用高压液相层析对31个属90株生孢囊放线菌及其它属菌的甲基萘醌进行了测定。其结果表明:游动放线菌属.小瓶菌属和小单孢菌属的甲基萘醌为MK-9(H4)MK-10(H4),无定形孢囊菌属和游动单孢菌属的甲基萘醌为MK-9(H4),指孢囊菌属的甲基萘醌为MK-9(H8)MK-9(H4),链孢囊菌属的甲基萘醌为MK-9(H4)或MK-9(H6),螺孢菌属的甲基萘醌为MK-9(H6)MK-9(H4),游动双孢菌属的甲基萘醌为MK-9(H2)MK-9(H4)。不同放线菌属的甲基萘醌组份是较稳定的,可作为属的分类特征之一。建议用甲基萘醌与形态和细胞壁化学组份相结合区分放线菌中的不同属。  相似文献

伦茨菌属(Lentzea)由Yassin等于1995年建立,是一个经典的丝状稀有放线菌类群,目前共包含23个有效描述种。伦茨菌的细胞壁含有meso-二氨基庚二酸,优势甲基萘醌成分为MK-9(H_4),磷酸类脂主要包括磷脂酰乙醇胺、双磷脂酸甘油、磷脂酰甘油和磷脂酰肌醇,基因组DNA的(G+C)含量为68.6mol%–79.6mol%。伦茨菌代谢产物具有显著的生物活性多样性,包括I型人体免疫缺损病毒整合酶的抑制活性、抗肿瘤活性、抗结核活性等,在生物医药研究领域逐步显示出潜在的应用价值。本课题组在研究贵州石灰岩风化壳放线菌多样性时分离获得若干株伦茨菌,采用多相分类方法研究Lentzea xinjiangensis DHS C013~T和Lentzea pudingi DHS C021~T两株菌的分类学地位,证实均为伦茨菌属的新物种。在此研究基础上,引用有关文献和最新研究成果,对伦茨菌属的建立、分类学特征、生态位和物种分布、功能基因组、天然活性产物应用开发等方面的研究进展进行了综述。  相似文献

河北九莲城淖尔可培养放线菌多样性及抗菌活性筛选   总被引:1,自引:1,他引:0  
【目的】勘探干涸的九莲城淖尔土壤放线菌多样性并进行活性筛选,以期发现药用微生物资源,为新抗生素的发现奠定基础。【方法】采用15种分离培养基,以稀释涂布法分离放线菌;根据分离菌株的16S rRNA基因序列同源性分析放线菌多样性;发酵液经乙酸乙酯萃取,菌丝体经丙酮浸提,获得提取浓缩物样品;样品通过纸片扩散法进行抗菌活性初筛;抗菌阳性菌株采用PCR技术进行Ⅰ型聚酮合酶(PKS I)KS域、Ⅱ型聚酮合酶(PKS II)KS域和非核糖体多肽合成酶(NRPS)A结构域抗生素生物合成基因的检测。【结果】从11份盐湖土壤样品中分离纯化到251株放线菌,其分布于放线菌纲的10个目15个科31个属,其中优势菌属为链霉菌属和拟诺卡氏菌属;251株放线菌中包括57株耐(嗜)盐放线菌,其优势菌属为拟诺卡氏菌属(22株)和涅斯捷连科氏菌属(15株)。基于16S r RNA基因序列的系统发育分析显示,菌株J11Y309为糖霉菌科潜在新属,菌株J12GA03为分枝杆菌科潜在新种。96株放线菌活性检测结果显示,56株至少对1株检定菌具有抗菌活性,阳性率为58.3%;56株有活性的放线菌中,47株至少含有1种抗生素生物合成基因,其中17株同时具有3种抗生素生物合成基因。【结论】干涸的九莲城淖尔土壤中含有较为丰富的药用放线菌资源,具有从中发现放线菌新物种和新抗生素的潜力。  相似文献

【目的】糖丝菌属(Saccharothrix)是一类丝状稀有放线菌,在生物医药、工业酶制剂和环境修复等领域展现出应用价值。本研究尝试建立以核糖体蛋白质为标志物,利用基质辅助激光解吸电离飞行时间质谱(matrix-assisted laser desorption/ionization-time of flight mass spectrometry,MALDI-TOF MS)技术鉴定糖丝菌属放线菌的方法。【方法】检索基因组数据库,提取糖丝菌属测序菌株15种核糖体蛋白质的序列并计算理论分子量;通过分子量比对分析糖丝菌属不同菌种之间及其模式菌株与邻近属菌种模式菌株之间15种核糖体蛋白质的匹配度,提出鉴定至菌种及属的核糖体蛋白质匹配数标准;选取目标属和非目标属菌种进行MALDI-TOF MS测试和分析并修正鉴定标准。【结果】将待测菌株的MALDI-TOF质谱峰与糖丝菌属各菌种模式菌株的15种核糖体蛋白质分别匹配,通过最大匹配数、质谱峰强度模式及特征质谱峰鉴定至属或种。【结论】本研究建立了基于15种核糖体蛋白质标志物及MALDI-TOF MS技术鉴定糖丝菌属放线菌的方法,可用于定向筛选和快速鉴...  相似文献

1株嗜盐、产丝放线菌新种TRM4064分离自四川大英盐湖。它的分类地位通过多相分类技术被证明。16S rRNA基因序列系统发育分析表明:菌株TRM4064在多孢放线菌属中形成一个独立的分支单元,同近缘物种Actinopolyspora mortivallis的同源性为98.8%,与它的DNA-DNA杂交值为23.2%,同Actinopolyspora菌属另外有效命名物种的相似性小于97%。菌株TRM4064的最适生长温度是37℃,最适生长pH为7.0,最适生长盐浓度为13%;细胞水解糖为木糖、葡萄糖、核糖和阿拉伯糖,主要的醌型是MK-10(H4)(38.2%)、MK-9(H4)(25.1%)、MK-9(H2)(28.6%)和MK-8(H4)(7.3%),主要的脂肪酸是anteiso-C17:0(36.9%)和iso-C17:0(19.3%),细胞磷脂是心磷脂、磷脂酰甘油、卵磷脂、磷脂酰肌醇和2个未知的磷脂类型。综合其表型、化学型及其分子遗传学等指标表明菌株TRM4064为多孢放线菌属的一个新成员。  相似文献

【背景】抗生素的滥用导致了耐药病原菌的不断增加和扩散,成为威胁人们健康的一个重要因素,因此挖掘新型抗生素显得尤其重要。新的物种很有可能会产生新的生物活性物质,特别是那些常规分离方法很少分离到的稀有放线菌。【目的】对2株小双孢菌潜在新种进行全基因组测序分析与抗菌活性测定,从多层次分子水平鉴定其新种分类地位,并预测次级代谢基因簇,为发现产生新颖活性物质的药用放线菌资源奠定基础。【方法】通过16S rRNA基因序列分析初步确定菌株H10836和H11081的种属分类;采用Illumina平台对2个菌株进行全基因组测序和注释,基于此构建全基因组系统发育树、计算ANI (Average Nucleotide Identity)和dDDH (Digital DNA-DNA Hybridization)值,最终确定菌株的新种分类地位;基于全基因组注释结果进行COG (Clusters of Orthologous Genes)聚类、KEGG (Koyto Encyclopedia of Genes and Genomes)代谢通路和antiSMASH次级代谢基因簇预测分析;采用琼脂打孔法进行抗菌活性测定。【结果】菌株H10836和H11081与小双孢菌属8个种两两之间的ANI值范围分别为85.3%-92.1%和85.2%-92.1%,dDDH值范围分别为33.0%-44.5%和31.3%-44.5%。ANI和dDDH值均低于物种判断的临界值,而且2株菌在全基因组系统发育树上与小双孢菌属聚成一簇,但独立成一亚支,从分子水平上综合确定菌株H10836和H11081是小双孢菌属的2个潜在新种。antiSMASH分析发现该2株菌基因组中有多种次级代谢产物合成基因簇,而且与已知抗生素合成基因簇相似度较低。抗菌活性测定显示该2株菌代谢产物均有抗耐甲氧西林金黄色葡萄球菌活性。【结论】菌株H10836和H11081是具有抗菌活性的新菌种,因此值得挖掘其新颖活性天然产物,试验结果将为菌株的进一步研究和应用提供参考。  相似文献

放线菌作为干旱、半干旱环境中生物土壤结皮(Biological Soil Crusts,BSCs)组成的重要生命存在形式之一,不仅是潜在临床有用天然产物化学多样性的重要来源,也是该生态系统物质循环与能量流动的重要参与者。以腾格里沙漠东南缘广泛分布的藻结皮和藓结皮为研究对象,通过宏基因组测序比较分析两种BSCs放线菌种群的分布特征、组成及其潜在代谢功能。结果表明,腾格里沙漠东南缘藻结皮与藓结皮土壤微生物组主要形成以地嗜皮菌属、红色杆菌属、类诺卡氏菌属、游动放线菌属、芽生球菌属、链霉菌属、贫养杆菌属、糖丝菌属、土壤红杆菌属、假诺卡氏菌属、小单孢子菌属、康奈斯氏杆菌属、大理石雕菌属、小月菌属以及弗兰克氏菌属等为主要类群的放线菌群落结构,在两种BSCs类型之间各属分布存在差异。藓结皮中放线菌参与的氨基糖与核苷酸糖代谢、原核生物中的碳固定途径、丁酸代谢、丙酸代谢、丙氨酸/天门冬氨酸和谷氨酸代谢、甲烷代谢、2-羰基羧酸代谢、肽聚糖生物合成、淀粉与蔗糖代谢以及缬氨酸/亮氨酸与异亮氨酸降解显著高于藻结皮。藓结皮中地嗜皮菌属和红色杆菌属对相对丰度前10的代谢功能分类的贡献度显著低于藻结皮,而类诺卡氏菌属、芽生球菌属、贫养杆菌属、游动放线菌属、链霉菌属、假诺卡氏菌属和糖丝菌属等对这些功能的相对贡献在藓结皮中具有重要作用。这些结果可为全面、深入理解腾格里沙漠东南缘藻结皮与藓结皮放线菌资源多样性及其潜在功能多样性提供科学数据,也为理解放线菌在不同类型BSCs中的生态功能提供参考。  相似文献

【背景】细菌耐药性问题日益严峻,新抗生素的研发速度远远落后于临床需要,从特殊生境中挖掘微生物药物资源有望解决以上问题。【目的】勘探西藏仲巴五彩沙漠土壤放线菌多样性并进行生物活性筛选,为发现药用放线菌资源、开发新型抗生素奠定基础。【方法】采用8种分离培养基,通过平板稀释涂布法分离放线菌;根据分离菌株的16S r RNA基因序列同源性分析放线菌多样性;采用PCR技术对分离的放线菌菌株进行II型聚酮合酶(PKS-II)酮缩酶结构域KS、非核糖体多肽合成酶(NRPS)腺苷酸化结构域A、安莎类抗生素生物合成前体3-氨基-5-羟基-苯甲酸合酶(AHBA)保守区、黄素腺嘌呤二核苷酸卤化酶(Halo)保守区抗生素生物合成基因检测;对生物合成基因检测阳性的菌株进行液体发酵,发酵液经乙酸乙酯萃取、菌体经丙酮浸提,获得提取浓缩物样品进行抑菌活性和抗氧化活性筛选。【结果】从4份土样中分离纯化到231株放线菌,分布于7个属,其中链霉菌为优势菌属。68株放线菌的生物合成基因分析显示至少具有1种生物合成基因簇,其中6株同时具有4种生物合成基因簇;进一步的抑菌活性检测显示所有检测的菌株至少表现为对1株检定菌具有抑菌活性,其中8株具有广谱抗菌活性;抗氧化活性筛选结果为13株显示总抗氧化能力阳性,10株具有较好的羟自由基清除能力,3株显示较强的氧自由基清除能力。【结论】西藏仲巴五彩沙漠土壤中含有较丰富的放线菌药用资源,具有从中发现放线菌新菌种和开发新抗生素的潜力。  相似文献

【目的】从3种红树林植物根际沉积物中分离和鉴定放线菌,进行抑菌活性初筛获得目标菌株并研究其次级代谢产物。【方法】使用5种培养基对红树林植物根际沉积物放线菌进行分离,采用16S rRNA基因序列比对的方法研究沉积物中的放线菌多样性,结合抑菌活性筛选获得目标菌株后进行放大规模发酵和分离鉴定次级代谢产物,根据生物合成基因簇定位和分析对化合物的生物合成途径进行推导。【结果】从3种红树林植物根际沉积物中分离得到放线菌49株,包括链霉菌属(Streptomyces)31株、小单孢菌属(Micromonospora)14株、小双孢菌属(Microbispora)、链孢子囊菌属(Streptosporangium)、野野村氏菌属(Nonomuraea)和糖单孢菌属(Saccharomonospora)各1株。获得粗浸膏抑菌活性较好的菌株Streptomyces sp. Scirtothrips perseae Nakahara was discovered attacking avocados in California, USA, in 1996. Host plant surveys in California indicated that S. perseae has a highly restricted host range with larvae being found only on avocados, while adults were collected from 11 different plant species. As part of a management program for this pest, a “classical” biological control program was initiated and foreign exploration was conducted to delineate the home range of S. perseae, to survey for associated natural enemies and inventory other species of phytophagous thrips on avocados grown in Mexico, Guatemala, Costa Rica, the Dominican Republic, Trinidad, and Brazil. Foreign exploration efforts indicate that S. perseae occurs on avocados grown at high altitudes (>1500 m) from Uruapan in Mexico south to areas around Guatemala City in Guatemala. In Costa Rica, S. perseae is replaced by an undescribed congener as the dominant phytophagous thrips on avocados grown at high altitudes (>1300 m). No species of Scirtothrips were found on avocados in the Dominican Republic, Trinidad, or Brazil. In total, 2136 phytophagous thrips were collected and identified, representing over 47 identified species from at least 19 genera. The significance of these species records is discussed. Of collected material 4% were potential thrips biological control agents. Natural enemies were dominated by six genera of predatory thrips (Aeolothrips, Aleurodothrips, Franklinothrips, Leptothrips, Scolothrips, and Karnyothrips). One genus each of parasitoid (Ceranisus) and predatory mite (Balaustium) were found. Based on the results of our sampling techniques, prospects for the importation of thrips natural enemies for use in a “classical” biological control program in California against S. perseae are not promising.  相似文献

Komárek has recently reviewed the various species assigned to the green algal genusNeochloris Starr (Chlorococcales, Chlorococcaceae) and removed those with uninucleate vegetative cells to a new genus,Ettlia. Watanabe & Floyd, unaware ofKomárek's work, also reviewed the species ofNeochloris and distributed them among three genera—Neochloris, Chlorococcopsis gen. nov., andParietochloris gen. nov.—on the basis of details of the covering of the zoospore and the arrangement of the basal bodies of the flagellar apparatus. This paper reconciles these two treatments and makes additional recommendations at the ranks of genus, family, order, and class.  相似文献

The cardinalfishes (Apogonidae) are a diverse clade of small, mostly reef-dwelling fishes, for which a variety of morphological data have not yielded a consistent phylogeny. We use DNA sequence to hypothesize phylogenetic relationships within Apogonidae and among apogonids and other acanthomorph families, to examine patterns of evolution including the distribution of a visceral bioluminescence system. In conformance with previous studies, Apogonidae is placed in a clade with Pempheridae, Kurtidae, Leiognathidae, and Gobioidei. The apogonid genus Pseudamia is recovered outside the remainder of the family, not as sister to the superficially similar genus Gymnapogon. Species sampled from the Caribbean and Western Atlantic (Phaeoptyx, Astrapogon, and some Apogon species) form a clade, as do the larger-bodied Glossamia and Cheilodipterus. Incidence of visceral bioluminescence is found scattered throughout the phylogeny, independently for each group in which it is present. Examination of the fine structure of the visceral bioluminescence system through histology shows that light organs exhibit a range of morphologies, with some composed of complex masses of tubules (Siphamia, Pempheris, Parapriacanthus) and others lacking tubules but containing chambers formed by folds of the visceral epithelium (Acropoma, Archamia, Jaydia, and Rhabdamia). Light organs in Siphamia, Acropoma, Pempheris and Parapriacanthus are distinct from but connected to the gut; those in Archamia, Jaydia, and Rhabdamia are simply portions of the intestinal tract, and are little differentiated from the surrounding tissues. The presence or absence of symbiotic luminescent bacteria does not correlate with light organ structure; the tubular light organs of Siphamia and chambered tubes of Acropoma house bacteria, those in Pempheridae and the other Apogonidae do not.  相似文献

The genusKarschia, in the earlier sense, including saprophytes and parasites on lichens, has been thought to be a non-lichenized parallel genus of the lichen genusBuellia. Modern workers included it on the one hand inBuellia, on the other hand combined it with bitunicate ascomycetes. It is now proved thatKarschia is heterogeneous and contains but superficially similar members both of the genusBuellia of theLecanorales and of typical or masked bitunicateAscomycetes. Therefore, it can not be regarded as a link betweenLecanorales andDothideales. The type species ofKarschia belongs to theDothideales.

Since 1999, four specific weevils (Coleoptera, Curculionidae) were released in the Republic of Congo against three exotic floating water weeds: Neochetina eichhorniae Warner and N. bruchi Hustache against water hyacinth, Neohydronomus affinis Hustache against water lettuce, and Cyrtobagous salviniae Calder and Sands against water fern. Recoveries of exotic weevils were made from all 24 release sites except one, and all four species have established and spread (up to 800 km for water hyacinth weevils). Within a few years of releases, control of water fern and water lettuce was such that fishing and navigation could be resumed, while reductions of water hyacinth populations were only beginning.  相似文献

A molecular phylogeny of Hebeloma species from Europe   总被引:2,自引:1,他引:1  
In order to widen the scope of existing phylogenies of the ectomycorrhizal agaric genus Hebeloma a total of 53 new rDNA ITS sequences from that genus was generated, augmented by sequences retrieved from GenBank, and analysed using Bayesian, strict consensus and neighbour joining methods. The lignicolous Hebelomina neerlandica, Gymnopilus penetrans, and two species of Galerina served as outgroup taxa. Anamika indica, as well as representatives of the genera Hymenogaster and Naucoria, were included to test the monophyly of Hebeloma, which is confirmed by the results. Hebeloma, Naucoria, Hymenogaster and Anamika indica cluster in a strongly supported monophyletic hebelomatoid clade. All trees largely reflect the current infrageneric classification within Hebeloma, and divide the genus into mostly well-supported monophyletic groups surrounding H. crustuliniforme, H. velutipes, H. sacchariolens, H. sinapizans, and H. radicosum, with H. sarcophyllum being shown at an independent position; however this is not well supported. The section Indusiata divides with strong support into three groups, the position of the pleurocystidiate Hebeloma cistophilum suggests the possible existence of a third subsection within sect. Indusiata. Subsection Sacchariolentia is raised to the rank of section.  相似文献

Synopsis In a fourteen month study (May 1976 – June 1977) I examined the following characteristics of an intertidal bay goby (Lepidogobius lepidus) in Morro Bay, California, U.S.A.: annual and seasonal patterns of abundance, age composition and growth rates, survivorship and mortality patterns, and the reproductive cycle for female gobies. Fishes were collected with the aid of quinaldine and otoliths and ovaries removed. Age and growth rates were estimated from otolith annuli using a back calculation formula and a Brody-Bertalanffy growth curve. Mortality rates were derived using the methods of Heincke (1913), Robson & Chapman (1960), mean age, and a catch curve (Ricker 1975). A gonad index was used to describe the annual reproductive cycle. Results indicated that abundance fluctuated seasonally and that these fluctuations appeared to be caused by reproductive emigrations. Bay gobies reached an age of 7+ and a standard length of 87 mm. Growth was relatively constant (6 mm yr−1) until age 5, at which point it began to decline. The mean rates of survivorship, mortality, and instantaneous mortality were 0.75, 0.25, and 0.29 respectively. Mortality rates for individual age classes ranged from 0.13 to 0.51 and increased with age. This stock appears to reproduce mainly during the winter.  相似文献

The mature embryo of seven species belonging to five genera of Indian bamboos is described. In all these the basic pattern of embryo organisation is same: the scutellar and coleoptilar bundles are not separated by an internode, the epiblast is absent, the lower portion of the scutellum and the coleorhiza are separated by a cleft and the margins of embryonic leaves overlap. The features unique to fleshy fruited bamboos are: presence of a massive scutellum, the juxtaposition of plumule and radicle and the occurrence of a bud in the axil of the coleoptile. The fleshy fruit bearing bamboos should be classified into one group, the tribeMelocanneae. Evidence is provided to recognise additional groups in the subfamilyBambusoideae.  相似文献
