用根瘤匀浆法,从粗枝木麻黄(Casuarina glauca)根瘤中分离到一株内生菌FCg77。生物学特性试验表明:该菌株的适宜分离培养基为BAP培养基,最佳碳源为吐温-80,最适氮源为牛肉膏,能耐5%的盐分,生理类型为AB型,细胞壁化学组分为Ⅲ型。结合回接试验结果,初步判定分离菌株FCg77应属于弗兰克氏菌(Frankia)的成员。  相似文献

Immunogold localization of hydrogenase in free-living Frankia CpI1   总被引:1,自引:0,他引:1  
Abstract The free-living Frankia strain CpI1 cultured under nitrogen-fixing and non-nitrogen-fixing conditions was investigated for occurrence of hydrogenase protein by Western blots. Transmission electron microscopy and immunocytological labelling were used to study the distribution of hydrogenase in the Frankia strain.
Western immunoblots revealed that a 72-kDa protein in the Frankia strain CpI1 was immunologically related to the large subunit of a dimeric hydrogenase purified from Alcaligenes latus . Immunolocalization showed that the hydrogenase protein is located both in vesicles and hyphae in Frankia strain CpI1 grown in a nitrogen-free medium. Earlier reports that nitrogenase is localized in the vesicles [1,2], together with this finding, point out a possible role for hydrogenase in increasing relative efficiency of nitrogen fixation. In CpI1 grown in media containing nitrogen (lacking vesicles), the enzyme was evenly distributed in the hyphae. The impact of this result has to be further analysed.  相似文献

从浙江省短枝木麻黄(Casuarina equisetifolia)和细枝木麻黄(C.curninghamiana的根瘤中共分离获得14株共生菌株.形态观察表明,菌株具有分枝状菌丝、多腔孢囊、泡囊等典型的Frankia结构、16S rDNA测序结果表明,供试菌株均为Frankia,其中4株属于生理类群A,7株属于生理类群B,3株属于生理类群AB.固氮效应研究表明,菌株均具有固氨酶生物学活性,但菌株之间存在显著差异,其中菌株ZCN192固氨酶活性最强,可达2.897 μmol·mg-1·h-1,菌株ZCN199最低,固氮酶活性为0.056 μmol·mg-1·h-1.活体固氮试验显示,与阴性对照相比,供试菌株能显著提高苗高、地径和干重,且一般情况下,离体固氮酶活性强的菌株在活体接种时能获得更明显的固氮效应.  相似文献

AIMS: In order to depict the fine interactions that lead to nodulation, absolute microbiological control of the symbiotic partners is required, i.e. the ability to obtain in vitro axenic nodulation, a condition that has never been fulfilled with the Casuarina-Frankia symbiosis. The effects of culture conditions on plant growth and nodule formation by Casuarina cunninghamiana were investigated. METHODS AND RESULTS: Axenic (capped tubes with different substrates), and nonaxenic cultures (Gibson tubes, pot cultures) were tested. In axenic conditions, C. cunninghamiana, inoculated with Frankia, had poor growth and did not form nodules at 6 weeks. Plants cultivated in Gibson tubes reached the four axillary shoots stage within 6 weeks and formed nodules 4 weeks after inoculation. Sand-pot cultures allowed us to relate the plant development stage at inoculation with nodulation. CONCLUSIONS: The sterile replacement of the cap by a plastic bag increased plant growth and enabled nodule formation 6 weeks after inoculation. The new system of plant culture allows the axenic nodule formation 6 weeks after inoculation. Nodulation behaviour is related to plant development and confinement. SIGNIFICANCE AND IMPACT OF THE STUDY: This axenic plant nodulation system is of major interest in analysing the roles of Frankia genes in nodulation pathways.  相似文献

Occurrence and activity of the hydrogen uptake enzyme were studied in root nodule homogenates made from plants of Alnus incana (L.) Moench collected from field sites in the northern part of Sweden. Nitrogenase (EC 1.7.99.2) activity (estimated by acetylene reduction) and hydrogen evolution were studied in excised nodules. All Frankia sources showed acetylene reduction activity, and possessed a hydrogen uptake system. Hydrogen uptake in nodule homogenates from the Frankia sources measured at 23.8 μM H2 ranged from 0.04 to 5.0 μmol H2 (g fresh weight nodule)−1 h−1. The H2 uptake capacity of nodule homogenates from one of the Frankia sources was almost 8 times higher than the hydrogen evolution from nitrogenase, both expressed on a nodule fresh weight basis. Frankia sources from field sites 6 and 11 showed Km for H2 of 13.0 and 23.6 μM H2, respectively. This indicates similarities in the hydrogen uptake enzymes in the two Frankia sources. It is concluded that hydrogen uptake is a common characteristic in Frankia.  相似文献

A mutant derivative of hycE, the gene for the large subunit of hydrogenase 3 from Escherichia coli, was constructed that lacks the 3′-terminal part encoding the C-terminal portion of the HycE polypeptide, which is proteolytically removed during maturation of the hydrogenase. The truncated gene was transferred to the in situ position on the chromosome. Although the mutant possessed HycE in its "mature" form, it was devoid of hydrogenase 3 activity. The activity was not restored by high nickel concentrations in the medium. The mutated HycE was not associated with detectable radioactivity when the strain was grown in the presence of 63Ni2+. These results indicate that the C-terminal extension in the precursor form of the large subunit keeps the protein in a conformation required for the coordination of the metal. Superoxide dismutase and catalase activities in purified Frankia vesicles   总被引:2,自引:0,他引:2  
Superoxide dismutase (EC 1.15.1.1.) and catalase (EC 1.11.1.6.) activities of Frankia cells grown in the presence of ammonium were very high in comparison with those of other prokaryaotes and particularly Rhizobium . Furthermore, these activities were significantly enhanced under nitrogen-fixing conditions where vesicles were produced. By using a single-step sucrose gradient, Frankia vesicles were isolated and appeared intact and free of hyphal contamination. The contents of superoxide dismutase and catalase in the purified vesicles were similar to those in preparations containing both vesicles and hyphae. These results suggest an important role of superoxide dismutase and catalase in the protection of the overall nitrogen-fixation process against O2 in Frankia vesicles. Beside the protective role played by the thick walls of the vesicles, the presence of specialized enzymes is emphasized.  相似文献

Isotope substitution of 57Fe (I = 12) for 56Fe has a pronounced effect on the two EPR signals of hydrogenase of Chromatium vinosum. It is proposed that signal 1, the intensity of which is increased several-fold by a deoxygenation-oxygenation cycle with a simultaneous increase of a signal from Fe3+, is due to a [3Fe-xS] cluster. It is further proposed that signal 2 is caused by a magnetic interaction of a [4Fe-4S]3+ cluster with an unidentified paramagnet. The addition of 10 μM Ni to the culture medium (already containing 1 μM Ni) increased the enzyme activity 3–6-fold, without effect on the growth of the bacterium. Addition of 61Ni (I = 32) to the medium did not change the EPR spectrum of hydrogenase. From a comparison of the EPR signal intensities and the enzyme activities it is concluded that, in the hydrogenase preparation as isolated, molecules containing a [3Fe-xS) cluster are not active, and that active molecules have a [4Fe-4S]3+(3+,2+) cluster plus an as yet unidentified paramagnetic redox component. The latter is thought to be the primary site of interaction of the enzyme with H2. Ni is considered as a possible candidate for this component.  相似文献

The H2 uptake activity (units/mg protein) of Clostridium pasteurianum cells with methylene blue as the electron acceptor increases with cell density independent of the growth conditions. The H2 evolution activity (units/mg protein) of the same cells with reduced methyl viologen as the electron donor remains fairly constant under all growth conditions tested. Cells grown under N2-fixing conditions have the highest H2 uptake activity and were used for the purification of hydrogenase II (uptake hydrogenase). Attempts to separate hydrogenase II from hydrogenase I (bidirectional hydrogenase) by a previously published method were unreliable. We report here a new large-scale purification procedure which employs a rapid membrane filtration system to fractionate cell-free extracts. Hydrogenases I and II were easily filtered into the low-molecular-weight fraction (Mr less than 100 000), and from this, hydrogenase II was further purified to a homogeneous state. Hydrogenase II is a monomeric iron-sulfur protein of molecular weight 53 000 containing eight iron atoms and eight acid-labile sulfur atoms per molecule. Hydrogenase II catalyzes both H2 oxidation and H2 evolution at rates of 3000 and 5.9 μmol H2 consumed or evolved/min per mg protein, respectively. The purification procedure for hydrogenase II using the filtration system described greatly facilitates the large-scale purification of hydrogenase I and other enzymes from cell-free extracts of C. pasteurianum.  相似文献

The research on the preparation of Frankia solution, seedling culture and inoculation was carried out. Results indicated that the collective amount of cells may be increased when the logarithm phase cells were used as the inoculum and regularly homogenizing and transplanting inoculum by the method of magnetic stirring were adopted during the cultural process of Frankia. The inoculative tests verified that the strain with the highest ability to infect Elaeagnus mollis Diels is SIB 1301118 isolated from Elaeagnus angustifolia L. other than SIB 1332281 isolated from Elaeagnus mollis Diels. The effectivities of inoculation for germinating seeds and the seedlings to be transplanted were seed soaking and root soaking in Frankia solution preparel respectively. Early inoculation was beneficial to the growth and nodulation of seedings. The fefectivity of late inoculation was bad. Sand-soil mixture (2:1) was properly suitable to the seed germination and seedling growth. The seedling growth and nodulative rate could be promoted when 1/4 Sileris-Yong nutrient solution was suppplimented regularly.  相似文献

In order to investigate the effects of Al on nodule formation and function in the Casuarina-Frankia symbiosis, inoculated plants were grown in sand culture at five nominal Al concentrations (0-880 M Al) at pH 4.0. There was an Al-free control at pH 6.0 to assess the effects of pH 4.0 treatments. Mean N concentration of nodules was significantly less at pH 4.0 (1.83%) than at pH 6.0 (2.01%). There were nodulated plants at all Al levels, though there were fewer nodulated plants at 440 and 880 M Al. Dry weights of nodules, shoots and roots were not reduced by Al concentrations at or below 220 M Al, but were decreased by Al concentrations at or above 440 M Al. Nodule weight expressed as a percentage of total weight did not differ significantly with respect to an Al-free control at pH 4. N concentrations of shoots and whole plants were significantly reduced at 440 M Al. Nodular specific acetylene reduction activity (ARA) did not differ significantly among Al treatments. However, N2-fixation efficiency was decreased from 0.20 to 0.10 mg N fixed mg nodule dry weight–1 at 880 M Al.  相似文献

It has been established that the hydrogenase from autotrophically cultured Bradyrhizobium japonicum contains selenium as a bound constituent. About 80% of the enzyme selenium remains bound during precipitation with 5% trichloroacetic acid (TCA). However, 85% of the selenium bound to the enzyme is released by a combined treatment of urea, heat and TCA. Neither selenomethionine nor selenocysteine could be detected on analysis of anaerobically hydrolyzed enzyme. These results are consistent with the report showing that the structural genes for this enzyme do not contain a TGA codon (Sayavedra-Soto et al. 1988) which has been reported to code for selenocysteine incorporation into several proteins (Chambers et al. 1986; Zinoni et al. 1986; Stadtman 1987). We have demonstrated that 75Se from the labeled hydrolyzed enzyme forms the derivative' selenodicysteine. The form of selenium resulting in the synthesis of this derivative apparently is SeO inf3 sup= or a compound such as Se= which is easily oxidized to SeO inf3 sup= . In a separate approach it was established that 12–16% of the total 75Se in the native enzyme reacted with 2,3-diaminonaphthalene indicating that this fraction was present as SeO inf3 sup= . The remaining 75Se was bound to the enzyme protein. From this research, we concluded that Se in Bradyrhizobium japonicum hydrogenase is present in a labile bound form. In this respect, this enzyme is similar to xanthine dehydrogenase and nicotinic acid hydroxylase, both of which contain labile Se constituents that have not been defined.Technical paper no. 8980 from the Oregon Agricultural Experiment Station  相似文献

An in vitro experiment was conducted to investigate the effects of different sources and levels of P supply on growth, viability and phosphatase activity of three tropical Frankia strains isolated from Casuarina. P concentration for optimum growth was between 0.1 and 10.0 M in the absence of external combined nitrogen. Specific viability was not influenced by P supply. Morphological features of Frankia, such as hyphal length and vesicle numbers, were found to largely mirror growth. Phosphatase activity was detected in all three Frankia strains and was highest when P was omitted from the culture solution. There were more than 10-fold differences between the Frankia strains in the level of phosphatase activities shown. This study suggested that soils low in P are unlikely to restrict micro-symbiont growth activity.  相似文献

Actinorhizal nodules do not usually evolve H2 due to the action of an uptake hydrogenase. We have found that nodules of several Frankia symbioses evolved large amounts of H2 gas when returned to air following exposure to 10 kPa C2HT2 during an acetylene reduction assay. Increased H2 evolution in air persisted for several days when intact root systems of Alnus incana (L.) Moench (inoculated with Frankia UGL 011101) were treated with 10 kPa C.H2 for 1 h. Full recovery of uptake hydrogenase activity required 4 to 8 days. Studies with crude homogenates of nodules of the same plants showed that hydrogenase (measured amperometrically with phenazine metho-sulfate as electron acceptor) was directly affected, since activity in treated nodules was only 10% of that in untreated nodules. A survey of actinorhizal symbioses revealed variation in the effect of an acetylene reduction assay on hydrogen metabolism. Nodules of three species, including Alnus rubra Bong, inoculated with Frankia HFPArD. showed complete inactivation of hydrogenase. H2 evolution in air was 25% of the C2H2 reduction rate and H, evolution in Ar/O2 was equal to the QH2 reduction rate. Two symbioses, Ceanothus americanus L. (soil inoculant) and Batista glomerata Baill. (soil inoculant) showed no change following an acetylene reduction assay. A third group of symbioses showed an intermediate response.  相似文献

Four species of Casuarina were raised in the glasshouse and inoculated with nodules collected from nine different geographical areas within Australia. Isolations ofFrankia were attempted from 10 of the Casuarina-Frankia nodule combinations using two methods, a nodule dissection and a filtration method. With both techniquesFrankia isolates were obtained from four of the 10Frankia sources. Spores were not observed in sections of nodules from the four sources from whichFrankia was isolated, whereas spores were observed in the remaining six nodule sources. For selected nodule sources a range of isolation media were tried, but no improvement in the isolation success rate was achieved. The effect of host species on ease of isolation was studied. The results obtained suggested it was theFrankia strain and not the host plant species which determined the ease of isolation from Casuarina nodules.  相似文献

Upon partial reduction of hydrogenase from Chromatium vinosum with ascorbate plus phenazine methosulphate, EPR signals due to Ni(III) and a [3Fe-xS] cluster appear simultaneously and with equal intensities. Since the intact enzyme shows no S = 12 signals, it is concluded that Ni(III) and a [4Fe-4S]3+ cluster interact magnetically in such a way as to prevent the detection of the two paramagnets as individual S = 12 systems. This interaction is thought to be the origin of a signal in which Fe is involved and which is not due to an S = 12 system (Albracht, S.P.J., Albrecht-Ellmer, K.J., Schmedding, D.J.M. and Slater, E.C. (1982) Biochim. Biophys. Acta 681, 330–334). A variable fraction of the enzyme preparation shows signals due to Ni(III) and a [3Fe-xS] cluster with equal intensities without any further treatment. These are thought to be derived from irreversibly inactivated enzyme molecules. The enzyme contains no selenium.  相似文献

An apparatus is described in which the root systems of up to ten plants can be simultaneously incubated with 15N2 gas mixtures, while the aerial environment remains undisturbed. The size of incubation chamber and the number of chambers can be adjusted depending on the size and growth stage of the plants being tested. The apparatus has the facility for the recovery of gas mixtures, sampling and adjustment of gas concentrations, and control of soil moisture status within a sealed system. Possible experimental applications are suggested and some preliminary results presented which demonstrate bacterial nitrogen fixation and uptake into Sorghum seedlings.  相似文献
