Summary Sample pretreatment is often the bottleneck of a tracelevel analytical procedure. In order to increase performance, increasing attention is therefore being devoted to combining sample pretreatment on-line with the separation technique that has to be used. In the present review, a variety of procedures in use today for sample treatment coupled on-line to capillary gas chromatography (GC) is briefly discussed. Special attention is devoted to coupled-column techniques such as SPE-GC and LC-GC (SPE, solid-phase extraction; LC, column liquid chromatography) which are topics of much current interest, also because of their frequent use in so-called hyphenated systems.  相似文献

Progress during the last 5 years in on-line LC-GC and related techniques is reviewed. In normal-phase LC-GC, the wire interface proved to have advantages over the loop type interface. Further investigations on the solvent evaporation process in an uncoated precolumn under conditions of an early vapour exit revealed that the rules for the transfer by the retention gap techniques must be modified. For reversed-phase LC-GC, approaches with a phase transfer compete with direct evaporation. Eluents were extracted into a bed of Tenax located in a programmed-temperature vaporiser and thermally desorbed. Direct evaporation is possible when a hot vaporising chamber is used and solvent/solute separation occurs in a separate compartment, a coated precolumn possibly in combination with packed beds. As a future strategy, LC-GC transfer techniques should be adjusted to those of large volume injection and involve a single device. It is believed that on-column injection/transfer is the choice. This requires that concurrent evaporation in LC-GC is performed by the on-column interface.  相似文献

建立了在线液相-气相二维色谱测定卷烟主流烟气中4-(N-甲基亚硝胺基)-1-(3-吡啶基)-1-丁酮(NNK)的方法。 NNK 的分析在在线凝胶气质联用仪上进行,采用自行装填的微型碱性氧化铝柱,并把仪器上的凝胶柱换成氧化铝柱,用于 NNK 的分析。捕集有主流烟气总粒相物的剑桥滤片用二氯甲烷提取,以 D4-NNK 为内标,提取液经微型氧化铝柱分离,含 NNK 的部位切割进入气相色谱,排干溶剂后启动气相色谱升温经毛细管柱进行分离,用质谱检测。本方法将烟气国标方法 NNK 测定中的氧化铝柱色谱净化和气相色谱-质谱分析在线连接起来,可不经样品前处理净化直接进样分析;每次进样可达40μL,是常规气相色谱-质谱分析最大进样量(2.0μL)的20倍,显著提高了分析灵敏度。方法线性范围达1.2~120 ng/ mL,相关系数为r=0.9998,回收率为93.9%~96.0%;检出限和定量限分别为0.25 ng/ mL 和0.9 ng/ mL,样品分析结果与中国烟草总公司企业标准方法进行对比,结果相符合。  相似文献

The present paper describes an investigation directed toward the development of a rapid heart-cutting LC-GC method for the analysis of mineral oil saturated hydrocarbons contained in vegetable oils. The automated LC-GC experiments were carried out by using a system equipped with a syringe-type interface, capable of both heart-cutting and comprehensive (LC × GC) two-dimensional analysis. The first dimension separation was achieved on a 100 mm × 3 mm ID × 5 μm d(p) silica column, operated under isocratic conditions (hexane). A single 30-s cut, corresponding to a 175 μL volume, was transferred to a programmed temperature vaporizer. After the large volume injection, the target analytes were separated in a rapid manner (~9 min) using a 15 m × 0.1mm ID × 0.1 μm micro-bore GC capillary. The overall LC-GC run time enables the analysis of ca. 4 samples/hour. Quantification was performed by using external calibration, in the 1-200 mg/kg range. The method was validated in terms of linearity, precision, limits of detection and quantification, and accuracy. A series of commercial samples were subjected to analysis. Various degrees of contamination were found in all samples, in the 7.6-180.6 mg/kg range.  相似文献

Phenolic compounds represent a class of highly complex naturally occurring molecules that possess a range of beneficial health properties. As a result, considerable attention has been devoted to the analysis of phenolics in a variety of samples. HPLC is the workhorse method for phenolic separation. However, conventional HPLC methods provide insufficient resolving power when faced with the complexity of real-world phenolic fractions. This limitation has been traditionally circumvented by extensive sample fractionation, multiple analysis methods and/or selective detection strategies. On the other hand, there is an increasing demand for improved throughput and resolving power from the chromatographic methods used for phenolic analyses. Fortunately, during the last decade, a number of important technological advances in LC have demonstrated significant gains in terms of both speed and resolution. These include ultra high-pressure liquid chromatography (UHPLC), high-temperature liquid chromatography (HTLC), multi-dimensional separations as well as various new stationary phase chemistries and morphologies. In recent years, these technologies have also found increasing application for phenolic analysis. This review seeks to provide an updated overview of the application of recent advances in HPLC to phenolic separation, with the emphasis on how these methodologies can contribute to improve performance in HPLC analysis of phenolics.  相似文献

Characterization of complex petrochemical samples has been a classical subject of comprehensive two-dimensional (2D) gas chromatography (GC x GC). Macroscopic properties of these samples can be described accurately by separation of compounds in classes of identical molecular functionality. Ring structures in the carbon backbone of these compounds, which can be divided in saturated and unsaturated, are amongst the foremost functionalities affecting samples properties. Unfortunately, GC x GC tuned for separation of both saturated and unsaturated ring structures is likely to result in convoluted chromatograms when a distribution of both molecular properties is present in the sample. An independent liquid chromatographic (LC) separation preceding GC x GC could be used to resolve the mixture based on unsaturated rings, allowing saturated rings to be resolved separately in the GC x GC separation. This three-dimensional separation (abbreviated LC-GC x GC) was performed after rigorous evaluation of LC as part of a multidimensional separation using LC x GC. Group-type separation was achieved using this separation for components with either saturated or unsaturated rings. Results of this separation were used to compare information obtained by GC x GC with LC-GC x GC.  相似文献

A fully automated on-line sample cleanup system based on the coupling of size exclusion chromatography to high resolution gas chromatography is described. The transfer technique employed is based on fully concurrent solvent evaporation using a loop-type interface, early vapor exit and co-solvent trapping. Optimization of the LC-GC transfer was done visually via an all-glass oven door. To circumvent the problem of mixing within the injection loop, an adaptation was made to the standard loop-type interface. The determination of a series of additives in a polymer matrix is presented as one example of the vast range of applications opened up by this technique.  相似文献

Gas chromatography is widely applied to separate, identify, and quantify components of samples in a timely manner. Increasing demand for analytical throughput, instrument portability, environmental sustainability, and more economical analysis necessitates the development of new gas chromatography instrumentation. The applications of resistive column heating technologies have been espoused for nearly thirty years and resistively heated gas chromatography has been commercially available for the last ten years. Despite this lengthy period of existence, resistively heated gas chromatography has not been universally adopted. This low rate of adoption may be partially ascribed to the saturation of the market with older convection oven technology, coupled with other analytical challenges such as sampling, injection, detection and data processing occupying research. This article assesses the advantages and applications of resistive heating in gas chromatography and discusses practical considerations associated with adoption of this technology.  相似文献

The enormous interest in proteomics research in recent years has inspired many developments in peptide chromatography. Different strategies have been developed to cope with the vast complexity of proteomics samples, trying to provide sufficient degree of separation to be able to exploit fully the potential of protein identification by mass spectrometry (MS). As reversed-phase liquid chromatography (RPLC) coupled to MS is still the method of choice for the analysis of protein digests, many efforts focus on the development of high-efficiency RP methods (e.g., monolithic columns and ultra-high-performance LC). This can also increase the speed and the sensitivity of the analysis of protein digests.As RPLC-MS alone is unlikely to provide sufficient resolution to unravel the composition of highly complex samples comprehensively, multidimensional methods will remain essential in proteome research. In this area, hydrophilic interaction chromatography (HILIC) seems to be a promising alternative to the traditional strong cation-exchange-based methods. Also, HILIC has found application in the analysis of post-translational modifications (e.g., phosphorylation and glycosylation).This review describes recent developments in LC methods for proteomics research, focusing on advances in column technology and the application of novel column materials. Illustrative examples show the possibilities of the new columns in proteomics research.  相似文献

Summary Normal-phase LC (NPLC) is a powerful method for the clean-up of fatty samples in the determination of organochlorine pesticides (OCPs). The injected sample deactivates the stationary phase and the triglyceride matrix therefore serves as a polarity modifier in the NPLC separation. Thus, the amount of sample injected is the key to both selectivity and sensitivity in matrixmodified LC coupled to capillary GC. In coupled LC-GC the NPLC separation becomes particularly critical because only a limited amount of the LC eluent can be transferred to the GC and the triglyceride matrix must be prevented from entering the GC, because it degrades the performance of the injector and the column. In previous applications method development was seriously hampered by these boundary conditions and tedious and lengthy trial-and-error experiments were required to determine suitable experimental conditions. In this study an empirical model was developed that describes the NPLC separation process in terms of column dimensions and fat loadability. The output is given as the probability of achieving successful LC-GC analysis of a particular set of analytes, thus furnishing a useful tool for the development of new applications in the field of exposure assessment and analysis of residues of apolar compounds in fatty samples. The limitations of current procedures—maximum transfer volumes and minimal separation—are also discussed.  相似文献

对石油地质样品的化学组成进行全面准确的剖析,可以获得丰富的地球化学信息,为油气勘探工作提供科学依据。然而,该类样品除了组成复杂之外,还易受到各种物理(如蒸发、乳化、扩散、溶解和吸附)、化学(如光降解)和生物(如微生物降解)过程的影响。这些特点给样品的分析研究工作带来了极大的困难,传统的一维气相色谱/质谱技术很难对其进行理想的分离。全二维气相色谱(GC×GC)作为新发展起来的一种分离技术,在复杂样品分析方面具有独特的优势,虽然在石油地质样品分析中的应用相对较晚,但也日益受到关注。本文主要综述了近5年来GC×GC在石油地质方面国内外的研究进展以及存在的主要问题,并对今后的研究进行了展望。  相似文献

The concept of unified chromatography has been in existence for 50 years after the work of Giddings proposing that all modes of chromatography (gas chromatography, liquid chromatography, supercritical fluid chromatography and so on) may be treated together under a single unified theory. His idea was partially fulfilled 23 years later by Ishii, Takeuchi and colleagues, who demonstrated for the first time the possibility to analyze a complex sample containing substances with a wide range of boiling points and polarities in the same instrument and column, just by varying the mobile phase pressure and temperature to change from one chromatographic mode to another. This approach has been demonstrated through application to the separation of complex mixtures in several areas including crude oil, edible oils and polymers. Still, unified chromatography has not yet been fully developed. In the present work, we will review the fundamentals, instrumentation and several applications of the technique. Also discussed are the drawbacks that still hinder development, as well as the recent developments and trends in instrumentation and columns that suggest the most feasible ways forward to the full development of unified chromatography.  相似文献

Retention prediction models for reversed-phase liquid chromatography (RPLC) have been extensively studied owing to the fact that RPLC remains the most widely used chromatographic technique especially in the field of pharmaceutical and biomedical analyses. However, RPLC is not always the method of choice for the analysis of some compounds that have high polarity. Hydrophilic interaction chromatography (HILIC) has been gaining interest in the last few years as an alternative option to RPLC for the analysis of polar and hydrophilic analytes. HILIC is a variant of normal-phase liquid chromatography, but utilizes water in a water-miscible organic solvent as the eluent in conjunction with a hydrophilic stationary phase. The present review aims to summarize recent contributions on the development of retention prediction models for a group of basic analytes, namely, the adrenoreceptor agonists and antagonists, on different polar stationary phases. The use of multiple linear regression and artificial neural networks in model building is highlighted.  相似文献

Counter-current chromatography is a chromatographic separation and purification technique being developed. The development of different elution modes has significantly contributed to this field. Multiple dual-mode elution is a method developed based on dual-mode elution, which consists of a series of changing cycles of the phase role and the direction by switching between normal and reverse elution modes of counter-current chromatography. This dual-mode elution method takes full advantage of the liquid nature of stationary and mobile phases of counter-current chromatography and effectively improves the separation efficiency. So, this unique elution mode has gained extensive attention for separating complex samples. This review mainly describes and summarizes in detail its development, applications, and characteristics in recent years. Meanwhile, its advantages, limitations, and future outlook also have been discussed in this paper.  相似文献

Design of experiment (DoE) techniques have been widely used in the field of chromatographic parameters optimization as a valuable tool. A systematic literature review of the available DoE techniques applied to the development of a chromatographic analysis method is presented in this paper. First, the most common available designs and the implementation steps of DoE are comprehensively introduced. Then the studies in recent 10 years for the application of DoE techniques in various chromatographic techniques are discussed, such as capillary electrophoresis, liquid chromatography, gas chromatography, thin-layer chromatography, and high-speed countercurrent chromatography. Current problems and future outlooks are finally given to provide a certain inspiration of research in the application of DoE techniques to the different chromatographic techniques field. This review contributes to a better understanding of the DoE techniques for the efficient optimization of chromatographic analysis conditions, especially for the analysis of complex systems, such as multicomponent drugs and natural products.  相似文献
