以鼠李糖乳杆菌LGG(Lactobacillus rhamnosus GG)菌株为对照,将几株乳杆菌进行体外抗氧化能力的比较,以DPPH自由基、·OH及·O2-清除率为指标综合筛选出抗氧化能力较全面的鼠李糖乳杆菌B6,并比较不同培养基条件下其代谢产物的抗氧化能力;然后通过建立PC12细胞氧化损伤模型,测定益生菌代谢产物处理后丙二醛、活性氧的含量,探究鼠李糖乳杆菌B6代谢产物对细胞氧化应激的影响。结果表明,鼠李糖乳杆菌B6发酵MRS上清液的DPPH自由基、·OH以及·O2-清除率分别达到63.23%、99.38%和26.35%,都显著高于LGG。B6发酵脱脂乳得到的代谢产物与发酵MRS得到的代谢产物相比较,DPPH自由基清除率和·OH清除率都有显著升高,两者·O2-清除率相当。在PC12细胞氧化损伤实验中B6发酵脱脂乳的代谢产物降低了PC12细胞的氧化应激损伤,提高了氧化应激状态下的抗氧化能力。  相似文献

以椰子皮为原料,在单因素试验的基础上,采用沸水浸提法优化椰子皮多糖的提取工艺。同时对椰子皮多糖进行紫外光谱、红外光谱、13C NMR和DEPT 135分析,并利用清除ABTS+自由基、DPPH自由基和O2-能力评价其体外抗氧化活性,同时也评价了体外抗HepG2增殖活性。结果表明,椰子皮多糖的最佳提取工艺条件为:浸提温度100℃,浸提时间3小时,料液比1∶5(w/w),在此条件下椰子皮多糖提取率为4.73%。体外抗氧化试验表明,椰子皮多糖对ABTS+自由基、DPPH自由基和O2-均有一定的清除效果,随着椰子皮多糖浓度的增加清除能力逐渐增强,当多糖浓度为3.2 mg/mL时,其对ABTS+自由基、DPPH自由基和O2-的清除率分别达到89.58%、94.62%和95.21%,此时抗氧化能力与维生素C相当。与此同时,体外抗细胞增殖试验表明,椰子皮多糖对HepG2细胞显示出明显的抗增殖活性。  相似文献

为研究野樱莓原花青素在模拟体外胃肠消化过程中抗氧化活性的变化。采用超声-微波协同萃取法提取原花青素并探讨了乙醇体积分数、提取时间、液料比和提取温度对原花青素提取率的影响。结果表明,超声-微波协同萃取野樱莓原花青素的最佳工艺条件为乙醇体积分数为60%,提取时间40min,液料比为20∶1mL/g,提取温度为50℃,在此条件下野樱莓原花青素提取率为5.77%±1.01%。模拟体外消化表明野樱莓原花青素消化产物具有较强的抗氧化活性。在模拟口腔唾液消化15min时,野樱莓原花青素的·OH和O2-·自由基清除能力达到最强,而O2-·在模拟唾液消化过程中无明显变化。在模拟胃液消化1h时·OH和O2-·自由基清除率最强,DPPH·自由基清除率在模拟胃液消化2h时达到最强;在模拟小肠消化4h时,野樱莓原花青素的·OH和O2-·自由基清除率最强,DPPH·自由基清除率在消化6h时最强。  相似文献

以水果蓝莓为原料,进行酒精发酵,所用酵母菌种为Fermivin酵母,并且调整其糖度至16%左右,在此条件下确定出蓝莓的酒精发酵最适温度20℃;将已完成发酵的蓝莓果酒与活化充分的醋酸菌种接种进行醋酸发酵,确定出蓝莓果醋最适宜的醋酸发酵温度。结果表明,蓝莓醋最适醋酸发酵温度为35℃。试验对已发酵好的蓝莓果醋的抗氧化性进行研究,并且与市售的苹果醋、食醋进行对比分析,测定3种醋对DPPH自由基、对羟自由基·OH-、超氧阴离子·O2-的清除能力并进行对比分析。结果表明:蓝莓果醋对DPPH自由基清除率为51.7%,对羟基自由基·OH-清除率达76.4%,对超氧阴离子·O2-清除率相对较低,仅27.5%;蓝莓果醋对3种自由基的清楚能力均显著高于市售的苹果醋及食醋,说明试验方法酿制的蓝莓果醋具有较好的抗氧化能力。  相似文献

为探讨热处理对黄花梨冷藏品质及其氧化还原水平的作用,黄花梨果实采后先经40℃热空气处理12h,再在1℃贮藏120d,随后20℃存放7d,监测果实硬度、可溶性固形物、电导率、丙二醛(MDA)含量、超氧阴离子自由基(O2-·)生成速率、可溶性蛋白含量和活性氧清除酶活性的变化。结果显示:热处理可显著抑制果实可溶性蛋白分解,提高超氧化物歧化酶(SOD)、过氧化氢酶(CAT)和过氧化物酶(POD)的活性,降低O2-·生成速率和MDA含量,同时可以保持果实硬度和可溶性固形物,延缓电导率上升。研究表明,贮前热空气处理(40℃,12h)可延缓黄花梨果实的衰老变质,其作用与较高的抗氧化水平有关。  相似文献

以草果为原料,测定5种不同低共熔溶剂提取物中总黄酮和总多酚含量,通过总还原能力及对DPPH·、ABTS+·、·OH、NO2-清除能力评价其抗氧化活性,并探讨总黄酮和总多酚与抗氧化能力的相关性。结果表明氯化胆碱—乙二醇低共熔溶剂草果提取物总黄酮含量最高,氯化胆碱—草酸低共熔溶剂草果提取物总多酚含量最高,不同低共熔溶剂提取物均有抗氧化活性,草果提取物中总多酚与ABTS+自由基清除率、羟基自由基清除率和NO2-清除率呈显著相关,为草果的高效利用提供依据。  相似文献

本研究以清除DPPH自由基(DPPH·)、羟基自由基(·OH)、超氧阴离子自由基(O2-·)能力和α-葡萄糖苷酶抑制活性能力为指标,初步评价不同产地青钱柳多糖的体外抗氧化及降血糖活性。结果表明:不同产地青钱柳的多糖含量存在显著差异(p<0.05),其中江西修水县青钱柳的多糖含量最高,为5.85%±0.02%,贵州榕江县平阳乡多糖含量最低,为3.50%±0.10%。不同产地青钱柳多糖的DPPH自由基(DPPH·)、羟基自由基(·OH)、超氧阴离子自由基(O2-·)能力及α-葡萄糖苷酶抑制活性有所差异,且与多糖的浓度呈一定剂量关系,即随多糖浓度的增大,多糖的抗氧化及抑制α-葡萄糖苷酶活性能力增强。青钱柳多糖具有良好的体外抗氧化和降血糖活性,可对其进一步开发利用。  相似文献

通过ABTS+自由基、DPPH自由基清除能力、铁还原能力、小鼠耳肿胀实验考查辣椒叶多酚提取物粗提物及纯化物的体外抗氧化及抗急性炎症活性.结果表明,辣椒叶多酚提取物具有一定的抗氧化能力,其纯化物清除ABTS+自由基、DPPH自由基和铁还原能力的IC50分别为0.700、0.643、0.758mg/mL,与对照品VC浓度处于同一数量级,说明辣椒叶多酚提取物是一种有效的天然抗氧化剂;600mg/kg剂量的辣椒叶多酚的粗提物及纯化物,均对二甲苯所致小鼠耳肿胀有极显著抑制作用,抑制率分别为59.84%、62.69%(p<0.01);炎症组织PGE2含量从32.41A/g可分别降至19.27、16.51A/g(p <0.05);实验小鼠血清肝损伤指标ALT(谷丙转氨酶)和AST(谷草转氨酶)的监测表明辣椒叶多酚提取物具有较高安全性.实验结果表明辣椒叶多酚提取物具有一定的抗氧化及抗急性炎症活性且安全性较高,为进一步开发利用辣椒资源提供了理论依据.  相似文献

目的:旨在分析大理白族地区使用频率最高的滇红玫瑰在发酵过程中提取物酚类物质含量及其抗氧化和抗炎活性变化。方法:采用民族植物学方法进行调查确定了药食同源玫瑰品种,以模仿传统的玫瑰糖发酵工艺为研究对象,利用DPPH和ABTS+法评价不同糖比例和发酵时间发酵滇红玫瑰提取物(Fermented Rosa rugosa‘Dianhong’,FDR)的抗氧化活性,建立脂多糖诱导RAW264.7细胞模型(NO抑制率)评价提取物抗炎活性,并将酚类物质含量与抗氧化和抗炎活性指标进行相关性分析。结果:7种玫瑰被当地用做药食同源植物,其中滇红玫瑰的使用频率值(f)最高。滇红玫瑰经鲁氏酵母发酵的过程中,FDR-2(35%红糖、65%花瓣)和FDR-3(25%红糖、75%花瓣)的酚类物质含量(总黄酮、总酚)、DPPH和ABTS+自由基清除能力、NO抑制率均呈先上升后下降的趋势,其中,在发酵7 d时以上指标均达到最高值,即与发酵前相比能够显著(P<0.05)增加酚类物质含量,提高DPPH和ABTS+自由基的清除活性以及NO生成抑制率。然而,F...  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
