Summary Antibodies against hyperglycemic hormone (CHH) ofCarcinus were raised in rabbits by injection of extract from sinus glands which contain high concentrations of CHH. The antiserum neutralizes the biological activity of CHH and binds125J-CHH. A RIA for CHH was established and was used to measure the hormone content of sinus glands.Dedicated to Prof. M. Immunocytochemical techniques applied at both light- and electron microscopical levels are valuable in the study of regulatory peptide distribution in normal and diseased tissue, whether in the form of sections or whole cell preparations. Successful immunolocalization depends on adequate preservation of the peptide antigen and the tissue structure in which it resides; a suitably specific and sensitive labelled antibody detecting system. In general, peptides are stable molecules, most of which retain their antigenicity after conventional cross-linking fixation and tissue processing, allowing standard immunocytochemical methods to be used for light- and electron microscopy. Regulatory peptides are derived from precursor molecules and several 'families' of structurally similar peptides are now generally recognised. Region-specific antibodies may be needed to overcome problems of cross-reactivity or to identify a bioactive form in the presence of its precursor. Multiple co-localisation of different related and unrelated peptides in the same cell or even storage granule is now recognised and can be identified by immunocytochemistry.  相似文献

The contact of long and medium chain fatty acids with the ileal mucosa inhibits basal or stimulated pancreatic protein secretion of Man, Dog, Cat and Rat. This inhibition is due to an inhibitory factor transmitted by cross-circulation. We isolated from partially purified extracts of Pig ileum a peptide which inhibits strongly volume and protein pancreatic output of the conscious Rat. We propose to call this factor ACP, anticholecystokinine peptide.  相似文献

Summary Human -atrial natriuretic peptide (h-ANP) makes the urine of dehydrated volunteers hypotonic to plasma despite high circulating concentrations of antidiuretic hormone. Urinary dilution with h-ANP also occurs in subjects receiving indomethacin. Therefore, h-ANP antagonises effects of antidiuretic hormone on distal tubular V2-receptors in man, probably without involving prostaglandins.  相似文献

Summary In view of a recent proposal that calcitonin injections may arrest the bony pathology of otosclerosis, we have tested the possibility of obtaining locally effective concentrations by giving salmon calcitonin in eardrops. Osteolysis of guinea-pig ossicles induced by injecting parathyroid hormone shortly before explantation was markedly inhibited by 3 days prior instillation of the calcitonin in an aqueous vehicle or in dimethyl sulphoxide, but not by a solution in propylene carbonate.We thank Mr R. Apo-carotenoid compounds such as retinol (vitamin A) are involved in a variety of cellular processes and are found in all kingdoms of life. Instead of being synthesized from small precursors, they are commonly produced by oxidative cleavage and subsequent modification of larger carotenoid compounds. The cleavage reaction is catalyzed by a family of related enzymes, which convert specific substrate double bonds to the corresponding aldehydes or ketones. The individual family members differ in their substrate preference and the position of the cleaved double bond, giving rise to a remarkable number of products starting from a limited number of carotenoid substrate molecules. The recent determination of the structure of a member of this family has provided insight into the reaction mechanism, showing how substrate specificity is achieved. This review will focus on the biochemistry of carotenoid oxygenases and the structural determinants of the cleavage reaction.  相似文献
