The sacS gene controls the expression of 2 saccharolytic enzymes in Bacillus subtilis (sucrase and levansucrase).This paper describes a recombinant plasmid containing a mutant allele, sacSc. The plasmid was isolated from a B. subtilis DNA bank established in Escherichia coli. Moreover, it was shown that the sacSc allele, placed on a high-copy plasmid, is dominant over the wild-type chromosomal sacS+ allele. This result strongly suggests that the sacS gene encodes a positive regulatory protein.  相似文献

枯草芽胞杆菌Bacillussubtilis在工业生物技术以及合成生物学领域作为一种重要的微生物可广泛用作代谢工程、重组蛋白表达以及新型基因电路的底盘。在B. subtilis中构建基于非编码RNA的高精准调节元件,能够实现不依赖蛋白质因子的基因表达调控,丰富B.subtilis基因表达通用工具。通过基因工程手段,设计了基于茶碱适体域的核糖开关E和适体核酶AZ调节元件,并与不同的B.subtilis内源组成型启动子适配,构建出茶碱激活型基因表达控制元件。测定这两种调节元件与6种组成型启动子组合匹配下报告基因GFP的荧光强度,鉴定并分析各调控元件的工作性能。并进一步以红色荧光蛋白mCherry和普鲁兰酶两种不同的异源蛋白验证核糖开关或适体核酶与启动子的最优组合。结果表明,同一种RNA调节元件与不同启动子组合呈现不同水平的调控效率。在核糖开关与启动子的组合中,启动子PsigW和核糖开关E组合(sigWE)对GFP表达的诱导率最高,达到16.8。在适体核酶与启动子的组合中,AZ与启动子P43、PrpoB组合(P43AZ和rpoBAZ)的诱导率最高,分别达到了6.1和6.2。进一步验证结果显示,sigWE调控mCherry的诱导率最高(9.2),而P43E调控普鲁兰酶的诱导率最高(32.8),产酶水平达到了81U/mL。核糖开关和适体核酶对GFP、mCherry、普鲁兰酶均能实现调控,但是不同元件组合的调控性能有所差异,对不同基因的调控效果也不尽相同。  相似文献

目的:建立一种简便、快速的木聚糖酶分离和提取方法。方法:采用活性聚丙烯酰胺凝胶电泳和均质提取法相结合,分离纯化枯草芽孢杆菌(Bacillus subtilis)固体培养基发酵产物中的木聚糖酶,进一步用薄层色谱和高压液相色谱对木聚糖酶进行鉴定。结果:采用活性聚丙烯酰胺凝胶电泳和均质提取法相结合,从枯草芽孢杆菌(Bacillus subtilis)固体培养基发酵产物中分离得到了两种内切木聚糖酶,酶解桦木木聚糖的产要产物以木二糖和木三糖为主。结论:活性聚丙烯酰胺凝胶电泳和均质提取法相结合是一种新的分离纯化木聚糖酶的简便、有效方法。  相似文献

Maltose uptake and its regulation in Bacillus subtilis   总被引:4,自引:0,他引:4  
Extracts prepared from cultures of Bacillus subtilis, grown on maltose as the sole carbon source, lacked maltose phosphotransferase system activity. There was, however, evidence for a maltose phosphorylase activity, and such extracts also possessed both glucokinase and glucose phosphotransferase system activities. Maltose was accumulated by whole cells of B. subtilis by an energy-dependent mechanism. This uptake was sensitive to the effects of uncouplers, suggesting a role for the proton-motive force in maltose transport. Accumulation of maltose was inhibited in the presence of glucose, and there was no accumulation of maltose by a strain carrying the ptsI6 null-mutation. A strain carrying the temperature-sensitive ptsI1 mutation accumulated maltose normally at 37 degrees C but, in contrast to the wild-type, was devoid of maltose transport activity at 47 degrees C. The results indicate a role for the phosphotransferase system in the regulation of maltose transport activity in this organism.  相似文献

In order to characterize the dynamics of the interaction between the emergent membrane translocated exoprotein and the components of Bacillus subtilis cell wall, we examined the kinetics of the in vitro refolding of levansucrase and alpha-amylase, at pH 7 and 37 degrees C, in the presence of polyphosphates (polyP) of various chain lengths (2/=16. In contrast, polyP modulate indirectly the rate of alpha-amylase refolding via their affinity for calcium. These differential effects might explain that the rate of the cell wall translocation of alpha-amylase secretion was found to be half that of levansucrase.  相似文献

用PCR方法从巨大芽孢杆菌的基因组DNA中扩增到青霉素G酰化酶基因,并装载到枯草杆菌质粒pPZW103中,将其转化到枯草杆菌DB104中进行了分泌表达,重组菌株产酶无需苯乙酸诱导。在37℃培养24h,菌液酶活力可达6u/ml。10天的连续传代实验表明重组菌株的稳定性很高。  相似文献

为了筛选出对植物病原菌具有拮抗作用的生物防治细菌,采用平板对峙法从蔬菜根际土壤中分离获得一株对多种病原真菌具有抑制作用的枯草芽孢杆菌,并对抑制后的病原菌菌丝进行了观察,结果表明该菌株对尖孢镰刀菌、串珠镰刀菌、层出镰刀菌以及腐皮镰刀菌均具有抑菌活性。通过形态特征、生理生化特征及16S rRNA序列分析,将该菌株鉴定为枯草芽孢杆菌(Bacillus subtilis),并对其发酵培养基进行了优化。  相似文献

脂肽(Lipopeptide)是由枯草芽孢杆菌(Bacillus subtilis)等微生物产生的一类具有较强表面活性的生物表面活性剂.枯革杆菌磷酸泛酰巯基转移酶基因(afp)是枯草芽孢杆菌中参与脂肽代谢的功能性基因.采用sfp基因PCR对从环境中得到的一组产生表面活性剂的微生物进行筛选,结合Tricine-SDS-PAGE电泳对PCR结果呈阳性的菌蛛的代谢粗初提物进行检测,初步鉴定得到两株枯草芽孢杆菌.进一步利用16S rDNA序列的系统发育学分析确定这两种菌株为枯草芽孢杆菌,并利用TLC、HPLC鉴定其产物为脂肽类表面活性剂,从而建立了一套快速分离检测产生脂肽类生物表面活性剂的枯草芽孢杆菌方法.  相似文献

采用同源重组法高效构建枯草芽孢杆菌转酮酶(tkt)缺失突变株。以大肠杆菌(E.coliDH5α)质粒pBlUSKM为框架,构建出基于枯草杆菌(B.S104)tkt基因位点的整合载体pb-Trs-n,将此载体重组到Bacillus subtilis104中,从新霉素抗性平板上挑取转化子,整合载体pb-Trs-n的测序结果与Kunst.F报道的tkt基因高度同源(98.9%)。同源重组后,B.S104染色体上的tkt基因(2 004bp)部分与载体pb-Trs-n的neo基因(1 197bp)发生了同源交换,确定了该转化子为枯草芽孢杆菌转酮酶缺失突变株(tkt-,neo),该方法构建枯草芽孢杆菌转酮酶(tkt)缺失突变株是可行的,为D-核糖工程菌的研究奠定了基础。  相似文献

作为一种食品安全级的典型工业模式微生物,枯草芽孢杆菌Bacillus subtilis由于具有非致病性、胞外分泌蛋白能力强以及无明显的密码子偏爱性等特点,现已被广泛应用于代谢工程领域。近年来,随着分子生物学和基因工程技术等的迅速发展,多种研究策略和工具被用于构建枯草芽孢杆菌底盘细胞进行生物制品的高效合成。文中从启动子工程、基因编辑、基因回路、辅因子工程以及途径酶组装等方面介绍枯草芽孢杆菌在代谢工程领域的研究历程,并总结其在生物制品生产中的相关应用,最后对其未来的研究方向进行展望。  相似文献

A cloned putative promoter region upstream of the 16S rRNA gene of the western X-disease phytoplasma was inserted behind the promoterless chloramphenicol acetyltransferase gene of plasmid pPL603. The DNA construct was used to transform Bacillus subtilis cells. The transformants were assayed for chloramphenicol acetyltransferase activity, showing that the phytoplasma promoter is efficiently expressed in a B. subtilis background.  相似文献

脂肽是一类具有特殊作用的生物表面活性剂。本实验将血红蛋白基因(vhb)置于RDR细菌启动子驱动下的质粒PSET中,构建PSET-RDR-vhb重组质粒,并通过电激作用转入脂肽代谢菌株-枯草芽孢杆菌株(bacillus subtilis)ZW-3中,转化菌株经酶切和PCR电泳检测鉴定,Southern-blot杂交显示部分转化菌株中外源基因插入基因组DNA,采用一氧化碳差异色谱法测定了血红蛋白的表达量。实验进一步对转化菌株的生长曲线、总蛋白量、过氧化氢酶活性、脂肽的产率进行了测定,结果显示,相比于原始菌株,转化菌株数据均有明显提高。  相似文献

Abstract Using promoter-probe plasmids, more than 200 promoter-containing fragments from Bacillus stearothermophilus and Bacillus subtilis were cloned in B. subtilis . Among these, 15 promoter fragments were highly temperature-dependent in activity compared to the promoter sequence (TTGAAA for the −35 region, TATAAT for the −10 region) of the amylase gene, amyT , from B. stearothermophilus . Some fragments exhibited higher promoter activities at elevated temperature (48°C), others showed higher activities at lower temperature (30°C). Active promoter fragments at higher and lower temperatures were obtained mainly from the thermophile ( B. stearothermophilus ) and the mesophile ( B. subtilis ), respectively. A promoter fragment active at high temperature was sequenced, and the feature of the putative promoter region was discussed.  相似文献

Abstract

Lipases are one of the most important catalysts for several industries such as detergent, dairy, and textile industry due to their bio-catalytic ability in aqueous and non-aqueous media. Stability to extreme conditions is an important property since it makes enzymes suitable to several industrial processes. In this study, lipase producing soil bacteria were screened and identified with 16S rDNA sequencing. A new hyper-thermophilic lipase named as Bacillus subtilis LP2 isolate was partially purified by ammonium sulphate precipitation with 17.8-fold purification and 583?U/mg specific activity. Maximum activity was exhibited at pH 7 and 80?°C with the substrate tween 80?KM and Vmax values were calculated as 18.3?mM and 680?U/mg with a catalytic efficiency (kcat/KM) of 307?s?1M?1. These results indicate that lipase from Bacillus subtilis LP2 can be a valuable candidate for industrial applications such as organic synthesis and fats and oils industry due to their efficient catalysis in higher temperatures.  相似文献

We have isolated a mutant of Bacillus subtilis, strain 590, which is blocked at stage III of sporulation. The spo mutation which is carried by this strain is linked to pheA by transformation and defines a previously unidentified locus, spoIIIF. The spoIIIF locus is contiguous with the spoVB locus, in which a mutation causes a block at stage V of sporulation. We also give a detailed genetic map of the pheA region of the chromosome.  相似文献

Abstract cAMP was detected in Bacillus subtilis SB 19 and in a temperature sensitive mutant ts 33-6 under oxygen limitation. Growth rate and cAMP content were negatively correlated. Dinitrophenol and α-methylglucoside elicited an increase of the synthesis of cAMP and an increase of the intracellular cAMP content. In response to a decrease was increased. This increase of cAMP concentration did not occur if nitrate was present in the growth medium as an alternative terminal electron acceptor.  相似文献
