BACKGROUND: The aim of the work presented was to synthesize a series of amphiphilic hyperbranched poly[(amine‐ester)‐co‐(D ,L ‐lactide)] (HPAE‐co‐PLA) copolymers and study the formation of copolymeric micelles. These copolymeric micelle systems are expected to be potential candidates for applications in protein drug delivery. RESULTS: The chemical structures of the copolymers were confirmed by Fourier transform infrared spectroscopy, 13C NMR and thermogravimetric analysis. Fluorescence spectroscopy and dynamic light scattering confirmed the formation of copolymeric micelles of the HPAE‐co‐PLA copolymers. The maintenance of stability of bovine serum albumin (BSA) during release from micelles in vitro was also measured using circular dichroism and fluorescence spectrometry. CONCLUSION: Novel hyperbranched HPAE‐co‐PLA copolymers have been synthesized. Conjugation of PLA to HPAE was proved to be an available method for the preparation of micelles for protein delivery. The BSA‐loaded micelles showed enhanced encapsulation efficiency and the structural stability of BSA was retained during the release process. The hyperbranched polymeric micelles could be useful as drug carriers for protein drug delivery systems. Copyright © 2008 Society of Chemical Industry  相似文献

This article describes the synthesis and characterization of 2‐hydroxylethyl methacrylate‐coN‐vinylpyrrolidone copolymers, (HEMA‐co‐NVP), via free radical polymerization followed by grafting of poly(lactide) onto (HEMA‐co‐NVP) copolymers, via ring opening polymerization using tin octoate as a catalyst. The copolymers and the grafted copolymers (i.e., amphiphiles) were subjected to sustained release studies using salicylic acid, as a model drug. Characterization of the formed copolymers was performed using 1H‐NMR, 13C‐NMR, FTIR, TGA, DSC, and SEM techniques. Derivative of TGA thermogram was used to determine %hydrophilicity and %hydrophobicity in the grafted and ungrafted copolymers. The SEM morphology revealed porous layers with crispy structure that were most likely due to the presence of poly(lactide) chains. At lower content of poly(lactide) moiety, grafted copolymers showed non‐Fickian diffusion release rate, whereas Fickian diffusion release rate at higher content of poly(lactide) was observed. The increase of poly(lactide) content (i.e., larger %hydrophobicity) in the copolymer increased the drug‐sustainability, due to the consistent but porous amphiphilic degradable structures that allow controllable release of drug in time interval. © 2011 Wiley Periodicals, Inc. Poly(N‐isopropylacrylamide‐co‐hydroxyethyl methacrylate) [P(NIPAM‐co‐HEMA)] copolymer was synthesized by controlled radical polymerization from respective N‐isopropylacrylamide (NIPAM) and hydroxyethyl methacrylate (HEMA) monomers with a predetermined ratio. To prepare the thermosensitive and biodegradable nanoparticles, new thermosensitive graft copolymer, poly(L ‐lactide)‐graft‐poly(N‐isoporylacrylamide‐co‐hydroxyethyl methacrylate) [PLLA‐g‐P(NIPAM‐co‐HEMA)], with the lower critical solution temperature (LCST) near the normal body temperature, was synthesized by ring opening polymerization of L ‐lactide in the presence of P(NIPAM‐co‐HEMA). The amphiphilic property of the graft copolymers was formed by the grafting of the PLLA hydrophobic chains onto the PNIPAM based hydrophilic backbone. Therefore, the graft copolymers can self‐assemble into uniformly spherical micelles ò about 150–240 nm in diameter as observed by the field emission scanning electron microscope and dynamic light scattering. Dexamethasone can be loaded into these nanostructures during dialysis with a relative high loading capacity and its in vitro release depends on temperature. Above the LCST, most of the drugs were released from the drug‐loaded micelles, whereas a large amount of drugs still remains in the micelles after 48 h below the LCST. © 2011 Wiley Periodicals, Inc. A series of triblock co‐polymers, consisting of a poly(ethylene glycol) (PEG) central block joined to two blocks of random p‐dioxanone‐co‐L ‐lactide copolymers were synthesized by ring‐opening polymerization of p‐dioxanone (PDO) and L ‐lactide (LLA) initiated by PEG in the presence of stannous 2‐ethylhexanoate catalyst. The resulting copolymers were characterized by various techniques including 1H and 13C NMR and FTIR spectroscopies, gel permeation chromatography, inherent viscosity, wide‐angle X‐ray diffractometry (WAXD) and differential scanning calorimetry (DSC). The conversion of PDO and L ‐lactide into the polymer was studied various mole ratios and at different polymerization temperature from 1H NMR spectra. Results of WAXD and DSC showed that the crystallinity of PEG macroinitiator was greatly influenced by the composition of PDO and L ‐lactide in the copolymer. The triblock copolymers with low molecular weight were soluble in water at below room temperature. © 2003 Society of Chemical Industry  相似文献

5‐Fluorouracil (5‐Fu) loaded poly(glycolide‐co‐lactide‐co‐caprolactone) (PGLC) nanoparticles were prepared by modified spontaneous emulsification solvent diffusion method (modified‐SESD method) and characterized by dynamic light scattering, scanning electron microscopy and 1H NMR determination. It was found that the obtained nanoparticles showed near spherical shape and was controllable with the radius range of 30–100 nm. Compared with the nanoparticles prepared by polylactide and poly (lactide‐co‐glycolide) (PLGA) under the similar preparation condition, yield of PGLC nanoparticles was the highest, which reached to about 100%. On the other hand, drug entrapment efficiency of PGLC nanoparticles was also higher than that of PLGA and PLLA nanoparticles. 5‐Fu release behavior of PGLC nanoparticles in vitro showed that 5‐Fu release of PGLC nanoparticles showed a near zero‐order release profile, and 5‐Fu release rate of PGLC nanoparticles was faster than that of PLLA and PLGA nanoparticles. According to degradation behavior of PGLC nanoparticles, it could be proposed that the kinetic of degradation controlled release played an important role in the release process of PGLC nanoparticles. Surfactant‐free nanoparticles of poly(DL ‐lactide‐co‐glycolide) (PLGA) nanoparticles were prepared with or without poly(L ‐lactide)‐poly(ethylene oxide) (LE) diblock copolymer (abbreviated as PLGA/LE and PLGA nanoparticles) by dialysis method. LE diblock copolymer was used to make PLGA nanoparticles to alternate conventional surfactant. The size of PLGA and PLGA/LE nanoparticles was 295.3 ± 171.3 and 307.6 ± 27.2 nm, respectively, suggesting LE diblock copolymer might be coated onto the surface of nanoparticles. Observation of scanning electron microscope (SEM) showed that PLGA/LE nanoparticles have spherical shapes ranging ~ 200–500 nm. In 1H‐NMR study, characteristic peaks of the methyl protons of PLGA disappeared in D2O, whereas characteristic peaks of the methyl proton of both PEG and PLGA were shown in both CDCl3 and D2O, indicating that LE diblock copolymer coated on the surface of the PLGA nanoparticles. The higher the initial content of drug, the higher the drug contents and the lower the loading efficiency. PLGA/LE nanoparticles at higher drug contents resulted in slower adriamycin·HCl (ADR) release rate than that of lower drug contents. Also, slower release rate of ADR was achieved by entrapped into the PLGA/LE nanoparticles, whereas LE polymeric micelles showed rapid ADR release. © 2003 Wiley Periodicals, Inc. Two enantiomeric triblock ABA copolymers composed of poly(L ‐lactide)–poly(ethylene glycol)–poly(L ‐lactide) (PLLA–PEG–PLLA) and poly(D ‐lactide)–poly(ethylene glycol)–poly(D ‐lactide) (PDLA–PEG–PDLA) were synthesized with two different middle‐block PEG chain lengths by ring‐opening polymerization of L ‐lactide and D ‐lactide in the presence of PEG, respectively. A pair of enantiomeric triblock copolymers were combined to form a stereocomplex by a solvent‐casting method. The triblock copolymers and their stereocomplexes were characterized by 1H‐ and 13C‐NMR spectroscopy and gel permeation chromatography. Their crystalline structures and crystalline melting behaviors were analyzed by the wide‐angle X‐ray diffraction method and differential scanning calorimetry. The stereocomplex formed between a pair of enantiomeric triblock copolymers exhibited a higher crystalline melting temperature with a distinctive 3/1 helical crystalline structure. PLLA–PEG–PLLA and its stereocomplex with PDLA–PEG–PDLA were used to fabricate a series of microspheres encapsulating a model protein drug, bovine serum albumin (BSA). They were prepared by a double‐emulsion solvent‐evaporation method. A diblcok copolymer monomethoxy poly (ethyleneglycol)‐block‐poly(L ‐lactide‐co‐2‐methyl‐2‐carboxyl‐propylene carbonate) (MPEG‐b‐P(LA‐co‐MCC)) was obtained by copolymerization of L ‐lactide (LA) and 2‐methyl‐2‐benzoxycarbonyl‐propylene carbonate (MBC) and subsequent catalytic hydrogenation. The pendant carboxyl groups of the copolymer MPEG‐b‐P(LA‐co‐MCC) were conjugated with antitumor drug docetaxel and tripeptide arginine‐glycine‐aspartic acid (RGD), respectively. 1H‐NMR spectra confirmed the structure of the copolymer MPEG‐b‐P(LA‐co‐MCC/docetaxel) and MPEG‐b‐P(LA‐co‐MCC/RGD). In vitro antitumor assay indicates that the MPEG‐b‐P(LA‐co‐MCC/docetaxel) conjugate shows high cytotoxic activity against HeLa cancer cells. Cell adhesion and spreading experiment shows that copolymer MPEG‐b‐P(LA‐co‐MCC/RGD) is of benefit to cell adherence and is a promising biodegradable material for cell and tissue engineering. © 2008 Wiley Periodicals, Inc. The preparation of poly(DL ‐lactide‐co‐glycolide) (PLGA) nanoparticles was performed by a dialysis method without surfactant or emulsifiers. The size of the PLGA nanoparticles prepared from dimethylacetamide (DMAc) as an initial solvent was smaller than that from acetone. The sizes of the PLGA nanoparticles from DMAc and acetone were 200.4 ± 133.0 and 642.3 ± 131.1 nm, respectively. The effects of the initial solvent selected to dissolve the copolymer and the lactide:glycolide ratio were investigated. The PLGA nanoparticles were spherical as revealed by the results of scanning electron microscopy and transmission electron microscopy observations. From these results it was shown that PLGA nanoparticles could be formed by the dialysis method without surfactant. The drug‐loading contents and efficiency were also dependent on the lactide:glycolide ratio and initial feeding amount of the drug. A higher lactide ratio resulted in higher drug loading and higher loading efficiency. However, a higher initial feeding amount of the drug resulted in higher drug loading and lower loading efficiency. Clonazepam was released for at least 2 days and the release rate was slower with a higher lactide:glycolide ratio and a larger amount of drug‐loading nanoparticles than that with a lower lactide:glycolide ratio and a smaller amount of drug‐loading nanoparticles. © 2001 John Wiley & Sons, Inc. Diblock copolymers of poly(L ‐lactide)‐block‐poly(methyl methacrylate) (PLLA‐b‐PMMA) were synthesized through a sequential two‐step strategy, which combines ring‐opening polymerization (ROP) and atom transfer radical polymerization (ATRP), using a bifunctional initiator, 2,2,2‐trichloroethanol. The trichloro‐terminated poly(L ‐lactide) (PLLA‐Cl) with high molecular weight (Mn,GPC = 1–12 × 104 g/mol) was presynthesized through bulk ROP of L ‐lactide (L ‐LA), initiated by the hydroxyl group of the double‐headed initiator, with tin(II) octoate (Sn(Oct)2) as catalyst. The second segment of the block copolymer was synthesized by the ATRP of methyl methacrylate (MMA), with PLLA‐Cl as macroinitiator and CuCl/N,N,N′,N″,N″‐pentamethyldiethylenetriamine (PMDETA) as catalyst, and dimethyl sulfoxide (DMSO) was chosen as reaction medium due to the poor solubility of the macroinitiator in conventional solvents at the reaction temperature. The trichloroethoxyl terminal group of the macroinitiator was confirmed by Fourier transform infrared spectroscopy (FTIR) and 1H‐NMR spectroscopy. The comprehensive results from GPC, FTIR, 1H‐NMR analysis indicate that diblock copolymers PLLA‐b‐PMMA (Mn,GPC = 5–13 × 104 g/mol) with desired molecular composition were obtained by changing the molar ratio of monomer/initiator. DSC, XRD, and TG analyses establish that the crystallization of copolymers is inhibited with the introduction of PMMA segment, which will be beneficial to ameliorating the brittleness, and furthermore, to improving the thermal performance. © 2010 Wiley Periodicals, Inc. Ring‐opening polymerization of L ‐ or D ‐lactide was realized at 140 °C for a period of 7 days in the presence of dihydroxyl poly(ethylene glycol) (PEG), with M?n = 4000 g mol?1, using zinc lactate as initiator. The resulting poly(L ‐lactide)–PEG–poly(L ‐lactide) and poly(D ‐lactide)–PEG–poly(D ‐lactide) triblock copolymers are water soluble with polylactide (PLA) block length ranging from 11 to 17 units. Both the tube inverting method and rheological measurements were used to evaluate the gelation properties of aqueous solutions containing single copolymers or L /D copolymer pairs. Stereocomplexation between poly(L ‐lactide) and poly(D ‐lactide) blocks is observed for mixed solutions. Hydrogel formation is detected in the case of relatively long PLA blocks (DP PLA = 17), but not for copolymers with shorter PLA blocks (DP PLA = 11–13) due to partial racemization of L ‐lactyl units. Racemization is largely reduced when the reaction time is shortened to 1 day. Under these conditions, DP PLA of 8 is sufficient for the stereocomplexation of PLA–PEG block copolymers, and DP PLA above 10 leads to the formation of hydrogels of PLA–PEG block copolymers. On the other hand, racemization appears as a general phenomenon in the (co)polymerization of L ‐lactide with Zn(Lac)2 as initiator, although it is negligible or undetectable in the case of high molar mass polymers. Therefore, racemization is the limiting factor for the stereocomplexation‐induced gelation of water‐soluble PLA–PEG block copolymers where the PLA block length generally ranges from 10 to 30. Novel amphiphilic ABA‐type poly(D ‐gluconamidoethyl methacrylate)‐b‐polyurethane‐b‐poly(D ‐gluconamidoethyl methacrylate) (PGAMA‐b‐PU‐b‐PGAMA) tri‐block copolymers were successfully synthesized via the combination of the step‐growth and copper‐catalyzed atom transfer radical polymerization (ATRP). Dihydroxy polyurethane (HO‐PU‐OH) was synthesized by the step‐growth polymerization of hexamethylene diisocyanate with poly(tetramethylene glycol). PGAMA‐b‐PU‐b‐PGAMA block copolymers were synthesized via copper‐catalyzed ATRP of GAMA in N, N‐dimethyl formamide at 20°C in the presence of 2, 2′‐bipyridyl using Br‐PU‐Br as macroinitiator and characterized by 1H‐NMR spectroscopy and GPC. The resulting block copolymer forms spherical micelles in water as observed in TEM study, and also supported by 1H NMR spectroscopy and light scattering. Miceller size increases with increase in hydrophilic PGAMA chain length as revealed by DLS study. The critical micellar concentration values of the resulting block copolymers increased with the increase of the chain length of the PGAMA block. Thermal properties of these block copolymers were studied by thermo‐gravimetric analysis, and differential scanning calorimetric study. Spherical Ag‐nanoparticles were successfully synthesized using these block copolymers as stabilizer. The dimension of Ag nanoparticle was tailored by altering the chain length of the hydrophilic block of the copolymer. A mechanism has been proposed for the formation of stable and regulated Ag nanoparticle using various chain length of hydrophilic PGAMA block of the tri‐block copolymer. © 2012 Wiley Periodicals, Inc. A new amphiphilic Pluronic (F68)‐PLGA copolymer, which can be used to prepare the stealth or long‐circulating nanoparticles, was synthesized with Pluronic (F68), DL ‐Lactide, and glycolide. The structures of Pluronic (F68)‐PLGA copolymer as the products were characterized with infrared spectrometry, nuclear magnetic resonance and their molecular weights were determined by gel permeation chromatography. Two methods, double emulsion (DE) and nanoprecipitation (NP), were employed to fabricate the polymeric nanoparticles. Bovine serum albumin (BSA) was loaded into nanoparticles as a model protein. Influence of the preparation conditions on the nanoparticles size, encapsulation efficiency, and release profile in vitro was investigated. They showed the entrapment efficiency of 42.9–63.4% and the average diameter of 119.1–342.8 nm depending on the fabrication technique of nanoparticles and the type of copolymer. The stability maintenance of BSA in the nanoparticle release in vitro was also measured via sodium dodecyl sulfate‐polyacrylamide gel electrophoresis (SDS‐PAGE), circular dichroism (CD), and fluorescence spectrometry. Well defined poly (styrene‐co‐methylstyrene) grafted polyaniline/organo‐modified MgAl layered double hydroxide (LDH) was produced through solution intercalation method. After LDH nanoparticles were modified by the anion exchange reaction of MgAl (Cl) LDH with sodium dodecyl benzene sulfonate, Poly (styrene‐co‐methylstyrene) copolymers were synthesized by “living” free radical polymerization and then brominated with N‐bromosuccinimide. Afterwards, 1,4‐phenylenediamine was linked to brominated copolymers and prepared functionalized copolymer with amine. Poly (St‐co‐MSt)‐g‐PANI, has been synthesized by adding solution of ammonium persulfate and p‐toluenesufonic acid in DMSO solvent. Finally, Poly (styrene‐co‐methylstyrene) grafted‐Polyaniline/LDH nanocomposites were prepared by solution intercalation method. Characterization of these well‐defined nanocomposites included FT‐IR, gel permeation chromatography, thermogravimetric analysis, differential scanning calorimeter, transmission electron microscopy, and X‐ray diffraction. © 2011 Wiley Periodicals, Inc. In this study, synthesis, characterization, partial hydrolysis, and salt formation of poly(2‐hydroxyethyl methacrylate)‐co‐poly(4‐vinyl pyridine), (poly(HEMA)‐co‐poly‐(4‐VP)) copolymers were investigated. The copolymers were synthesized by free radical polymerization using K2S2O8 as an initiator. By varying the monomer/initiator ratio, chain lengths of the copolymers were changed. The copolymers were characterized by gel permeation chromatography (GPC), viscosity measurements, 1H and 13C NMR and FTIR spectroscopies, elemental analysis, and end group analysis methods. The effect of poly(D ,L ‐lactide‐copara‐dioxanone) (PLADO) as the compatibilizer on the properties of the blend of poly(para‐dioxanone) (PPDO) and poly(D ,L ‐lactide) (PDLLA) has been investigated. The 80/20 PPDO/PDLLA blends containing from 1% to 10% of random copolymer PLADO were prepared by solution coprecipitation. The PLADO component played a very important role in determining morphology, thermal, mechanical, and hydrophilic properties of the blends. Addition of PLADO into the blends could enhance the compatibility between dispersed PDLLA phase and PPDO matrix; the boundary between the two phases became unclear and even the smallest holes were not detected. On the other hand, the position of the Tg was composition dependent; when 5% PLADO was added into blend, the Tg distance between PPDO and PDLLA was shortened. The blends with various contents of compatibilizer had better mechanical properties compared with simple PPDO/PDLLA binary polymer blend, and such characteristics further improved as adding 5% random copolymers. The maximum observed tensile strength was 29.05 MPa for the compatibilized PPDO/PDLLA blend with 5% PLADO, whereas tensile strength of the uncompatibilized PPDO/PDLLA blend was 14.03 MPa, which was the lowest tensile strength. © 2010 Wiley Periodicals, Inc. Temperature‐responsive polymers have become increasingly attractive as carrier for the injectable drug delivery systems. In the present work, we have studied the preparation of poly(N‐isopropylacrylamide‐acrylamide‐vinilpyrrolidone) (NIPAAm‐AAm‐VP terpolymer) nanoparticulated terpolymer and its blend with poly(lactide‐co‐glycolide, PLGA; molar ratio of lactide/glycolid 1/3). Thermosensitive terpolymer, poly(NIPAAm‐AAm‐VP) was prepared by free‐radical polymerization in aqueous solution. The nanoparticles of poly(NIPAAm‐AAm‐VP) and its blend with PLGA containing naltrexone were prepared using the evaporation and w/o emulsion‐solvent evaporation methods, respectively. Nanoparticles prepared from terpolymer‐PLGA blend at low polymer concentration (5%) shows larger particle size (>300 nm) and higher drug content%. Various types of nanoparticles showed a burst release of less than 10% after 24 h . The results suggest that by regulating different variables, desired release profiles of naltrexone can be achieved using a blend of PLGA‐poly(NIPAAm‐AAm‐VP) nanoparticulate system. © 2008 Wiley Periodicals, Inc. Polystyrene terminated with benzyl alcohol units was employed as a macroinitiator for ring‐opening polymerization of ε‐caprolactone and L ‐lactide to yield AB‐ and ABC‐type block copolymers. Even though there are many reports on the diblock copolymers of poly(styrene‐block‐lactide) and poly(styrene‐block‐lactone), this is the first report on the poly(styrene‐block‐lactone‐block‐lactide) triblock copolymer consisting of two semicrystalline and degradable segments. The triblock copolymers exhibited twin melting behavior in differential scanning calorimetry (DSC) analysis with thermal transitions corresponding to each of the lactone and lactide blocks. The block derived from ε‐caprolactone also showed crystallization transitions upon cooling from the melt. In the DSC analysis, one of the triblock copolymers showed an exothermic transition well above the melting temperature upon cooling. Thermogravimetric analysis of these block copolymers showed a two‐step degradation curve for the diblock copolymer and a three‐step degradation for the triblock copolymer with each of the degradation steps associated with each segment of the block copolymers. The present study shows that it is possible to make pure triblock copolymers with two semicrystalline segments which also consist of degradable blocks. Narrow‐distribution, well‐defined comb‐like amphiphilic copolymers are reported in this work. The copolymers are composed of poly(methyl methacrylate‐co‐2‐hydroxyethyl methacrylate) (P(MMA‐co‐HEMA)) as the backbones and poly(2‐(dimethylamino)ethyl methacrylate) (PDMAEMA) as the grafted chains, with the copolymer backbones being synthesized via atom‐transfer radical polymerization (ATRP) and the grafted chains by oxyanionic polymerization. The copolymers were characterized by gel permeation chromatography (GPC), Fourier‐transform infrared (FT‐IR) spectroscopy and 1H NMR spectroscopy. The aggregation behavior in aqueous solutions of the comb‐like amphiphilic copolymers was also investigated. 1H NMR spectroscopic and surface tension measurements all indicated that the copolymers could form micelles in aqueous solutions and they possessed high surface activity. The results of dynamic light scattering (DLS) and scanning electron microscopy (SEM) investigations showed that the hydrodynamic diameters of the comb‐like amphiphilic copolymer aggregates increased with dilution. Because of the protonizable properties of the graft chains, the surface activity properties and micellar state can be easily modulated by variations in pH. Poly(ethylene glycol)‐poly(L ‐lactide) diblock and triblock copolymers were prepared by ring‐opening polymerization of L ‐lactide with poly(ethylene glycol) methyl ether or with poly(ethylene glycol) in the presence of stannous octoate. Molecular weight, thermal properties, and crystalline structure of block copolymers were analyzed by 1H‐NMR, FTIR, GPC, DSC, and wide‐angle X‐ray diffraction (WAXD). The composition of the block copolymer was found to be comparable to those of the reactants. Each block of the PEG–PLLA copolymer was phase separated at room temperature, as determined by DSC and WAXD. For the asymmetric block copolymers, the crystallization of one block influenced much the crystalline structure of the other block that was chemically connected to it. Time‐resolved WAXD analyses also showed the crystallization of the PLLA block became retarded due to the presence of the PEG block. According to the biodegradability test using the activated sludge, PEG–PLLA block copolymer degraded much faster than PLLA homopolymers of the same molecular weight. © 1999 John Wiley amp; Sons, Inc.
