新疆荒漠昆虫光滑鳖甲cDNA文库的构建及功能基因筛选   总被引:6,自引:1,他引:5  
以过冬后早春的新疆荒漠拟步甲属昆虫光滑鳖甲(Anatolica Polita borealis)成虫为研究材料,应用SMARTTM cDNA Library Construction技术,通过总RNA提取,反转录合成cDNA,定向构建至噬菌体载体λTripEx2,经体外包装构建了光滑鳖甲的cDNA表达文库。测试结果表明库容量为2.2×106,重组率为84.8%。通过对cDNA文库克隆的序列测定和初步生物信息学分析,获得28个光滑鳖甲表达序列标签(ESTs),包括17个EST(60.7%)与NCBI中已注册的已知功能基因相似性较高,另外的11个EST(39.3%)则没有发现与之相似的序列,推测可能是功能未知的新基因。同时,利用PCR扩增文库技术从cDNA文库中克隆获得了光滑鳖甲的抗冻蛋白基因,初步表明光滑鳖甲cDNA表达文库构建的成功,为深入研究新疆荒漠昆虫的抗冻机理及发现新的昆虫功能基因奠定了基础。  相似文献

以西藏八角莲(Dysosma tsayuensis Ying)和桃儿七[Sinopodophyllum hexandrum(Royle)Ying]为材料,构建其根及根茎的SSH文库,从中筛选鬼臼类植物属种间与鬼臼毒素生物合成相关的差异表达基因。从文库中随机挑取201个阳性克隆测序后得到183条ESTs。去除载体序列和冗余序列,聚类拼接得到17个西藏八角莲的unique ESTs。经BLAST同源比较和功能查寻,有功能注释的unique ESTs共12个,占70.6%,所编码的蛋白涉及光合作用、合成代谢、转录调控等功能;无功能注释和匹配结果的共5个,占29.4%。该研究成功构建了西藏八角莲和桃儿七SSH文库,为进一步揭示鬼臼毒素生物合成途径及其调控机制奠定了基础。  相似文献

通过构建香猪肌肉组织cDNA文库,并在文库中随机挑选克隆进行测序的方法,获得了131个香猪肌肉EST序列.在这131个EST序列所代表的109个单一克隆中,有99个为人类及其他物种的同源序列,3个为已知的猪的ESTs,7个为未知ESTs.对这10个已知、未知ESTs进行开放阅读框预测并进行B1ast分析,没有找到高度同源的氨基酸序列.对上述EST所对应的基因功能分析结果表明,除去27.27%的EST未能分类外,克隆到的EST大多来自与基因/蛋白的表达调控相关的基因(占45.46%).来自具有其他功能的基因的EST依次是细胞代谢占10.10%、细胞结构/迁移占10.10%、细胞/机体防御占5.05%和细胞信号/传导占2.02%.没有发现和细胞分裂相关的已知功能基因.本研究结果为中国地方品种香猪提供了第一个骨骼肌的基因表达谱,为今后寻找猪肌肉生长和肉用品质的候选基因奠定了基础.  相似文献

大白菜霜霉菌诱导抑制性消减杂交cDNA文库的构建和分析   总被引:2,自引:0,他引:2  
以抗霜霉病大白菜双单倍体系(DH)‘T12-19’为材料,构建了霜霉病诱导表达的正向抑制性消减文库,并利用反向Northern斑点杂交技术对768个阳性克隆进行了筛选,共获得57个病原菌诱导上调表达的克隆。测序后得到55条通读表达序列标签(ESTs),对这些ESTs序列进行聚类和拼接分析,共获得50个unigenes。Blast分析表明,37个unigenes与已知基因高度同源,占全部非重复序列的67.3%。对已知功能基因按MIPS的分类方法进行功能分类,发现这些基因的功能主要涉及物质与能量代谢、转录调控、蛋白质合成与代谢、膜及转运、信号转导、抗病防御等。为了验证文库筛选结果的可靠性,采用实时荧光定量PCR技术分析了其中2个克隆BFCH10和BFIA7的表达谱。结果表明,这2个克隆在接种病菌6h后明显上调表达,与反向Northern斑点杂交结果基本一致。  相似文献

白粉病是我国小麦的主要病害之一。尝试用表达序列标签 (expressedsequencetags,EST)技术 ,研究了经白粉病菌诱导后的小麦基因表达。从构建的普通cDNA文库中随机挑取约 15 0 0个阳性克隆并进行测序 ,获不重复ESTs序列 387条。不重复序列均获GenBank的存储号。其中 4 9.4 %的序列与已知基因同源 ,196条序列功能未知 ,84条序列为新ESTs。将不重复序列制备成高密度点阵膜 ,用差示杂交法筛选到几个抗病相关序列。  相似文献

为筛选东方山羊豆盐诱导差异性基因,以250 mmol/L NaCl处理的东方山羊豆cDNA为实验组,未经诱导刺激的为驱动组,利用抑制性消减杂交技术(SSH)构建消减文库并对其部分阳性克隆进行了ESTs序列分析。该消减文库克隆的重组率为92%,插入片段大部分集中在0.2~1kb之间。随机挑取500个克隆进行测序及同源性分析,获得258个cleanESTs,经聚类、拼接,去除冗余序列,共获得132个Unigene,其中含有32个contigs和100个singlets,该文库的冗余度为24%。对其进行功能预测及分类,得到大量参与信号转导、转录调控、渗透和代谢调节、机体防御以及光作用等过程的相关基因。随机选择非重复的4个差异表达的序列设计引物,以半定量PCR方法验证其消减效率,结果显示,诱导组的表达量显著高于非诱导组,表明该文库有较高质量,且所采用的技术手段有助于快速发现东方山羊豆新功能基因。  相似文献

目的:为了获得功能基因的信息,构建烤烟品种南江3号的均一化cDNA文库.方法:用RNeasy Plant Mini Kit提取烤烟南江3号叶片和花的RNA,用反转录酶逆转录合成第一链eDNA.以LD-PCR扩增获得的双链cDNA为模板,采用基于双链特异性核酸酶(Duplex-Specific Nuclease,DSN)的均一化cDNA文库技术,构建南江3号盛花期的均一化cDNA文库.结果:构建了南江3号盛花期的均一化cDNA文库,文库重组率大为97.47%,库容量约为1.26×10<'6>,插入片段平均长度大于1.2kb.从文库中随机48个克隆进行PCR检测,挑取20个克隆进行测序,序列通过BLAST比对结果显示文库可能包含大量基因和ESTs序列.结论:该文库为研究南江3号的基因功能和资源提供材料来源.  相似文献

Darwinian theory predicts that organisms will display traits that benefit themselves rather than other individuals; exceptions to this rule usually are explicable by kin selection. Our studies on an insular population of venomous snakes in north-eastern China reveal a different situation. Only one species of snake (Gloydius shedaoensis, Viperidae) occurs on the island of Shedao, and displays altruism between size (age) classes. First, small snakes frequently kill prey items larger than they can swallow themselves. This behaviour enhances rates of feeding of larger conspecifics, which scavenge the birds' carcasses. Second, large snakes kill raptorial birds (sparrowhawks Accipiter nisus) that pose little or no threat to themselves. This behaviour reduces predation risk for smaller snakes. These effects are presumably accidental consequences of the high venom toxicity of the pit-vipers, which enable them to kill inedible prey and non-threatening predators at little cost. Nonetheless, this accidental altruism may have significant ecological consequences. For example, these behaviours may contribute to the remarkably high population densities of snakes on Shedao.  相似文献

采用荧光染料(Cy5)标记中国辽宁蛇岛蝮(Gloydius shedaoensis shedaoensis,GSS)蛇毒(snake venom,SV,GSS-SV)蛋白质,获得了该蛇毒的双向十二烷基硫酸钠聚丙烯酰胺凝胶电泳(2D SDS-PAGE)图谱,经DeCyder软件分析,分辨出1000多个蛋白点,分子量范围在10~150ku间,等电点在4~7的蛋白质点占78.8%。凝胶后染色(post-staining)采用蛋白荧光染料Deep Purple,选取的5个蛋白点经胶内酶解,产生的肽段经高效液相色谱-电喷雾串联质谱(high performance liquid chromatography/HPLC-electro-spray ionization tandem mass spectrometry/ESI-MS/MS,HPLC-ESI-MS/MS)进行序列测定,质谱数据经Sequest Bioworks软件分析,为蛇毒L-氨基酸氧化酶、金属蛋白酶、类凝血酶、纤溶酶原激活物和磷脂酶A2的同源蛋白。本研究采用的荧光标记2DSDS-PAGE结合HPLC-ESI-MS/MS的技术适于高通量研究蛇毒蛋白组成。  相似文献

目的进一步了解蛇岛蝮蛇的种群动态,深入探讨全岛的种群密度和分布格局对捕食率的影响。方法结合蛇岛蝮蛇种群数量调查,对全岛所有区域的各种生境蝮蛇数量进行标记统计,每1~2天进行一个沟溪全面踏查,分别记录春秋两季不同区域、不同生境的蛇岛蝮蛇不同年龄组的分布数量和捕食状况,进行比较。结果2005~2006年蛇岛蝮蛇种群密度为0.0278条/m2,春秋两季是候鸟迁徙和捕食高峰时期,灌木林是蛇岛蝮蛇捕食栖息的主要场所,占全部种群的63.38%,其次是稀树草地27.04%,乔木林和裸地分别为5.86%、3.72%。蛇岛蝮蛇春季捕食持续时间短,捕食率10.92%,秋季捕食持续时间长,捕食率为8.61%。结论应密切关注候鸟迁徙的动态。  相似文献

目的了解影响蛇岛蝮蛇树栖分布的7个相关因子,探讨其间的关系。方法2006年10月12~15日,在蛇岛二沟南坡、北坡、西坡,测定不同年龄组蛇岛蝮蛇树栖环境的7个相关因子,进行比较分析。结果三种树栖环境中,老成体、成体、亚成体、幼体对因子的选择基本都有差异;不同年龄组的树栖高度与冠幅负相关不显著,与枝长(成体和亚成体中)负相关不显著,与其它正相关不显著;老成体和成体在林中和林缘,对树栖高度的选择差异显著,亚成体和幼体差异不显著;相同年龄组两两因子间线性关系明显。结论影响蛇岛蝮蛇树栖分布的因子具有一定的规律性。  相似文献

Snake venom thrombin-like enzymes (SVTLEs) are widely applied in the treatment of thrombotic diseases, however, the molecular mechanism of its inhibition by synthetic and natural proteinaceous inhibitors is not yet understood. Here we investigated effects of protease inhibitors including phenylmethylsulfonil fluoride (PMSF), benzamidine (BMD) and its derivates on the activity of recombinant gloshedobin, a SVTLE from the snake Gloydius shedaoensis. The molecular inhibition mechanism was postulated by separately docking inhibitors into three-dimensional model of gloshedobin using protein C activator from Agkistrodon contortrix contortrix venom (ACC-C, which bear 78% identity with gloshedobin) as template. The analysis indicated that the strongest inhibitor, PMSF, was via a covalent bond with the catalytic Ser195, while other inhibitors showing weaker inhibitory activity were via hydrogen bond with Ser195 or non-catalytic residues.  相似文献

采用ABC(avidin-biotin-peroxidase complex)免疫组织化学方法,观察蛇岛蝮蛇(Gloydius shedaoensis)消化道内5-羟色胺(5-HT)免疫阳性内分泌细胞的分布及形态.结果显示,5-羟色胺细胞从食管到直肠各段均有分布.细胞分布密度呈波浪式,其中胃贲门部分布密度最高(7.15±2.38),直肠部次之(4.55±3.14),食管部最低(1.2±0.71).5-HT阳性细胞广泛分布于消化道上皮细胞之间、上皮基部、腺泡上皮细胞之间以及固有膜内.形态多样,呈圆形、锥体形、梭形等.分析认为蛇岛蝮蛇消化道5-HT细胞具有内、外分泌两种作用途径,并且其密度分布可能与其食性、生存环境有关.  相似文献

The total RNA was extracted from the venom gland of snake, Gloydius shedaoensis. The defibrase cDNA was amplified by RT-PCR. Assaying the nucleotide sequence of the cDNA allowed the complete amino acid sequence for defibrase of Gloydius shedaoensis to be determined. The defibrase cDNA was inserted into the vector, pPIC 9K, and expressed in Pichia pastoris which then produced 10 mg target protein l–1.  相似文献

  • 1.Do thermal factors influence foraging-site selection by ectothermic predators? Snake species that obtain their prey from ambush must remain immobile for long periods, precluding overt behavioural thermoregulation; and some “ambush” snakes use thermal cues to detect endothermic prey. Plausibly, alternative ambush sites might differ either in equilibrial body temperatures available to snakes, or in the thermal “background” against which prey items must be detected.
  • 2.We examined this topic with field data on pit-vipers (Gloydius shedaoensis) on a small island in northeastern China. Adult snakes feed only on migrating passerine birds. The snakes ambush birds both from arboreal perches (branches of small trees) and from the ground.
  • 3.Arboreal versus terrestrial ambush sites differed both in operative temperatures and thermal “backgrounds” available to the snakes. Operative temperatures inside copper models were lower in trees than on the ground (because of wind), and snakes in arboreal ambush sites were cooler than those in terrestrial sites. Thermal backgrounds from arboreal perches were cooler (and thus, provided more contrast against prey items) than did backgrounds available from terrestrial ambush-sites.
  • 4.Thermal factors thus modify the suitability of alternative ambush locations for these pit-vipers, but with a trade-off: a snake in a tree can “see” its prey more clearly, but may not be warm enough (and hence, able to strike fast enough) to capture it. Further work is required to determine whether or not snakes actually use such thermal differences as criteria for the selection of ambush sites.

A protein with the activity of phospholipase A2 named asAPLA2 was purified to homogeneity from the venom of Agkistrodon shedaoensis Zhao through DEAE-Sepharose CL-6B anion exchange column, Source S and Mono Q FPLC. Its molecular weight was estimated as 19 kD by SDS-PAGE and its pI was about 3.5 by IEF analysis. It inhibits the platelet aggregation that was induced by 1 μmol/ L ADP, and the IC50 was determined to be 6 μmol/L. Degenerate primer was designed and synthesized according to the N-terminal amino acid sequence of asAPLA2. Its full-length cDNA was cloned by RT-PCR from the total RNA extracted from the snake venom gland. According to the deduced amino acid sequence, its molecular weight and pI are determined to be 13,649 and 4.39 respectively as calculated by DNAclub and DNAstar softwares. The gene was then cloned into the expression plasmid pET-40b( ) and expressed in E. coli BL21(DE3). Western blot analysis indicated that the expressed protein cross-reacted with the antibody against the native  相似文献
