蔓荆子中木脂素类成分的分离与结构鉴定

[目的]对蔓荆子中的木脂素类成分进行研究。[方法]采用正相硅胶、Sephadex LH-20、MCI GEL(MCI)、反相ODS(ODS)等柱色谱及高效液相色谱手段进行分离纯化,并通过理化性质与波谱分析方法鉴定化合物的结构。[结果]从蔓荆子95%乙醇提取物中分离得到7个木脂素类单体成分,分别为dihydrodehydrodiconiferyl alcohol(1),ficusal (2),(7R,8S)-dihydrodehydrodiconiferyl alcohol 9-O-β-D-glucopyranoside (3),dihydrodehydrodiconiferyl alcohol-β-D-(2′-O-p-hydroxybenzoyl)glucoside (4),(7R,8R)-7,8-dihydro-9′-hydroxyl-3′-methoxyl-8-hydroxymethyl-7-(4-hydroxy-3-methoxyphenyl)-1′-benzofuranpropanol 9′-O-β-D-glucopyranoside (5),松脂醇(pinoresinol)(6),1-syringaresinol(7)。[结论]化合物5为首次从牡荆属中分离得到,化合物6和7为首次从该植物中分离得到的化合物。

MGMT表达缺失在胃癌发生中的作用

[目的]探讨DNA直接修复蛋白MGMT表达缺失在胃癌发生中的作用和意义.[方法]免疫组织化学方法检测66例胃癌、34例胃炎(慢性浅表性胃炎25例,慢性萎缩性胃炎9例)标本MGMT表达.[结果]MGMT表达率在慢性浅表性胃炎黏膜(CS)、慢性萎缩性胃炎黏膜(CA)和胃癌(GC)组分别为48.0%(12/25)、33.3%(3/9)和19.7%(13/66)(CS v.s.CA,P=0.697;CA v.s.GC,P=0.392;CS v.s.GC,P=0.007).胃癌组,MGMT表达与患者性别、年龄、胃癌分化程度及HP感染间均无显著相关关系;胃炎组,HP感染与MGMT表达显著正相关(r=0.408,P=0.017).[结论]MGMT表达缺失引起的胃黏膜细胞DNA损伤修复障碍可能是胃癌发生的原因之一.     

长柱十大功劳木生物碱提取物的质量标准研究

[目的]建立长柱十大功劳木生物碱提取物的质量标准。[方法]采用高效液相色谱法,色谱柱为Kromasil ODSBP C18(4.6 mm×250 mm,5μm),以乙腈-0.05 mol/L磷酸二氢钾缓冲液(磷酸调pH值至3.0)(3∶7)为流动相;检测波长为265 nm;柱温:室温;流速:1 ml/min。[结果]盐酸小檗碱在0.0697~1.3944μg范围内线性关系良好(r=0.9997),平均回收率为100.7%,RSD为1.35%(n=6);盐酸巴马汀在0.2070~4.1400μg范围内线性关系良好(r=0.9997),平均回收率为102.8%,RSD为0.91%(n=6);盐酸药根碱在0.1086~2.172μg范围内线性关系良好(r=0.9997),平均回收率为103.0%,RSD为1.21%(n=6)。[结论]该方法简便、准确,专属性强,重现性、回收率好,可作为长柱十大功劳木生物碱提取物的质量控制方法。
