采用多溶剂筛选最佳提取溶剂;以紫外分光光度法、高效液相色谱法、红外光谱法系统比较石楠红、绿色素成分的差别;同时考察红、绿色素对DPPH·清除能力、还原Fe3+能力和清除·OH自由基能力。结果以90%乙醇和乙酸乙酯-乙醇(1∶1)提取红色素、绿色素效果较好;通过显色反应、紫外光谱、红外光谱鉴别和高效液相色谱分析,基本确定红色素成分为黄酮类,绿色素成分为叶绿素类。抗氧化实验结果表明,在达到最大浓度0.34mg·m L-1时,石楠叶红、绿色素和维生素C对DPPH·清除率依次为79.6%,70.8%,66.9%,红、绿色素对·OH自由基的清除率依次为67.1%,51.2%,且红色素还原Fe3+能力较强。  相似文献

Anthocyanins are natural pigments displaying different attractive colors ranging from red, violet, to blue. These pigments present health benefits that increased their use in food, nutraceuticals, and the cosmetic industry. However, anthocyanins are mainly extracted through conventional methods that are time-consuming and involve the use of organic solvents. Moreover, the chemical diversity of the obtained complex extracts make the downstream purification step challenging. Therefore, the growing demand of these high-value pigments has stimulated the interest in designing new, safe, cost-effective, and tunable strategies for their extraction and purification. The current review focuses on the potential application of compressed fluid-based (such as subcritical and supercritical fluid extraction and pressurized liquid extraction) and deep eutectic solvents-based extraction methods for the recovery of anthocyanins. In addition, an updated review of the application of counter-current chromatography for anthocyanins purification is provided as a faster and cost-effective alternative to preparative-scale HPLC.  相似文献

The present study aimed to maximize the conventional extraction and compare it with the ultrasound-assisted method for extracting bioactive compounds obtained from the red araçá peel. The behavior of anthocyanins related to the pre-treatment of the vegetal matrix, employed solvent, extraction kinetics of both methods, the levels of total phenolic compounds, flavonoids and carotenoids, as well as the antioxidant activity were evaluated. The ultrasound-assisted extraction (40 KHz −154 W and 90 min) had an increase of 12% in the levels of anthocyanins (121.85 Eq. mg of cyanidin-3-glycoside/100 g of peel) and a 25% reduction in time extraction compared to conventional extraction by maceration (116.81 Eq. mg of cyanidin-3-glycoside/100 g of peel) using 90% ethanol, for 2 h, pH 1.5, at 40 °C and mass/volume ratio 1 g/10 mL). Analyses of the total phenolic compounds, flavonoids and carotenoids presented promising results for the ultrasound-assisted and conventional extractions, respectively. Analyzes of total phenolic compounds, flavonoids and carotenoids, show promising results for ultrasound-assisted extractions, respectively, indicating that red araçá is rich in bioactive compounds beneficial to human health, in addition to being considered natural pigments that can be used in food.  相似文献

The objective of this study was to develop a method for the determination of red beetroot betalains based on the dispersive solid‐phase extraction and modified QuEChERS methods followed by micro‐high performance liquid chromatography coupled with a mass spectrometer that was equipped with a quadrupole and time‐of‐flight detector. Currently, new techniques for the extraction of the pigments are necessary and in this study, an extraction of beetroot betalains based on the QuEChERS method was developed for the first time. Twelve variants of the methods with different sorbent combinations were tested. The extraction with 15% methanol and with 0.05% formic acid was performed as a reference method to compare the obtained results. In all of the samples with the addition of sorbents, a lower noise was demonstrated in the obtained results. The betalain concentrations obtained using the tested methods were 0.32–0.54 mg g?1, while the value of the reference method was 0.44 mg g?1. The method that used the strong ion exchange sorbent (0.44±0.05 mg g?1) was the most adequate in terms of analyzed content, related standard deviation value and interference compared to the reference method. It was concluded that the properly modified QuEChERS method can be successfully applied for the determination of red beetroot betalains.  相似文献

In recent years, more attention has been paid to natural sources of antioxidants. Flavonoids are natural substances synthesized in several parts of plants that exhibit a high antioxidant capacity. They are a large family, presenting several classes based on their basic structure. Flavonoids have the ability to control the accumulation of reactive oxygen species (ROS) via scavenger ROS when they are formed. Therefore, these antioxidant compounds have an important role in plant stress tolerance and a high relevance in human health, mainly due to their anti-inflammatory and antimicrobial properties. In addition, flavonoids have several applications in the food industry as preservatives, pigments, and antioxidants, as well as in other industries such as cosmetics and pharmaceuticals. However, flavonoids application for industrial purposes implies extraction processes with high purity and quality. Several methodologies have been developed aimed at increasing flavonoid extraction yield and being environmentally friendly. This review presents the most abundant natural flavonoids, their structure and chemical characteristics, extraction methods, and biological activity.  相似文献

The adsorption and desorption capacities of 11 different solid-phase extraction sorbents were tested for the preconcenration of pigments of various Hungarian red wines. The concentrates were evaluated by multiwavelengh spectrophotometry combined with a spectral mapping technique (SPM) and by reversed-phase high-performance liquid chromatography. The highest (10-fold) concentration of pigments was achieved on octadecylsilica sorbent. It can be used five times without losing adsorption and desorption characteristics. SPM indicated that multiwavelength spectrophotometry can be employed for the differentiation of red wines. Comparison of the chromatograms of pigments with and without preconcentration showed that preconcentration makes possible the separation and detection of pigments present in low concentration in red wines.  相似文献

Colour is an organoleptic characteristic of virgin olive oil and an important attribute that affects the consumer perception of quality. Chlorophylls and carotenoids are the main pigments responsible for the colour of virgin olive oil. A simple analytical method for the quantitative determination of chlorophylls and carotenoids in virgin olive oils has been developed. The pigments were isolated from small samples of oil (1.0 g) by solid-phase extraction using diol-phase cartridges (diol-SPE), and the extract was analysed by reverse-phase HPLC with diode-array UV detection. Chromatographic peak resolution, reproducibility (coefficient of variation (C.V.) <4.5%) and recovery (>98.4%) for each component were satisfactory. A comparative study of the proposed method was performed versus classical liquid-liquid extraction (LLE) with N,N′-dimethylformamide and solid-phase extraction using a C18 column (C18-SPE). While 96.4% of the pigments were recovered by LLE, only 51.3% were isolated by C18-SPE in comparison to diol-SPE. Likewise, a higher alteration of pigment composition was observed when such LLE and C18-SPE procedures were used. In this sense, a higher ratio of pheophytin in comparison to that isolated by the diol-SPE procedure was achieved with both extraction procedures, indicating a greater extent of the pheophytinization reaction. Therefore, quantification of pigments from virgin olive oil by diol-SPE followed by RP-HPLC was found to be rapid, simple, required only a small amount of sample, consumed only small amounts of organic solvents, and provided high recoveries, accuracy and precision.  相似文献

Nutraceuticals and functional foods are gaining more attention amongst consumers interested in nutritious food. The consumption of foodstuffs with a high content of phytochemicals has been proven to provide various health benefits. The application of biostimulants is a potential strategy to fortify cultivated plants with beneficial bioactive compounds. Nevertheless, it has not yet been established whether the proposed higher plants (St. John’s wort, giant goldenrod, common dandelion, red clover, nettle, and valerian) are appropriate for the production of potential bio-products enhancing the nutritional value of white cabbage. Therefore, this research examines the impact of botanical extracts on the growth and nutritional quality of cabbage grown under field conditions. Two extraction methods were used for the production of water-based bio-products, namely: ultrasound-assisted extraction and mechanical homogenisation. Bio-products were applied as foliar sprays to evaluate their impact on total yield, dry weight, photosynthetic pigments, polyphenols, antioxidant activity, vitamin C, nitrates, micro- and macroelements, volatile compounds, fatty acids, sterols, and sugars. Botanical extracts showed different effects on the examined parameters. The best results in terms of physiological and biochemical properties of cabbage were obtained for extracts from common dandelion, valerian, nettle, and giant goldenrod. When enriched with nutrients, vegetables can constitute a valuable component of functional food.  相似文献

Anthocyanins are the largest group of polyphenolic pigments in the plant kingdom. These non-toxic, water-soluble compounds are responsible for the pink, red, purple, violet, and blue colors of fruits, vegetables, and flowers. Anthocyanins are widely used in the production of food, cosmetic and textile products, in the latter case to replace synthetic dyes with natural and sustainable alternatives. Here, we describe an environmentally benign method for the extraction of anthocyanins from red chicory and their characterization by HPLC-DAD and UPLC-MS. The protocol does not require hazardous solvents or chemicals and relies on a simple and scalable procedure that can be applied to red chicory waste streams for anthocyanin extraction. The extracted anthocyanins were characterized for stability over time and for their textile dyeing properties, achieving good values for washing fastness and, as expected, a pink-to-green color change that is reversible and can therefore be exploited in the fashion industry.  相似文献

A simple and rapid microwave-assisted extraction (MAE) procedure was developed and optimized for two common color pigments, alizarin and purpurin, in various samples of Rubiaceae plants. Several variables that can potentially affect the extraction efficiency, namely temperature, methanol concentration in the extractant mixture, time, and solvent volume were optimized by means of a central composite design approach. The results suggest that temperature and methanol concentration in the solvent mixture are statistically the most significant factors. The separation and quantitative determination of the pigments was carried out in less than 6 min by a developed high-performance liquid chromatographic method with UV detection at 250 nm. Under optimum operating conditions, MAE showed significantly higher recoveries than those obtained by the conventional extraction methods (ultrasonic and reflux extraction), ranging from 84 to 94%. In addition, a drastic reduction of the extraction time (20 min versus 6 h) and solvent consumption (20 versus 100 mL) was achieved with a reproducibility (RSDs < 10%) comparable with that provided by the reflux extraction as a reference method.  相似文献

古菌是一类兼具真菌及细菌细胞特征、大多生活在极端环境下的单细胞微生物。由于膜结构及代谢通路的特殊性,研究古菌脂质对深入探究极端环境下生命的正常活动具有重要意义。前处理方法在组学分析中会极大影响最终结果的准确度及可靠性。该文基于超高效液相色谱-高分辨质谱联用(ultra-performance liquid chromatography-high resolution mass spectrometry,UPLC-HRMS)技术,以嗜热嗜压古菌Pyrococcus yayanosii为模式生物,采用Bligh-Dyer加酸法、Folch法、甲基叔丁基醚(methyl tert-butyl ether,MTBE)法及固相萃取(SPE)法提取脂质成分,从提取效率、重复性、歧视性等角度进行方法评价,并最终确定SPE法和MTBE法的提取重复性和提取效率最好,适合高通量古菌全脂质提取。利用SPE提取、高分辨质谱分析对P.yayanosii的常规脂质进行了全面分析,共鉴定到了1402种脂质。本研究旨在为古菌和其他极端微生物的非靶向脂质组学分析和脂质代谢研究提供方法参考。  相似文献
