Objective To explore the expression and the value of HIF-1α,and ET-1 in judging the prognosis of gastrointestinal stromal tumors (GISTs). Methods The expression of HIF-1α, and ET-1 protein was examined in 76 GISTs by immunohistochemistry S -P methods. Results There was a positive correlation between the expression of HIF-1 α and ET-1 ( P < 0.05 ). The positive expression rate of HIF-1 α and ET-1 was 73.68% (50/76) ,and 65.79% (50/76) respectively,which was related with histologicial grade, tumor diameter, infiltration and metastasis, nuclear division rating of GISTs ( P < 0.05 ), but had no relationship with patients' age, gender, initial position of the tumor ( P > 0.05 ). There was statistically sig-nificant difference in the expression of HIF-1 α and ET-1 in the following groups:among the three classes of very low-risk and low-risk, middle-risk, high-risk, bewteen the diameter < 2 cm and > 5 cm (P < 0.05). The more malignant degree and larger diameter, the more highly positive expression rate ( P < 0.05 ). The positive expression in the groups with infiltration and metastasis, and nuclear division ≥5/50 HP was sig-nificantly higher than the groups without infiltration and metastasis, and nuclear division < 5/50 HP (P < 0.05). Conclusion The expression of HIF-1α had a significant correlation with ET-1. HIF-1α,and ET-1 expression was closely related with the prognosis of GISTs,and can serve as important predictors for survival.  相似文献

目的 探讨缺氧诱导因子(HIF)-1α,血管内皮素(ET)-1在胃肠道间质瘤(GISTs)中的表达及其在预后判断中的价值.方法 应用免疫组织化学SP法对76例GISTs检测,观察HIF-1α、ET-1在GISTs中的表达情况.结果 HIF-1α、ET-1在GISTs中的阳性表达率分别为73.68%(56/76)、65.79(50/76),两者的表达呈正相关(P<0.05),其与胃肠道间质瘤的组织学分级、肿瘤直径、浸润转移和核分裂数有关(P<0.05),而与患者的年龄、性别、肿瘤原发部位无关(P>0.05);HIF-1α、ET-1在极低和低危险,中度危险和高危险及直径<2 cm和直径>5 cm组间差异有统计学意义(P<0.05),阳性表达率随恶性程度的增加和肿瘤直径的增大而增加(P<0.05),在有浸润转移和核分裂数≥5/50HP组中的表达明显高于无浸润转移和核分裂数<5/50HP组中的表达(P<0.05).结论 HIF-1α、ET-1在GISTs中的表达有联合倾向,可能提示肿瘤的恶性程度及其预后,并可作为判断预后的有用标志物.  相似文献

目的探讨葡萄糖转运蛋白1(Glut1)和缺氧诱导因子-1α(HIF-1α)在原发性肝细胞癌中的表达,以及在肝细胞癌发生发展过程中相互关系。方法应用免疫组织化学方法(SP法),检测108例肝细胞癌及其癌旁组织、15例正常肝组织中Glutl,HIF-1α的表达。结果Glutl和HIF-1α在108例肝细胞癌组织中的阳性表达率分别为74.1%和78.7%,明显高于其在癌旁组织(χ^2=15.27,χ^2=8.727)和正常肝组织(χ^2=31.78,χ^2=27.19)中的表达率(P〈0.05)。Glutl的表达强度与HCC的分化程度(χ^2=27.80),Edmondson分级(χ^2=6.260),有无转移(χ^2=11.54)等显著相关(P〈0.05)。HIF-1α在原发性肝癌中的表达强度与HCC分化程度(χ^2=13.05)和肿瘤大小(χ^2=11.74)相关(P〈0.05)。统计学分析显示Glutl和HIF-1α在HCC中的表达呈正相关(χ^2=18.81)。结论Glutl和HIF-1α的异常高表达在原发性肝细胞癌的发生发展中起重要作用,可能促进肝细胞癌的增殖和侵袭转移。Glutl和HIF-1α的表达强度对肝癌的诊断具有临床应用价值。  相似文献

目的 探讨缺氧诱导因子-1α(HIF-1α)、血管内皮素-1(ET-1)和血管内皮生长因子(VEGF)在胃肠道间质瘤(GIST)中的表达关系及其对患者预后判断的价值.方法 收集2003年1月至2006年12月中南大学湘雅医院(50例)、湖南省邵阳市中医院(36例)行手术治疗的86例GIST患者肿瘤标本,采用免疫组织化学染色方法检测标本中HIF-1 α、ET-1和VEGF的表达.计数资料采用x2检验,应用Spearman等级相关分析处理HIF-1α、ET-1和VEGF之间的相关性.Kaplan-Meier法计算患者生存率,生存分析比较采用Log-rank检验.结果 86例患者中,HIF-1α阴性23例、阳性30例、强阳性33例,阳性表达率为73.3%(63/86);ET-1阴性29例、阳性28例、强阳性29例,阳性表达率为66.3% (57/86);VEGF阴性24例、阳性26例、强阳性36例,阳性表达率为72.1%(62/86).HIF-1α、ET-1和VEGF的表达两两均呈正相关(r=0.594,0.655,0.347,P<0.05);上述3种检测指标与GIST的美国国立卫生研究院(NIH)危险度分级、浸润转移、肿瘤直径和核分裂象有关(x2=15.565、10.841、12.574、21.125,8.171、10.002、10.354、17.317,14.710、16.230、11.116、18.886,P<0.05);而与患者的性别、年龄、肿瘤原发部位无关(x2=0.059、0.071、5.070,0.468、0.074、1.665,0.231、0.370、3.712,P>0.05);进一步分析发现:上述3种检测指标阳性表达率随GIST NIH危险度分级的升高、出现浸润转移和肿瘤直径的增大而增加.86例患者随访率为91.9%(79/86),3、5年生存率分别为82%和69%.中位随访时间为40.2个月(2~66个月).HIF-1α、ET-1和VEGF阳性患者的5年生存率分别为42%、45%和42%,显著低于阴性患者的64%、65%和63%(x2=6.325,6.124,6.287,P<0.05).结论 HIF-1α、ET-1和VEGF在GIST中的表达两两呈正相关,这3种检测指标提示肿瘤的恶性程度及患者预后.  相似文献

目的本研究旨在探讨缺氧诱导因子1α(HIF-1α)和P53蛋白在人肾透明细胞癌组织中的表达及其对患者预后的影响。方法应用免疫组化方法检测65例肾透明细胞癌组织中HIF-1α和P53蛋白的表达情况,通过生存曲线法比较阳性组和阴性组患者的预后情况。结果 HIF-1α和P53蛋白表达的阳性率分别为40.0%(26/65)和52.3%(34/65);HIF-1α表达与肾透明细胞癌的组织学分级、临床分期和淋巴结转移有关(P〈0.05);HIF-1α蛋白阳性组患者的生存期明显较阴性组短(P=0.004);P53蛋白表达与肾透明细胞癌患者性别、年龄、肿瘤大小、组织学分级、临床分期、淋巴结转移及生存期均无关(P〉0.05);HIF-1α蛋白P53蛋白的表达之间存在相关性(r=0.402,P=0.001)。结论 HIF-1α的表达与肾透明细胞癌患者的预后相关,其阳性表达提示患者预后不良;检测HIF-1α表达水平有助于肾细胞癌预后评估。  相似文献

目的探讨缺氧诱导因子-1α(HIF-1α)及血管内皮生长因子C(VEGF-C)在食管癌中的表达及其与食管癌淋巴转移的关系。方法采用免疫组织化学法检测55例食管鳞状细胞癌(ESCC)组织及癌旁组织中HIF-1α、VEGF-C表达及微淋巴管密度(MLVD)的分布情况;运用SPSS12.0软件包进行统计学分析,率的比较采用χ2检验、两组均数的比较采用t检验,显著性水准取α=0.05。结果HIF-1α、VEGF-C在食管癌组织和癌旁组织中的表达阳性率分别为65.5%/10.9%、70.9%/9.1%,差异均有统计学意义(P<0.01);癌组织和癌旁组织的微淋巴管密度分别为4.9±1.3和2.6±0.7,差异有统计学意义(P<0.05);食管癌组织中HIF-1α与VEGF-C表达有统计学关联(P<0.05)。结论HIF-1α、VEGF-C在食管癌的发生发展和淋巴转移中起重要作用,有望成为食管癌治疗和判断预后的重要指标。微淋巴管的形成可能会促进食管癌经淋巴结转移的发生,MLVD可作为判断食管癌淋巴转移和预后的分子指标。HIF-1α可能通过上调VEGF-C的表达,进而通过VEGF/VEGFR信号系统,对肿瘤淋巴管形成调控,促进食管癌淋巴管生成,并导致肿瘤浸润和淋巴结转移。  相似文献

目的:探讨缺氧诱导因子-1α(HIF-1α)和血管内皮细胞生长因子(VEGF)在前列腺癌中的表达及其与病理分级、临床分期的关系。方法:采用免疫组化方法检测前列腺癌和良性前列腺增生(BPH)组织中HIF-1α和VEGF的表达。结果:前列腺癌中HIF-1α和VEGF的表达均明显高于BPH;HIF-1α和VEGF在前列腺癌中的表达水平与其病理分级和临床分期均呈正相关;前列腺癌中的HIF-1α表达水平和VEGF的表达水平呈正相关。结论:HIF-1α和VEGF是检测前列腺癌的较好分子标志物,可望用于前列腺癌的辅助诊断和预后判断,HIF-1α是VEGF表达的调控因子之一。  相似文献

目的 探讨缺氧诱导因子-1α(HIF-1α)与基因系尾型同源盒基因2(CDX2)蛋白在结直肠腺癌组织中的表达及其与结直肠癌发生、发展的关系,并且研究HIF-1α与CDX2表达之间的相关性.方法 收集62例结直肠腺癌及20例正常结直肠组织标本,采用免疫组化SABC法检测标本中HIF-1α、CDX2蛋白的表达,并采用Spearman等级相关分析分析HIF-1α与CDX2表达之间的相关性.结果 在20例正常结直肠黏膜组织中HIF-1α及CDX2蛋白表达阳性率分别为5.0%(1/20)及95.0%(19/20),在62例结直肠腺癌组织中HIF-1α及CDX2蛋白表达阳性率分别为62.9%(39/62)及69.4%(43/62),不同组织中蛋白表达的差异均有统计学意义(P<0.05).结直肠腺癌中,HIF-1α、CDX2蛋白的表达与肿瘤的分化程度、淋巴结转移及Dukes分期有关(P<0.05);Spearman等级相关分析表明,HIF-1α与CDX2的表达呈负相关(r=-0.293 2,P<0.05).结论 HIF-1α表达上调、CDX2表达下调可能与结直肠腺癌的发生有关,且这两者间呈负相关.HIF-1α可能参与调控CDX2的表达,进而加速了结直肠癌的恶性进展.  相似文献

目的:探讨缺氧诱导因子-1α(HIF-1α)和葡萄糖转运蛋白-1(GLUT-1)在膀胱移行细胞癌中的表达及与膀胱移行细胞癌生物学行为的关系.方法:应用免疫组化方法对50例膀胱移行细胞癌及正常膀胱组织中HIF-1α和GIUT-1的表达进行检测,并用计算机图象分析系统对其表达水平进行定量分析.结果:正常膀胱组织中HIF-1α和GLUT-1均不表达,50例膀胱移行细胞癌中,HIF-1α和GLUT-1的阳性表达率分别为52%和76%.HIF-1α的表达水平与膀胱移行细胞癌的分级和复发密切相关(P<0.05),但与分期无关;GLUT-1的表达水平与膀胱移行细胞癌的分级、分期和复发均密切相关(P<0.05).HI-1α和GIUT-1在膀胱移行细胞癌中的表达水平亦呈正相关(r=0.381,P=0.029).结论:膀胱移行细胞癌中HIF-1α和GLUT-1的表达与肿瘤的恶性进展密切相关.联合检测HIF-1α和GLUT-1可作为了解膀胱移行细胞癌生物学行为的参考指标,并有可能为膀胱移行细胞癌的基因治疗提供新的靶点和新的思路.  相似文献
