阐明生物内各种相关活性物质及其实时变化、生理分布与生物功能是探索自身与生命奥秘的重要组成部分。化学荧光探针是该领域研究的重要工具之一。含磷荧光探针因具有较强的还原性、良好的水溶性与生物相容性,可利用其特异性化学反应或者物理作用检测细胞内各种特定的分析底物。近几年基于不同含磷官能团荧光探针的研究取得了较大进展,本文对相关研究进行了梳理,对含磷官能团类的荧光探针进行归纳总结:主要包括2-(二苯基磷)苯甲酸酯类、磷酸酯类、二苯基磷酰类和三苯基磷阳离子等类型荧光探针,为进一步探索构建新的含磷基团荧光探针提供借鉴与参考。 相似文献

相比于基于非共价键的超分子相互作用的传统荧光探针,反应型荧光探针的识别过程不是通过配位和氢键的相互作用,而是通过与目标物发生化学反应,使得光谱性质或颜色改变,生成不同结构的光活性化合物。近年来,反应型荧光探针主要有三种化学反应类型:底物与受体通过共价键连接;底物作为催化剂,与受体发生不可逆的反应;基于置换反应使底物与受体络合。正是反应型荧光探针利用选择反应的优势识别特定物质,因而提供了方便、快捷、专一的分析检测目标物的方法,具有很高的灵敏度和选择性,受到越来越多的关注。本文综述了近六年来基于反应型罗丹明荧光探针用于检测金属阳离子、活性氧化物和阴离子诱导β-内酰胺开环的最新研究进展,评述其结构和检测性能之间的关系,并展望这类荧光探针的应用前景与发展趋势。 相似文献

A NIR fluorescent probe (DDAA) derived from fluorophore DDAO with alanine as the recognition group was developed for sensing aminopeptidase N (APN) in gut microbiota. Aminopeptidase N (APN) is an important drug target and biomarker for various tumors. The current work characterizes a novel APN-targeted fluorescent probe (Bes-Green, 2) that manifests comparable inhibitory activity with Bestatin. Fluorescent amphiphilic benzoxazole derivatives were synthesized and used to produce photoactive phosphatidylcholine (PC) liposomes by reserve-phase evaporation. The dyes absorbed in the UV region and were fluorescent in the blue-green region (determined by solvent polarity). The alkyl chain length seemed to play a fundamental role in the photophysics of the benzoxazole fluorophore in reverse liposomes, and despite the same ESIPT core and phospholipid building block, each amphiphilic dye had a particular emission profile related to the dye location in the liposome. The fluorescence emission spectra from dye 5 showed that its fluorophore experienced a polar environment, due to the single normal emission, while dyes 6–7 had (in part) a normal emission, and the main fluorescent band ascribed to the ESIPT emission indicated a more hydrophobic environment. Despite the complex fluorescent profiles, the benzoxazole derivatives could be successfully introduced into the reverse liposome structure due to the interaction between the alkyl chain and PC bilayer. 相似文献

The vicinal dithiol motif is widely present in proteins, and is critical for proteins' structures and functions.In recent years, a variety of fluorescent probes with high specificity and outstanding optical properties for sensing protein vicinal dithiols have been developed. In this review, we summarized the fluorescent probes of protein vicinal dithiols in literature. These probes are classified into four types based on their acceptor sites, i.e., biarsenical probes, monoarsenical probes, dimaleimide probes and diacrylate probes.Through analyzing the properties of different probes, we expect that this review would help readers further understand the structural factors of these probes and provide the design strategy for novel fluorescent probes with improved properties. 相似文献
