶剂90%乙醇,微波温度50℃,微波功率300W,微波时间15min,料液比1∶40g·mL-1,按此工艺条件进行重复性验证实验得到蕨藻红素的平均提取率为92.93%,RSD为0.5003%。结果表明微波提取率高,稳定性好,且大大缩短了提取时间。  相似文献

探索花生衣原花色素微波辅助提取的最佳条件,在单因素试验的基础上,应用响应面法优化花生衣中原花色素的提取条件;研究发现料液比和萃取温度对花生衣原花色素的提取率都有显著影响,且非线性关系,最佳提取工艺参数为微波输出功率600W、乙醇体积分数40%、料液比1:24.5、提取时间91s、提取温度40℃。在此条件下原花色素的提取率为12.59%。表明响应面法对花生衣原花色素提取条件的优化是可行的,得到的原花色素提取条件具有实际应用价值。  相似文献

为提高铁皮石斛的综合利用率,建立一种绿色高效的铁皮石斛多糖提取方法。本研究以铁皮石斛多糖提取率为指标,通过单因素考察了低共熔溶剂浓度、提取温度及液料比对铁皮石斛多糖提取率的影响,采用响应面设计优化铁皮石斛多糖的提取工艺,并对纯化后的多糖进行结构分析。结果表明:响应面优化后得到最佳工艺为低共熔溶剂(deep eutectic solvent,DES)浓度40%、提取温度80℃、液料比110:1(mL/g),在此条件下实际提取率为33.2%±0.28%,与预测值33.5%接近,多糖的纯度为56.95%±1.2%。多糖经阴离子交换柱及葡聚糖凝胶柱纯化后,纯度可达90.8%,其单糖主要由葡萄糖和甘露糖构成,质量比约为43:37,此外还含有少量的木糖、鼠李糖、核糖等,结构中同时含有α糖苷键和β糖苷键。本研究提供了一种低共熔溶剂提取铁皮石斛多糖的高效绿色提取方案,具有多糖提取率高及绿色的特点,为后续铁皮石斛多糖的开发提供了借鉴。  相似文献

研究酶-微波联用法提取枸杞总黄酮的最佳工艺条件。采用纤维素酶提取枸杞总黄酮,在单因素试验基础上利用正交试验研究乙醇浓度、酶解温度和酶解p H值对提取率的影响,确定最佳酶解条件。在单因素试验基础上,通过正交试验确定微波提取的最佳工艺参数,采用微波法进一步提取酶解液中的枸杞总黄酮。酶解阶段最佳工艺参数为:乙醇浓度45%,酶解温度50℃,酶解p H4.0;微波阶段的最佳工艺参数为:液固比30 m L/g,微波温度65℃,微波时间4 min。该酶-微波联用法提取枸杞总黄酮的提取率达1.560%。  相似文献

以香菇多糖为研究对象,采用单因素分组实验法对香菇多糖提取工艺(热水法和微波提取法)进行了初步的探讨,比较了时间,浸取温度,浸取次数,液固比等因素对多糖提取率的影响。其中传统热水提取法的最佳工艺条件为:液固比10,加热温度45℃,加热时间5h,提取二次。微波法提取香菇多糖的最佳工艺条件:液固比10,功率40%,加热时间180s,pH值1。通过两种最佳工艺条件的结果比较可知,微波萃取法具有节能、省时、环保、操作便利且提取率高等优点。  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
