A novel four-step methodology for the synthesis of 5-acyl-1,2-dihydropyrimidin-2-ones has been developed. The reaction of readily available N-[(1-acetoxy-2,2,2-trichloro)ethyl]ureas with Na-enolates of 1,3-diketones or β-oxoesters followed by heterocyclization-dehydration of the oxoalkylureas formed gave 5-acyl-4-trichloromethyl-1,2,3,4-tetrahydropyrimidin-2-ones. The latter, in the presence of NaH, eliminate CHCl3 to give the target compounds. 相似文献

The regioselective synthesis of cis and trans stereoisomers of variously functionalized octahydro[1,2,4]triazolo[4,3-a]quinazolin-5-ones was performed. The 2-thioxopyrimidin-4-ones used in the synthesis reacted with hydrazonoyl chlorides in a regioselective manner to produce the angular regioisomers [1,2,4]triazolo[4,3-a]quinazolin-5-ones rather than the linear isomers [1,2,4]triazolo[4,3-a]quinazolin-5-ones. The synthesis process took place with electronic control. The angular regiochemistry of the products was confirmed by X-ray experiments and two-dimensional NMR studies. 相似文献

One-pot efficient synthesis of functionalized 5,6-dihydropyran-2-ones has been delineated by reacting 2H-pyran-2-ones and acetol in the presence of a base at room temperature. The formation of 5,6-dihydropyran-2-ones revealed that the reaction proceeded in a unique ‘ring transformation-rearrangement’ sequence. 相似文献

Azetidin-2,3-diones have been used as synthons for the synthesis of C-3 alkylidene/alkylazetidin-2-ones. Some of the 3-alkylazetidin-2-ones are well known for their cholesterol absorption inhibitor activity. A regio and stereoselective Grignard reaction on a keto group followed by dehydration using PPh3/CCl4 reagent is a key step in this synthesis. Hydrogenation of the 3-alkylideneazetidin-2-ones provided stereoselectively cis-3-alkylazetidin-2-ones in very good yields. 相似文献

The composition and yields of the products from the reaction of 4,4-dialkyl-5-methylene-1,3-dioxolan-2-ones with amidines depend on the structure of the initial amidine and on the reaction conditions. 2-Aminopyridines lead to 3-substituted 4-hydroxy-4-methyloxazolidin-2-ones and 4-methylene-oxazolidin-2-ones and also to sym-carbamides. 2-Amino-4,6-dimethylpyrimidine leads to the corresponding 4-methyleneoxazolidin-2-one. 3-Aminothiazoles give linear oxourethanes and sym-carbamides. 相似文献

The interaction of sulfonamides with 4,5-dihydroxyimidazolidin-2-ones was studied for the first time. The earlier unknown
4,4′-sulfonyldiiminobis(1,3-dialkylimidazolidin-2-ones) and 4(5)-aryl(alkyl)-sulfonyliminoimidazolidin-2-ones were synthesized.
A probable pathway of the reaction was proposed. 1,3-Diethyl-4,5-dihydroxyimidazolidin-2-one was isolated and described.

An efficient synthesis of two new series of 6-aryl-4-trifluoromethyl-2H-pyran-2-ones and 6-aryl-4-carboxyethyl-2H-pyran-2-ones, obtained through the self-condensation reaction of 5-aryl-5-methoxy-3-(trifluoromethyl)penta-2,4-dienenitriles 3 and ethyl 4-aryl-2-(cyanomethylene)-4-methoxybut-3-enoates 4 respectively, is reported. The self-condensation reaction of the enoates 4 was performed in water in the presence of hydrochloric acid whereas the self-condensation reaction of the penta-2,4-dienenitriles 3 required the use of zinc bromide and hydrochloric acid in order to give the respective 2H-pyran-2-ones. Products were obtained up to 97% yield. 相似文献

The Michael condensation in the series of 5-aryl-3-arylidene-3H-pyrrol-2-ones and 5-aryl-3-arylidene-3H-furan-2-ones, containing
an activated C=C bond, with cyclohexanone was investigated. It was shown that the condensation products were 1,5-dicarbonyl
compounds containing a heterocyclic fragment. The enolization of one of the oxo groups, leading to the formation of hydroxypyrrole
or hydroxyfuran structures, was demonstrated by the spectral data.

The one-pot substituent-directed regioselective synthesis of 1,7-diaryl-2-methyl-4H,5H-pyrano[3,4-c]pyran-4,5-diones 3 as the major and 3,4-diaryl-2-methyl-6-methylsulfanylbenzonitriles 4 as the minor products has been delineated through ring transformation of suitably functionalized 2H-pyran-2-ones 1 with aryl acetones 2. A new synthesis of 2,3-dialkyl-4-carbomethoxyisoquinolin-1(2H)-ones and 6,7-dialkyl-8-carbomethoxy-1,6-naphthyridin-5(6H)-ones is reported. The process involves treatment of a β-enaminoester with 2-fluoro-5-nitrobenzoyl chloride, 2-fluorobenzoyl chloride or 2-chloronicotinoyl chloride followed by heating in the presence of base. The conversion, which proceeds by an N-acylation-SNAr reaction sequence, affords 50–86% yields when R1 is n-alkyl but ≤30% yields when R1 is α-branched. 相似文献

A five-step synthesis of 4-chloromethyl- and 4-mesyloxymethyl-5-tosyl-1,2,3,4-tetrahydropyrimidin-2-ones has been developed. The reaction of N-[(2-benzoyloxy-1-tosyl)ethyl]urea with sodium enolates of α-tosylketones followed by cyclization-dehydration, and debenzoylation gave 4-hydroxymethyl-5-tosyl-1,2,3,4-tetrahydropyrimidin-2-ones, which were transformed into the 4-chloromethyl- or 4-mesyloxymethyl-derivatives. Treatment of the latter with nucleophilic reagents, such as sodium cyanide, sodium diethyl malonate, sodium thiophenolate, or potassium phthalimide, afforded the corresponding 4,7-disubstituted 6-tosyl-2,3,4,5-tetrahydro-1H-1,3-diazepin-2-ones as a result of ring expansion. The effect of the leaving group and the substitution at the position C6 on the reactivity of the pyrimidines is discussed. 相似文献

on treatment with carboxylic acids and POCl3. 3-Arylmethylidenamino-2-thioxo-1,2,3,4-tetrahydroquinazolin-4-ones also cyclized to 2-aryl-5H-[1,3,4]thiadiazolo[2,3-b]quinazolin-5-ones
when oxidized with potassium chlorate in acetic acid, but on heating they were deaminated to give 2-thioxo-1,2,3,4-tetrahydroquinazolin-4-one
and aryl nitriles.

A three-stage diastereoselective synthesis of (4R∗,5R∗,6S∗)-5-mercapto-4-methyl-6-phenylhexahydropyrimidin-2-one has been developed. The first stage involves the formation of 4-hydroxy-5-(4-methoxybenzylthio)-4-methyl-6-phenylhexahydropyrimidin-2-one by the reaction of N-[(phenyl)(tosyl)methyl]urea with the sodium enolate of 1-(4-methoxybenzylthio)propan-2-one. Reduction of the obtained compound by NaBH4-CF3COOH and removal of the p-methoxybenzyl protecting group results in the target compound. 相似文献

The interaction of different 3-acetylfuran-2(5H)-ones (1, 4) with dimethylformamide dimethyl acetal (DMF/DMA) has been investigated, and the condensed furopyridine system 3 was obtained. By means of interaction of compounds 4 and DMF/DMA via the 3-acetylfuran-2(5H)-one enamines, the heterocyclic substituted furanones are obtained. The condensation of 3-acetyl-4,5,5-trimethylfuran-2(5H)-one (1a) with veratraldehyde was realized. Through the interaction of 1a with malononitrile and further intramolecular condensation of the product, 6-amino-dihydroisobenzofuran-5-carbonitrile was also synthesized. 相似文献
