目的:系统地评价火棘果粗分体系抗氧化活性,同时探究抗氧化能力与总多酚含量之间的关系。方法:用75%乙醇冷浸提取火棘果,分别用石油醚、乙酸乙酯、正丁醇和水分为四个不同极性部位,然后测定了各部位萃取物对DPPH自由基、ABTS+自由基的清除能力,并且测定了总还原力、FRAP值和总多酚含量,同时考察总酚含量与抗氧化活性的关系。结果:火棘果提取物的不同极性部位均有抗氧化活性,其中水、乙酸乙酯和正丁醇部位提取物表现出较好的抗氧化活性,石油醚部位的抗氧化活性最弱;各部位提取物的抗氧化活性与总多酚含量呈现较好的相关关系。其中,水部位提取物对DPPH和ABTS+自由基清除率最高,IC50值分别为(0.76±0.03)mg/m L和(1.71±0.10)mg/m L;乙酸乙酯部位FRAP值最大,为(382.20±4.72)μmol Fe2+/g干样;正丁醇部位总酚含量最高,为(2763±3.91)mg GAE/100g干样。  相似文献

研究定心藤75%乙醇提取物不同极性部位的体外抗氧化活性,采用5种不同极性溶剂萃取(石油醚、氯仿、乙酸乙酯、正丁醇、水)得到5种萃取物,分别测定黄酮含量,并通过还原能力测定、DPPH自由基清除能力、羟自由基清除能力和超氧阴离子自由基清除能力4种测定方法,对各萃取物抗氧化能力进行研究。结果显示,各提取物对Fe3+均有一定的还原能力,乙酸乙酯萃取物还原效果最为明显;定心藤不同极性部位均具有一定的抗氧化活性,乙酸乙酯萃取物抗氧化能力最强,DPPH·的半数清除浓度IC50为0.28mg/m L,羟自由基半数清除浓度IC50为0.41mg/m L,在0.2mg/m L浓度下,对超氧阴离子自由基的清除率达到42.88%,而其黄酮含量最高,达到609.96mg/g,这也许与它具有较高的抗氧化活性有关。说明乙酸乙酯萃取物具有较好的开发天然抗氧化剂的前景。  相似文献

以乳苣全草为原料,75%乙醇为溶剂提取乳苣,提取液用不同极性溶剂依次萃取,得石油醚萃取物、乙酸乙酯萃取物、正丁醇萃取物和水萃取物,同时测定了各相提取物对羟自由基、超氧阴离子自由基、DPPH自由基的清除能力,并与VC进行比较。结果表明:各萃取物均具有一定的抗氧化活性,且随着浓度的增加而增强;对羟自由基的清除能力大小依次为乙酸乙酯萃取物>正丁醇萃取物>VC>水萃取物>石油醚萃取物;对超氧阴离子自由基清除能力大小依次为VC>正丁醇萃取物>乙酸乙酯萃取物>石油醚萃取物>水萃取物;对DPPH自由基清除能力大小依次为VC>乙酸乙酯萃取物>正丁醇萃取物>水萃取物>石油醚萃取物;各萃取物还原能力大小依次为VC>正丁醇萃取物>乙酸乙酯萃取物>水萃取物>石油醚萃取物。  相似文献

为比较覆盆子不同极性溶剂提取物的抗氧化活性,分别用正丁醇、乙酸乙酯、丙酮、去离子水、50%乙醇和石油醚进行萃取,测定各提取物总多酚和总黄酮的含量,并通过清除DPPH自由基、ABTS自由基、羟基自由基和总还原能力4种体系对其体外抗氧化活性进行评价。结果显示,乙酸乙酯提取物中总多酚含量最高,50%乙醇提取物中总黄酮含量最高;50%乙醇提取物清除DPPH自由基能力和ABTS自由基能力最强,水提取物清除羟基自由基能力最强,丙酮提取物总还原能力最强。结果表明覆盆子不同极性溶剂的提取物均有抗氧化活性,可作为天然抗氧化剂开发。  相似文献

对芒果叶不同极性部位提取物的抗氧化活性进行研究。用DPPH法考察最优反应条件,综合评价其抗氧化活性,测定其黄酮含量,以IC50值作为评价清除DPPH自由基能力的指标,用芒果苷和抗坏血酸作为对照品进行自由基清除率的比较。试验结果表明:芒果叶石油醚、乙酸乙酯、正丁醇萃取物及水部位提取物的黄酮含量依次为:(33.616±3.208)、(292.063±1.100)、(180.469±2.067)、(21.717±1.157)mg/g,IC50值分别为:102.877、9.014、19.494、221.784μg/mL。由此可以看出,芒果叶各类提取物均具有一定的抗氧化活性,其中乙酸乙酯萃取物抗氧化活性最强,其次为正丁醇、石油醚,最后为水部位提取物。相关性分析结果表明,芒果叶各极性部位提取物的黄酮含量与清除DPPH自由基的能力呈负相关,芒果叶各部位提取物的黄酮含量是影响其抗氧化活性的主要因素。  相似文献

目的筛选乌饭树果乙醇提取物中抗氧化活性最强的极性分部。方法采用石油醚、乙酸乙酯、氯仿、正丁醇和水等5种不同极性溶剂对乌饭树果乙醇提取物进行进一步梯度萃取,并分析比较其总抗氧化能力(total antioxidant capacity,T_(AC))、抗脂质过氧化能力及对羟基自由基OH·、超氧阴离子自由基O_2~-·的清除效果。结果乌饭树果乙醇提取物的5个不同极性分部均具有一定的抗氧化活性,但其活性大小差异极显著(P0.01),T_(AC)大小依次为:乙酸乙酯部氯仿部正丁醇部石油醚部水部。同时,在OH·、O_2~-·自由基清除能力和抗脂质过氧化能力上,乙酸乙酯部均优于氯仿部和正丁醇部,且呈明显的量效关系。结论乌饭树果乙醇提取物的乙酸乙酯部具有最强的抗氧化活性,且对OH·、O_2~-·的清除能力及抗脂质过氧化能力均强于L-抗坏血酸及其它部位,可作为天然抗氧化剂的潜在有效来源。  相似文献

为了研究大蒜不同极性萃取物的抗氧化活性,确定大蒜的活性部位,促进大蒜资源在天然抗氧化剂和药品等领域的应用,文中采用体积分数80%乙醇回流提取、梯度萃取得到石油醚萃取物、乙酸乙酯萃取物、正丁醇萃取物和水萃取物4个不同极性部位。以清除二苯代苦味酰基(DPPH)自由基、还原力和对肝脂质过氧化抑制力为评价指标研究其体外抗氧化活性。结果表明:大蒜不同极性萃取物均有一定的抗氧化活性,其抗氧化能力与不同极性萃取物的质量浓度呈现出良好的线性依赖关系,抗氧化效果因反应体系不同而有所不同。正丁醇萃取物在3个体外抗氧化活性模型中均表现出较好的抗氧化活性,其清除DPPH自由基和肝脂质过氧化抑制活性的半数抑制浓度分别为0.46 mg/m L和0.36 mg/m L。大蒜正丁醇萃取物得率较高(7.86%),抗氧化活性较好,可以作为新型有效的天然抗氧化物质进行开发利用。  相似文献

研究玫瑰茄的体外抗氧化活性。采用不同极性的有机溶剂萃取玫瑰茄40%乙醇提取物(粗提物),得到石油醚萃取物、氯仿萃取物、乙酸乙酯萃取物、正丁醇萃取物,然后采用Folin-Ciocalteu比色法测定粗提物以及各萃取物的多酚含量,同时应用DPPH法、试剂盒法、ABTS法分别测定粗提物和各萃取物的DPPH自由基清除能力、还原Fe3+能力以及ABTS自由基抑制能力。DPPH法、试剂盒法、ABTS法测定各提取物抗氧化能力的结果为粗提物>正丁醇萃取物>乙酸乙酯萃取物>氯仿萃取物>石油醚萃取物。玫瑰茄提取物具有较高的多酚含量和较强的抗氧化能力;玫瑰茄提取物的多酚含量与抗氧化能力之间存在较好的相关性。  相似文献

研究香椿子不同极性萃取物体外抗氧化活性。采用75%乙醇提取、不同极性有机溶剂萃取,得到石油醚、乙酸乙酯、正丁醇、水4个萃取物,考察其总还原力、DPPH·清除能力,比较香椿子不同极性萃取物抗氧化活性;UV法初步判断所含官能团和化合物类别。结果表明,香椿子不同极性萃取物具有一定抗氧化作用,总还原力顺序为:石油醚乙酸乙酯正丁醇水;DPPH·清除能力顺序为:乙酸乙酯水石油醚正丁醇,其中石油醚和正丁醇线性关系较好,EC50值分别为0.490 7 mg/m L和0.609 1 mg/m L;UV法推断香椿子中可能含蒽醌类、苯丙素类、木脂素类、香豆素类等酚羟基成分。  相似文献

研究两种真姬菇不同溶剂提取物的抗氧化活性和多酚含量。制备真姬菇粗提物、正己烷萃取物、乙酸乙酯萃取物、正丁醇萃取物与水萃取物,分析它们的抗氧化活性与测定它们的多酚含量。结果表明:闽真2号正己烷萃取物的总抗氧化能力最好,乙酸乙酯萃取物清除DPPH自由基能力最强,正丁醇萃取物清除ABTS自由基能力最强;闽真3号乙酸乙酯萃取物的总抗氧化能力、清除DPPH与ABTS自由基能力均最好。相关性分析结果表明,真姬菇多酚含量与DPPH自由基清除能力具有一定的相关性。综合考虑抗氧化活性与萃取率,正己烷萃取物与乙酸乙酯萃取物可被作为天然抗氧化剂应用于食品科学领域。  相似文献

Since grapevine ( Vitis spp .) rootstock material is being traded increasingly as disbudded woody material a lack of distinctive morphological features on such material necessitates an alternative and reliable means of identification. Methods described here were developed for rapid and efficient extraction of DNA from woody samples rich in phenolic compounds and polysaccharides, and for subsequent identification of varieties by RAPD PCR. Using these methods, and with the application of only one selected RAPD primer, we were able to differentiate sixteen rootstock varieties, including the seven varieties most commonly used in Germany. Problems commonly encountered with reproducibility of RAPD patterns were avoided by choosing primers with a dinucleotide sequence and a high G/C content that allowed a rather high annealing temperature of 45°C. Methods described here should also be useful for other horticultural crops, especially those with woody tissues rich in phenolic compounds and polysaccharides.  相似文献

The characterization of the aromatic profile of several apricot cultivars with molecular tracers in order to obtain objective data concerning the aromatic quality of this fruit was undertaken using headspace–solid phase microextraction (HS–SPME). Six apricot cultivars were selected according to their organoleptic characteristics: Iranien, Orangered, Goldrich, Hargrand, Rouge du Roussillon and A4025. The aromatic intensity of these varieties measured by HS–SPME–Olfactometry were defined and classified according to the presence and the intensity of grassy, fruity and apricot like notes. In the six varieties, 23 common volatile compounds were identified by HS–SPME–GC–MS. Finally, 10 compounds, ethyl acetate, hexyl acetate, limonene, β-cyclocitral, γ-decalactone, 6-methyl-5-hepten-2-one, linalool, β-ionone, menthone and (E)-hexen-2-al were recognized by HS–SPME–GC–O as responsible of the aromatic notes involved in apricot aroma and considered as molecular tracers of apricot aromatic quality which could be utilized to discriminate apricot varieties.  相似文献

The advent of the functional barrier concept in food packaging has brought with it a requirement for fast tests of permeation through potential barrier materials. In such tests it would be convenient for both foodstuffs and materials below the functional barrier (sub-barrier materials) to be represented by standard simulants. By means of inverse gas chromatography, liquid paraffin spiked with appropriate permeants was considered as a potential simulant of sub-barrier materials based on polypropylene (PP) or similar polyolefins. Experiments were performed to characterize the kinetics of the permeation of low molecular weight model permeants (octene, toluene and isopropanol) from liquid paraffin, through a surrogate potential functional barrier (25 μm-thick oriented PP) into the food simulants olive oil and 3% (w/v) acetic acid. These permeation results were interpreted in terms of three permeation kinetic models regarding the solubility of a particular model permeant in the post-barrier medium (i.e. the food simulant). The results obtained justify the development and evaluation of liquid sub-barrier simulants that would allow flexible yet rigorous testing of new laminated multilayer packaging materials.  相似文献

This paper describes the second part of a project undertaken to develop certified mussel reference materials for paralytic shellfish poisoning toxins. In the first part two interlaboratory studies were undertaken to investigate the performance of the analytical methodology for several PSP toxins, in particular saxitoxin and decarbamoyl-saxitoxin in lyophilized mussels, and to set criteria for the acceptance of results to be applied during the certification exercise. Fifteen laboratories participated in this certification study and were asked to measure saxitoxin and decarbamoyl-saxitoxin in rehydrated lyophilized mussel material and in a saxitoxin-enriched mussel material. The participants were allowed to use a method of their choice but with an extraction procedure to be strictly followed. The study included extra experiments to verify the detection limits for both saxitoxin and decarbamoyl-saxitoxin. Most participants (13 of 15) were able to meet all the criteria set for the certification study. Results for saxitoxin.2HCl yielded a certified mass fraction of <0.07 mg/kg in the rehydrated lyophilized mussels. Results obtained for decarbamoyl-saxitoxin.2HCl yielded a certified mass fraction of 1.59+/-0.20 mg/kg. The results for saxitoxin.2HCl in enriched blank mussel yielded a certified mass fraction of 0.48 +/- 0.06 mg/kg. These certified reference materials for paralytic shellfish poisoning toxins in lyophilized mussel material are the first available for laboratories to test their method for accuracy and performance.  相似文献
